Clays can act as osmotic membranes and thus give rise to osmotically induced hydrostatic pressures. The magnitude of generated osmotic pressures in geologic systems is governed by the theoretical osmotic pressure calculated solely from solution properties and by value of the membrane's three phe-nomenological coefficients: the hydraulic permeability coefficient, Lp; the reflection coefficient, σ; and the solute permeability coefficient, ω. Generally, low values of Lp correspond to highly compacted membranes in which σ is near unity and ω approaches zero. Such membrane systems should give rise to initially high osmotic fluxes and gradual dissipation of their osmotic potentials.

The high fluid pressures in the Dunbarton Triassic basin, South Carolina, are a good example of osmotically induced potentials. A unique osmotic cell is created by the juxtaposition of fresh water in the overlying Cretaceous sediments against the saline pore water housed within the membrane-functioning sediments of the Triassic basin. Because wells penetrating the saline core of the basin show anomalously high heads relative to wells penetrating the basin margins, the longevity of this osmotic cell is probably dictated by the rate at which salt diffuses out into the overlying fresh water aquifer.

Sodium and chloride are the key factors maintaining normal osmotic pressure (OSM) and volume of the extracellular fluid, and influencing the acid–base balance of body fluids. The experiment was conducted to investigate the effects of dietary Na+ and Cl− level on growth performance, excreta moisture, blood biochemical parameters, intestinal Na+–glucose transporter 1 (SGLT1) messenger RNA (mRNA), and Na+–H+ exchanger 2 (NHE2) mRNA, and to estimate the optimal dietary sodium and chlorine level for yellow-feathered chickens from 22 to 42days. A total of 900 22-day-old Lingnan yellow-feathered male chickens were randomly allotted to five treatments, each of which included six replicates of 30 chickens per floor pen. The basal control diet was based on corn and soybean meal (without added NaCl and NaHCO3). Treatments 2 to 5 consisted of the basal diet supplemented with equal weights of Na+ and Cl−, constituting 0.1%, 0.2%, 0.3% and 0.4% of the diets. Supplemental dietary Na+ and Cl− improved the growth performance (P<0.05). Average daily gain (ADG) showed a quadratic broken-line regression to increasing dietary Na+ and Cl− (R2=0.979, P<0.001), and reached a plateau at 0.1%. Supplemental Na+ and Cl− increased (P<0.05) serum Na+ and OSM in serum and showed a quadratic broken-line regression (R2=0.997, P=0.004) at 0.11%. However, supplemental Na+ and Cl− decreased (P<0.05) serum levels of K+, glucose (GLU) and triglyceride. Higher levels of Na+and Cl− decreased duodenal NHE2 transcripts (P<0.05), but had no effect on ileal SGLT1 transcripts. The activity of Na+ /K+-ATPase in the duodenum decreased (P<0.05) with higher levels of dietary Na+ and Cl−. In conclusion, the optimal dietary Na+ and Cl− requirements for yellow-feathered chickens in the grower phase, from 22 to 42 days of age, to optimize ADG, serum Na+, OSM, K+ and GLU were 0.10%, 0.11%, 0.11%,0.17% and 0.16%, respectively, by regression analysis.

Water flow in plant tissues takes place in two different physical domains separated bysemipermeable membranes: cell insides and cell walls. The assembly of all cell insides andcell walls are termed symplast and apoplast,respectively. Water transport is pressure driven in both, where osmosis plays an essentialrole in membrane crossing. In this paper, a microscopic model of water flow and transportof an osmotically active solute in a plant tissue is considered. The model is posed on thescale of a single cell and the tissue is assumed to be composed of periodicallydistributed cells. The flow in the symplast can be regarded as a viscous Stokes flow,while Darcy’s law applies in the porous apoplast. Transmission conditions at the interface(semipermeable membrane) are obtained by balancing the mass fluxes through the interfaceand by describing the protein mediated transport as a surface reaction. Applyinghomogenization techniques, macroscopic equations for water and solute transport in a planttissue are derived. The macroscopic problem is given by a Darcy law with a force termproportional to the difference in concentrations of the osmotically active solute in thesymplast and apoplast; i.e. the flow is also driven by the local concentration differenceand its direction can be different than the one prescribed by the pressure gradient.

Culture characteristics of two species of piscine trypanosomes were compared. Trypanosoma catostomi from Catostomus commersoni was isolated in hypo-osmotic blood agar media but not in hyper-osmotic media. However, T. phaleri from Amia calva was isolated in both media. Only T. catostomi was stimulated to divide by diluting infected blood with water. The maximum number of T. phaleri and percentage of trypomastigotes were not affected by NaCl concentration. The maximum number of T. catostomi was inversely related to osmotic pressure and the percentage of trypomastigotes was directly related to osmotic pressure. This was confirmed by using equi-osmotic concentrations of NaCl, NaBr, Na acetate, KCl, choline-Cl or sucrose. Supplementation with urea had no effect on the number or morphology of trypanosomes. The survival of T. catostomi following cryopreservation was enhanced by prior incubation in hyper-osmotic media. Differential culture characteristics of these species may reflect adaptation to different host–vector systems.

To optimize IVF conditions for BALB/c mice, which are known to have poor in vitro fertilizability, the requirements for sperm–ova interaction were studied by use of modified simplex optimization medium (mKSOM) as a basic medium. Modified human tubal fluid (mHTF) was used for sperm preincubation and acted as a positive control. When the two media were compared, neither capacitation nor fertilization was supported in mKSOM. Increasing the calcium concentration in mKSOM to 5 mM or more during sperm: ova coincubation improved zona penetration but not male pronuclear (MPN) formation to the same level as those cells incubated in mHTF. When medium osmolarity was varied from 230–305 mOsmol by NaCl at 5 mM CaCl2, MPN formation improved at 280 mOsmol or higher osmolarity to the same level as that found when using mHTF. When NaCl equivalent to 25–75 mOsmol was substituted with trehalose, no significant reduction in fertilization was observed. Substitution of NaCl equivalent to 75 mOsmol with other osmotic reagents (sucrose, choline chloride and sorbitol) resulted in similar levels of fertilization as found with mHTF, except for sorbitol, which reduced fertilization significantly caused by its detrimental effect on sperm viability. At isotonic osmolarity (305 mOsmol), maximum fertilization was observed at 5 mM CaCl2; lower or higher concentrations of CaCl2 resulted in reduced fertilization. Calcium and osmolarity, therefore, are important for sperm : ova interaction in BALB/c mice and the increases in calcium to 5 mM and osmolarity to 305 mOsmol are optimal for BALB/c sperm to penetrate through the zona and to form MPN.

To elucidate requirements for in vitro sperm capacitation in inbred BALB/c mice, osmolarity, calcium and lactate were optimized using modified simplex optimization medium (mKSOM). Modified human tubal fluid (mHTF), a capacitation-supporting medium, was used as a control. In the first series of experiments, the effects of calcium and osmolarity were studied in the presence of lactate. Although preincubation with ≥5 mM CaCl2 improved fertilization after insemination significantly, it was still significantly lower than incubation with mHTF. To obtain fertilization at the equivalent levels to that of mHTF, isotonic osmolarity (305 mOsmol) was required. Trehalose, an osmotic reagent, could substitute for NaCl partially. In the second series of experiments, the effects of lactate were examined using a concentration of 5 mM calcium and isotonic osmolarity. Preincubation with ≤2.5 mM lactate increased fertilization significantly (>75%), as well as the percentages of B (capacitated) pattern sperm (≥40%) in chlortetracycline (CTC) staining, as compared with incubation in mHTF (46% and 28%, respectively; p < 0.05). In the third series of experiments, the effects of osmolarity and calcium in the absence of lactate were examined. An increase in osmolarity during sperm preincubation increased both fertilization and B-pattern sperm significantly in a dose-dependent manner. Trehalose, sucrose and choline chloride could substitute for NaCl. An increase in CaCl2 concentration during preincubation had no effect on fertilization, but this increase reduced the percentages of B-pattern sperm. In vitro capacitation of inbred BALB/c mice is sensitive to lactate and osmolarity, but that sensitivity for calcium varies depending on the presence or absence of lactate.

In the first experiment, osmotic pressure of semen and seminal plasma in a semen sample from each of the 20 mature Nili-Ravi buffalo bulls was determined. In the second experiment, effects of osmotic pressure on motility (%), plasma membrane integrity (%) and viability (%) in fresh and frozen-thawed semen samples from each of the seven mature Nili-Ravi buffalo bulls was determined. In the first experiment, seminal plasma was harvested by centrifuging semen at 400 × g for 10 min at 37°C and osmotic pressure was determined using an osmometer. In the second experiment, motility (%) was assessed in fresh and frozen-thawed (37°C for 30 s) semen samples using a phase-contrast microscope (×400). Plasma membrane integrity (%) was determined by mixing 50 μl each of fresh and frozen-thawed semen with 500 μl of solution having an osmotic pressure of 50, 100, 150, 190 or 250 mOsm/l (hypotonic treatments of fructose + sodium citrate) and incubating at 37°C for 1 h. Viability (%) of fresh and frozen-thawed spermatozoa before and after challenging them to osmotic pressure (hypotonic treatments) was assessed using supravital stain under a phase-contrast microscope (×400). In the first experiment, the mean ± s.e. osmotic pressures of the buffalo semen and seminal plasma were 268.8 ± 1.17 and 256.0 ± 1.53 mOsm/l, respectively. In the second experiment, motility (%) decreased (P < 0.05) in frozen-thawed semen samples as compared with fresh semen (60.1 ± 1.34 v. 81 ± 1.57, respectively). The plasma membrane integrity (%) and magnitude of osmotic stress in fresh and frozen-thawed semen samples was higher (P < 0.05) at 50, 100, 150 and 190 mOsm/l as compared with 250 mOsm/l. Loss of viability (%) in fresh and frozen-thawed semen samples was higher (P < 0.05) at 50 mOsm/l (59% in fresh, 70% frozen thawed) as compared with other osmotic pressures, while it was lowest at 250 mOsm/l (4.1% for fresh, 9.7% frozen thawed). In conclusion, osmotic pressure of Nili-Ravi buffalo semen and seminal plasma is determined. Furthermore, variation in osmotic pressure below 250 mOsm/l is not favorable to fresh and frozen-thawed buffalo spermatozoa.

The Escherichia coli tryptophan repressor protein (TR) represses the transcription of several genes in response to the concentration of tryptophan in the environment. In the co-crystal structure of TR bound to a DNA fragment containing its target very few direct contacts between TR and the DNA were observed. In contrast, a number of solvent mediated contacts were apparent. NMR solution structures, however, did not resolve any solvent mediated bonds at the complex interface. To probe for the role of water in TR operator recognition, the effect of osmolytes on the interactions between TR and a target oligonucleotide bearing the operator site was examined. In the absence of specific solvent mediated hydrogen bonding interactions between the protein and the DNA, increasing osmolyte concentration is expected to strongly stabilize the TR operator interaction due to the large amount of macromolecular surface area buried upon complexation. The results of our studies indicate that xylose did not alter the binding affinity significantly, while glycerol and PEG had a small stabilizing effect. A study of binding as a function of betaine concentration revealed that this osmolyte at low concentration results in a stabilization of the 1:1 TR/operator complex, but at higher concentrations leads to a switching between binding modes to favor tandem binding. Analysis of the effects of betaine on the 1:1 complex suggest that this osmolyte has about 78% of the expected effect. If one accepts the analysis in terms of the number of water molecules excluded upon complexation, these results suggest that about 75 water molecules remain at the interface of the 1:1 dimer/DNA complex. This value is consistent with the number of water molecules found at the interface in the crystallographically determined structure and supports the notion that interfacial waters play an important thermodynamic role in the specific complexation of one TR dimer with its target DNA. However, the complexity of the effects of betaine and the small or negligible effects of the other osmolytes could also arise from osmolyte induced competition between antagonistic coupled reactions.

Juice was extracted from a ryegrass silage after fermentation times of 0, 51 or 291 h. Three solutions of D/L lactic acid were prepared to have the same neutralizing values (NV) as the silage juices. Experiment A measured the mtakes by sheep of dried silage residues remaining after juice extraction. Experiment B measured the effect of adding each of the juices and lactic acid solutions (1·5 l/kg) on intake of unmolassed sugar-beet nuts. Intakes were measured daily and also over 3 h following an overnight fast.

Fermentation time had no effect on the composition or intake of the residues (P < 0·05). Increasing fermentation time from 0 to 51 to 291 h reduced silage juice pH (5·07, 4·96 and 4·10), increased lactic acid (gfl 6·2, 7·1, 18·7), NV (meq/l 29, 40, 157) and osmolality (mosmol/l 1000, 1010, 1390). Corresponding values for the lactic acid solutions were 2·82, 2·74, 2·41 (pH), 3·1, 4·3,18·3 (lactic), 26, 38,154 (NV) and 32, 42,176 (osmolality).

Total daily DM intake (g/kg live weight1·473) was unaffected by NV but was significantly higher overall (P < 0·05) for the silage juices (7·49) than for the lactic acid solutions (6·64) although this difference was mostly due to the extra dry matter (DM) contained in the added silage juice. Short-term DM intake (g per 3 h per kg live weight1·473) decreased with increasing NV, being significantly lower (P < 0·05) for the highest NV than for the others (3·58, 3·34 and 3·11) but lactic acid and silage juice gave similar responses (P > 0·05). Effects on short-term intake were influenced mainly by NV and suggested effects on immediate feedback mechanisms or palatability.

The effects of potassium deficiency (KD) and all-macronutrient deficiency (MD) on elongation of tap and lateral roots were studied on maritime pine seedlings (Pinus pinaster Ait.) in hydroponic culture.

Tap root elongation was unaffected by either of the two deficiencies. By marked contrast, lateral root elongation was strongly reduced. The analyses of cell turgor pressure and relative elemental growth rate (REGR) profile in the growing zone allowed us to determine the effects of the nutrient stresses on cell-wall properties. For both deficiency treatments, elongation rate, REGR profile (measured only for control and KD) and turgor pressure in the fastest growing cells were unaffected in the tap root, suggesting that KD and MD did not modify cell-wall properties in the growing zone. In lateral roots, KD shortened the growing zone and significantly reduced REGR. However, turgor pressure remained unaffected in this region. The absence of turgor pressure change suggests that KD reduced elongation of lateral roots by tightening cell walls. In mature cells of the two types of roots, turgor and osmotic pressures tended to be reduced by the nutrient deficiencies, indicating that these parameters were better maintained in the growing cells.

Cell turgor and osmotic pressures of control plants were 0·1 MPa lower at 30 mm (mature cells) than at 2–4 mm (expanding cells) from the meristem. Moreover, these parameters were 0·1 MPa lower in expanding cells of lateral roots than in those of tap the root. Turgor and osmotic pressures were not homogeneous throughout the root system and were affected differently by the nutrient deficiencies depending on the location in the root system.

A well fermented grass silage was mixed with 9·3 g lactic acid per kg silage supplied either as the free acid (lactic acid) or partially neutralized to pH 5·2 with sodium hydroxide (sodium lactate) to give three dietary treatments. Treatment with lactic acid reduced silage pH (P < 0·05) from 4·03 to 3·80 and increased (P < 0·01) the amount of sodium hydroxide required to raise silage pH to 6·5 (neutralizing value) from 167 to 261 mmol/kg silage and osmolality (P < 0·01) from 712 to 854 mosmoljkg silage; sodium lactate treatment increased (P < 0·05) silage pH to 4·15 and osmolality (P < 0·01) to 964 mosmoljkg silage but did not change neutralizing value.

Wlien fed in a randomized-block design to either six intact lambs or to four rumen fistulated sheep lactic acid treatment reduced (P < 0·05) silage dry matter (DM) intake (g/kg weight0·75) from 34·7 to 27·8 for intact sheep and from 35·5 to 29·9 for fistulated sheep. Intakes of the sodium lactate-treated silage tended to be higher (39·2, intact and 40·1, fistulated sheep, g/kg weight0·75) than the control silage. Silage DM intake was correlated more closely with neutralizing value (r2 = 0·34) than pH (rz = 0·24).

Treatment of silage with lactic acid or sodium lactate did not change rumen pH, volatile fatty acid patterns or osmolality. Blood pH, pCO2 and bicarbonate concentrations were not changed by the diets offered and were within normal ranges. Urine acid-base balance was not affected by lactic acid treatment whereas urine pH, and bicarbonate and sodium excretion were (P < 0·01) increased by sodium lactate treatment of the silage.

Four sheep sustained by intragastric nutrition were used to study saliva secretion and the relationship between osmotic pressure in the rumen and net water transport across the rumen wall. Different concentrations of buffer were infused into the rumen to change the rumen osmotic pressure. Salivary secretion was estimated from entrance of P into the rumen. Net water transport across the rumen wall was calculated as the difference between water inflow and water outflow from the rumen. A negative linear relationship between the rumen osmotic pressure (X, mOsm/kg) and the water absorption across the rumen wall (Y, ml/h) was found: Y = (394 SE 8·3)–(l·22 SE 0·03) X, r20·83, (P < 0·001), and a positive linear relationship was found between the rumen osmotic pressure (X, mOsm /kg) and the outflow rate of rumen fluid (Y, ml/h): Y = (34·0 SE 8·0) + (0·97 SE 0·03), X, r2 0·56, (P < 0·001). The implication is that rumen osmotic pressure can be a key factor in the control of the net water transport across the rumen wall, the outflow of rumen fluid to omasum and the rumen liquid dilution rate. A method is suggested by which salivary secretion in sheep may be calculated from the water balance in the rumen.

The effects of changing rumen osmotic pressure (OP) upon water kinetics and volatile fatty acid (VFA) absorption in the rumen of sheep were studied in two 4 × 4 Latin square experiments, each using four lambs with a rumen cannula and an abomasal catheter. In both experiments the lambs were sustained by the intragastric infusion of all nutrients (VFA, Ca, P, Mg and a buffer solution into the rumen, and casein, vitamins and trace elements into the abomasum). On experimental days, which were at least 1 week apart, drinking water and the casein infusion were withdrawn, and the ruminal OP was changed and held constant for 9·5 h, by incorporating NaCl at different concentrations in the buffer solution being infused. In Expt 1 the target OP values were 300, 340, 380 and 420 mosmol/kg, and in Expt 2 were 261 (no saline addition), 350, 420 and 490 mosmol/kg. Using soluble non-absorbable markers (PEG in continuous infusion and Cr-EDTA injected in pulse doses) rumen volume, liquid outflow rates, apparent water absorption through the rumen wall and VFA absorption rates were estimated at six sampling times corresponding to the 1·5 h intervals during the last 7·5 h following the change in rumen OP. Liquid outflow rate (F; ml/h) showed a significant and positive linear relationship with the rumen OP (mosmol/kg), resulting in the equation F = 1·24 OP (SE 0·096)–36·5 (SE 36·6) (r2 0·96). Similarly, water absorption rate (W; ml/h) was significantly affected by rumen OP, and this relationship was given by W = 395 (SE 39·9) −1·16 OP (SE 0·105) (r2 0·95), which means that for an OP of 341 mosmol/kg the net movement of water across the rumen wall would be zero, and either a net efflux or a net influx of water would be observed with lower or higher OP respectively. In Expt 2 there was a significant linear effect of OP on rumen volume (P <0·01), with higher OP being associated with increases in rumen liquid contents of about 10–20%. As rumen OP was increased there was also a decline in the absorption rate of VFA (from 232 mmol VFA/h for OP 350 to 191 mmol/h for OP 490 mosmol/kg), resulting in the accumulation of VFA (especially acetate) in the rumen and a consequent fall in rumen pH. Rumen OP seems to be important in defining water movement across the rumen wall and, hence, partitioning between absorption and outflow.
