About 400 subglacial lakes are known from Antarctica. The question of whether life unique of subglacial lakes exists has been paramount since their discovery. Despite frequent evidence of microbial life mostly from accretion ice, subglacial lakes are characterized by physiologically hostile conditions to metazoan life, as we know it. Pure water (salinity ≤0.4–1.2%), extreme cold (−3°C), high hydrostatic pressure, areas of limited or no oxygen availability and permanent darkness altogether require physiological adaptations to these harsh conditions. The record of gene sequences including some associated with hydrothermal vents does foster the idea of metazoan life in Lake Vostok. Here, we synthesize the physico-chemical environment surrounding sub-glacial lakes and potential sites of hydrothermal activity and advocate that the physico-chemical stability found at these sites may be the most likely sites for metazoan life to exist. The unique conditions presented by Lake Vostok may also offer an outlook on life to be expected in extra-terrestrial subglacial environments, such as on Jupiter's moon Europa or Saturn's moon Enceladus.

The influence of hydrostatic compression on the charge transport properties of an excellent 2,6-diphenylanthracene (2,6-DPA) semiconducting single crystal was investigated up to 10 GPa by performing density-functional calculations together with the tight binding approximation. In this pressure region the lattice constants a, b and c decrease by up to 0.948 Å (5.23%), 1.30 Å (17.26%), and 0.711 Å (11.34%), respectively, while the monoclinic angle β increases by 3.4°. The unit-cell volume decreases by increasing pressure, and the volume decreases by 30.5% at 10 GPa. In comparison, the C–C and C–H intermolecular distances within and between the herringbone layers reduced by 16–19% and 16–24%, respectively, in the same pressure ranges. The results indicate that under high pressure, the molecular planes of the crystal become more and more parallel to each other due to molecular rearrangement in the 2,6-DPA crystal. The band gap decreases with increasing pressure due to decreasing intermolecular separation between neighboring molecules. Finally, the results indicate an improvement of the hole mobility of 2,6-DPA single crystals under hydrostatic pressure.

At present, there is a huge interest in finding new prebiotics from agrofood industrial waste, such as the soyabean by-product Okara, rich in insoluble dietary fibre. A previous treatment of Okara with high hydrostatic pressure assisted by the food-grade enzyme Ultraflo® L achieved a 58·2 % increment in its soluble dietary fibre (SDF) contents. Therefore, potential prebiotic effect of both treated and native Okara was assayed using 48 h, pH-controlled, anaerobic batch cultures inoculated with human faecal slurries, which simulate the human gut. Changes in faecal microbiota were evaluated using 16S rRNA-based fluorescence in situ hybridisation, whereas release of SCFA and lactic acid was assessed by HPLC. Both Okara samples exhibited potential prebiotic effects but Okara treated to maximise its SDF content showed higher SCFA plus lactic acid, better growth promotion of beneficial bacteria, including bifidobacteria after 4 and 48 h and lactobacilli after 4 h of fermentation, and a greater inhibition of potentially harmful bacterial groups such as clostridia and Bacteroides. Differences found between fructo-oligosaccharides and Okara substrates could be attributed to the great complexity of Okara’s cell wall, which would need longer times to be fermented than other easily digested molecules, thus allowing an extended potential prebiotic effect. These results support an in vitro potential prebiotic effect of Okara.

The effect of hydrostatic pressure treatment on the induction of mitogynogenesis in the eggs of Japanese flounder Paralichthys olivaceus (Temminck et Schlegel) by using heterospecific sperm were studied. Before treatment, the eggs were at metaphase of the first mitosis. The spindle was disassembled by the treatment and then resembled in its pretreatment position, and the chromosomes were rearranged, i.e., the first mitosis was not blocked. During the second mitotic cycle, only a monopolar spindle was assembled in each blastomere and the chromosomes doubled, but cell cleavage was blocked. In the third cycle, mitosis proceeded normally with a bipolar spindle in each blastomere. Flow cytometric analysis of ploidy demonstrated that mitogynogenetic larvae were all diploid. The ultraviolet light-irradiated sperm of the red sea bream (Pagrus major) was condensed, formed a dense chromatin body, and randomly entered one blastomere.

This paper studies the buckling analysis for a ring-stiffened cylindrical shell consisted of functionally graded material (FGM) subjected to hydrostatic pressure and thermal loads. Material properties of the ring-stiffened FGM cylindrical shell are assumed to be temperature-dependent, and vary smoothly through the thickness direction of the structure according to a volume exponent. Based on the Donnell assumptions, buckling loads of the ring-stiffened FGM cylindrical shell are presented by utilizing the Galerkin method. Numerical results reveal that thermal loads, volume exponent and geometric parameters have significant effects on the buckling behavior of the ring-stiffened cylindrical shell.

This article presents the results from a study of drainage rates and groundwater pressure distributions surrounding a horizontal drain pipe. The pipe is assumed to be applied on a retaining wall and reached a steady state condition. The optimized design pattern for horizontal drain pipes on a retaining wall is deeply related to the efficiency of dissipation of groundwater and the pore pressure behind the wall. Therefore, simulations of groundwater flow through a single horizontal drain pipe with different sizes (diameters and lengths) and depths by a 3-D finite element program model would lead us to find out the mechanism of the drainage pattern. The study has revealed the equalized water pressure contours surrounding the horizontal drain pipe exhibited horn shapes distributions. The biggest but still has a closed shape of horn was named Characterized Iso-pressure Surface (CIPS) in this research. It was found to be a good object to reflect the effects of single drain pipe so as to explore the mechanisms of drainage pattern. This research conducted a series study of dimension factor influence. It could be a help in developing assessment and design methods for horizontal drain pipes applied to the retaining wall.

The aim of this study was to analyse the tolerance of the planktonic stages of Psammechinus miliaris to hydrostatic pressure and temperature. Embryos of Psammechinus miliaris were subjected to different combinations of pressure and temperature for 3, 6 and 12 h. The percentage of embryos at each stage and the percentage of embryos developing abnormally were measured. Larvae at the gastrula and prism stages were subjected to pressure and temperature combinations for 24 h and the larval survival was calculated measuring the percentage of swimming larvae. Both embryos and larvae could survive at greater pressures than the known adult depth limits. Larvae showed a much greater potential than embryos for surviving deeper, with approximately 100% of both gastrulae and prisms surviving up to 200 atm at 5°C. These results are similar to other shallow-water species of Echinoida. Thus larval tolerance of high pressure and low temperature may have been important for the success of this group in colonizing the deep-sea.

Balloon dilatation is well established in the management of enteric strictures. The use of this technique in the pharynx has been reported under radiological and endoscopic guidance. We describe the hydrostatic dilatation of a benign pharyngeal stricture in a laryngectomy patient under local anaesthetic, without radiological guidance, in the ENT out-patient department. This procedure was effective and well tolerated in a patient who required regular dilatations.

The effect of pressure on the unfolding of bovine α-lactalbumin was investigated by ultraviolet absorption methods. The change of molar volume associated with unfolding, ΔV, was measured in the presence or absence of guanidine hydrochloride at pH 7. The ΔV was estimated to be −63 cm3/mol in the absence of a chemical denaturant. While in the presence of guanidine hydrochloride (GuHCl), it was found that ΔV was −66 cm3/mol at 25 °C and was independent of the concentration of GuHCl, despite the fact that the molten globule fraction in the total unfolding product decreased with the increase of GuHCl concentration. The results indicate that the volume of α-lactalbumin only changes at the transition from a native to a molten globule state, and almost no volume change has been found during the transition from a molten globule to the unfolded state.

Native defects and some common dopants (Mg, Zn, and C) in cubic GaN and AlN are examined by means of ab initio theoretical calculations using two methods: i) the Green's function technique based on the linear muffin-tin orbital method in the atomic-spheres approximation; ii) a supercell approach in connection with the full-potential linear muffin-tin-orbital method. We apply the first method to look mainly at the energetic positions of the defect and impurity states in different charge states and their dependence on hydrostatic pressure. The second method allows us to study lattice relaxations. Whereas small relaxations are found near vacancies and substitutional Mg and Zn, the calculations predict large atomic displacements around antisite defects and the substitutional carbon impurity on the cation site.
