Nonspecific esterase (NSE), acetylcholinesterase (AChE) and pseudocholinesterase (ChE) have been localized by histochemical methods in various tissues of a cestode, Raillietina (Raillietina) johri obtained from the intestine of pigeon.

NSE has been found in the rostellum, suckers, hooks, tegument, subtegumental muscle, excretory canal, cirrus sac, vagina and eggs. Two types of cells have been recognized on the scolex surface—some are NSE positive and others are NSE negative. AChE, besides being localized in nerves, has also been visualized in almost all the structures as in case of NSE except in hooks, excretory canal and eggs. Additionally AChE has been observed in the vas deferens and sperm ductules. ChE has been observed only in nerves, vas deferens, cirrus sac and vagina; the intensity of enzyme activity being low when compared with AChE. Possible involvement of these enzymes in the physiology of the parasite has been discussed.

A method is described by means of which trichostrongylid spicules are separated from the surrounding soft tissues and their examination made easier. NaOH and KOH are the most satisfactory substances for this purpose, though both substances eventually act on the chitin of the spicules themselves, causing distortion. The reaction takes place at a temperature of 37°C. with reasonable speed. Spicules of Ostertagia can also be freed satisfactorily by this method but those of Nematodirus are quickly affected by the alkali, with consequent distortion.

A controlled study was carried out to determine the effect of some known anti-filarial drugs on the pattern of emergence of Onchoccrca volvulus microfilariae from skin snips into saline. The results show that before treatment, 81·7 to 92·1% of microfilariae emerge in all the groups within 24 hours. The differences between the groups were not significant. After treatment with oral diethylcarbamazine, metrifonate and topical diethylcarbamazine (DEC) the percentage of microfilariae emerging from the snips dropped significantly to 48·4,560 and 60·1 respectively, whereas no significant change occurred following treatment with levamisole (88·1%) and yeast tablets (89·1%). A pair-wise comparison of the post-treatment microfilarial emergence in all the groups showed statistically significant differences when (i) oral DEC is compared to levamisole, topical DEC and yeast, (ii) metrifonate is compared to levamisole and yeast and (iii) topical DEC is compared to yeast. It is suggested that microfilariae which fail to emerge from snips after treatment are those that have been killed or paralysed by the action of the drugs. The study has thus established that drug action in vivo is one of the factors which affects the proportion of microfilariae which emerge from skin snips and provides an additional parameter for measuring the antimicrofilarial potential of drugs in clinical chemotherapeutic trials.

In “Veterinary Medicine” for March, 1924, Dr. M. C. Hall contributes the third instalment of his paper, “Developments in Veterinary Parasitology during the past Fifteen Years,” and on pages 190 and 191, under the heading Lungworms (Metastrongylidæ), considers work on the life histories of the lung-worms of sheep and cattle, Dictyocaulus filaria and D. viviparus, and then refers to the paper under review. His words are as follows: “The life-histories of the swine lung-worms Metastrongylus elongatus and M. brevivaginatus have been ascertained by an American worker, Zebrowski. In general it is similar to that of Dictyocaulus filaria above, but the moisture requirements of the larvæ in cultures are rather exact. Zebrowski's work, published in 1921 in the proceedings of the Indiana Academy of Science, has been quite generally overlooked.”

Clonorchis sinensis and Capillaria hepatica are zoonotic parasites that mainly infect the liver and cause serious liver disorders. However, immunological parameters induced by co-infection with these parasites remain unknown. In this study, for the first time, we investigated immunological profiles induced by co-infection with C. hepatica (CH) in C. sinensis (CS)-infected rats (Sprague–Dawley). Rats were infected primarily with 50 metacercariae of C. sinensis; 4 weeks later, they were subsequently infected with 1000 infective C. hepatica eggs. Significantly higher levels of C. sinensis- or C. hepatica-specific IgG antibodies were found in the sera of rats. Interestingly, no cross-reacting antibody was observed between C. sinensis and C. hepatica infections. Significantly raised eosinophil levels were found in the blood of C. sinensis/C. hepatica co-infected rats (CS + CH) compared to the blood of rats infected singly with C. sinensis. Co-infected rats showed significantly higher levels of lymphocyte proliferation and cytokine production compared to a single C. sinensis infection. The worm burden of C. sinensis was significantly reduced in co-infected rats compared to the single C. sinensis infection. These results indicate that the eosinophils, lymphocyte proliferation and cytokine production induced by subsequent infection with C. hepatica in C. sinensis-infected rats might contribute to the observed C. sinensis worm reduction.

The performance of loop-mediated isothermal amplification (LAMP) for detection of Schistosoma mansoni DNA from stool and urine samples in comparison with Kato–Katz and real-time polymerase chain reaction (PCR) was studied. After obtaining informed consent, 50 children participated in the present study and agreed to submit stool and urine samples. Stool samples were examined by Kato–Katz. Both real-time PCR and LAMP techniques were applied on stool and urine samples. The overall prevalence of S. mansoni was 46% in stool and urine samples as detected by the employed techniques, and 90% of cases had light infection intensity. The highest percentage of infection was diagnosed by real-time PCR (44%), followed by Kato–Katz (42%) and LAMP in the stool (36%), while the lowest percentages of infection were diagnosed by real-time PCR and LAMP in urine samples (24% and 14%, respectively). Kato–Katz, real-time PCR and LAMP showed 100% specificity where the sensitivity was 91.3%, 95.7% and 78.3%, respectively, in stool samples. Real-time PCR and LAMP showed lower sensitivity in urine samples. The LAMP assay is a promising technique for S. mansoni diagnosis in endemic countries of moderate and high-intensity infection. Yet, it needs further optimization, particularly in urine samples.

Angiostrongylus costaricensis is a parasite that infects rodents, including the wild cotton rat Sigmodon hispidus and pygmy rice rats Oligoryzomys spp., among others. However, urban Rattus norvegicus and Mus musculus have not been identified as important hosts of A. costaricensis. In this study, Swiss mice (SW), Wistar R. norvegicus (RN), wild Oligoryzomys nigripes (ON) and a local strain of M. musculus (RGS) were experimentally infected with A. costaricensis. Survival, elimination of L1 (total sum per group, A0), and the number of adult worms recovered divided by the dose of each L3 inoculum (yield ratio, YR) were examined for each group after a 40-day post-infection period. The survival rates, A0 and YR values were: 27%, 207,589 and 0.42 for the SW group; 81%, 8691 and 0.01 for the RN group; and 63.6%, 26,560 and 0.16 for the RGS group, respectively, in each case. The survival rate for the ON group was 100% and the A0 value was 847,050. A YR was not calculated for the ON group since the ON group was maintained up to 565 days post-infection (pi) to examine long-term mortality. At 500 days pi (16 months), 50% of the ON group had died, while one animal (10%) survived 595 days pi (20 months). Taken together, these data indicate that A. costaricensis has undergone a greater degree of adaptation to the wild rodent, O. nigripes, than to R. norvegicus or a local M. musculus strain. In addition, titre curve (A0) modelling of adaptation status proved to be useful in evaluating A. costaricensis–rodent interactions.

Factors regulating gastrointestinal nematode infrapopulations are to a great extent density-dependent. Here, 23 natural infrapopulations (41–281 individuals) of the viviparous nematode Procamallanus elatensis (Camallanidae) from the fish Siganus rivulatus were found distributed in a well-defined fundamental niche (posterior 55% of the intestine). Immature worms were mostly found in the anterior third of this niche, while mature worms were found in aggregations posterior to them and followed by gravid females in the posterior 20% of the intestine. This distribution strongly suggests that worms migrate towards the posterior intestine while they mature, copulate and reproduce. In small infrapopulations, the sex ratios were distinctly female-biased and the number of gravid females was low. In large infrapopulations, the sex ratios were distinctly male-biased and the number of gravid females was high. However, the mean lengths of both immature males and females and mature females decreased dramatically as the infrapopulation size increased, while those of mature males increased significantly. These results strongly suggest intraspecific competition and density-dependent regulation of mean worm length, and the increase in the mean length of mature males strongly suggests intense sexual selection and competition between mature males. Production of larvae by female worms decreased significantly as the infrapopulation size increased, suggesting a density-dependent reduction in female worm fertility. The results are statistically significant and strongly suggest that infrapopulation self-regulation is through density-dependent mechanisms, in which development of immature worms, infrapopulation size, sex ratio, sexual selection and carrying capacity of the fundamental niche play essential roles in shaping and regulating the infrapopulations.

The life cycle of Pleurogenoides wayanadensis Shinad & Prasadan, 2018, infecting the frogs Hoplobatrachus tigerinus and Euphlyctis cyanophlyctis, is elucidated in this study. All the life cycle stages from egg to egg-producing adults were elucidated under natural conditions and successfully established in the laboratory. The life cycle took about 58 to 65 days for completion. Miracidia were released by teasing the eggs with fine needles. Sporocysts were found in the freshwater snail, Bithynia (Digoniostoma) pulchella, collected from paddy fields at Payode, Western Ghats, Wayanad region, in the months of October and November 2019. Cercariae were of the virgulate xiphidiocercous type. Metacercariae were recovered from the eyes of the damselfly naiads of the species Ischnura sp. and Copera sp., and the thorax and abdomen of the dragonfly naiads, Orthetrum sp. The metacercariae showed progenetic development. The growth and development of the metacercariae in the naiads that were exposed to cercariae, and development of the trematode in frogs that were force-fed with encysted metacercariae, have been studied at regular intervals. The prepatent period is 14–19 days. The present life cycle study of a Pleurogenoides spp. forms the seventh report from the world, fourth report from India and the third from Kerala.

The nematode genus Acrobeles is composed of two morphological groups distinguished by the presence (‘double’ cuticle) or absence (‘single’ cuticle) of the refringent inner layer of the cuticle. In the present study, four species of this genus, two with ‘single’ cuticle (Acrobeles ciliatus and Acrobeles cylindricus) and two with ‘double’ (Acrobeles aenigmaticus and Acrobeles complexus) are studied from coastal dunes in Spain. This study provides detailed morphological and morphometrical analyses for the four species, while molecular analysis, based on 18S and 28S ribosomal DNA, is provided for A. complexus. The four species are studied with scanning electron microscopy, which is obtained for the first time for A. cylindricus. These analyses revealed morphological and molecular differentiations between both groups, appearing as two related monophyletic entities. The subgenera Acrobeles and Seleborca, formerly considered as separate genera, are proposed to accommodate both groups.

The aim of the study was to compare the relative gene expression of Haemonchus contortus P-glycoprotein genes (Hco-pgp) between fourth (L4), infective (L3), and transitory infective (xL3) larval stages as laboratory models to study ivermectin (IVM) resistance. The H. contortus resistant to IVM (IVMr) and susceptible to IVM (IVMs) strains were used to develop xL3 in vitro culture and to infect Meriones unguiculatus (gerbils) to collect L4 stages. Morphometric differences were evaluated from 25 individuals of H. contortus from each strain. Relative gene expression from xL3 and L4 was determined between comparison of IVMr stages and from IVMr vs IVMs stages. Seven Hco-pgp genes (1, 2, 3, 4, 9, 10, and 16) were analysed by RT-qPCR using L3 stage as control group, per strain, and GAPDH and β-tubulin as constitutive genes. Morphological changes were confirmed between xL3 and L4 developing oral shape, oesophagus, and intestinal tube. In addition, the body length and width showed statistical differences (p < 0.05). The Hco-pgp1, 2, 3, and 4 genes (p < 0.05) were upregulated from 7.1- to 463.82-fold changes between IVMr stages, and Hco-pgp9 (13.12-fold) and Hco-pgp10 (13.56-fold) genes showed differences between L4 and xL3, respectively. The comparative study between IVMr vs IVMs strains associated to xL3 and L4 displayed significant upregulation for most of the Hco-pgp genes among 4.89–188.71 fold-change. In conclusion, these results suggest the use of H. contortus xL3 and L4 as suitable laboratory models to study IVMr associated with Hco-pgp genes to contribute to the understanding of anthelmintic resistance.

As part of surveillance of snail-borne trematodiasis in Knowsley Safari (KS), Prescot, United Kingdom, a collection was made in July 2021 of various planorbid (n = 173) and lymnaeid (n = 218) snails. These were taken from 15 purposely selected freshwater habitats. In the laboratory emergent trematode cercariae, often from single snails, were identified by morphology with a sub-set, of those most accessible, later characterized by cytochrome oxidase subunit 1 (cox1) DNA barcoding. Two schistosomatid cercariae were of special note in the context of human cercarial dermatitis (HCD), Bilharziella polonica emergent from Planorbarius corneus and Trichobilharzia spp. emergent from Ampullacaena balthica. The former schistosomatid was last reported in the United Kingdom over 50 years ago. From cox1 analyses, the latter likely consisted of two taxa, Trichobilharzia anseri, a first report in the United Kingdom, and a hitherto unnamed genetic lineage having some affiliation with Trichobilharzia longicauda. The chronobiology of emergent cercariae from P. corneus was assessed, with the vertical swimming rate of B. polonica measured. We provide a brief risk appraisal of HCD for public activities typically undertaken within KS educational and recreational programmes.

Unusual non-human parasitic nematodes and eggs were detected in the faeces of an 8-year-old Japanese female suffering from Henoch-Schönlein purpura. The worms were adult female rhabditiform nematodes measuring 325.6–441.2 μm in length and 18.3–26.5 μm in width. One pair of the labia oris was notched with many spiny projections, while the other pair was strongly curved outwards. The worms were identified using light and scanning electron microscopy as the free-living nematode Diploscapter coronata (Cobb) based on their characteristic morphology. The patient's faeces containing worms and eggs were cultured using a filter-paper culture technique and after 7 days of culture, male as well as female worms were recovered. Worm survival time and hatchability of the eggs were examined in vitro after treatment with an artificial gastric or intestinal fluid. Although adult worms survived for less than one minute, eggs hatched after treatment with artificial gastric fluid. This suggests that eggs accidentally ingested or produced by adult D. coronata could develop in the human gastro-intestinal tract. Some morphological features of male D. coronata are also described.

Liquid culture is the most scalable technology for the industrial production of entomopathogenic nematodes. Variability of the recovery after inoculation into cultures of Photorhabdus luminescens remains a persistent problem in the mass production of Heterorhabditis sp. In order to enhance infective juvenile (IJ) recovery and improve nematode population management, we analysed the correlation between the nematode Heterorhabditis megidis (strain KV – 136) development in liquid cultures, the density of bacteria of P. luminescens and the culture agitation speed. Analyses focused on the impact of different agitation speeds (160 rpm and 200 rpm) on the dynamics of population growth of H. megidis in liquid cultures at constant biotic and abiotic parameters (initial dose of nematodes introduced to the culture 2300 IJs/ml, temperature 25°C, the number of bacterial colonies 0.3 × 107/ml). The performed experiments showed that the agitation speed of 200 rpm favourably affected the density of bacteria of P. luminescens (24.14 × 107/ml). High density of bacteria at this agitation speed resulted in an earlier (on the fifth day of the culture) maximum increase in the number of hermaphroditic individuals (1239.6 H/ml) than in the culture at an agitation speed of 160 rpm.

New Zealand’s kākāpō parrot, once widespread, is now critically endangered due to habitat loss and introduced mammalian predators. Prior to major population decline, a unique kākāpō cestode, Stringopotaenia psittacea, was found in the 1880s and first described in 1904. Here we report the discovery of eggs of this cestode in kākāpō coprolites of pre-human settlement age from the Honeycomb Hill cave system, north-west Nelson. Analysis of 52 samples, including coprolites of post-human settlement age, from nine sites within six South Island locations across a wide geographic range, yielded only eight infected samples in this single cave system. Results suggest that prior to human settlement, S. psittacea was not widespread within and between kākāpō populations, in marked contrast to other parasite types of the extinct moa spp. Intense management of the last remaining kākāpō has endangered or possibly caused the extinction of this cestode. This is the first confirmed record of S. psittacea since its discovery in 1884.

The occurrence, location and sex ratio of Corynosoma strumosum (Acanthoceophala: Polymorphidae) from 26 juvenile grey seals, Halichoerus grypus, by-caught from around the Inishkea Islands, Co. Mayo, from March to June 1997 were recorded. The location of C. strumosum within the seal did not vary, with all worms being found in the small intestine. The prevalence of infection was 100%, with no other acanthocephalan species being recorded. The mean abundance was 416 (range 80–846) C. strumosum per seal. There was no significant change in abundance by month. The sex ratio of mature female to mature male parasites was 1.2:1. The lengths of female and male worms did not differ significantly.

While much attention has been paid to vector-borne filariasis, diseases that threaten millions of people in tropical and subtropical countries, the literature on host–parasite associations and transmission strategies of filarial nematodes in wildlife is scarce. Here, we report the co-occurrence of chigger mites (Eutrombicula alfreddugesi) and onchocercid nematodes (Oswaldofilaria chabaudi) parasitizing the lizard Tropidurus torquatus in the State of Minas Gerais, Brazil. Examination of chiggers established, for the first time, the occurrence of microfilariae in trombiculid mites (Trombiculidae). These larvae were morphologically similar to those recovered from adult females of O. chabaudi. The current evidence suggests that chiggers do not play a role in the transmission of filarioid nematodes, but rather act as accidental or dead-end hosts. Nevertheless, considering the polyphagous nature of trombiculid mites, similar to blood-sucking insects involved in the transmission of several infectious diseases, further studies may shed light on the potential role of chiggers as vectors of filarioids.

It is concluded that Bearup's cercaria, described from Pyrazus australis in Australia, is different from Cercaria variglandis; the former develops into Austrobilharzia terrigalensis and the latter into A. variglandis (Syn. M. Chapini Price, 1929; M. variglandis Stunkard and Hinchliffe, 1952).

Bearup's cercaria should be henceforth treated as that of A. terrigalensis and not as C. variglandis.

The histochemical site and distribution of hexokinase, glycogen phosphorylase (GP Rylase), lactate dehydrogenase (LDH) (key enzymes of glycolysis), glucose-6-phosphate dehydrogenase (GPD) and 6-phosphogluconate dehydrogenase (6PGD) (pentose phosphate shunt enzymes), isocitrate dehydrogenase (IDH), succinate dehydrogenase (SDH), malate dehydrogenase (MDH), and α-ketoglutarate dehydrogenase (α-KDH) (key enzymes of Krebs' cycle), malate synthetase (MS) and isocitrate lyase (IL) (enzymes of glyoxylate shunt) in various tissues of Ganeo tigrinum from hibernating and non-hibernating Rana cyanophlyctis and R. tigrina were studied. Differences in their intensities were revealed. Weak activity of GP Rylase and strong activity of hexokinase in flukes from non-hibernating hosts indicates that they utilize glucose through glycolysis for energy turnover. Intense GP Rylase and weak hexokinase activity in worms from hibernating hosts indicates the utilization of glycogen. Strong activity of IDH, SDH, MDH, α-KGD, MS and IL was demonstrable in the tissues of flukes from non-hibernating hosts, suggesting that Krebs' cycle and glyoxylate shunt, respectively, were operating. Tissues of the fluke from hibernating hosts, on the other hand, displayed positive activity only for SDH and MDH; no activity for MS and IL, the enzymes of glyoxylate shunt, was observed, The activity of the above enzymes was found to be relatively low in worms from hibernating hosts.
