This study provides a morphological and phylogenetic characterization of two novel species of the order Haplosporida (Haplosporidium carcini n. sp., and H. cranc n. sp.) infecting the common shore crab Carcinus maenas collected at one location in Swansea Bay, South Wales, UK. Both parasites were observed in the haemolymph, gills and hepatopancreas. The prevalence of clinical infections (i.e. parasites seen directly in fresh haemolymph preparations) was low, at ~1%, whereas subclinical levels, detected by polymerase chain reaction, were slightly higher at ~2%. Although no spores were found in any of the infected crabs examined histologically (n = 334), the morphology of monokaryotic and dikaryotic unicellular stages of the parasites enabled differentiation between the two new species. Phylogenetic analyses of the new species based on the small subunit (SSU) rDNA gene placed H. cranc in a clade of otherwise uncharacterized environmental sequences from marine samples, and H. carcini in a clade with other crustacean-associated lineages.

The European shore crab, Carcinus maenas, is one of the most successful marine invasive species. Its success has been in part attributed to the loss of parasites, rekindling an interest in host-parasite interactions and impacts on host fitness in this crab. In the present study, we investigated C. maenas populations from Europe, South Africa and Australia for parasites, and assessed their impact on the fitness of male crabs. For the shore crab, testes weight along with success in mating competition is traded off against other life-history traits. We therefore used this parameter as an indicator both for reproductive fitness and a possible resource trade-off in response to parasite infestation. In the native range, crabs infested with Sacculina carcini showed significantly lower testes weight than uninfected crabs. However, helminth parasites did not generally cause reduced testes weights. Crab populations from South Africa and Australia were either parasitized at very low prevalences, or were completely parasite free. However, no population level effect of this parasite release was reflected in testes weight. These findings do not support a severe fitness impact of helminth parasites on C. maenas, which questions the role of parasites on its population dynamics, both in the native area and for invasive success.

The temporal and spatial variability in population dynamics of the European crab Carcinus maenas (L.) was studied between June 2003 and September 2007 in the Mondego estuary. An extreme climate event (extreme drought) occurred in 2004 and 2005 with a low freshwater flow discharge into the estuary, leading to an increase in salinity. Juveniles' recruitment was continuous throughout each year with a high proportion of young recruits being recorded in the spring of 2005. The proportion of green crabs clearly increased from the mouth to the upstream areas and the proportion of crabs actually in moult followed the same pattern. Parasitization by the endoparasitic barnacle, Sacculina carcini was observed, principally at the mouth of the estuary. The C. maenas population showed a regular size–frequency distribution and structure for both sexes during the study. Secondary production (P) of C. maenas estimated for the 4-year study period was different between years though, P/B ratios were similar during the whole study period. During the drought period, catches of the crab recruits were higher, particularly in the spring, probably due to the increase in salinity, which emphasizes the importance of these extreme events in controlling the abundances of coastal populations.

This investigation reports for the first time the establishment of a trematode life-cycle in the laboratory using in ovo cultivation of the adult stage. Microphallus primas metacercarial cysts were removed from Carcinus maenas crabs and chemically excysted. Using a modification of the technique developed by Fried (1962) the released metacercariae were grown in fertile hen's eggs to ovigerous adult flukes. These were removed from the under surface of the chorio-allantoic membrane of chick embryos 5 days post-infection and the fluke eggs recovered were used to infect laboratory-reared Hydrobia ulvae snails. When challenged against parasite-free crabs the cercariae released from the experimentally infected snails gave rise to metacercarial cysts that were again identified as M. primas.

Habitat modelling requires incorporation of both biotic and abiotic information. For juvenile flatfish the factors that most influence growth are water temperature, food abundance and predatory pressure. This study focuses on the impact the predator, the shore crab, Carcinus maenas, has on the foraging activity of sole, Solea senegalensis. The results show that in the presence of both prey (ragworm, Nereis diversicolor) and predator, ≃10% decrease in foraging activity is observed when compared to the sole in the presence of only food. This suggests that when the shore crab is present, Senegalese soles are not only affected by direct predation but sub-optimal foraging is also found. Behaviours most correlated with foraging were crawling and tapping and these activities were also affected by the presence of the predator. This study also provides further support for visual recognition of predators and olfactory prey recognition in the Senegalese sole. Predator–prey encounters shape species behaviours profoundly and have to be looked at from different levels of ecology, behaviour and modelling.

Landmark-based geometric morphometric methods were used to test which chela is most suitable to use in population differentiation of brachyuran crabs. Two species were analysed: Pachygrapsus marmoratus, a homochelous species, and Carcinus maenas, a heterochelous species. Variance analysis of shape variables and of each chela was calculated to determine which claw presents less variation within populations. The results showed that the right chela of P. marmoratus is more appropriate to use when the goal is to differentiate populations. In C. maenas, the males major chela and females right chela are more suitable to use, probably due to its heterochely. To test these results, specimens from different geographical locations were compared. Pachygrapsus marmoratus populations presented significant differences in manus shape, and C. maenas populations had differences in polex size and shape. Differences in carapace shape were also encountered between populations of both species. This phenotypic variation was not corroborated with genetic data, which were obtained using the mitochondrial DNA gene cytochrome oxidase I as a marker. These opposite results suggest that environmental factors, such as diet and parasitism, can be determining shape differentiation.

The ecdysteroids ecdysone (E), 20-hydroxyecdysone (20E) and ponasterone A (PoA) were measured in the haemolymph, hepatopancreas and oocytes of female shore crabs Carcinus maenas during the moulting cycle and during oocyte maturation using HPLC-MS. In the haemolymph, ecdysteroid titres varied over the moulting cycle with high levels during premoult and low levels during postmoult and intermoult, however, no significant change in haemolymph ecdysteroid titres was observed in relation to oocyte development. In the hepatopancreas, PoA levels were high during premoult but low during postmoult and intermoult. This is in contrast to E and 20E where levels remained high from early intermoult (C1) until late premoult (D3) and only decreased during postmoult. In the oocytes, ecdysteroid levels were low during postmoult and for 20E and PoA also during late premoult D2 and D3. In contrast, all three ecdysteroids were observed to increase in the oocytes during oocyte development, in particular E and PoA. The present study demonstrates that changes in haemolymph ecdysteroid titres relate to changes in moulting status and not to changes in oocyte development. Also, the study indicates that the hepatopancreas is involved in the metabolism of ecdysteroids related to the moulting cycle but may also be involved in ecdysteroid metabolism during oocyte development. Furthermore, the pronounced increase in oocyte ecdysteroids during oocyte development during periods where haemolymph ecdysteroids titre is low, indicates that the oocytes are capable of de novo synthesis of the three ecdysteroids investigated.

This study established the mechanisms by which Microphallus primas cercariae penetrate the crab Carcinus maenas in which they form metacercarial cysts. Light and electron microscopy were used to investigate cercarial features and to follow the fate of cercariae released in close proximity to crabs. It was shown that cercariae were carried in respiratory currents into crabs' branchial chambers where each enveloped itself in a transparent penetration cyst on the gill lamellae. When cercariae were present the number of respiratory current reversals performed by crabs increased. Using an ‘artificial branchial chamber’ it was possible to observe how cercariae attached to crab gills during breaks in current flow that preceded each current reversal. Inside the penetration cysts the now tail-less larvae used their stylets to pierce holes through which they levered themselves into underlying haemolymph channels in the gills. Histochemical tests demonstrated that the penetration cysts were products of glands in the cercariae and that penetration of the crabs was achieved by mechanical means. The importance of crab respiratory current reversals to the success of cercarial penetration is discussed as it represents the exploitation by a parasite of a host behavioural response to an unrelated stimulus.
