Avian trichomonosis, caused by the flagellated protozoan Trichomonas gallinae, is a recently emerged infectious disease of British passerines. The aetiological agent, a clonal epidemic strain of the parasite, has caused unprecedented finch mortality and population-level declines in Britain and has since spread to continental Europe. To better understand the potential origin of this epidemic and to further investigate its host range, T. gallinae DNA extracts were collected from parasite culture and tissue samples from a range of avian species in Britain. Sequence typing at the ITS1/5.8S rRNA/ITS2 region resolved three distinct ITS region types circulating in free-ranging British birds. Subtyping by sequence analyses at the Fe-hydrogenase gene demonstrated further strain variation within these ITS region types. The UK finch epidemic strain was preponderant amongst columbids sampled, however, wide strain diversity was encountered in isolates from a relatively small number of pigeons, suggesting further strains present in columbid populations across the UK are yet to be identified. Fe-hydrogenase gene sequence data in isolates from birds of prey with disease were predominantly identical to the UK finch epidemic strain, demonstrating its presence as a virulent strain in UK birds of prey since at least 2009.

Killing of intramolluscan schistosomes by host haemocytes is mediated by reactive oxygen metabolites. Hence, defence against oxidative damage is essential for the parasite to survive. In this study, expression of three key antioxidant enzymes, superoxide dismutase (EC 1.15.1.1), glutathione peroxidase (EC 1.11.1.9) and glutathione-S-transferase (EC 2.5.1.18) was determined in Schistosoma mansoni miracidia, sporocysts and cercariae. Stage-dependent expression of these enzymes was shown to be regulated at the transcriptional level. Second, the influence on enzyme expression of reactive oxygen species (ROS) and of haemocytes from schistosome-resistant and -susceptible host snails was determined. Generation of ROS by xanthine/xanthine oxidase resulted in increased transcript levels for all three enzymes. Addition of hydrogen peroxide induced a significantly increased expression of GPx and SOD but not GST. Snail haemocytes induced an up-regulation of SOD and GPx at 12 and 18 h post-exposure, respectively. Susceptible haemocytes elicited a stronger induction of transcript expression than resistant haemocytes. After 36–48 h, SOD remained up-regulated in sporocysts encapsulated by haemocytes from susceptible hosts, whereas a down-regulation of SOD and GPx occurred in schistosomes encapsulated by haemocytes from resistant snails. These observations indicate that schistosomes express elevated levels of antioxidant enzymes in interaction with haemocytes from susceptible snail hosts in which they survive. On the other hand, haemocytes of resistant snails may interfere with reactive oxygen detoxification via down-regulation of schistosome antioxidant enzymes, thus shifting the balance towards parasite killing.

Experiments were carried out to investigate the effects of Ascaris suum infection on the growth, food intake, nitrogen and fat utilization and intestinal function of young pigs receiving diets low in protein. An infection procedure was developed which provided relatively uniform and heavy infections in pigs under study. Ascaris-infected pigs showed statistically significant reduction in growth rate and food intake compared to uninfected controls. The reduction in growth rate was observed after the A. suum were mature and eggs were observed in the faeces of pigs. The degree of growth retardation was significantly correlated with worm burden. Infected pigs showed no consistent reduction in nitrogen and total solids digestibility compared to controls but fat digestion was reduced. The infected pigs showed reduced nitrogen retention compared to uninfected controls. Ascaris-infected pigs had heavier intestinal tracts with increased size of the tunica muscularis. Lactase activity in mucosa from infected pigs was significantly lower than in controls, whereas no consistent effect was observed in mucosal sucrase and maltase activity. When Ascaris-infected pigs were given an oral lactose load, blood glucose levels rose less than in control pigs, suggesting that the infection resulted in impaired lactose tolerance.

1. The cycle of vertical migration of the infective larvae of Haemonchus contortus on experimental plots of Lolium perenne S 24, kept out of doors, has been related to meteorological and micrometeorological factors.

2. The three main factors concerned with vertical migration are temperature, humidity and light intensity.

3. In the majority of cases the greatest number of larvae are on the grass blades in the early morning and in the evening, less being recovered at night and during the middle of the day. The time of the morning maximum becomes progressively earlier passing from winter to summer, and the time of the evening maximum progressively later. The reverse is true in the second half of the year.

4. Low and high temperatures accompanied by low humidity inhibit vertical migration on the grass.

5. High humidity (continuous precipitation) does not prevent migration, the two daily maxima are apparent, but the numbers climbing are less than in the absence of rain.

Infection by a parasite often induces behavioural changes in the host and these changes may benefit either the host or the parasite. However, whether these changes are active host defence mechanisms or parasitic manipulations or simply incidental byproducts of the infection is not always clear. It has been suggested that understanding the proximate mechanisms of these changes as well as comparative studies could help distinguish these alternatives better. Behavioural fever is a common response to an infection in many animals and we investigated the phenomenon in the novel host-parasite relationship between the honeybee and the temperature-sensitive microsporidian Nosema ceranae. Our results show that infected bees prefer higher temperatures and even though this seems to benefit the pathogen, the proximate mechanism underlying this change is the pathological stress underlying the infection. Especially because it is a new host-parasite relationship, it is best to label the observed behavioural change as a case of incidental benefit although this does not rule out selection acting on it. We discuss the importance of looking at the behavioural outcomes of host-parasite relationships and the importance of studying them at multiple levels for understanding their origin and maintenance.

To clarify the taxonomy of trichomonads associated with human respiratory diseases, we examined a collection of axenic trichomonad strains isolated from the oral cavity and bronchi of patients from pulmonary diseases clinics in Tallin, Estonia. The oral and bronchial strains were compared mutually as well as with a reference strain of Trichomonas tenax, a common inhabitant of the human oral cavity, and other trichomonad species from humans and animals. Unexpectedly, the morphological studies, as well as DNA sequencing of ITS1-5.8S rRNA-ITS2 regions revealed that the Estonian strains belong to the genus Tetratrichomonas, with a high similarity to the avian species Tetratrichomonas gallinarum. None of the strains belonged to Trichomonas tenax. DNA fingerprinting using the RAPD method separated Estonian strains into 2 distinct groups: ‘bronchial’ consisting of 5 and 2 strains isolated from bronchi and ‘oral’ cavity, respectively, and oral consisting of 3 oral strains. Consistent differences between ‘bronchial’ and ‘oral’ groups were confirmed by analysis of ITS1-5.8S rRNA-ITS2 sequences. Our results have revealed novel trichomonad species of the human oral cavity and bronchi.

Dikerogammarus villosus is an invasive amphipod that recently colonized the main rivers of Central and Western Europe. Two frequent microsporidian parasites were previously detected in this species, but their taxonomic status was unclear. Here we present ultrastructural and molecular data indicating that these two parasites are in fact a single microsporidian species. This parasite shares numerous characteristics of Nosema spp. It forms elongate spores (cucumiform), developing in direct contact with host cell cytoplasm; all developmental stages are diplokaryotic and the life cycle is monomorphic with disporoblastic sporogony. Initially this parasite was described as Nosema dikerogammariOvcharenko and Kurandina 1987. However, phylogenetic analysis based on the complete sequence of SSU rDNA places the parasite outside the genus Nosema and it is therefore ascribed to a new genus Cucumispora. The key features characteristic to this genus are: presence of a very well-developed, umbrella-shape anchoring disk covering the anterior part of polaroplast; arrangement of isofilar polar filament into 6–8 coils convoluted with different angles, voluminous diplokaryon, thin spore wall and relatively small posterior vacuole containing posterosome. The parasite infects most host tissues but mainly muscles. It showed high rates of horizontal trophic transmission and lower rates of vertical transmission.

We review the literature on parameter values relevant to the epidemiology of strongyle nematode infections of domestic sheep. Information is subdivided by parasite genus, country of origin and climate type. While field observations have been made in a large number of countries, the bulk of studies under controlled conditions have been conducted in Australia, New Zealand and the UK. For these countries, experiments and parameters are interpreted in terms of a previously published model of nematode dynamics, and are used to calculate the basic reproduction number. Average values range from less than 6 for Haemonchus contortus in New Zealand and a winter rainfall region of Australia, to more than 16 for Ostertagia circumcincta in New Zealand and the UK. Additional considerations of the effects of climate and the annual replacement of host stock show that for conditions favourable for parasite transmission this is a robust indicator of parasite epidemiology. When climate variation and annual replacement are added to the model, it is shown to reasonably describe the qualitative behaviour of an experimental data set, indicating it to be a useful tool for further investigation of some of the underlying assumptions of sheep–nematode dynamics.

A decade of genome sequencing has transformed our understanding of how trypanosomatid parasites have evolved and provided fresh impetus to explaining the origins of parasitism in the Kinetoplastida. In this review, I will consider the many ways in which genome sequences have influenced our view of genomic reduction in trypanosomatids; how species-specific genes, and the genomic domains they occupy, have illuminated the innovations in trypanosomatid genomes; and how comparative genomics has exposed the molecular mechanisms responsible for innovation and adaptation to a parasitic lifestyle.

The double isotope labelling method of Arias, Doyle & Schimke (1969) and Dehlinger & Schimke (1971) was used to determine the relative rates of turnover of proteins in various tissue fractions and in the culture medium of adult Schistosoma mansoni. The majority of proteins in the surface membrane turned over at about the same rate. This is consistent with a model for membrane assembly and degradation involving multi-protein units. The proteins in the other fractions examined, except frozen-thawed supernatant fluid, also turned over at about the same rate. In the frozen and thawed supernatant fluid, the higher molecular weight (>40000 daltons) proteins turned over at a much greater rate than the lower molecular weight proteins (<40000 daltons). The antigens in the culture medium, both particulate and soluble, had a higher turnover rate than the worm tissue proteins. It is concluded that the culture medium antigens are released as the result of two distinct processes: (a) surface membrane turnover and (b) a rapid secretory process. It is possible that the high molecular weight proteins found in the frozen-thawed supernatant fluid are involved in the rapid secretory process. The culture media of 6-day and 16-day schistosomula were also examined by the double isotope method. The culture medium of the adult worm and 16-day schistosomula contains more material presumed to be secreted than does that from the 6-day schistosomula. This may explain the poor immunogenicity of young schistosomula.

The surface lipophosphoglycans (LPG) of Leishmania promastigotes express stage- and species-specific polymorphisms that are defined by variations in the type and number of phosphorylated oligosaccharide repeats. We have studied how these polymorphic structures control the development of transmissible infections in the sandfly vector as well as the species-specificity of vectorial competence. Procyclic promastigotes displayed an inherent capacity to bind to midgut epithelial cells of a competent vector. This capacity was lost during their transformation to metacyclic promastigotes, permitting the selective release and anterior migration of infective-stage parasites for subsequent transmission by bite. Midgut attachment and release were found to be controlled by developmental modifications in terminally exposed saccharides on LPG, which, depending on the species of Leishmania, involved either substitution or capping of terminal side-chain sugars, loss of terminal side-chain sugars, substitution or loss of neutral capping sugars. The stage-specific terminal sugars involved in midgut adhesion are, in some cases, also species-specific, and the extent to which these differences affect midgut attachment, forcefully predicted vectorial competence.

Relationships between ascariasis and lactose digestion and between ascariasis and food transit time from mouth to caecum were investigated in young children from Chiriqui Province, Republic of Panama. The breath hydrogen method was used in both studies. Ascaris-infected children showed a significantly poorer degree of lactose digestion following a test oral load than uninfected children. Recovery of the capacity of the children to digest lactose was still not fully complete for at least 3 weeks following anthelmintic treatment. On average, the mouth-to-caecum transit time was similar in infected and uninfected children, but among the Ascaris-infected children the transit time tended to be shorter in relation to the intensity of infection. Evidence from a cross-sectional survey indicated that ascariasis was significantly associated with reduced plasma vitamin A and carotenoid concentrations. This relationship remained after controlling for a range of socio-economic variables. Ascaris-infected children were frequently found to have lower haematocrits and blood haemoglobin concentrations than uninfected children, but these relationships could not be attributed to ascariasis alone.

Avian trypanosomes are widespread parasites of birds, the transmission of which remains mostly unclear, with various blood-sucking insects mentioned as possible vectors. A search for vectors of trypanosomes of sparrowhawk (Accipiter nisus), buzzard (Buteo buteo), lesser-spotted eagle (Aquila pomarina) and kestrel (Falco tinnunculus) was performed in Czech and Slovak Republics. Black flies (Eusimulium spp.), hippoboscid flies (Ornithomyia avicularia), mosquitoes (Culex pipiens pipiens) and biting midges (Culicoides spp.), trapped while attempting to feed on raptor nestlings, were found to contain trypanosomatids in their intestine. Trypanosomes from the raptors and blood-sucking insects were isolated, and their 18S rRNA sequences were used for species identification and for the inference of intra- and interspecific relationships. Together with the trypanosome isolated from a black fly, the bird trypanosomes formed a well-supported Trypanosoma avium cladeThe GenBank accession numbers of the determined sequences are: AF416559 (Trypanosoma ‘avium’ APO1), AF416560 (Herpetomonas sp. CER1), AF416561 (Trypanosoma sp. CUL1), AF416562 (Trypanosoma sp. OA6), AF416563 (Trypanosoma ‘avium’ SIM3).. The isolates derived from hippoboscid flies and mosquitoes are most likely also avian trypanosomes infecting birds other than the studied raptors. Analysis of the kinetoplast, that has features characteristic for the avian trypanosomes (minicircle size; dimensions of the kinetoplast disc), provided further evidence for the identification of vectors. It is suggested that all trypanosomes isolated from raptors included in this study belong to the T. avium complex and are transmitted by the ornithophilic simuliids such as Eusimulium securiforme.

The sex ratio may be influenced by many factors, such as stress and immunosuppression, age of parents, parity and sex of preceding siblings. In animal systems, parasitism often changes the sex ratio of infected hosts, which can increase the probability of their transmission. The most common human protozoan parasite in developed countries, Toxoplasma gondii (prevalence 20%−80%), is known to change the behaviour of its intermediate hosts, thereby increasing the probability of transmission to its definitive host (the cat) by predation. The intermediate hosts, which under natural conditions are rodents, serve as the vector for Toxoplasma. Therefore, we speculate that Toxoplasma can alter the secondary sex ratio (i.e. male to female ratio in the offspring) of infected females to increase the proportion of (congenitally infected) male offspring, which are the more migratory sex in most rodent species. Here we studied the sex ratio of experimentally infected laboratory mice, expressed here as the proportion of males in the litter. In accordance with our hypothesis and results of previous retrospective cohort studies on human subjects, mice with toxoplasmosis produced a higher sex ratio than controls, in the early phase of latent infection. In the later phase of infection, mice with congenital toxoplasmosis had a lower sex ratio than controls, which is in accord with the Trivers-Willard hypothesis of sex ratio manipulation, suggesting that females in poor physical condition give birth to more female offspring.

The studies presented in this paper describe the effects of control exerted by the immunological response of the host (sheep) on the developmental stages and fecundity of the parasite Nematodirus spathiger. Thirty Merino lambs, maintained worm-free, were randomly divided among three treatment groups each containing ten animals. The group 1 lambs were each given a single dose of 50,000 infective larvae, the group 2 animals were given a single dose of 130,000 infective larvae, and the group 3 animals were given twenty-five consecutive daily doses of 2000 infective larvae. Group fecundity was estimated by performing worm egg counts on daily faecal samples and development of the parasite burden was followed by carrying out differential worm counts on animals slaughtered at crucial stages during the infections. Total fecundity was greatest in group 1 and least in group 3 with group 2 intermediate. An inverse relationship between total fecundity and percentage retardation of development (percentage of 4th-stage larvae in the total worm burden) was demonstrated between the three treatment groups.

The results of the studies also show that control of the parasitic infection is mediated about threshold levels of immunological responsiveness and is expressed by (1) elimination at the infective (L3) stage, (2) retardation of development at the L4 stage, (3) reduction in egg production of the adult female, (4) elimination of adult worms, and (5) discrimination against the female in both the degree of retardation in the L4 stage and the extent of elimination in the adult stage.

Great variation in response was observed between individuals within treatment groups. The source of variation is attributed to differences in degree of antigenic disparity of the parasite with individual members of the heterozygous host population in which antigenic characters are not uniformly segregated.

The concept of ‘fitness’ of the parasitic species was introduced into the discussion to provide a rational interpretation of the results.

Virus diseases have been reported from more than 800 species of insects and mites. Isolates of the baculovirus and cytoplasmic polyhedrosis virus groups have biological properties which should lead to their successful use as microbial control agents in integrated pest management programmes. These viruses infect the larval stages of many lepidopterous and hymenopterous pests, producing a chronic or lethal infection and the release of large quantities of relatively stable infective inclusion bodies (IBs). The IBs serve as the means by which the viruses are transmitted and persist outside the host. Younger larvae are more susceptible to infection than older stages, and this difference influences the timing of application and doses of virus needed for practical pest control. The high degree of host specificity of many isolates reduces their potential ecological hazard but also limits their use, particularly on crops where a complex of pests is established. Environmental persistence is also a limiting factor as virus is rapidly inactivated by ultra-violet light even when contained within IBs. The viruses persist for longer periods when transmitted within the host population, a feature of virus infections restricted to the insect gut.

The practical use of insect viruses in horticulture and agriculture does not utilize their full epizootic potential, but takes advantage of their high pathogenicity and specificity. The baculoviruses of codling moth, and Heliothis spp. provide satisfactory pest control, but for their most cost-effective use it is important to determine the minimum dosage rates of virus required. It is encouraging that studies of the virus control of Pieris spp. have suggested that control achieved by the insecticidal use of a virus can be closely predicted from information on dosage-mortality responses, larval feeding rates and virus persistence. The stability of forest and grassland, and their high economic thresholds makes them ideal candidates for longer-term control. Viruses of the coconut rhinoceros beetle and european spruce sawfly provide examples of classical biological control where the viruses persist for long periods, are efficiently transmitted and act as natural regulators of their hosts. Virus control of pasture, and some forest, pests may be possible by manipulating enzootic viruses without the need for direct control measures. More frequently insecticidal applications are needed, providing control of limited duration which requires periodic ‘topping-up’.

Few viruses are commercially-available; their selectivity and often small potential market, may limit industrial interest. However, improvements in virus production, formulation and a better understanding of virus epizootiology should lead to an increasing role for this group of insect pathogens in biological control.

Immunocytochemical techniques used in conjunction with confocal scanning laser microscopy (CSLM) and electron microscopy have been used to demonstrate, for the first time, the distribution of the parasitic platyhelminth neuropeptide, neuropeptide F (NPF) in the cestode, Moniezia expansa. Antisera were raised to intact NPF(1–39) and to the C-terminal decapeptide of NPF(30–39). These antisera were characterized and validated for use in both immunocytochemistry and radioimmunoassay (RIA). NPF immunoreactivity (IR) was detected using both antisera throughout all of the major components of the central and peripheral nervous systems of the worm. The pattern of NPF-IR was found to mirror the IR obtained using a C-terminally directed pancreatic polypeptide (PP) antiserum and FMRFamide antisera; blocking studies using these antisera revealed that FMRFamide and PP antisera cross-react with NPF(M. expansa). RIA of acid-alcohol extracts of the worm measured 114 ng/g using the C-terminal NPF antiserum and 56 ng/g using the whole-molecule-directed antiserum. While the C-terminally-directed NPF antiserum cross-reacts with NPF-related peptides from other invertebrates, the whole-molecule-directed NPF antiserum is specific for NPF(M. expansa). The C-terminal NPF antiserum has potential for use in the identification and purification of NPF analogues from other platyhelminth parasites.

This study investigated the ability of the Pacific oyster, Crassostrea gigas, to act as a carrier or reservoir of the protistan Bonamia ostreae. Studies were carried out independently in Ireland and in Spain. Naïve C. gigas were exposed to B. ostreae both in the field and in the laboratory via natural exposure or experimental injection. Naïve flat oysters, Ostrea edulis, were placed in tanks with previously exposed C. gigas. Oysters were screened for B. ostreae by examination of ventricular heart smears and by polymerase chain reaction (PCR) screening of tissue samples (gill and/or heart) and shell cavity fluid. PCR-positive oysters were further screened using histology and in situ hybridization (ISH). B. ostreae DNA was detected in the tissues and/or shell cavity fluid of a small number of C. gigas in the field and in the laboratory. B. ostreae-like cells were visualized in the haemocytes of 1 C. gigas and B. ostreae-like cells were observed extracellularly in the connective tissues of 1 other C. gigas. When C. gigas naturally exposed to B. ostreae were held with naïve O. edulis, B. ostreae DNA was detected in O. edulis; however, B. ostreae cells were not visualized. In Spain, B. exitiosa DNA was also detected in Pacific oyster tissues. The results of this study have important implications for C. gigas transfers from B. ostreae-endemic areas to uninfected areas and highlight B. ostreae and B. exitiosa's ability to survive extracellularly and in other non-typical hosts.

Patterns of nematode fecundity were investigated for infections of the caecal worm Heterakis gallinarum in the ring-necked pheasant (Phasianus colchicus). Worm length was a good predictor of parasite fecundity. After controlling for worm length no other factors, including parasite intensity, were related to worm fecundity. Density dependence in worm size was detected in natural infections at parasite intensities above a threshold of 96 worms (worm size decreased with increasing parasite intensity). However, below this threshold, worm size actually decreased with decreasing parasite intensity (inverse density dependence). The interaction between density dependence and inverse density dependence in regulating the development and subsequent fecundity of H. gallinarum worms in ring-necked pheasants was demonstrated in an infection experiment. Density dependence was observed in the stunted growth of worms in heavily infected hosts, relative to worms in lightly infected hosts. Inverse density dependence in worm size was the common pattern across hosts by the end of the experiment, when parasite intensities were below the density dependence threshold. This is the first study to document both density dependence and inverse density dependence in parasite fecundity in the same host–helminth system.

In areas where malaria is endemic, helminthic infections, caused by intestinal or filarial parasites, commonly coexist with malaria in the same individual. This study investigates the course of Plasmodium berghei malaria infection in CBA/J mice inoculated with irradiated attenuated 3rd-stage larvae (L3) of Brugia pahangi. Peripheral eosinophil counts, serum IgE levels and cytokine production revealed that the filarial antigen induced T-helper type 2 (Th2) cell predominance in these mice, which protected them against the development of cerebral malaria. These mice significantly prolonged their survival, compared with the control mice after P. berghei infection. All of the mice not inoculated with irradiated L3 died within 12 days with acute neurological manifestations unrelated to the level of parasitaemia after infection of P. berghei. Conversely, most of the inoculated mice lived more than 3 weeks following infection with P. berghei, dying in the fourth week of severe anaemia and overwhelming parasitaemia. This suggests that Th2-dominant responses lead to the downregulation of susceptibility to murine cerebral malaria.
