The potential of Momordica charantia var. muricata, a little-known wild form, as a source of favourable alleles in breeding and improvement of bitter melons (M. charantia var. charantia), is demonstrated. The M. charantia var. charantia cultivar Priyanka and M. charantia var. muricata accession IC634896 were crossed with each other and cross ability and performance of F1 plants were studied. These botanical varieties were fully cross-compatible on either direction. Surprisingly, cross of Priyanka × IC634896 produced 80.0 fruits compared to 17.0 in Priyanka and fruit yield per plant (2287.4 g) was more than double that in Priyanka (1042.3 g). This hybrid has to be back crossed with commercial cultivar to improve its fruit size, with no compromise on fruit number. This is the first study showing the promise of M. charantia var. muricata as a potential donor in bitter melon breeding.

Single nucleotide polymorphism (SNP) genotyping tools, which can analyse thousands of SNPs covering the whole genome, have opened new opportunities to estimate the inbreeding level of animals directly using genome information. One of the most commonly used genomic inbreeding measures considers the proportion of the autosomal genome covered by runs of homozygosity (ROH), which are defined as continuous and uninterrupted chromosome portions showing homozygosity at all loci. In this study, we analysed the distribution of ROH in three commercial pig breeds (Italian Large White, n = 1968; Italian Duroc, n = 573; and Italian Landrace, n = 46) and four autochthonous breeds (Apulo-Calabrese, n = 90; Casertana, n = 90; Cinta Senese, n = 38; and Nero Siciliano, n = 48) raised in Italy, using SNP data generated from Illumina PorcineSNP60 BeadChip. We calculated ROH-based inbreeding coefficients (FROH) using ROH of different minimum length (1, 2, 4, 8, 16 Mbp) and compared them with several other genomic inbreeding coefficients (including the difference between observed and expected number of homozygous genotypes (FHOM)) and correlated all these genomic-based measures with the pedigree inbreeding coefficient (FPED) calculated for the pigs of some of these breeds. Autochthonous breeds had larger mean size of ROH than all three commercial breeds. FHOM was highly correlated (0.671 to 0.985) with FROH measures in all breeds. Apulo-Calabrese and Casertana had the highest FROH values considering all ROH minimum lengths (ranging from 0.273 to 0.189 and from 0.226 to 0.152, moving from ROH of minimum size of 1 Mbp (FROH1) to 16 Mbp (FROH16)), whereas the lowest FROH values were for Nero Siciliano (from 0.072 to 0.051) and Italian Large White (from 0.117 to 0.042). FROH decreased as the minimum length of ROH increased for all breeds. Italian Duroc had the highest correlations between all FROH measures and FPED (from 0.514 to 0.523) and between FHOM and FPED (0.485). Among all analysed breeds, Cinta Senese had the lowest correlation between FROH and FPED. This might be due to the imperfect measure of FPED, which, mainly in local breeds raised in extensive production systems, cannot consider a higher level of pedigree errors and a potential higher relatedness of the founder population. It appeared that ROH better captured inbreeding information in the analysed breeds and could complement pedigree-based inbreeding coefficients for the management of these genetic resources.

Dongxiang common wild rice (Oryza rufipogon Griff., DXWR) is an important genetic resource for the improvement of cultivated rice. For the past three decades, great achievements have been made in the field of molecular marker development. Although structural variations (SVs) had been studied between DXWR and Nipponbare (Oryza sativa L. ssp. japonica), the development and application of SV markers in DXWR has not been reported. In this study, based on the genome-wide SV loci, we developed and synthesized a total of 195 SV markers that were evenly distributed across the 12 rice chromosomes. Then, these markers were tested for their stabilities and polymorphisms. Of these 195 markers, 147 (75.4%) were successfully amplified and displayed abundant polymorphisms between DXWR and Nipponbare. Meanwhile, through the genotyping of 20 rice varieties from 13 countries and areas, we concluded that these SV markers have a wide application prospect in the analysis of cultivated rice. Therefore, these molecular markers greatly enrich the number of markers available for DXWR, which will facilitate genomic research and molecular breeding for this important and endangered germplasm resource.

In this Research Communication we evaluate the fatty acid (FA) profile of Wallachian sheep milk. The study was performed on 38 ewes in Beskydy Mountains. Samples were collected 4 times during the lactation, at monthly intervals. FA profile as well as groups of saturated, monounsaturated (MUFA), and polyunsaturated (PUFA) FAs were investigated. Considerable increase over the lactation was detected for lauric, myristic, myristoleic and palmitic acids, while stearic acid showed the opposed tendency. Variability, supported by significant differences among particular days of milk collection, was demonstrated for oleic acid; its highest distribution occurred at the beginning and at the end of the trial. The highest distribution of CLA was at the second sample day. Milk of Wallachian sheep naturally grazed at permanent pasture areas showed higher content of PUFA and MUFA in contrast with intensive or semi-intensive sheep breeds reported in the literature.

Shortawn foxtail (Alopecurus aequalis Sobol.) is an invasive and highly troublesome weed species originating from North America that has become widespread across China. Since its proliferation seriously threatens crop production worldwide, understanding its genetic diversity is critical for developing a forecasting system for integrated pest management plans. To accelerate the application of molecular markers in A. aequalis, this study aimed to develop a set of expressed sequence tag-simple sequence repeat (SSR) markers using previous high-throughput sequencing data. In this study, a total of 1411 SSR loci were identified from 95,479 unigenes. Tri-nucleotide repeat motifs were the most abundant type with a frequency of 66.27%, followed by di- (24.95%) and tetra-nucleotide (8.78%). Among the loci, 584 primer pairs were successfully designed for marker development. Subsequently, a subset of 36 primer pairs was randomly selected and synthesized, of which 12 (33.33%) pairs successfully revealed abundant allelic polymorphism. Additionally, to investigate their utility, the genotypes of 160 individuals from 20 natural populations representing diverse wild genotypes of A. aequalis were analysed by using these 12 polymorphic markers. These novel SSR markers developed here are reliable and useful for genetic analysis on this invasive plant and will greatly enrich its genetic resource.

Dongxiang wild rice (Oryza rufipogon, DXWR), one of the species of common wild rice, is regarded as an important genetic resource for the improvement of cultivated rice (Oryza sativa). Molecular markers are reliable tools that can greatly accelerate the breeding process and have been widely used in various species. In the present study, a total of 3681 genic simple sequence repeat (SSR) markers were developed for DXWR based on transcriptome sequencing technology. Additionally, 25 primer pairs were randomly selected and synthesized for the verification. Among them, 18 (72%) primer pairs were successfully amplified in PCR amplification with genomic DNA of DXWR and also had abundant polymorphisms between DXWR and cultivated rice. These novel genic SSR markers will enrich current genomic resources for DXWR, and provide an effective tool for genetic study and molecular marker assisted breeding for this valuable and endangered germplasm.

It is known that genetic diversity is the most important factor in classical and modern plant breeding. The considerable increase in the number of transgenic crops reveals the value of new plant genetic resources. In this study, a set of 12 wheat progenitors were screened for tissue culture parameters such as callus induction, callus weight, regeneration capacity of callus and callus efficiency using mature embryos. Embryos were excised from imbibed seeds of the progenitors. The excised embryos were placed scutellum upwards in dishes containing 2 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D) for callus induction. The developed calli and regenerated plants were maintained on 2,4-D free MS medium. When mature embryos of 12 wheat progenitors (Aegilops sp. and Triticum sp.) were compared, significant differences were detected in callus induction frequency, weight of callus, regeneration capacity and culture efficiency. A significant genotypic effect was observed on the culture responses. Of the 12 wheat progenitors tested, Aegilops umbellulata had the highest regeneration capacity of callus. Aegilops biuncialis created the most regenerable calli because of the highest callus induction and culture efficiency. In the experiment, callus induction was significantly correlated with callus weight (r= 0.820) and regeneration capacity (r= 0.955). Weight of callus was significantly correlated with regeneration capacity (r= 0.740), while there was no significant correlation between callus induction frequency and culture efficiency (r= 0.350). Our results showed that, generally, mature embryos of some Aegilops and Triticum species have a high regeneration capacity, and therefore, can be used as an effective explant source for the successful application of biotechnology in crop improvement.

The objective of this paper was to estimate the farmers' preferences based on the non-market traits in selection, breeding and conservation of indigenous chicken. The study was carried out in four selected counties of Kenya. Data were collected using structured questionnaires with the use of multi-attribute elicitation techniques applied to 450 rural households. Conjoint analysis through an ordered probit model was used to analyse the data. The study identified traits preferred by the farmers based on their current low-input production circumstances. Using these traits, indigenous chicken can be selected for higher productivity and performance while retaining their diversity and adaptability. In general, the determination of the importance of various attributes elucidates to the stakeholders some valuable aspects that would assist them in overcoming some of the major constraints of indigenous chicken production in rural areas and at the same time provides vital information for selection, conservation and development of breeding programmes.

Leicester Longwool sheep are pivotal in the history of standardised sheep breeds. The breed was imported into the United States in 1990, and it was never imagined as more than a minor sideline to the array of breeds in the United States. Eighteen sheep were originally imported from Tasmania, with further contributions via semen from 12 rams in Australia, New Zealand, and the United Kingdom. In subsequent years the population of this breed in those source countries has plummeted, leaving the United States with the largest national flock (800). The decline in numbers worldwide have made the genetic management of the North American population increasingly important. The wool is sought by handcrafters and brings a premium price. Attention to maintaining the breed with its original fleece characteristics is an important priority, especially given the breed's role in the development of multiple longwool breeds. One divergence of the American flock from the other countries is the registration of coloured animals, and these make up 6% of the current flock.

On-farm surveys and characterisation of six indigenous north Ethiopian cattle breeds (Afar, Arado, Begait, Fogera, Medenes and Raya) was carried out with the objective of determining the current status of these breeds. Information from a structured questionnaire distributed to 450 farmers, 37 group discussions, field measurements of heart girth and height on 25 males and 25 females from each breed and secondary information were used to carry out the study. Population size of the Arado breed increased by more than three fold between 1992 and 1999, while the Raya, Fogera and Begait population decreased by57, 27 and 67%; between 1981 and 1999 respectively. Three distinctive types were identified within the Afar and Begait breeds with different adaptation and threat levels in relation to changes to the bio-physical and social environment. Overall production and reproduction performance of the breeds in the pastoral production system (Afar and Begait) was higher than the Raya and Medenes, and Arado and Fogera breeds of the agro-pastoral and mixed crop/livestock production systems respectively. Extinction probability for most ofthe breeds was high, the highest (0.67) being for the Begait breed. On the other hand, except for the initiative taken to evaluate, improve and conserve the Fogera breed at the Metekel and Andasa cattle breeding ranches, there are no institutionalized attempts towards improving and/or conserving the other breeds.

North Bengal Grey (NBG) cattle are an important indigenous cattle genetic resource found mainly in the northern part of Bangladesh. The study was undertaken at Bogra Sadar, Shibgonj and Kahalu Upazila (sub-district) in the Bogra district. The physical and morphological characteristics, and the productive and reproductive performances of NBG cattle were studied. The coat colour of these animals is deep grey to white. The coat colour of the neck region in adult bulls was found to be generally ashy with a range of shades.

The body is small, compact and less fleshy. Ear length and ear width were 18.0±0.17 and 11.0±0.21 cm, respectively. The head length average was 38.0±0.56 cm, the head width 16.0±0.17 cm, the foreleg length average 65.0±0.64 cm, the hind leg length 71.0±0.64 cm, the tail length average 71.0±0.67 cm, the horn length average 9.0±0.39 cm, the horn diameter 10.0±0.37 cm, the average teat length 5.0±0.18 cm, the teat diameter 6.0±0.22 cm, the distance betweenthe front teats 7.0±0.13 cm and the distance between the rear teats 7.0±0.13 cm. Body length, height at wither and heart girth in adult cows were 105.0il.20, 94.0+1.12 and 127.0±1.52 cm, respectively.

The recorded highest peak milk production per day was 3.5±0.18 kg, lactation length was 219±6.1 days, and the dry period was 180±6.8 days. The average birth weight of calves was 18.4±0.52 kg and mature live weight of cows 241.0±4.0 kg. The age at first heat was 869±29.6 days, age at first calving 1191±19.7 days, gestation length 281±1.3 days, calving interval 442±7.4 days, postpartum heat period 110±4.2 days and the number of services per conception 1.4±0.6. About 54% of total cattle population was NBG cattle in the surveyed area of Bangladesh. The results indicated that the productive and reproductive performance of NBG cattle was better than other non-descript indigenous cattle of Bangladesh. The study further revealed an obvious need for more in-depth and objective information on wider samples of this type of indigenous cattle in order to assess the future need for conservation and improvement programs to be undertaken.

A field survey was carried out between January and June 2002 in order to describe and evaluate two local cattle breeds, Abergelle and Irob, found in the Tigray region of North Ethiopia with little previous description available. Information from structured questionnaire of 175 farmers, twenty-one focused group discussions, field measurements of hearth girth and height at withers of 25 males and 25 females of each breed, and secondary information were utilised to carry out an on-farm characterisation and description of these breeds. The two breeds are found to be the smallest breeds in the region with an average body weight of 234 ± 13 and 153 ± 15 kg and average height at withers of 109 ± 3 and 97 ± 5 cm for males and females of the Abergelle breed. For the Irob breed the corresponding figures were 245 ± 36 and 200 ± 36 kg body weight and 106 ± 3 and 105 ± 5 cm height at withers. The Abergelle breed is recognized by farmers to have adaptive advantages to the hotter and drier low lands. Its tolerance to diseases and parasites and ability to cope with feed shortages during the long dry periods are favourably rated by the majority of farmers. The Irob cattle breed is adapted to a mountainous production environment and is highly rated by farmers for its capacity to thrive on the difficult terrain and its utilization of cactus (Opantia ficus indica) as a major source of nutrition.
