The most common way to test seed quality is to use a simple and reliable but time- and space-consuming germination test. In this paper we present a fast and simple method to analyse cabbage seed deterioration by measuring ethanol production from partially imbibed seeds. The method uses a modified breath analyser and is simple compared to gas chromatographic or enzymatic procedures. A modified method using elevated temperatures (40°C instead of 20°C) shortened the assay time and improved its sensitivity. The analysis showed an inverse correlation between ethanol production and seed quality (e.g. the final percentages or speed of germination and the number of normal seedlings). The increase in ethanol production was observed when cabbage seeds were deteriorated by storage under ambient conditions or hot water treatments, both of which reduced the number of normal seedlings. Premature seeds produced more ethanol upon imbibition than mature seeds. Ethanol production occurred simultaneously with oxygen consumption, indicating that lack of oxygen is not the major trigger for ethanol production.

A recent hypothesis states that the accumulation of amino-carbonyl reaction products of reducing sugars with proteins (Maillard products) is related to the loss of seed vigour and viability during ageing. Since Maillard products are biochemical end products, their accumulation should provide an index of seed deterioration. A simple fluorescence assay has been used to estimate the quantity of Maillard products in aqueous seed extracts. We tested whether an accumulation of fluorescent compounds correlates with the loss of seed viability during ageing. Fluorescence of extracts from lettuce (Lactuca sativa) embryonic axes increased after controlled deterioration, but was not correlated with ageing period. Fluorescence of extracts from whole broccoli (Brassica oleracea var. italica) seeds was either unchanged or declined during controlled deterioration. Extracts from naturally aged whole seeds of carrot (Daucus carota), tomato (Lycopersicon esculentum), and cauliflower (B. oleracea var. botrytis) had higher fluorescence than controls, but there was no consistent increase in fluorescence in proportion to the loss of viability. Extracts of naturally aged onion (Allium cepa) seeds exhibited less fluorescence than extracts from seeds with higher viability. We conclude that the fluorescence assay does not have general utility as an index of seed deterioration.

In view of the finding that dry seeds may exist in the glassy state, a brief review of the commonest methods of detection and quantification of the glass transition is presented. While the glassy state may contribute to seed tolerance of desiccation, it does not appear to account for desiccation tolerance. Its major function in dry seeds may be its contribution to the stability of the seed components during storage.

Some of the seed-evolved volatiles, which were mainly composed of methanol, acetaldehyde, ethanol and acetone, caused the loss of seed germinability during storage. In general, the deleterious effects of the volatiles increased with increasing RH and temperature during storage. Acetaldehyde had the strongest deleterious effect regardless of RH and temperature, while ethanol caused seed deterioration only at high RHs. Acetone was slightly deleterious to some species, while methanol was almost inert in most seeds. Various aldehydes applied during storage showed some toxicity to seed germinability, which decreased, except for 3-methylbutanal, with increasing molecular size, suggesting that the endogenous volatile compounds with an aldehyde group cause seed deterioration. On the other hand, the contents of volatile compounds in seeds were higher when they were stored at 44% RH (water sorption zone 2), than when stored at 12% RH (water sorption zone 1). Acetaldehyde, the most deleterious volatile, was more abundantly accumulated within the seeds stored at −3.5°C than at 23°C. Based on these facts, it is suggested that endogenous volatiles, especially acetaldehyde, may be an important factor that accelerates seed deterioration which often occurs under lower RHs and/or temperatures throughout long-term storage.

DNA polymerase 2 holoenzyme is an α-type polymerase present as a major activity in 24-h germinated maize embryo axes. At this time, several of the polypeptides that compose this multisubunit enzyme are phosphorylated. During the germination of artificially deteriorated maize embryo axes, total DNA polymerase activity decreases by around 50% when compared with activity in non-deteriorated axes. DNA polymerase 2 appears to be damaged by the deterioration treatment, greatly reducing its activity. This decrease in activity is related to protein degradation during germination since the amount of both the holoenzyme and the different polymerase 2 subunits is reduced as imbibition progresses. The decrease in amount is more dramatic for some of the subunits than for others, i.e., the amount of the 90-kDa catalytic subunit is reduced by about half, whereas all other subunits totally disappear at different times during germination. The phosphorylation state of the different polypeptides is also greatly affected.

Chromosome integrity and DNA polymerase activity are negatively affected by improper storage of maize seeds. We have extended our studies on the effect of improper storage on DNA metabolism by analysing the effect of hot–dry and hot–humid conditions on DNA ligase activity. An assay to measure ligase activity was established by using linearized plasmid DNA as substrate and protein extracts from maize axes. Optimal conditions were established for the in vitro assay. Enzyme activity was importantly reduced under both storage conditions. Activity recovered 6–9 h after imbibition. A model for loss of viability and vigour due to damage of DNA metabolism is proposed.

Storage of tomato seeds under deteriorating conditions reduced the rate of germination, the uniformity of germination, total germination and the proportion of normal seedlings. Pre-storage humidification and hydropriming of seeds resulted in a significant increase in resistance to deterioration. In contrast, osmoprimed seeds were more deterioration sensitive. Humidification or hydropriming for one day did not allow nuclei to enter the S phase of the cell cycle. Osmopriming strongly increased the percentage of nuclei with replicated DNA in the embryonic root tip, indicating initiation of the cell cycle and progression towards the G2 phase. The interaction between the prestorage treatments, cell cycle progression and deterioration resistance, is discussed. Based on the changes in moisture content equilibrium and cell cycle activity it is hypothesized that the beneficial effects of pre-storage humidification and hydropriming were related to metabolic activities induced by the partial hydration and that the adverse effects of osmopriming were caused by a decrease in DNA repair activity due to progression in the cell cycle.
