This study was designed to measure responses of four-year-old black-lip pearl oysters(Pinctada margaritifera) to different temperatures and foodconcentrations and to identify the energy requirements of each sex. Oysters were fed amonospecific microalgal diet of Isochrysis affinis galbana(T-iso). Measurements of oxygen consumption and ingestion rates were carried outat 24 °C and 27 °C and at two algal (T-iso) concentrations: 5000 and 30 000 cellml-1. Glycogen content in adductor muscle, absorption efficiency and scopefor growth were also estimated. Females and males responded differently to environmentalfactors, with food level being the most influential parameter. Oxygen consumption andabsorption efficiency were significantly higher in females than in males, but males hadsignificantly higher glycogen content than females. At high food concentration, glycogencontent, ingestion rate, oxygen consumption, and scope for growth were significantlyhigher than at the low food concentration. Only absorption efficiency was significantlyhigher at the low food concentration. Oxygen consumption was significantly higher at 27 °Cthan at 24 °C. These results indicate that females and males have different bioenergeticfunctioning and that energy demands for reproduction are higher in females.

Pinctada margaritifera mollusc is cultivated in French Polynesia for the production of black pearls. For this study, the colour of juvenile samples (48 days old) was investigated in the visible range spectra (430–700 nm) using spectrophotometry. A first measurement was done with the soft parts still inside the shell (entire animal). Then, the soft parts were removed in order to do a second measurement on the growing edge of the shell. Comparison of the two measurements shows that the estimation of the living animal colour with unaided eye is strongly influenced by the colour of the soft parts. The use of the International Commission on Illumination (ISI) chromaticity diagram shows that at this growth stage, the shells are “white”; i.e. present no absorptions to the visible part. Multivariate statistical analyses based on the intensities of 6 wavelengths show that the shell colour is less variable than the colours of the entire animals. Wavelength intensities of the shells are similar, so no colour trend is visible. On the other hand, the colours of the entire animals are darker, and more variable. At this growth stage, the shell colour is not predictable for a potential selection of receiver or donor.

Cryopreservation is a valuable tool for genetic improvement programs. Several bivalvemollusc species have already been the subject of such programs and the Tahitian blackpearl oyster industry is now planning the development of selective breeding for desirabletraits in Pinctada margaritifera. The ability to cryopreserve spermatozoawould, therefore, offer significant benefits to the cultured black pearl industry.Spermatozoa were cryopreserved with cryoprotectant agent (CPA) 0.7 M trehalose in 0.8 MMe2SO and a two-step freezing process was used: straws were first maintainedin nitrogen vapour for 10 minutes, then directly plunged into liquid nitrogen and storedfor one week before use. The viability of thawed sperm was 23% lower than that of freshsperm. When using thawed sperm, therefore, a higher sperm/egg ratio of 100 000:1 wasrequired to reach 80% oocyte fertilization, compared with 100:1 for fresh sperm.Nevertheless, this first demonstration of cryopreserved sperm fertility in black pearloyster confirms the hatchery applicability of the cryopreservation technique defined here.Monitoring for larval viability during the first 23 days of life revealed no significantdifferences between the progeny produced with cryopreserved sperm and that produced usingfresh sperm.

In French Polynesia, the black-lip pearl oyster Pinctada margaritifera has been farmed to produce pearls since the 1980s, forming the basis of a major industry. The sustainability of this activity relies on spat collection in the lagoons. However, pearl oyster spat can be difficult to identify for the evaluation of stock variations. It is especially hard to distinguish Pinctada spp. larvae at a very early stage of development. In the present study, a whole-mount in situ hybridisation (ISH) technique was developed to allow the discrimination of larvae of closely-related pearl oyster species found in the French Polynesian atolls. Using specific ribosomal 16S-DNA sequence data, we were able to successfully differentiate between Pinctada margaritifera and Pinctada maculata larvae from 5 to 13 days old. This is the first description of a non-destructive method allowing bivalve larvae discrimination between species within this genus. The method allowed us to successfully identify P. margaritifera larvae in natural plankton samples. This result is a key step needed to develop monitoring of P. margaritifera larval distribution in French Polynesian lagoons, a procedure which will increase spat collection efficiency and ensure sustainable development of pearl oyster farming.

Pearl oyster grafting is a complex surgical operation that should lead to pearl formation after approximately eighteen months. Although this technique has been used for many years in French Polynesia, the grafting process is still not standardised. While studies have been carried out in order to improve graft performance and yield, these remain highly variable due to post-grafting mortality, nucleus rejection and unreliable pearl quality, all of which constrain pearl farm profitability. The present study uses histological analysis to monitor oysters that either rejected or retained their nuclei. Both groups of oysters are compared in terms of evolution of the graft, which could influence retention, and the development of a pearl sac in cases where grafting was successful. Data show that rejection phenomena are linked to a number of causes, notably an inflammatory reaction in the “receiving” oyster, the presence of numerous tissue lesions and the quality of the grafted tissue. These results suggest that study is needed on the different concomitant elements of the grafting process: the graft “donor” oysters, the nucleus and the “receiving” oyster and their interactions.

Hatchery produced pearl oyster spat are generally transferred to the ocean on spat collectors which may, or may not, be covered with fine mesh to prevent spat loss. This study examined the effectiveness of mesh bag covers on culture units for spat of the blacklip pearl oyster, Pinctada margaritifera. It assessed spat retention and growth in culture units without mesh bags (REMOVE) and in those where mesh bags were replaced weekly (REPLACE), washed weekly (WASH) or left unwashed (NO WASH) for the duration of the six-week experiment. Culture units in the REPLACE treatment had the greatest number of spat at the end of the experiment and 41% more spat than in the REMOVE treatment. Grading of the spat from each treatment at the end of the experiment showed that the REPLACE treatment had the highest number of spat in large and medium size classes with mean (± SE) dorso-ventral height of 23.9 ± 0.5 mm and 17.4 ± 0.5 mm, respectively. Mean wet weight of the mesh bags at the end of the experiment differed between treatments with bags from the WASH and NO WASH treatments having significantly greater mean wet weights than those from the REPLACE treatment. The results show that the presence of a mesh bag around spat culture units may improve retention of spat by between 18–41%. However, protective mesh bags may foul quickly and, to maximise retention and spat growth, weekly replacement of mesh bags should be undertaken.

An infection of pearl oysters, Pinctada maxima, attributed to a Haplosporidium sp. by Hine and Thorne (1998) has been detected on 3 occasions and is considered to represent a serious concern to the pearling industry in Australia. The spore ornamentation of the parasite was determined by scanning electron microscopy and transmission electron microscopy. Spores of the parasite were pleomorphic, or elongated 3·5–4 μm×2·5–3·0 μm in size. Two filaments were wound around the spore and originated from 2 ‘knob-like’ posterior thickenings. Both filaments passed up one side of the spore together until just below the operculum whereupon each split and passed obliquely under the lip of the opercula lid. Each filament wrapped around the spore 4 times. The posterior thickenings seem to appear late in the development of the spore and were composed of spore wall material. A second set of branching tubular filaments composed of a different material was observed on the spore body although not on mature spores possessing a ‘knob-like’ posterior thickening. The ornamentation on the spores of the pearl oyster parasite was unique amongst described haplosporidian species where spore ornamentation is known. The parasite is named in this manuscript as Haplosporidium hinei n. sp.

The characteristics of the phytoplankton of Takapoto Atoll are reviewed from the studies conducted between 1974 and 1998. These studies mainly concerned the biomass and primary production of phytoplankton while the taxonomic composition received far less attention. The mean biomass is 0.2–0.3 μg chl a·L–1. The phytoplankton is homogeneously distributed on a year scale although an higher biomass (0.8 μg chl a·L–1) may temporarily exist in the south part of the atoll under moderate tradewinds or calm weather. The gross primary production reached 0.8 g C·m–2·day–1 whereas the net primary production is estimated to be 0.7 g C·m–2·day–1. No significant long-term changes of the biomass or primary production can be observed. The implications of this stability are discussed in the context of the mother-of-pearl mariculture. Size fractionated samples revealed the predominance of picophytoplankton which represented more than 60% of the phytoplankton biomass and achieved > 50% of the primary production. The taxonomic composition observed in 1974 showed the predominance of three algal groups: diatoms, dinoflagellates and coccolithophorids. The diatoms were the most diversified group, while the dinoflagellates were the most abundant. No further examination of the phytoplankton was undertaken until 1996. At that time, the microplankton was quite absent, and the phytoplankton communities were dominated by the pico- and nanophytoplankton, mainly chlorophytes, prymnesiophytes and dinoflagellates. This drastic shift of the phytoplankton communities towards smaller size is not clearly understood. It emphasises the need of taxonomic studies for a better understanding of the lagoon ecology.

Between 1994 and 1999, several sampling designs were used to measure the growth of the black-lip pearl oyster, Pinctada margaritifera var. cumingi at different sites in French Polynesia. Using a common statistical method, growth data were analysed for nine sites and showed significant geographic variability. Parameters of the Von Bertalanffy model, fitted on the nine data sets, ranged from 147 to 186.5 mm for the H∞ parameter and from 0.42 to 0.58 year–1 for the k parameter. The φ parameter (combination of H∞ and k) provided a ranking classification of growth in these nine sites: growth was low in closed atoll lagoons, such as Takapoto, whereas the island lagoons and ocean habitat supported a very promising shell growth. Calculated on the basis of these models, two parameters of interest for pearl farming were computed: 1) the time for pearl oysters to reach a size of 100 mm (size at which they are suitable for nucleus implantation), which ranged from 21 to 26 months, and 2) the annual shell growth increment at this size (correlated to the rate of nacreous deposition on the pearl), which varied from 19.7 to 31.8 mm·year–1. The combination of these two sets of results demonstrated that the time necessary to produce a comparable pearl varied significantly according to the site. Several hypotheses to explain the differences observed in the growth of P. margaritifera in these nine sites are proposed. The most reasonable would appear to be: 1) the negative effect of high temperature (> 30°C), and 2) the degree of water renewal and food supply around the bivalve. Although this paper demonstrated significant variability in growth performance in the nine investigated sites, further investigations are needed, especially concerning the flesh growth rate. Indeed, this highly relevant parameter, which reflects both bivalve health and the suitability of farming sites, has received very little attention to date.
