Shelled acanthors (=eggs) of two isolates of Moniliformis moniliformis (Acanthocephala) were tested for their capacity to respond to a hatching stimulus in vitro and for their retention of infectivity to a natural intermediate host (Periplaneta americana). The shelled acanthors were stored for more than 120 weeks (Australian isolate) and for 104 weeks (Texan isolate) together with rat faeces in an incubator maintained at 22·2±0·1°C. In both cases, infectivity to P. americana was shown to have been retained. In vitro tests of the hatching response were carried out on many occasions during this period of faecal storage. Shelled acanthors continued to respond and no differences were detected either between isolates or within an isolate through time.

The assay system for antibody responses against Em2, the most specific antigen for serodiagnosis of alveolar echinococcosis (AE), has been established by enzyme-linked immunosorbent assay (ELISA) but not by Western blot assay, since Em2 antigen is not protein but carbohydrate in nature. Recently we reported that previously undescribed protein epitopes, designated Em18 and Em16 due to their molecular weights, were good serologic markers for AE by Western blot analysis. It has been shown that Em18 and Em16 are the only two epitopes recognized by commercially available weak positive (cut off) sera for the Em2plus-ELISA.

Early in 1938 the writer received a tube of nematodes from Mr. Colin Smee, Entomologist, Department of Agriculture, Zomba, Nyasaland, collected from a rotting peach fruit in which he had been trying to hatch fruit-flies. According to Mr. Smee's letter the worms had formed masses in the fruit and had also appeared on the surface as waving threads or tendrils composed of numerous eelworms massed together. The material had been fixed in glycerine alcohol and consisted of numerous adult males and females as well as large numbers of larval forms. Fixation was not good as although the gross morphology and anatomy was fairly easily discernible, some of the finer structural details were not as clear as one could have wished.

Four male specimens of this species were once collected from the white necked Stork, Dissoura episcopus episcopus. The worms occurred under the horny layer of the gizzard.

The cuticle is smooth. The lateral alae extend from the middle of the oesophagus to the region of the spicules in the males, but in females they are extended further posterior to the middle of the tail or more. The anterior short "pharynx", the procorpus, is poorly differentiated from the remaining cylindrical portion of the oesophagus. Three sclerotized structures are present at the anterior end of the oesophagus. The posterior flaps of the valves of the bulb extend slightly into the dilated lumen of the intestine. The intestine at its commencement is broader than the bulb, and gradually narrows down towards the anus. The excretory pore is situated anterior to the usual pre-bulbar position, and is located at the beginning of the posterior third of the cylindrical oesophagus. It is surrounded by a crown of chitinous rod-like structures. The tail is sharply pointed in both sexes.

In his “Monograph on the Anguillulidae, or Free Nematoids, Marine, Land and Freshwater; with Descriptions of 100 New Species,” published in 1865, Bastian described a nematode which he found in the yellow lichen Xanthoria (then known as Parmelia) parietina. He named it Aphelenchus parietinus, giving a short description and two drawings. His drawings are reproduced in Fig. 1 and his description is as follows:

In a paper recently published in the Philosophical Transactions of the Royal Society of London, the writer describes the discovery of certain small Tylenchoid nematodes in the swollen stems of the seedling oats suffering from frit-fly attack at Winches Farm, St. Albans. Adults of both sexes and larvæ undergoing the last two moults were found in the destroyed plant tissues surrounding the fly larvæ. The spermatized female worms were next discovered within the body-cavity of the frit-fly larvæ. In due course, they were obtained from pupæ of the fly, having increased in size, and finally were found within adult flies of both sexes lying coiled within the abdomen as comparatively large sausage-shaped worms. As a result of the presence of the worm, the flies are sterilised, being unable to develop their reproductive organs. The worms become viviparous and shed large numbers of larvæ into the body-cavity of the fly and these, after undergoing a certain amount of growth, make their way into the gut of the host and pass to the exterior via the anus. In the present communication a brief account is given of the principal observations on the new parasite, its life-history and the effects on the host, described in detail in the original paper.

Information concerning worm control practices of sheep and goat farmers in the region of Trikala (central Greece) was collected through a questionnaire survey by visiting farms and interviewing farmers. Questionnaires from 57 farmers residing in 23 rural communities were collected. Anthelmintics were used by 89% of the farmers. On average, lambs, kids and goats were treated once annually, while sheep were treated either once or twice annually. Only 2% of farmers reported treatment of animals with anthelmintics when moving to new pastures. The most common broad-spectrum anthelmintics used were those belonging to the benzimidazoles and probenzimidazoles. Fifty nine percent of the farmers used the same anthelmintic group for 3 or more years and 34% used two or more anthelmintic groups in the same year. Almost all farmers reported estimating live weights for calculating anthelmintic doses through visual perception on the basis of an average weight (96%). Tablets and boluses were the most preferred anthelmintic formulation used by 96% of farmers. The selection of an anthelmintic was based for 58% of farmers on recommendation by a veterinarian and for 39% of farmers on the cost of the drug. The most common occasions for deworming the animals were at turn out (86%) and after parturition (31%). Only 6% of farmers reported deworming new animals before introducing them onto the farm. Farmers preferred to seek information about the use of anthelmintics and worm control strategies from veterinarians (63%) and other farmers (37%).

The present investigation aimed to evaluate the extent to which maternal filarial infection influences IgG subclass immune responses in the cord blood of neonates. Prevalence of antigenaemia was detected using an Og4C3 assay. Filaria-specific IgG subclasses against excretory/secretory antigens were measured by ELISA. Transplacental transfer of circulating filarial antigen (CFA) was observed from 34.8% of CFA-positive mothers to their respective cord bloods. Filaria-specific IgG1, IgG2 and IgG4 responses were significantly higher among cord bloods of infected mothers compared to cord bloods of uninfected mothers. In contrast, the IgG3 response was significantly higher among cord bloods of uninfected mothers. The study shows that transplacental transfer of filarial antigens and filaria-specific IgG4 occurs more in mothers having high worm burdens, and transfer of filaria-specific IgG3 occurs more in the cord blood of uninfected mothers. The findings of the study provide evidence for the development of prenatal sensitization to filarial antigens in utero, and high filaria-specific IgG4 in cord blood may serve as a marker for in-utero sensitization.

Sheep reared worm free were used as indicators of the availability of sheep nematode infective larvae during the summer on pasture grazed by ordinary flock ewes and their lambs.

Data were collected in terms of worms found in the test sheep at post mortem examination after periods of three weeks' exposure in the field.

A new genus and species of nematode, Tziminema unachi n. gen., n. sp. is described from the caecum and colon of Baird's tapir Tapirus bairdii (Gill, 1865), found dead in the Reserva de la Biósfera El Triunfo, Chiapas State, in the Neotropical realm of Mexico. Tziminema n. gen. differs from the other nine genera included in the Strongylinae by two main characteristics: having 7–9 posteriorly directed tooth-like structures at the anterior end of the buccal capsule, and the external surface of the buccal capsule being heavily striated. Phylogenetic analyses of the DNA sequences of the mitochondrial cytochrome c oxidase and nuclear DNA, including a partial sequence of the internal transcribed spacer 1, 5.8S and a partial sequence of the internal transcribed spacer 2 of the new taxon, confirmed its inclusion in Strongylinae and its rank as a new genus.

Susceptibility and tolerance to parasite infection vary greatly between host species. The congeneric isopod species Austridotea annectens and Austridotea lacustris often occur in sympatry and can both serve as intermediate hosts for the trematode Maritrema poulini. However, the intensity and prevalence of infections vary greatly in natural populations, with A. annectens often being heavily infected and A. lacustris rarely being infected. To shed light on the factors that may be involved in infection avoidance in A. lacustris, 100 isopods were collected and experimentally exposed to M. poulini cercariae. To examine for potential behavioural mechanisms, we used carbonated water as a paralysing agent to temporarily stop any movement by the isopods, and exposed paralysed isopods to cercariae. In the experiments, none of the individuals that were exposed to the parasite were found to be infected, although some cercariae seemed to have penetrated isopod hosts. Behavioural defence mechanisms do not seem to explain the different infection levels between A. lacustris and A. annectens, suggesting a physiological, possibly immunological, factor may be present. At the ecosystem level, this suggests a potential dilution effect caused by this low-competency host, and its effects on parasite transmission dynamics should be investigated.

Schistosomiasis remains a major neglected tropical disease that afflicts over 200 million people globally. Schistosomes, the aetiological agent of schistosomiasis, are parasitic flatworms that propagate between molluscan and mammalian hosts. Inside the mammalian host, schistosomes rapidly grow over 100-fold in size and develop into a sexually mature male or female that thrives in the bloodstream for several decades. Recent work has identified schistosome stem cells as the source that drives parasite transmission, reproduction and longevity. Moreover, studies have begun to uncover molecular programmes deployed by stem cells that are essential for tissue development and maintenance, parasite survival and immune evasion. Such programmes are reminiscent of neoblast-driven development and regeneration of planarians, the free-living flatworm relative of schistosomes. Over the last few decades, research in planarians has employed modern functional genomic tools that significantly enhanced our understanding of stem cell-driven animal development and regeneration. In this review, we take a broad stroke overview of major flatworm organ systems at the cellular and molecular levels. We summarize recent advances on genetic regulators that play critical roles in differentiation and maintenance of flatworm cell types. Finally, we provide perspectives on how investigation of basic parasite biology is critical to discovering new approaches to battle schistosomiasis.

Amino acid neurotransmitters play an important role in regulating neuromuscular activity of helminth parasites. The present study aimed to investigate the effects of different amino acid neurotransmitters [l-glutamate, glycine, gamma-aminobutyric acid (GABA)] on spontaneous muscular activity of isometrically mounted Gastrothylax crumenifer. l-Glutamate caused a significant increase in the amplitude and frequency of spontaneous contractions of rumen fluke at 10− 7–10− 4 m and at 10− 5–10− 4 m concentrations, respectively. Glycine application (10− 7–10− 3 m) produced a significant decrease in the amplitude and frequency of spontaneous muscular contractions in a concentration-dependent manner, as compared to control amplitude (0.53 ± 0.02 g) and frequency (51 ± 4.65/5 min). Similarly, GABA produced a significant (P < 0.05) decrease in amplitude, baseline tension and frequency of spontaneous muscular contractions of G. crumenifer. To further substantiate the GABA effect, GABAA receptor antagonists, picrotoxin and bicuculline were applied. Picrotoxin (10− 5–10− 3 m) caused a significant (P < 0.05) increase in amplitude, baseline tension and frequency of the rumen fluke as compared to control; whereas bicuculline did not elicit any observable effect in these attributes in isometrically mounted rumen flukes. These observations suggested that l-glutamate has an excitatory, whereas GABA and glycine have an inhibitory, effect on the spontaneous muscular activity of G. crumenifer.

Taenia solium is the most common parasite infection of the brain, causing neurocysticercosis and typically found in rural communities with free-ranging pigs. Identification of transmission in rural areas is essential for its control. Risk factors and transmission of the parasite were evaluated in three rural Venezuelan communities (Valle del Rio and Potrero Largo, Cojedes state; and Palmarito, Portuguesa state) by a questionnaire (112 households) and coprological (492 samples) and serological (433 human and 230 porcine sera) analysis, respectively. Typical risk factors were found in all three communities: free-foraging pig husbandry, deficient sanitary conditions, high open defecation and ignorance of the parasite life cycle. Coprological examinations revealed a high level of soil-transmitted parasites. Importantly, two T. solium adult worm carriers were identified in each of the three communities. Anti-metacestode antibodies and the HP10 secreted metacestode glycoprotein were detected at significant levels in human and porcine sera in Valle del Rio, Potrero Largo and Palmarito. In conclusion, these communities may be considered to be endemic for taeniasis/cysticercosis, and the instigation of an appropriate control programme is recommended.

Between 1992 and 1996, 95 rabbits from the immediate locality of Malham Tarn, North Yorkshire, UK were examined for the presence of helminth parasites. All the examinations took place in late September or October. Three species of nematodes, Graphidium strigosum, Passalurus ambiguusand Trichostrongylus retortaeformis and two species of cestodes, Taenia pisiformis and Cittotaenia pectinata were identified. There were no associations between helminth species richness and year of sampling, host weight or sex. A logistic model was fitted to the prevalence data from these helminths as was an over-dispersed Poisson model to the worm burden data. Graphidium strigosum was the most frequently identified species with an average prevalence of 78%. The mean prevalence and intensity of Graphidiuminfection were significantly effected by sampling year. The lower than normal rainfall recorded at the Tarn during the years 1995 and 1996 may have be one reason for this pattern. The worm burden of G. strigosum was significantly positively associated with rabbit body weight. The intensity of infection with P. ambiguus was significantly higher in female rabbits. There was a significant non-linear relationship between P. ambiguus worm burden and rabbit weight (P = 0.002) with worm burdens being highest in the 1000 g to 1499 g weight cohort. Trichostrongylus retortaeformis was only identified in 1994 and male rabbits harboured significantly higher worm burdens than females (48 vs. 7, P = 0.022). Over the five years, the average Taenia pisiformis prevalence was 31% and there was a significant positive association between worm burden and rabbit weight (P = 0.001). Cittotaenia pectinata had a prevalence of 37% over the whole study period with no interactions between prevalence or intensity and body weight, year of sampling or rabbit sex. All five helminths showed an overdispersed distribution with k values less than 1.

Host–parasite coevolution may result in life-history changes in hosts that can limit the detrimental effects of parasitism. Fecundity compensation is one such life-history response, occurring when hosts increase their current reproductive output to make up for expected losses in future reproduction due to parasitic infection. However, the potential trade-offs between this increase in quantity and the quality of offspring have been relatively unexplored. This study uses the trematode, Schistosoma mansoni, and its snail intermediate host, Biomphalaria glabrata, to better understand how this host life-history response, fecundity compensation, impacts host reproduction. Measures of host reproductive output as well as offspring hatching success and survival were collected to assess the reproductive consequences of infection. Infected snails exhibited fecundity compensation by increasing the number of eggs laid and the overall probability of laying eggs compared to uninfected snails. Parental infection status did not play a significant role in hatching or offspring survival to maturity. Offspring from a later reproductive bout demonstrated a higher hatching success rate. Overall, the lack of an apparent trade-off between quantity and quality of offspring suggests that infected parental snails invest more resources towards reproduction not only to increase reproductive output, but also to maintain the fitness of their offspring, possibly at the expense of their own longevity.

Trichinellosis is a re-emerging worldwide foodborne zoonosis. Oxidative stress is one of the most common detrimental effects caused by trichinellosis. In addition, Trichinella infection poses an infinite and major challenge to the host’s immune system. Resistance and side effects limit the efficiency of the existing anti-trichinella medication. Given that concern, this work aimed to investigate the anti-helminthic, antioxidant, anti-inflammatory and immunomodulatory effects of resveratrol and zinc during both phases of Trichinella spiralis infection. Sixty-four Swiss albino mice were divided into four equal groups: non-infected control, infected control, infected and treated with resveratrol, and infected and treated with zinc. Animals were sacrificed on the 7th and 35th days post-infection for intestinal and muscular phase assessments. Drug efficacy was assessed by biochemical, parasitological, histopathological, immunological, and immunohistochemical assays. Resveratrol and zinc can be promising antiparasitic, antioxidant, anti-inflammatory, and immunomodulatory agents, as evidenced by the significant decrease in parasite burden, the significant improvement of liver and kidney function parameters, the increase in total antioxidant capacity (TAC), the reduction of malondialdehyde (MDA) level, the increase in nuclear factor (erythroid-derived 2)-like-2 factor expression, and the improvement in histopathological findings. Moreover, both drugs enhanced the immune system and restored the disturbed immune balance by increasing the interleukin 12 (IL-12) level. In conclusion, resveratrol and zinc provide protection for the host against oxidative harm and the detrimental effects produced by the host’s defense response during Trichinella spiralis infection, making them promising natural alternatives for the treatment of trichinellosis.

Methods are discussed for greatly increasing the cyst content of Helerodera-intested soil, so that very little soil need be “floated” to provide the number of cysts necessary to form a replicate in bioassay. Such a “float” contains very little debris and may be used directly, thus avoiding the laborious and time-consuming step of separating the cysts from the debris. An apparatus for quick recovery of the “float” is described.
