Blattisocius keegani (Fox) is a predatory mite in the family Ascidae (Acari), noted for potential biological control of Coleopteran stored product pests. Performance of B. keegani on eggs of navel orangeworm, Amyelois transitella Walker (Lepidoptera: Pyralidae), was investigated. Mites completed development from egg to adult in 9.2±0.22 days at 25°C and 50–60% relative humidity, and in 6.33±0.29 days at 32.2°C, 30% relative humidity. Mites provisioned with three or five eggs consumed a median of 1.25 to 1.5 eggs, with a maximum of three eggs consumed over 24 h. Regression analyses indicated egg-laying by B. keegani was significantly correlated with the number of A. transitella eggs consumed, and female mites laid an average of 5.82±0.44 eggs over 72 h. Blattisocius keegani, developed on fresh and frozen eggs, laid significantly more eggs when provided with fresh eggs (F3,26=6.16, P=0.0026) and were able to develop on frozen Ephestia kuehniella Zeller (Lepidoptera: Pyralidae) eggs as an alternative host. Mites were equally fecund when fed eggs stored at 0° or −20°C. Provisioning of adult moth bodies in addition to egg prey items increased mite fecundity, although it was demonstrated that B. keegani are phoretic on adult moths as well. The results are the first experimental evidence of B. keegani as a predator of Lepidopteran eggs, as a phoretic species, and of their potential for biological control of navel orangeworm.

Molecular approaches for identifying aquatic macroinvertebrate species are increasingly being used but there is ongoing debate about the number of DNA markers needed to differentiate species accurately. Here, we use two mitochondrial genes (cytochrome oxidase I, cytochrome b) and a nuclear gene (carbamoylphosphate synthetase) to differentiate species variation within the taxonomically challenging chironomid genus Procladius from southern Australia, a genus which is important for pollution monitoring. The mitochondrial genes indicated cryptic species that were subsequently linked to morphological variation at the larval and pupal stage. Two species previously described based on morphological criteria were linked to molecular markers, and there was evidence for additional cryptic species. Each genetic marker provided different information, highlighting the importance of considering multiple genes when dissecting taxonomically difficult groups, particularly those used in pollution monitoring.

The invasive larger grain borer Prostephanus truncatus (Horn) is the most important pest of farm-stored maize in Africa. It was introduced into the continent from Mesoamerica in the late 1970s and by 2008 had spread to at least 18 countries. Classical biological control using two populations of the predator Teretrius nigrescens Lewis achieved long-term and cost effective control in warm-humid areas, but not in cool and hot-dry zones. The present study investigated the phylogenetic relationships between geographical populations of the predator. Ten populations of T. nigrescens were studied using randomly amplified polymorphic DNA polymerase chain reaction (RAPD-PCR), sequence analysis of mitochondrial Cytochrme oxydase 1 (mtCOI) gene and ribosomal internally transcribed spacers (ITS) 1, 5.8S and ITS2. The mtCOI variation revealed two clades associated with geographical regions in Central America. It also reveals a significant isolation by distance between populations and considerable genetic shifts in laboratory rearing. RAPD-PCR did not reveal any potential SCAR diagnostic markers. The ITS variation mainly involved insertions and deletions of simple sequence repeats even within individuals. This study reveals the existence of two different mitochondrial lineages of the predator, associated with the geographical origin of populations distinguishable by fixed mutations on the mtCOI gene. The populations of T. nigrescens released in Africa belonged to two different clades from Meso America, namely south (released in West Africa) and north (released in eastern Africa). However, more polymorphic markers are required to clarify the observations in demographic time scales.

The ‘paddy paradox’, the occurrence of large populations of vectors but low amounts of malaria transmission where irrigated rice is grown, was investigated in a village in Ghana where M form Anopheles gambiae are common. Peridomestic and indoor host-seeking mosquitoes were collected in tent traps and light traps over 21 consecutive nights at the start of the rainy season in June 2009 when the population increased exponentially from less than 100 per night to over 1000. Infection rates in the overall mosquito population were 0.3% and in the estimated parous population were 1.9%. Numbers of An. gambiae in the tent trap peaked between midnight and 02:40 am. The majority of insects were taking their first blood meal, as virgins or shortly after mating. More than expected were collected in the light trap during a rainstorm at the start of the rains but overall numbers were not affected. Fewer than expected were collected after a subsequent storm. Recruitment to the adult population decreased over the following days. It is hypothesised that the ‘paddy paradox’ is due to young pre-gravid insects dispersing more widely than gravid ones, not necessarily to low survival in the mosquito.

1-Octen-3-ol is a kairomone for many haematophagous insects including mosquitoes. Numerous studies have examined the effects of racemic 1-octen-3-ol; however, few studies have investigated the role of individual enantiomers in relation to mosquito attraction. In the present study, we investigated the behavioural and electrophysiological responses of two mosquito species, Aedes aegypti and Culex quinquefasciatus, to individual enantiomers and mixtures of 1-octen-3-ol, employing a laboratory Y-tube olfactometer and single sensillum recordings. The olfactory receptor neurons of both Ae. aegypti and Cx. quinquefasciatus had a significantly higher response to the (R)-1-octen-3-ol enantiomer compared to the (S)-1-octen-3-ol enantiomer at 10−9 g μl−1 to 10−6 g μl−1. Behaviourally, Ae. aegypti was more responsive to the (R)-1-octen-3-ol enantiomer, showing an increase in flight activity and relative attraction compared to Cx. quinquefasciatus. The (R)-1-octen-3-ol enantiomer caused an increase in activation for Cx. quinquefasciatus. However, the most notable effect was from an (R:S)-1-octen-3-ol mixture (84:16) that caused significantly more mosquitoes to sustain their flight and reach the capture chambers (demonstrated by a reduced non-sustained flight activity), suggesting that it may have a behaviourally excitatory effect. For Cx. quinquefasciatus, a reduced relative attraction response was also observed for all treatments containing the (R)-1-octen-3-ol enantiomer, either on its own or as part of a mixture, but not with the (S)-1-octen-3-ol enantiomer. This is the first time enantiomeric selectivity has been shown for Ae. aegypti using electrophysiology in vivo. The implications of these results for exploitation in mosquito traps are discussed.

The analysis of microsatellite loci has allowed significant advances in evolutionary biology and pest management. However, until very recently, the potential benefits have been compromised by the high costs of developing these neutral markers. High-throughput sequencing provides a solution to this problem. We describe the development of 13 microsatellite markers for the eusocial ambrosia beetle, Austroplatypus incompertus, a significant pest of forests in southeast Australia. The frequency of microsatellite repeats in the genome of A. incompertus was determined to be low, and previous attempts at microsatellite isolation using a traditional genomic library were problematic. Here, we utilised two protocols, microsatellite-enriched genomic library construction and high-throughput 454 sequencing and characterised 13 loci which were polymorphic among 32 individuals. Numbers of alleles per locus ranged from 2 to 17, and observed and expected heterozygosities from 0.344 to 0.767 and from 0.507 to 0.860, respectively. These microsatellites have the resolution required to analyse fine-scale colony and population genetic structure. Our work demonstrates the utility of next-generation 454 sequencing as a method for rapid and cost-effective acquisition of microsatellites where other techniques have failed, or for taxa where marker development has historically been both complicated and expensive.

The diversity and abundance of wild bees ensures the delivery of pollination services and the maintenance of ecosystem diversity. As previous studies carried out in Central Europe and the US have shown, bee diversity and abundance is influenced by the structure and the composition of the surrounding landscape. Comparable studies have so far not been carried out in the Mediterranean region. The present study examines the influence of Mediterranean landscape context on the diversity and abundance of wild bees. To do this, we sampled bees in 13 sites in olive groves on Lesvos Island, Greece. Bees were assigned to five categories consisting of three body size groups (small, medium and large bees), the single most abundant bee species (Lasioglossum marginatum) and all species combined. The influence of the landscape context on bee abundance and species richness was assessed at five radii (250, 500, 750, 1000 and 1250 m) from the centre of each site. We found that the abundance within bee groups was influenced differently by different landscape parameters and land covers, whereas species richness was unaffected. Generally, smaller bees’ abundance was impacted by landscape parameters at smaller scales and larger bees at larger scales. The land cover that influenced bee abundance positively was olive grove, while phrygana, conifer forest, broad-leaved forest, cultivated land, rock, urban areas and sea had mostly negative or no impact. We stress the need for a holistic approach, including all land covers, when assessing the effects of landscape context on bee diversity and abundance in the Mediterranean.

The effect of four temperatures (18, 20, 25 and 30°C) on pupa development and sexual maturity of Anastrepha obliqua adults was investigated under laboratory conditions. The results showed that the duration of the pupal stage decreased with an increase in temperature (29, 25, 13 and 12 days, respectively), and maintaining the pupae at 18°C and 20°C results in a low percentage of pupation, pupa weight loss and lesser flying ability. However, it significantly favored sexual behavior, a higher proportion of sexual calls and matings. While enhanced pupa development was observed at a temperature of 30°C, adults had low sexual efficiency, as well as a lower proportion of calls and matings. Gas chromatography-mass spectrometry (GC-MS) analysis of male volatiles showed that the amount of (Z,E)-α-farnesene did not vary among males from pupae reared at different temperatures; however, less (E,E)-α-farnesene was emitted by males obtain from pupa reared at 30°C. Male flies kept at 30°C during their larval stage had more (Z)-3-nonenol and, also, an unknown compound was detected. The fecundity of the females was higher at low temperatures. Regarding fertility, no significant differences were found between temperatures. The optimal temperature on pupa development was 25°C when males displayed ideal attributes for rearing purposes.

We, herein, report evidence that wing fanning by the aphid parasitoid Aphidius ervi Haliday (Hymenoptera: Braconidae) constitutes a courtship song. Complete removal of the forewings or only the distal half of them reduced male copulation success in comparison to intact males. Males that achieved copulation within the observation period produced wing fanning at a higher rate than males that did not copulate. Playback of wing fanning sound altered the behaviour of virgin females, increasing the time they devoted to grooming, as compared with subjects that were exposed to silence or white noise. Our findings contribute to the understanding of the acoustic dimension of the sensory modalities employed by this aphid parasitoid in sexual signalling.

Solenopsis invicta Buren (red imported fire ant) are invasive pests that have the capability of major destructive impacts on lifestyle, ecology and economy. Control of this species is dependent, in part, upon ability to estimate the potential spread from newly discovered nests. The potential for spread and the spread characteristics differ between monogyne and polygyne social forms. Prior to this study, differentiation of the two social forms in laboratory test samples commonly used a method involving restriction endonuclease digestion of an amplified Gp-9 fragment. Success of this assay is limited by the quality of DNA, which in the field-collected insects may be affected by temporary storage in unfavourable conditions. Here, we describe an alternative and highly objective assay based upon a high resolution melt technique following preamplification of a significantly shorter Gp-9 fragment than that required for restriction endonuclease digestion. We demonstrate the application of this assay to a S. invicta incursion in Queensland, Australia, using field samples from which DNA may be partially degraded. The reductions in hands-on requirements and overall duration of the assay underpin its suitability for high-throughput testing.

Most of the current knowledge about African tephritids originates from studies performed in agricultural areas, while information about their distribution in pristine or moderately disturbed environments is extremely scarce. This study aims at (i) describing levels of spatial variability of frugivorous tephritids in tropical forests and small rural villages of the Congo River basin and (ii) verifying if human-mediated activities, such as small-scale agriculture and trade, can affect their distribution patterns. Four locations were sampled along a 250 km stretch of the Congo River. At each location, pristine and disturbed habitats (i.e. tropical forests and small rural villages, respectively) were sampled, with three replicate sites in each combination of habitat and location. Sampling with modified McPhail traps baited with four different attractants yielded 819 tephritid specimens of 29 species from seven genera (Bactrocera, Carpophthoromyia, Ceratitis, Dacus, Celidodacus, Perilampsis, Trirhithrum). The three most abundant species sampled (Dacus bivittatus, D. punctatifrons, Bactrocera invadens) showed significant variations in abundance across locations and sites and accounted for 98.29% of the overall dissimilarity between habitats. Assemblages differed among locations and sites while they showed significant differences between pristine and disturbed habitats in two out of the four locations. This study shows that frugivorous tephritids in central Congo have remarkably patchy distributions with differences among locations and sites representing the main source of variability. Our data show that, in rural villages of central Democratic Republic of Congo, human activities, such as small-scale agriculture and local commerce, are not always sufficient to promote differences between the tephritid assemblages of villages and those of the surrounding tropical forests.

Necrophagous flies that colonize human and animal corpses are extremely efficient at locating and utilizing carrion. Adult flies deposit eggs or larvae on the ephemeral food resource, which signals the beginning of intense inter- and intra-species competition. Within a short period of time after egg hatch, large larval aggregations or maggot masses form. A period of intense larval feeding ensues that will culminate with consumption/decomposition of all soft tissues associated with the corpse. Perhaps the most distinctive feature of these feeding aggregations is heat production; that is, the capacity to generate internal heat that can exceed ambient temperatures by 30°C or more. While observations of maggot mass formation and heat generation have been described in the research literature for more than 50 years, our understanding of maggot masses, particularly the physiological ecology of the aggregations as a whole, is rudimentary. In this review, an examination of what is known about the formation of maggot masses is presented, as well as arguments for the physiological benefits and limitations of developing in feeding aggregations that, at times, can represent regions of intense competition, overcrowded conditions, or a microclimate with elevated temperatures approaching or exceeding proteotoxic stress levels.

In this study, silkworm moth (Bombyx mori L.) larvae were regarded as an animal protein source for astronauts in the bioregenerative life support system during long-term deep space exploration in the future. They were fed with mulberry and stem lettuce leaves during the first three instars and the last two instars, respectively. In addition, this kind of environmental approach, which utilised inedible biomass of plants to produce animal protein of high quality, can likewise be applied terrestrially to provide food for people living in extreme environments and/or impoverished agro-ecosystems, such as in polar regions, isolated military bases, ships, submarines, etc.

Respiration characteristics of the larvae during development under two main physiological conditions, namely eating and not-eating of leaves, were studied. Nutrient compositions of silkworm powder (SP), ground and freeze-dried silkworms on the 3rd day of the 5th instar larvae, including protein, fat, vitamins, minerals and fatty acids, were measured using international standard methods.

Silkworms’ respiration rates, measured when larvae were eating mulberry leaves, were higher than those of similar larvae that hadn't eaten such leaves. There was a significant difference between silkworms fed on mulberry leaves and those fed on stem lettuce in the 4th and 5th instars (P<0.01). Amounts of CO2 exhaled by the silkworms under the two physiological regimes differed from each other (P<0.01). There was also a significant difference between the amount of O2 inhaled when the insects were under the two physiological statuses (P<0.01). Moreover, silkworms’ respiration quotient under the eating regime was larger than when under the not-eating regime. The SP was found to be rich in protein and amino acids in total; 12 essential vitamins, nine minerals and twelve fatty acids were detected. Moreover, 359 kcal could be generated per 100 gram of SP (dry weight).
