Twenty-nine Leishmania infantum strains characterized by different host source, tropism and belonging to 6 zymodemes, were examined by restriction enzyme analysis of kinetoplast DNA (kDNA) using 15 endonucleases. The enzymes which produced only one fragment revealed full identity between all the strains examined, while those producing many bands gave different electrophoretic patterns. They were interpreted with the aid of numerical analyses (cluster and multifactorial analysis). The results show a cline of genetic variability among the strains, the highest similarity being observed between most of the viscerotropic strains isolated from man, dog, black rat and sandflies. The strain agents of human cutaneous leishmaniasis show a varying degree of genetic divergence from this group, which appears more evident when characters from isoenzymes are considered.

Two domestic guinea-pigs (Cavia porcellus), bought in Pinheiros, São Paulo State, Brazil, were taken by their owners a farm in the rural district of Capão Bonito, close to the Atlantic Forest, São Paulo, where they both developed tumour-like and ulcerating lesions on the ears. The causative agent was identified as Leishmania (L.) enriettii, based on biological characters and isoenzyme profiles. Sources of the parasite in wild mammals, and the possible sandfly vector species discussed.

The variability of larval excretion impedes the parasitological diagnosis of Strongyloides stercoralis in infected individuals. We assessed the number of larvae excreted per gram (LPG) stool in 219 samples from 38 infected individuals over 7 consecutive days before and in 470 samples from 44 persons for 21 consecutive days after ivermectin treatment (200 μg kg−1 BW). The diagnostic sensitivity of a single stool sample was about 75% for individuals with low-intensity infections (⩽1 LPG) and increased to 95% for those with high-intensity infections (⩾10 LPG). Doubling the number of samples examined per person increased sensitivity to more than 95%, even for low-intensity infections. There was no indication of a cyclic excretion of larvae. After treatment, all individuals stopped excreting larvae within 3 days. Larvae were not detected during any of the following 18 days (total 388 Baermann and 388 Koga Agar tests). Two stool samples, collected on consecutive days, are recommended in settings where low or heterogeneous infection intensities are likely. In this way, taking into account the possible biological variability in excretion, the efficacy of ivermectin treatment can be assessed as soon as 4 days after treatment.

Y strain parasites (Wellcome stock) were cloned as epimastigotes by limiting dilution and one clone, Wel Tryp A2 was used to infect cell cultures and irradiated mice. The fibrosarcoma line, M4, was susceptible to infection with Wel Tryp A2 trypomastigotes but, after the first round of intracellular infection, proliferation and differentiation, failed to support parasite growth in long-term continuous culture. In contrast, a muscle-derived cell line, S2, produced regular waves of extracellular trypomastigotes and amastigotes during 80 days of continuous culture reported here. These infected S2 cultures have remained in continuous culture for up to 18 months over a 5-year period and have yielded in excess of 108 parasites/20 ml culture at 4–6 day intervals. Extracellular amastigotes produced in these cultures have a close morphological, immunological and biochemical analogy to intracellular parasites derived from rodents, and have maintained an absolute dependence on host cells for their growth and proliferation. These amastigotes produced low-grade chronic infections in normal CBA/T6 mice but produced acute, fatal infections with high parasitaemia in mice of the same inbred strain given 900 rad. whole-body irradiation prior to infection. Although present findings relate to a single parasite clone, the parent stock from which the clone was derived showed similar growth characteristics when co-cultured with S2 cells.

This paper consists of two parts: (I) a complete account of the life-history and a description of early stages of Melinda cognata Meigen which I found here in Cambridge living as parasites on a common snail, Helicella virgata da Costa, and (II) a review of all observations made hitherto on the life-history of different flies feeding upon living or dead molluscs.

Sarcocystis species have been described in a wide variety of mammals, birds and reptiles throughout temperate and tropical countries of the world. Although early workers (Alexeieff, 1913; Wenyon, 1926) considered it impossible to differentiate species on the morphology, there are such clear-cut differences in the structure of the cyst wall, morphology of spores and other details, that there at present seems every justification for giving specific rank to these different parasites in their various hosts. Furthermore, with one or two exceptions, attempts to infect laboratory animals have met with little or no success and it would appear that at least some species of Sarcocystis may be highly host-specific.

The structure, method of feeding and relationship with the host cells of the tissue parasitizing stages of Histomonas meleagridis have been studied by means of light microscopy, cytochemistry and electron microscopy. The invasive stage is amoeboid, lacks flagella but has what appears to be remnants of the flagella apparatus, and feeds by phagocytosis. The vegetative stage is a round quiescent form. It apparently feeds by secreting proteolytic enzymes, which carry out extra-corporeal digestion of the host's tissues, and then taking up small particles of disrupted host cells by means of pinocytosis and probably by diffusion. The vegetative stage also contains remnants of flagella apparatus. Neither the invasive stage nor the vegetative stage contains mitochondria and succinic dehydrogenase is absent. It is concluded that there is no resistant stage.

The reaction of the host's tissues to the parasites has been briefly described.

The major surface glycoprotein of Leishmania, referred to as GP63, is a zinc metalloproteinase of 63000 Mr present on promastigotes and amastigotes from diverse species of Leishmania. GP63 shares several characteristics with the members of the matrix metalloproteinase family including degradation of at least one component of the extracellular matrix, location at the cell surface, requirement for Zn2+ for proteinase activity and inhibition of the proteinase activity by chelating agents and α2–macroglobulin. Site-directed mutagenesis of the cloned L. major GP63 genes was carried out to determine whether the proposed active site of Leishmania GP63 was homologous to those of other zinc metalloproteinases. The codon encoding the catalytic glutamic acid was modified to encode an aspartic acid and when expressed in COS–7 cells the resulting mutant GP63 had no demonstrable proteinase activity compared to wild type GP63. GP63 was predicted to be synthesized as a precursor protein containing a pro region at the NH2–terminus of GP63 implicated to be involved with the regulation of proteinase activity. As with many other proteinases, including matrix metalloproteinases, these enzymes are synthesized as latent proteinases that require activation for full proteinase activity. L. major recombinant GP63 (rGP63) has been produced in the baculovirus expression system where rGP63 was secreted as a latent proteinase. To study the activation of baculovirus rGP63, purified rGP63 was incubated with the mercurial compound, HgCl2, at concentrations previously shown to result in activation of other latent matrix degrading metalloproteinases and resulted in a significant enhancement of GP63 proteinase activity. The similarity of GP63 to the family of matrix-degrading proteinases suggests that the proteinase activity of GP63 maybe involved with the pathology of lesion formation in the mammalian host and may also be involved with the promastigote life stage in the sandfly vector. To study the functional role of GP63 proteinase, mutant strains of L. major, deficient in the expression of GP63, are currently being derived by targeted gene deletion. Using this strategy results have demonstrated the deletion of an entire L. major GP63 locus, containing in total six GP63 genes. Strategies to delete the second GP63 gene locus are developed and will determine whether deletion of both loci results in viable promastigotes. L. major strains deficient in the expression of GP63 may then be used to address the function of GP63 glycoprotein in the life cycle of Leishmania.

Plasmodium knowlesi is a simian malaria of primarily the macaque species of South East Asia. While it was known that human infections could be induced during the years of malariotherapy, naturally occurring P. knowlesi human infections were thought to be rare. However, in 2004, knowlesi infections became recognized as an important infection amongst human populations in Sarawak, Malaysian Borneo. Since then, it has become recognized as a disease affecting people living and visiting endemic areas across South East Asia. Over the last 12 years, clinical studies have improved our understanding of this potentially fatal disease. In this review article the current literature is reviewed to give a comprehensive description of the disease and treatment.

Genomic DNAs from 3 oocyst isolates of Hammondia sp. from foxes (Vulpes vulpes and V. lagopus) and 1 oocyst isolate of Hammondia heydorni from a dog, were examined by PCR and sequence analysis of 6 loci in order to determine whether the isolates were conspecific. Consistent genetic differences were found between the fox and dog isolates, respectively, at the ITS-2 region, the lsu rRNA gene, the alpha tubulin gene and the HSP70 gene, but not at the ssu rRNA gene or ITS-1 locus. Infection experiments established that dogs were unsuitable as definitive hosts for Hammondia sp. of foxes; hence this species is regarded as separate from H. heydorni of wolf-like canids, but probably identical with Isospora triffittae (syn. Isospora triffitti) previously reported from foxes. This species has therefore been named Hammondia triffittae n. comb. Reindeer, moose, sheep, goats, foxes and rabbits may act as intermediate hosts for H. triffittae. Muscle tissues of inoculated intermediate hosts were infectious for foxes from 16 days post-infection. Oocysts of H. triffittae were subspherical, averaging 12·5×10·9 μm in size. The lsu rRNA gene and the alpha tubulin gene seem to be suitable genetic markers for differentiating between H. triffittae and H. heydorni.

It was suggested that the unlimited proliferative capacity of the Echinococcus multilocularis metacestode may be related to overproduction of the 14-3-3 protein. As is known, the proliferative capacities of E. granulosus and E. multilocularis metacestodes are very different. By comparing the expression levels of the 14-3-3 gene between in vitro-obtained E. granulosus and E. multilocularis metacestodes, we were able to provide experimental evidence of the potential relation between 14-3-3 over-expression and tumour-like growth in E. multilocularis metacestodes. RT–PCR and Northern blot experiments indicated that 14-3-3 expression level is about 4-fold higher in the E. multilocularis metacestode. This differential expression was confirmed both by immunoblotting and immunocytochemistry experiments, which allowed detection of the protein in the cyst wall from E. multilocularis but not in the cyst wall from E. granulosus. The alignment of the Echinococcus 14-3-3 cDNA sequence with known 14-3-3 isoforms from other organisms, grouped the parasite sequence into the tumour growth-related isoforms.Nucleotide sequence data reported in this paper are available in the EMBL, GenbankTM and DDJB databases under the acession numbers AF207904 and AF207905. The known relation between over-expression of some 14-3-3 isoforms and tumour-related processes, together with the present results, suggest that the Echinococcus 14-3-3 protein could be one of the molecules responsible for the differences between E. granulosus and E. multilocularis metacestode growth behaviour.

Besnoitia besnoiti is an obligate intracellular apicomplexan protozoan parasite, which causes bovine besnoitiosis. Recently increased emergence within Europe was responsible for significant economic losses in the cattle industry due to the significant reduction of productivity. However, still limited knowledge exists on interactions between B. besnoiti and host innate immune system. Here, B. besnoiti bradyzoites were successfully isolated from tissue cysts located in skin biopsies of a naturally infected animal, and we aimed to investigate for the first time reactions of polymorphonuclear neutrophils (PMN) exposed to these vital bradyzoites. Freshly isolated bovine PMN were confronted to B. besnoiti bradyzoites. Scanning electron microscopy (s.e.m.)- and immunofluorescence microscopy-analyses demonstrated fine extracellular networks released by exposed bovine PMN resembling suicidal NETosis. Classical NETosis components were confirmed via co-localization of extracellular DNA decorated with histone 3 (H3) and neutrophil elastase (NE). Live cell imaging by 3D holotomographic microscopy (Nanolive®) unveiled rapid vital NETosis against this parasite. A significant increase of autophagosomes visualized by specific-LC3B antibodies and confocal microscopy was observed in B. besnoiti-stimulated bovine PMN when compared to non-stimulated group. As such, a significant positive correlation (r = 0.37; P = 0.042) was found between B. besnoiti-triggered suicidal NETosis and autophagy. These findings suggest that vital- as well as suicidal-NETosis might play a role in early innate host defence mechanisms against released B. besnoiti bradyzoites from tissue cysts, and possibly hampering further parasitic replication. Our data generate first hints on autophagy being associated with B. besnoiti bradyzoite-induced suicidal NETosis and highlighting for first time occurrence of parasite-mediated vital NETosis.

Antibodies against the retrovirus envelope glycoprotien (gp70) of mouse xenotropic retrovirus, BALB virus 2 (Bv2) reacted with the adult worms of Schistosoma japonicum and S. mansoni. This reaction was completely inhibited after adsorption of the antibodies with virions of retrovirus. The reactive schistosome antigen was located in the subtegumental layer of the adult male fluke and in the vitelline gland of the adult female of S. japonicum and S. mansoni. Proteins extracted from both parasites were examined by immunoblot analysis. Anti-Bv2 gp70 antiserum reacted with those proteins from both schistosomes and the band patterns were different among sexes and species. Southern hybridization of the DNA extracted from adults of S. japonicum and S. mansoni demonstrated the presence of sequences homologous to the env gene of mouse ecotropic and xenotropic retroviruses. DNA sequences homologous to the gag and pol regions of the ecotropic murine leukaemia virus were also detected in the DNAs of schistosomes

Hymenolepis microstoma was found to live for at least 728 days in a mouse, but an initial infection of 10 worms per mosue decreased exponentially for approximately the first 110 days to a level of 3–5 worms, after which little further loss occurred.

A primary infection of six H. microstoma, expelled with anthelmintic on day 21, strongly protected against a single worm H. diminuta challenge; worms in 80 ‘immunized’ mice weighed less than 5% of the weight of worms from 80 control mice. However, a primary (5-worm) H. diminuta infection led only to a small, though statistically significant, decrease in weight of the 3-worm H. microstoma challenge.

The results are interpreted as indicating that H. microstoma evokes a strong immunological response which cross-reacts with H. diminuta. The failure of H. diminutato protect strongly against a 3-worm challenge by H. microstoma suggests, as does the long survival of a primary infection when it is reduced to approximately 4 worms, that H. microstoma survives in low infection because it is able to avoid the immune response. This could be due to antigen camounflage, ability to repair immune damage or to the low permeability of the bile duct wall to components of the immune response. The worm loss which occurs from heavy infection until only 3–5 worms remain may be physiologically rather than immunologiacally mediated.

Seven dipterous parasites have been reared and identified from woodlice: Plesina maculata Fall., Melanophora roralis L., Phyto melanocephala Meig., Styloneuria discrepans Pand., Frauenfeldia rubricosa Meig., Cyrillia angustifrons Rond., and Rhinophora lepida Meig. The biology and larval forms of all of these except R. lepida are described and figured, together with those of two undetermined species, “A” and “B.”

Chagas' disease is one of the few functional gastrointestinal disorders for which a causative agent has been identified. However, some pathological aspects of the chagasic megasyndromes are still incompletely understood. Chagasic megacolon is characterized by an inflammatory process, organ dilatation and neuronal reduction in both plexuses of the enteric nervous system (ENS). Although some studies on the ENS in Chagas' disease have been performed, the process of neuronal destruction and neuronal regeneration still remains unclear. Our hypothesis is that the regeneration process of the ENS may be involved with the mechanisms that prevent or retard organ dilatation and chagasic megacolon development. For that reason, we evaluated the neuronal regeneration with the marker GAP-43 in the colon's neuronal plexuses from chagasic patients with megacolon, and from non-infected individuals. Visual examination and quantitative analysis revealed an increased neuronal regeneration process in the dilated portion from chagasic patients when compared with the non-dilated portion and with non-infected individuals. We believe that this increased regeneration can be interpreted as an accentuated neuronal plasticity that may be a response of the ENS to avoid megacolon propagation to the entire organ and maintain the colon functional innervation.

The primate malaria Plasmodium knowlesi has a long-standing history as an experimental malaria model. Studies using this model parasite in combination with its various natural and experimental non-human primate hosts have led to important advances in vaccine development and in our understanding of malaria invasion, immunology and parasite–host interactions. The adaptation to long-term in vitro continuous blood stage culture in rhesus monkey, Macaca fascicularis and human red blood cells, as well as the development of various transfection methodologies has resulted in a highly versatile experimental malaria model, further increasing the potential of what was already a very powerful model. The growing evidence that P. knowlesi is an important human zoonosis in South-East Asia has added relevance to former and future studies of this parasite species.

Human helminthiases are common in China, especially in rural areas where sanitation conditions are poor. Soil-transmitted helminths (STHs) are predominantly found in the southern provinces. Schistosoma japonicum is also endemic to southern China. Here we review the prevalence of helminth infections and polyparasitism in China, and discuss the interactions between helminth parasites in the co-infected host. It is clear that STHs are more prevalent in rural China than previously suggested emphasizing the need for systematic control of STHs. Further, the need for improved sanitation and hygiene conditions to prevent parasite transmission is highlighted. We provide supporting evidence for human genetic susceptibility to both single helminth infection and polyparasitism, and suggest that susceptibility to helminths infections may not be independent of one or the other. We demonstrate an association between single nucleotide polymorphism (SNP) variants in IL-5 and symptomatic S. japonicum infection and discuss the potential role of IL-5 in other helminth infections. Fundamental to disease and morbidity control is adequate and effective diagnosis and surveillance of disease. We discuss the role of sICAM-1 and TNFR-I and -II as candidate markers for schistosome-induced hepatomegaly and fibrosis, and their potential for assessing disease stage and progression in schistosomiasis.

To test the hypothesis that the relatively high developmental temperature threshold of the parasitic nematode Elaphostrongylus rangiferi in the intermediate snail host is an adaptation to minimize larval mortality during winter, an experiment was set up in which snails of the species Arianta arbustorum were experimentally infected with the parasite. The snails were divided into 3 groups known to contain 1st, 2nd or 3rd-stage larvae, and incubated at 3 °C for an experimental period of 18 weeks. First-stage larvae showed a significantly higher survival rate within snails than 2nd or 3rd-stage larvae. We also found that snails carrying 1st-stage larvae survived better than snails with other larval stages. It is concluded that if the nematode has started development before the hibernation, this has a real and significant effect on the risk of dying. The high developmental threshold is therefore likely to be an adaptation to reduce the chance of hibernating as developing larvae during long periods of low temperatures.
