It has been thirty years since the first genetically engineered animal with altered milk composition was reported. During the intervening years, the world population has increased from 5bn to 7bn people. An increasing demand for protein in the human diet has followed this population expansion, putting huge stress on the food supply chain. Many solutions to the grand challenge of food security for all have been proposed and are currently under investigation and study. Amongst these, genetics still has an important role to play, aiming to continually enable the selection of livestock with enhanced traits. Part of the geneticist's tool box is the technology of genetic engineering. In this Invited Review, we indicate that this technology has come a long way, we focus on the genetic engineering of dairy animals and we argue that the new strategies for precision breeding demand proper evaluation as to how they could contribute to the essential increases in agricultural productivity our society must achieve.

The present study was conducted in order to investigate if selected minor milk components would be indicative for the nutritional situation of the cow. Forty-eight dairy cows were offered a high digestible ration vs. a lower digestible ration combined with 2 protein levels in a 4 × 4 Latin square design. Milk glucose, glucose-6-phosphate, cholesterol, triacylglycerides (TAG), uric acid and β-hydroxybutyrate (BHBA) were measured and correlated mutually and towards other milking parameters (yield, h since last milking, days in milk (DIM), urea, etc). The variation range of the suggested variables were broad, a fact that may support their utilisation as predictive parameters. The content of milk metabolites was significantly affected by the change in rations as milk glucose, glucose-6-phosphate, uric acid, and the ratio cholesterol: triacylglycerides increased with higher energy intake while BHBA and TAG decreased. The content of some of the milk metabolites changed during 24 h day/night periods: BHBA, cholesterol, uric acid and TAG increased whereas free glucose decreased in the night period. Certain associations between milk metabolites and calculated energy parameters like ECM, body condition score (BCS), and body weight gain were found, however, these associations were to some extent explained by an interaction with DIM, just as changes in milk metabolites during a 24 h period seems to interfere. It is concluded that the practical use of the suggested milk variables should be based on more than one metabolite and that stage of lactation and possibly time of the day where the milk is collected should be incorporated in predictive models.

Immune system and inflammatory responses are affected by α-linolenic acid (αLA: 18:3 ω-3). The objective of this study was to determine the effects of αLA-enriched rations on gene expression of systemic (blood) and local (mammary gland) inflammatory markers in Holstein dairy cattle. Further, the effect of dietary treatments was evaluated on the concentration of αLA in serum phospholipids. Camelina (Camelina sativa) meal (containing 24·2% αLA) was fed at 0, 3, 6, and 9% (dry matter basis) replacing canola meal (rich in 18:1 ω-9) to provide rations with incremental concentrations of αLA. Lactating primiparous Holstein cows (n = 18) were randomly assigned to a treatment sequence in a 4 × 4 Latin square design. Each period lasted 16 d and milk and blood samples were collected during the final 2 d of each period. Peripheral blood mononuclear cells (PBMC) and milk cells (MC) were harvested, and RNA extracted and converted to complementary DNA for quantitative real time PCR analysis. The effect of dietary treatments (αLA) on the relative abundance of pro- and anti-inflammatory genes in the PBMC and MC was tested by the MIXED procedure of SAS. Expression of pro-inflammatory tumour necrosis factor (TNF)-α in MC was linearly reduced (up to 40%) as dietary αLA increased. Expression of pro-inflammatory markers interleukin (IL)-1β, IL-8, and TNF-α was reduced (29, 20, and 27%, respectively) in PBMC isolated from cows fed 6% camelina meal ration as compared with cows fed 0% (control). Expression of IL-6 was, however, increased with inclusion of camelina meal. Greater dietary αLA linearly increased serum phospholipids αLA contents, and when fed up to 6% DM down-regulated expression of some of the local (milk) and systemic (blood) pro-inflammatory markers in vivo.

This study investigated the effects on goat milk yield and composition of a diet supplemented with Echium plantagineum oil (EPO). Twenty-four mid-lactation multiparous Camosciata goats were divided into two balanced groups and fed for 44 d a diet based on hay and concentrate, supplemented (EPO group, Echium) or not (CON group, control) with 40 ml of ruminally unprotected EPO. Individual milk yield was recorded and individual milk samples were collected at 11, 22, 33, and 44 d after supplementation. Milk samples were analysed for milk components and fatty acids (FA). Data were statistically analysed by repeated-measures analysis of variance. Milk yield, protein and lactose contents were significantly higher in EPO than CON group. The inclusion of EPO significantly decreased total saturated FA and total branched-chain FA, and contemporarily sharply increased trans biohydrogenation intermediates (P ⩽ 0·001). Milk concentration of α-linolenic, stearidonic and γ-linolenic acids increased by 23, 1000 and 67%, respectively (P ⩽ 0·001). Due to extensive ruminal biohydrogenation, their apparent transfer rate was less than 3%. As a consequence, the milk concentrations of very long-chain (VLC) polyunsaturated fatty acids (PUFA), such as eicosapentaenoic (20:5 n-3) and dihomo-γ-linolenic (20:3 n-6) acids, significantly increased with EPO treatment, but values remained very low. Docosahexaenoic acid (22:6 n-3) was undetectable in all analysed milk samples. Results show that ruminally unprotected EPO can enhance milk yield and protein and improve the overall goat milk FA profile. However, this kind of supplementation cannot be considered a valuable strategy to develop goat functional dairy products enriched with VLC n-3 PUFA for human consumption.

Based on a previous ensiling study in glass silos of various pomegranate pulp (PP) mixtures, fresh pomegranate pulp (PP) was mixed with drier feeds including soy hulls and corn silage (40:35:25 on DM basis) and ensiled in 32 pressed bales (700 kg each) wrapped with stretch polyethylene film. This ensiled pomegranate pulp mixture (PPM) was included in lactating cow total mixed ration (TMR) at a level of 20% of DM (PPM-TMR). Performance and digestion experiment was conducted with two groups of 21 milking cows each, fed individually one of the two TMR: 1. Control TMR without ensiled PPM; 2. Experimental TMR which contained 20% ensiled PPM, including 8% PP as corn grain replacer. Voluntary DM intake of cows fed the control TMR was 5·04% higher than that of the PPM cows. In vivo digestibility of DM, OM, NDF, CP and fat were significantly higher in the control cows compared with the PPM group, but methane production in the rumen fluid was 25% lower in the PPM cows. A slightly higher milk yield (by 2·2%) observed in the control cows; however, milk fat content was 5·9% higher in the PPM cows. This was reflected in similar yield of energy corrected milk (ECM) and 3·97% increase in production efficiency (ECM/DM intake) of the PPM cows compared with the control ones. Welfare of the cows, as assessed by length of daily recumbence time, was in the normal range for both groups. Body weight gain was also similar in both groups. Data suggest that the level of 8% PP in the PPM-TMR used in this study was probably too high for lactating cows and should be lowered to 4% in order to achieve better performance.

Milk and Minas frescal cheese were evaluated from crossbred Holstein × Gir cows that were fed diets enriched with 0, 33, 66 and 100% inclusion levels of palm kernel cake in a concentrated supplement in replace of soybean meal. Eight crossbred lactating cows were distributed (four animals × four treatments × four periods) in the experimental design of double 4 × 4 Latin squares. The capric (C : 10, P = 0·0270), undecylic (C : 11, P = 0·0134), and lauric (C : 12, P = 0·0342) saturated fatty acid concentrations and CLA (C18 : 2c9t11, P = 0·0164) of the milk fat decreased linearly with an increasing percentage of peanut cake in the diet. The increased peanut cake content (100%) in the diet was associated with a linear decrease in C : 10 (P = 0·0447), C : 12 (P = 0·0002), mirystic (C : 14, P < 0·0001) and palmitic (C : 16, P < 0·0001) saturated fatty acid concentrations and an increase in arachidic, lignoceric, palmitoleic and elaidic acid levels in the Minas frescal cheese fat made from the milk. Both the milk and the Minas frescal cheese showed a linear decrease in the concentration of monosaturated fatty acids (P < 0·0001), atherogenicity index, and thrombogenicity index (P < 0·05), while the hypocholesterolaemic: hypercholesterolaemic and omega 6: omega 3 (P > 0·05) ratios were not influenced by the different peanut cake levels. The inclusion of up to 100% peanut cake as a substitution for soybean meal in the concentrate of grazing lactating cows resulted in changes in the nutritional quality of their milk products, as indicated by the increase in polyunsaturated fatty acids and the decrease of saturated fatty acids (lauric, myristic, and palmitic).

Elevated levels of blood interleukin-1β (IL-1β), interleukin-6 (IL-6) and tumour necrosis factor–α (TNF–α) increase insulin resistance and result in inflammation. It is not clear whether elevated blood level of acetoacetate (ACAC) and decreased blood level of glucose, which are the predominant characteristics of clinical biochemistry in ketotic dairy cows, increase proinflammatory cytokines and subsequent inflammation. The objective of this study was to test the hypothesis that ACAC and glucose activate the NF-κB signalling pathway to regulate cytokines expression in bovine hepatocytes. Bovine hepatocytes were cultured with ACAC (0–4·8 mm) and glucose (0–5·55 mm) with or without NF-κB inhibitor PDTC for 24 h. The secretion and mRNA levels of cytokines were determined by enzyme-linked immunosorbent assay (ELISA) and real-time fluorescence quantitative polymerase chain reaction (qRT-PCR). The NF-κB signalling pathway activation was evaluated by western blotting. Results showed that the secretion and expression of IL-1β, IL-6 and TNF-α increased in an ACAC dose-dependent manner. Additionally, there was an increase in the secretion and mRNA expression of these three cytokines in glucose treatment group, which increased significantly when the glucose concentrations exceed 3·33 mm. Furthermore, both ACAC and glucose upregulated NF-κB p65 protein expression and IκBα phosphorylation levels. However, these effects were reduced by PDTC. These results demonstrate that elevated levels of ACAC and glucose increase the synthesis and expression of proinflammatory factors by activating NF-κB signalling pathway in hepatocytes, which may contribute to inflammation injury in ketotic dairy cows.

On-farm instruments for assessing colostrum quality are needed in order to ensure that the calf is supplied with enough IgG to avoid failure of passive transfer. The aim of this study was to evaluate methods for estimating the IgG concentration in cows' colostrum. This research included 126 colostrum samples from 21 Danish farms with different breeds, ensuring a broad variation pattern in IgG, total protein and fat concentration. Approximately one third of the samples did not fulfil the recommendation of >50 g IgG/l colostrum, and the IgG concentration decreased with time from calving to milking. The ratio of IgG to total protein varied from 6 to 61%, however IgG and total protein were correlated with r2 = 0·70. The variation in fat was independent of variations in protein and IgG. The IgG concentration was measured by ELISA and compared to fast measurements by specific gravity by colostrometer, Brix by refractometer and prediction from infrared spectroscopy. The three fast methods were all correlated to the total protein concentration of colostrum; however specific gravity was also influenced by the fat concentration. Furthermore, specific gravity generally overestimated the IgG concentration, and the cut-off level should be raised to 1050 in order to ensure adequate IgG in colostrum. None of the methods estimated IgG concentration better than the correlation of total protein and IgG, meaning that they all depended on the indirect correlation between total protein and IgG. The results suggest that using a refractometer for quality control of colostrum is an easy and feasible method, and a cut-off level of Brix 22 seems sufficient to assure adequate IgG concentration in colostrum fed to the calf.

Vitamin D has become one of the most discussed nutrients in human nutrition, which has led to an increased interest in milk as a vitamin D source. Problems related to fortifying milk with synthetic vitamin D can be avoided by securing a high content of natural vitamin D in the milk by supplying dairy cows with sufficient vitamin D. However, choosing the most efficient route and form of supplementation requires insight into how different vitamin D metabolites are transported in the body of cattle. There are two forms of vitamin D: vitamin D2 (D2) and vitamin D3 (D3). Vitamin D2 originates from fungi on roughage. Vitamin D3 originates either from endogenous synthesis in the skin or from feed supplements. Vitamin D2 is chemically different from, and less physiologically active than, D3. Endogenous and dietary D3 is chemically similar but dietary D3 is toxic, whereas endogenous D3 appears well regulated in the body.

The objective of this study was to determine whether the occurrence of a previous case of pathogen-specific clinical mastitis (CM) protects Holstein dairy cows against a recurrent case. Pathogens studied were Escherichia coli, Staphylococcus aureus, Staphylococcus spp., Streptococcus spp., Klebsiella spp., and Trueperella pyogenes. A total of 40 864 lactations (17 265 primiparous and 23 599 multiparous) from 19 835 cows from 5 large, high milk producing New York State dairy herds were analysed. We estimated the effects of parity, calving diseases, milk yield, current season and number of CM cases in the previous lactation on the risk of a first CM case using generalised linear mixed models with a log link and Poisson error distribution. The aforementioned risk factors and the occurrence of previous cases of pathogen-specific CM within the current lactation were evaluated as risks for second and third cases of pathogen-specific CM. Cows with more CM cases in the previous lactation were at greater risk of pathogen-specific CM in the current lactation. Multiparous cows were at greater risk of a second CM case if they had suffered from a first CM case that was caused by the same pathogen as the second case. In contrast, a second CM case generally put cows at greater risk of a third case, irrespective of whether the third case was caused by the same or a different pathogen. Our results showed that a previous case of pathogen specific CM does not generally protect against a recurrent case.

The aim of this study was to identify the differences between the main macro and micronutrients including proteins, fat, minerals and vitamins in cow and goat dehydrated fermented milks. Fermented goat milk had higher protein and lower ash content. All amino acids (except for Ala), were higher in fermented goat milk than in fermented cow milk. Except for the values of C11:0, C13:0, C16:0, C18:0, C20:5, C22:5 and the total quantity of saturated and monounsaturated fatty acids, all the other fatty acid studied were significantly different in both fermented milks. Ca, Mg, Zn, Fe, Cu and Se were higher in fermented goat milk. Fermented goat milk had lower amounts of folic acid, vitamin E and C, and higher values of vitamin A, D3, B6 and B12. The current study demonstrates the better nutritional characteristics of fermented goat milk, suggesting a potential role of this dairy product as a high nutritional value food.

In the current study the microbiological, sensory and chemical properties of 24 kefirs (12 producers) from Estonian, Latvian and Lithuanian retail market were determined using gas chromatography (GC), high performance liquid chromatography (HPLC-MS/MS-Q-TOF and LC-ion trap MS/MS), spectrophotometry and other methods. Antihypertensive, angiotensin-converting enzyme (ACE) inhibiting, antioxidant and antibacterial peptides were found in the kefir samples. According to the results of principal component analysis of 200 most abundant compounds obtained with HPLC-MS/MS-Q-TOF analysis, Estonian kefirs differed from the rest. Kefirs of Latvian and Lithuanian origin showed similarities in several characteristics, probably related to the starter cultures and technological processes. The fatty acids composition of all Baltic kefirs was uniform. The antioxidant capacity of the kefirs varied slightly, whereas intermediate positive correlation (r = 0·32, P < 0·05) was found between antioxidativity and total bacterial count. The lipid oxidation level, estimated as the content of linoleic and oleic acid primary oxidation products, oxylipins, was very low in all studied kefirs. Only one third of analysed kefirs met the requirements of the minimum sum of viable microorganisms, indicated in the Codex Standard for Fermented Milks.

Sweet cheese whey has been used to obtain whey cheese without the addition of milk. Pre-treated whey was concentrated by nanofiltration (NF) at different concentration ratios (2, 2·5 and 2·8) or by reverse osmosis (RO) (2–3 times). After the concentration, whey was acidified with lactic acid until a final pH of 4·6–4·8, and heated to temperatures between 85 and 90 °C. The coagulated fraction (supernatant) was collected and freely drained over 4 h. The cheese-whey yield and protein, fat, lactose and ash recoveries in the final product were calculated. The membrane pre-concentration step caused an increase in the whey-cheese yield. The final composition of products was compared with traditional cheese-whey manufacture products (without membrane concentration). Final cheese yields found were to be between 5 and 19·6%, which are higher than those achieved using the traditional ‘Requesón’ process.

In the present work, the use of scotta as substrate for bacterial fermentation was studied with the objective of obtaining a drink from transformation of this by-product. Scotta retains most of the lactose of the milk and it is normally colonized by a natural microbiota. A treatment was devised to reduce the autochthonous microbial populations in order to reduce competition towards the inoculated bacterial strains. Nine lactic acid bacteria (LAB) were assessed for their capability to develop in scotta. They evidenced different behaviors regarding growth rate, acidification capability and nitrogen consumption. A co-inoculum of three LAB, namely a Streptococcus thermophilus, a Lactobacillus delbrueckii subsp. bulgaricus and a Lb. acidophilus strains, chosen among those giving the best performances in single-strain fermentation trials, gave abundant (close to 109 cfu/ml) and balanced growth and lowered pH to 4·2, a value similar to that of yogurt. These results show that scotta may have potential as a substrate for bacterial growth for the production of a fermented drink. Further studies are needed to optimize the organoleptic aspects of the final product.

Protein nanofibrils with 10–20 nm diameters were formed by heating whey protein solution at pH 2·0. Nanofibrils solution was deacidified slowly through dialysis followed by adding different amounts of CaCl2 (0–80 mM) into the dialysis water resulting in formation of a soft viscoelastic gel over time. The gel fabricated from the nanofibrils solution dialyzed against distilled water with 0 mM CaCl2 had zero ash content. Fourier transform infra-red spectroscopy revealed a change in the pattern of hydrogen bond formation in gel network by calcium chloride. The higher the ash content of gels, the lower was the storage modulus and fracture stress of samples. Gels with higher ash contents had a more porous microstructure which was attributed to the diminished hydrophobic interactions and hydrogen bonding among nanofibrils by the action of chloride. Higher ash contents also led to higher water holding capacity of gels which was attributed to the influence of the strongly hydrated calcium ions that interacted with the non-charged regions of proteins via site-specific interactions.

Lactic acid bacteria (LAB) can be isolated from different sources such as milk and cheese, and the lipolytic, proteolytic and glycolytic enzymes of LAB are important in cheese preservation and in flavour production. Moreover, LAB produce several antimicrobial compounds which make these bacteria interesting for food biopreservation. These characteristics stimulate the search of new strains with technological potential. From 156 milk and cheese samples from cow, buffalo and goat, 815 isolates were obtained on selective agars for LAB. Pure cultures were evaluated for antimicrobial activities by agar antagonism tests and for proteolytic activity on milk proteins by cultivation on agar plates. The most proteolytic isolates were also tested by cultivation in skim milk followed by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) analysis of the fermented milk. Among the 815 tested isolates, three of them identified as Streptococcus uberis (strains FT86, FT126 and FT190) were bacteriocin producers, whereas four other ones identified as Weissella confusa FT424, W. hellenica FT476, Leuconostoc citreum FT671 and Lactobacillus plantarum FT723 showed high antifungal activity in preliminary assays. Complementary analyses showed that the most antifungal strain was L. plantarum FT723 that inhibited Penicillium expansum in modified MRS agar (De Man, Rogosa, Sharpe, without acetate) and fermented milk model, however no inhibition was observed against Yarrowia lipolytica. The proteolytic capacities of three highly proteolytic isolates identified as Enterococcus faecalis (strains FT132 and FT522) and Lactobacillus paracasei FT700 were confirmed by SDS–PAGE, as visualized by the digestion of caseins and whey proteins (β-lactoglobulin and α-lactalbumin). These results suggest potential applications of these isolates or their activities (proteolytic activity or production of antimicrobials) in dairy foods production.

The proteolytic stage of the digestion process of white cheese curd was optimised to maximise the angiotensin I-converting enzyme (ACE)-inhibitory activity of the final enzyme-modified cheese (EMC) paste. It was found that bioactive peptides generation in EMC paste was of multi-variable dependent nature and could be optimised by targeted selection of specific component variables. Maximum ACE-inhibitory was obtained by proteolysis at 48 °C for 25 h with 1 g Flavourzyme/kg cheese curd. This bioactive EMC paste was subsequently spray-dried. The drying conditions were optimised to obtain a highly soluble powder to warrant quick and complete hydration, with the lowest water activity to maximise long term storage. The higher the inlet drying air temperature, the greater was the solubility of resultant EMC powder. Differential scanning calorimetry analysis revealed that the highest drying air temperature (200 °C) resulted in a lower glass transition temperature for the potentially bioactive EMC powder.
