Animal models are a key component of translational medicine, helping transfer scientific findings into practical applications for human health. A fundamental principle of research ethics involves weighing the benefits of the research to society against the burden imposed on the animals used for scientific purposes. The utilisation of wild animals for research requires evaluation of the effects of capture and invasive sampling. Determining the severity and duration of these interventions on the animal’s physiology and behaviour allows for refining study methodology and for excluding or accounting for biased data. In this study, 39 Scandinavian brown bears (Ursus arctos) captured either while hibernating in winter or via helicopter in summer and that underwent surgery as part of a human health project had their movement, body temperature and timing of onset of hibernation compared with those of 14 control bears that had not been captured during the same period. Bears captured in winter and summer showed decreased movement from den exit until late summer, compared to those in the control group. Bears captured in summer showed reduced movement and body temperature for at least, respectively, 14 and 3 days, with an 11% decrease in hourly distance, compared to pre-capture levels, but did not differ in the timing of hibernation onset. We reveal that brown bear behaviour and physiology can be altered in response to capture and surgery for days to months, post-capture. This has broad implications for the conclusions of wildlife studies that rely upon invasive sampling.

Textbook recommendations for gavaging rats vary between 1-5 ml for an adult rat. Rats weighing either 130 g or 250 g were gavaged with varying dosages of barium sulphate (BaSO4). After dosing, radiographs were taken at 0, 15 and 60 min. Animals showing a section of the small intestine totally filled with BaSO4 were scored as displaying spontaneous release. Other rats of the same sizes were gavaged with similar doses and subsequently tested in an open-field arena for behavioural abnormalities that might indicate stress or pain resulting from the procedure. Body temperature before and after treatment was recorded using microchip transponders. None of the 250 g rats in the 1 ml dosage group showed spontaneous release through the pyloric sphincter. In the 2 ml and 4 ml dosage groups, only one out of five animals showed spontaneous release. In the 6 ml dosage group, half of the animals showed spontaneous release. In the 8 ml and 10 ml dosage groups, five out of six and four out of five, respectively, showed spontaneous release. If doses were higher than 12 ml, no animal was able to keep all of the BaSO4 in its stomach. In the rats weighing 130 g, the 3 ml dosage group showed only one out of four rats with spontaneous release, whereas in the 5 ml and 7 ml dosage groups, all animals showed spontaneous release. After 15 min, all of the rats in both weight groups showed BaSO4 in the duodenum. Ambulation, rearing up onto the hind legs and defecation, as well as body temperature immediately after dosing, correlated very strongly with the dose (ml kg−1); increasing the dose resulted in reduced ambulation, rearing, defecation and body temperature. However, 10 min after performance of the open-field test, neither body temperature, serum corticosterone nor serum glucose showed any correlation with dose. This study indicates that high doses (ie doses up to 10 ml for a 250 g rat) might be safe to use; however, if an adverse impact on the rat is to be avoided, use of much lower doses should be considered—for example, doses that do not enforce opening of the pyloric sphincter in any rat. This would be less than 4 ml kg−1 in a 250 g rat.

The effect of transport on core and peripheral body temperatures and heart rate was assessed in ten 18-month-old Coopworth ewes (Ovis aries) Manual recordings of core (rectal) temperatures were obtained, and automated logging of peripheral (external auditory canal and pinna) temperatures and heart rate was carried out on the day prior to (day 1) and during (day 2) a standardised transport procedure. Transport produced a significant increase in the rectal temperature, which declined following unloading. Peripheral measures of body temperature also exhibited changes with transport. However, both ear-canal and pinna temperatures declined during actual transport, reflecting to some extent the decline in ambient temperatures recorded externally by sensors on the ear tags of the animals. Peripheral measurement of temperature, particularly at the readily accessible ear canal, may offer potential as a technique for the long-term monitoring of thermal responses to stress. However, further research is required into the potentially confounding effects of ambient temperature and wind chill factors.

Since the World Health Organization’s (WHO’s) pandemic declaration on March 11, 2020, coronavirus disease 2019 (COVID-19) outbreaks have occurred on numerous maritime vessels and the containment measures, travel restrictions, and border closures continue to make it increasingly difficult for ship operators world-wide to be granted pratique, conduct trade, and conduct crew changes.

Knowledge of Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2) circulating on-board a ship prior to its arrival has significant implications for the protection of shore-based maritime workers (ie, pilots, stevedores, and surveyors), the broader community, and trade. A useful approach is a graded assessment of the public health risk. The Western Australia (WA) experience and associated observed pitfalls in implementing the prediction equation for the potential presence of SARS-CoV-2 on-board based on five COVID-19 outbreaks on commercial and cruise vessels during 2020 is described.

Despite best efforts, the qualitative and quantitative predictors of SARS-CoV-2 circulating on-board commercial vessels are failing to deliver the required certainty, and to date, the only accepted method of ascertaining the presence of SARS-CoV-2 remains the real-time reverse transcription polymerase chain reaction (rRT-PCR) testing reported by an accredited laboratory.

Based on legal or regulatory requirements, germane processes, underpinned by robust and auditable processes and procedures, must be put in place to inform the risk assessment of SARS-CoV-2 circulating on-board vessels.

Pre-weaning mortality represents a major economic loss and welfare concern for the global pork industry. Caffeine administration prior to, or after, parturition positively affects metabolic parameters associated with survival in newborn animals. However, its effects on piglet viability and survival when administered within the first 24 h of life have not been evaluated. This study determined the effect of caffeine treatment during the 24 h postpartum period on piglet viability, growth and survival. Piglets received 30 or 0 mg of caffeine orally at birth or 8 to 12 h following birth and again at 24 h. Vitality, meconium staining and blood lactate were assessed at birth. Time to first reach the udder and suckle was recorded. Body weight and rectal temperature were measured at 10 min, 24 and 72 h after birth. A blood sample for analysis of serum immunoglobulin G was collected at 24 h, and BW and survival were monitored to 20 days of age. Caffeine had no effect on body temperature, blood glucose, serum immunoglobulin G concentration or weight (P > 0.05). However, when caffeine was administered at birth, it significantly increased 24 h mortality in piglets (P < 0.05). Piglet mortality between birth and weaning also tended to be higher for piglets receiving caffeine at birth (P = 0.063). These data demonstrate that caffeine administration to piglets at birth, but not 8 to 12 h after birth, impaired piglet survival. Further research into caffeine dosage and timing postpartum is required to establish its efficacy.

Heat stress has a significant impact on all livestock and poultry species causing economic losses and animal well-being concerns. Providing shade is one heat-abatement strategy that has been studied for years. Material selected to provide shade for animals greatly influences the overall stress reduction provided by shade. A study was conducted to quantify both the environment and animal response, when cattle had no shade access during summertime exposure or were given access to shade provided by three different materials. A total of 32 Black Angus heifers were assigned to one of the four treatment pens according to weight (eight animals per pen). Each pen was assigned a shade treatment: No Shade, Snow Fence, 60% Aluminet Shade Cloth and 100% Shade Cloth. In the shaded treatment pens, the shade structure covered ~40% of the pen (7.5 m2/animal). Animals were moved to a different treatment every 2 weeks in a 4×4 Latin square design to ensure each treatment was applied to each group of animals. Both environmental parameters and physiological responses were measured during the experiment. Environmental parameters included dry-bulb temperature, relative humidity, wind speed, black globe temperature (BGT), solar radiation (SR) and feedlot surface temperature. Animal response measurements included manual respiration rate (RRm), electronic respiration rate (RRe), vaginal temperature (body temperature (BT)), complete blood count (CBC) and plasma cortisol. The environmental data demonstrated changes proportional to the quality of shade offered. However, the animal responses did not follow this same trend. Some of the data suggest that any amount of shade was beneficial to the animals. However, Snow Fence may not offer adequate protection to reduce BT. For some of the parameters (BT, CBC and cortisol), 60% Aluminet and 100% Shade Cloth offers similar protection. The 60% Aluminet lowered RRe the most during extreme conditions. When considering all parameters, environmental and physiological, 60% Aluminet Shade Cloth offered reductions of BGT, SR, feedlot surface temperature and the best (or equal to the best) overall protection for the animals (RRe, RRm, BT, blood parameters).

Seasonal affective disorder (SAD) is characterised by recurrent episodes in autumn and winter of depression, hypersomnia, augmented appetite with carbohydrate craving, and weight gain, and can be successfully treated with bright light. Circadian rhythm hypotheses (summarized in) have stimulated research into the pathophysiology of SAD, postulating that:

1. 1.The illness is a consequence of delayed phase position,

2. 2.It is correlated with diminished circadian amplitude, or

3. 3.It results from changes in the nocturnal duration between dusk and dawn e.g. of low core body temperature or melatonin secretion. Light is considered to act directly on the circadian pacemaker (‘Process C’) and not on sleep dependent processes (‘Process S’). Thus successful treatment of SAD must act via mechanisms within known retinohypothalamic pathways. Conversely, emergence of SAD symptoms may reflect inappropriate neurobiological response to decreasing daylength.

In this prospective observational study rectal and vaginal temperature of 82 (26 primiparous, 56 multiparous) early post-partum healthy dairy cows that calved without intervention within 3 months and did not show clinical signs of infectious and metabolic diseases were continuously measured and evaluated for associations with plausible factors during the first 10 days in milk (DIM). During May, June and July mean (±sd) temperature humidity index (THI) was 60·1±5; 66·8±5·6 and 74·2±4·3, respectively. Environmental conditions had a negligible effect on body temperature (BT) during May (P<0·05). During June and July, however, the ambient temperature and THI influenced BT (P<0·05). Furthermore, plausible factors like parity, DIM, months and time of day had an effect on BT (P<0·05). Overall, primiparous cows demonstrated 0·2°C greater BT during the first 10 DIM than multiparous cows. The effect of parity, however, on BT varied between DIM according to month (P<0·001). During this 3-month study period all cows demonstrated BT rhythms; however, the amplitude of BT increased from May to July (0·3 to 0·7°C). A greater proportion of the vaginal temperature measurements exceeded a threshold tested (⩾39·5°C) during July (46·8%) than in June (33·9%) and May (19·3%). Overall the percentage of BT values above a threshold of ⩾39·5°C was lower during the period 6.00–10.00 compared with the remaining 20 h (P<0·05). Therefore this study concluded that the BT of healthy post-partum dairy cows during the period 1–10 DIM post partum is greater compared with the reference range of 38·6 to 39·5°C reported by others and is influenced by parity, DIM, time of day and THI. When the association between BT and THI increased the reliability of threshold levels of BT (⩾39·5°C) decreased.

The study of mammals suffering intense poaching in remote areas poses an increasingly difficult conservation challenge, in part because the extreme flightiness of such species complicates safe capture. The benefits of handling (an opportunity to obtain biological information and attach radio collars) must be weighed against stress to the animals and potential capture-related mortality. In parts of Central Asia this problem is not trivial, as populations have been heavily harvested and opportunities for restraint are often limited. Mongolian saiga Saiga tatarica mongolica, being both Endangered and poached, typifies these issues. Here we describe capture protocols for adult females handled quickly and without anaesthesia. Using multiple vehicles driven at high speed, individual saiga were isolated from groups and herded into nets. Chase time was linearly associated with rectal temperature (P < 0.03), with maximum pursuits and temperatures of 9 minutes and 43.1°C, respectively; time to release averaged < 7 minutes. Given that rural residents often chase saiga and other desert and steppe-dwelling ungulates, for photography or for amusement, our results offer conservationists and government officials an empirical basis for recommending prudence on chase times and/or for recommending that the practice be prevented.

This study examined concurrent stresses of nematode infection and pregnancy using pregnant and non-pregnant CD1 mice infected 3 times with 0, 50 or 100 Heligmosomoides bakeri larvae. Physiological, energetic, immunological and skeletal responses were measured in maternal and foetal compartments. Resting metabolic rate (RMR) was elevated by pregnancy, but not by the trickle infection. Energy demands during pregnancy were met through increased food intake and fat utilization whereas mice lowered their body temperature during infection. Both infection and pregnancy increased visceral organ mass and both altered regional bone area and mineralization. During pregnancy, lumbar mineralization was lower but femur area and mineralization were higher. On the other hand, infection lowered maternal femur bone area and this was associated with higher IFN-γ in maternal serum of heavily infected pregnant mice. Infection also reduced foetal crown-rump length which was associated with higher amniotic fluid IL-1β.

Angora goats are known to be vulnerable to cold stress, especially after shearing, but their thermoregulatory responses to shearing have not been measured. We recorded activity, and abdominal and subcutaneous temperatures, for 10 days pre-shearing and post-shearing, in 10 Angora goats inhabiting the succulent thicket of the Eastern Cape, South Africa, in both March (late summer) and September (late winter). Within each season, environmental conditions were similar pre-shearing and post-shearing, but September was an average 5°C colder than March. Shearing resulted in a decreased mean (P < 0.0001), minimum (P < 0.0001) and maximum daily abdominal temperature (P < 0.0001). Paradoxically, the decrease in daily mean (P = 0.03) and maximum (P = 0.01) abdominal temperatures, from pre-shearing to post-shearing, was greater in March than in September. Daily amplitude of body temperature rhythm (P < 0.0001) and the maximum rate of abdominal temperature rise (P < 0.0001) increased from pre-shearing to post-shearing, resulting in an earlier diurnal peak in abdominal temperature (P = 0.001) post-shearing. These changes in amplitude, rate of abdominal temperature rise and time of diurnal peak in abdominal temperature suggest that the goats’ thermoregulatory system was more labile after shearing. Mean daily subcutaneous temperatures also decreased post-shearing (P < 0.0001), despite our index goat selecting more stable microclimates after shearing in March (P = 0.03). Following shearing, there was an increased difference between abdominal and subcutaneous temperatures (P < 0.0001) at night, suggesting that the goats used peripheral vasoconstriction to limit heat loss. In addition to these temperature changes, mean daily activity increased nearly two-fold after March shearing, but not September shearing. This increased activity after March shearing was likely the result of an increased foraging time, food intake and metabolic rate, as suggested by the increased water influx (P = 0.0008). Thus, Angora goats entered a heat conservation mode after shearing in both March and September. That the transition from the fleeced to the shorn state had greater thermoregulatory consequences in March than in September may provide a mechanistic explanation for Angora goats’ vulnerability to cold in summer.

Summary

Background and objective: Cardiopulmonary bypass is associated with temperature pertubations that influence extubation time. Common extubation criteria demand a minimum value of core temperature only. The aim of this prospective study was to test the hypothesis that changes in core and skin surface temperature are related to extubation time in patients following normothermic and hypothermic cardiopulmonary bypass. Methods: Forty patients undergoing cardiac surgery were studied; 28 patients had normothermic cardiopulmonary bypass (nasopharyngeal temperature >35.5°C) and 12 had hypothermic cardiopulmonary bypass (28–34°C). In the intensive care unit, urinary bladder temperature and skin surface temperature gradient (forearm temperature minus fingertip temperature: >0°C =vasoconstriction, ≤0°C =vasodilatation) were measured at 30-min intervals for 10 h postoperatively. At the same intervals, the patients were evaluated for extubation according to common extubation criteria. Results: On arrival in the intensive care unit the mean urinary bladder temperature was 36.8 ± 0.5°C in the normothermic group and 36.4±0.3°C in the hypothermic group ( P = 0.014). The skin surface temperature gradient indicated severe vasoconstriction in the both groups. The shift from vasoconstriction to vasodilatation was faster in normothermic cardiopulmonary bypass patients (138±65 min) than in patients after hypothermic cardiopulmonary bypass (186±61 min, P = 0.034). There was a linear relation between the time to reach a skin surface temperature gradient = 0°C and extubation time (r2 = 0.56, normothermic group; r2 = 0.82, hypothermic group). Conclusions: The transition from peripheral vasoconstriction to vasodilatation is related to extubation time in patients following cardiac surgery under normothermic as well as hypothermic cardiopulmonary bypass.

Aim: To evaluate the reliability of infrared tympanic thermometry in children who have undergone myringotomy with grommet insertion.

Method: Forty children who had undergone myringotomy with at least one grommet insertion had the tympanic temperature of each ear and the axillary temperature measured on admission and 30 minutes post-operatively.

Result: No difference was found between the pre- and post-operative temperatures measured by either method (p > 0.05, paired t-test, hypothesized difference of 0).

Conclusion: Infrared tympanic thermometry is reliable in monitoring body temperature in children who have had minor ear surgery.

In highly seasonal environments animals that use daily torpor usually do so during the rest phase. In subtropical and tropical environments, however, daily torpor may not necessarily be confined to the animals' rest phase. Animals may display torpor during the active phase. We investigated the daily rhythms and seasonal rhythms of body temperature (Tb) in normothermic and torpid free-ranging rock elephant shrews Elephantulus myurus at Weenen Game Reserve, KwaZulu-Natal, South Africa. Tbs were recorded using miniature dataloggers from May 2001 to May 2002. Daily patterns of Tb in normothermic E. myurus suggested polyphasic Tb patterns that nevertheless suggested a rest phase that was coincident with the daytime. When the elephant shrews used torpor, the lowest Tbs were observed during the night-time, such that torpor was used during the coldest part of the night and arousal was associated with the 3-h period around sunrise. We suggest that the principal benefit of a flexible daily rhythm of Tb was that it facilitated arousal by passive exogenous heating using ambient temperature cycles.

Natterer's bat Myotis nattereri is one of the least known European bats, using roosts inaccessible to humans and shifting to alternative roosts every few days. The characteristics of day and night roosts used by maternity colonies during summer was assessed. Roosts were located by radio-tracking. Roost sites (which may contain more than one roost) were concentrated mainly in core areas of up to 2 km2, within which roost site density ranged from 7 to 15 per km2. Day roosts included tree cavities, apices of attics, mortises in attics and in barns, soffit boxes at eaves, tops of gable walls, crevices in stone walls and a modern cavity wall. Trees comprised 67% of roost sites. Two maternity colonies each used between 21 and 31 roosts distributed across 15 to 25 roost sites. During late summer, the temperature of roosts adjacent to a roof covering averaged 22.5 °C during the day and were significantly warmer than roosts in trees, which averaged 17.3 °C. Attic mortise roosts were intermediate at 20.0 °C. The maximum ambient roost temperature in which bats remained was 32 °C and the maximum body surface temperature recorded while roosting was 37.6 °C. Thus, bats seem to require access to a large number of roosts with a range of temperatures that they can use depending on their reproductive state and energy requirements. The study colonies also exhibited high inter-annual fidelity to roosts. Attic maternity roosts used during late gestation and early lactation were reused for the same function in successive years, as were heavily timbered barns during late lactation and post-lactation. This continuing dependence upon established roosts indicates that these types of maternity roost should be given high conservation priority. Field boundaries should also be managed to maintain mature trees with cavities for potential day roosts and hedges suitable as night perches.

The discovery of Trichinella zimbabwensis in farm crocodiles of Zimbabwe has opened up a new frontier in the epidemiology of the Trichinella genus. The objective of the present study was to investigate the infectivity of encapsulated species (T. spiralis, T. nativa, T. britovi, T. murrelli and T. nelsoni) and non-encapsulated species (T. pseudospiralis, T. papuae and T. zimbabwensis) in caimans (Caiman crocodilus), varans (Varanus exanthematicus), pythons (Python molurus bivittatus) and turtles (Pelomedusa subrufa) raised at their natural temperature range (26–32 °C). Mice and chickens were used as controls. At 6 days post-infection (p.i.), adult worms were detected in the small intestine of reptiles infected with T. papuae and T. zimbabwensis, of chickens infected with T. pseudospiralis and of mice infected with all encapsulated and non-encapsulated species. At 60 days p.i., T. papuae and T. zimbabwensis adult worms were collected from the intestine of varans and caimans and larvae from muscles of the four reptile species, T. pseudospiralis larvae from muscles of chickens, and larvae of all Trichinella species from mouse muscles. The highest reproductive capacity index of both T. papuae and T. zimbabwensis was observed in varans. The results show that T. papuae and T. zimbabwensis are able to complete their entire life-cycle in both poikilothermic and homoiothermic animals.

Dairy operations located in many temperate climates experience a decline in milk production associated with short-term moderate heat stress during the summer months. The duration and intensity of this heat stress differs dramatically from the sustained, intense heat experienced in many tropical and subtropical countries where the vast majority of heat stress research has been conducted. As such, many of the strategies utilized to ameliorate production losses associated with sustained, intense heat may not be effective in temperate climates. The objective of this research was to characterize the production responses of lactating dairy cows during and after short-term, moderate heat exposure and, to determine if feeding a fungal culture, Aspergillus oryzae, during a 5-day heat stress period, could effectively alleviate the associated production losses. In a two-period, cross-over design, eight mature lactating cows were given a total mixed ration with or without Aspergillus oryzae. Each 15-day period consisted of a 5-day thermoneutral phase, a 5-day heat stress phase and a 5-day thermoneutral recovery phase. When exposed to moderate heat stress for a 5-day period, cows experienced a rise in vaginal temperature and a decline in dry-matter intake. Following the 5-day heat stress phase, milk yield declined by a factor of 0·09. Supplementation with Aspergillus oryzae had no effect on vaginal temperature, dry-matter intake, water intake, milk yield or milk components. These data indicate that short-term, moderate heat stress, which occurs during the spring and summer months in temperate climates, will significantly decrease production in the lactating cow. Addition of fungal cultures to the diet during the period of imposed heat did not ameliorate production losses associated with this type of heat stress.

Background and objective: Clinical sedation assessment becomes insufficient in deeply sedated patients. Bispectral index as a processed electroencephalogram parameter provides a continuous and observer-independent value reported to change with sedation. The aim of this prospective observational study was to determine the reliability and possible confounding factors of the bispectral index to assess sedation in surgical intensive care patients.

Methods: Following major surgery, bispectral index, body temperature and electromyographic activity of 44 ventilated patients were recorded. Sedation levels were assessed with Ramsay sedation score.

Results: Although bispectral index correlated with Ramsay sedation score (-0.64; P < 0.01) we found that in deeply sedated patients temperature instability and electromyographic activity increased bispectral index. Bispectral index correlated significantly with electromyographic activity (0.80; P < 0.01) and with an increase of body temperature (0.55; P < 0.01) not only in all patients but also in clinically deeply sedated patients (0.57; P < 0.01 and 0.46; P < 0.05).

Conclusions: Only under certain conditions, such as low muscular activity and body temperature stability, may the bispectral index be a useful addition to clinical scoring in the sedation assessment of critically ill patients.
