Adult Fasciola gigantica are leaf-shaped with tapered anterior and posterior ends and measure about 35 mm in length and 15 mm in width across the mid section. Under the scanning electron microscope its surface appears rough due to the presence of numerous spines and surface foldings. Both oral and ventral suckers have thick rims covered with transverse folds and appear spineless. On the anterior part of the ventral surface of the body, the spines are small and closely-spaced. Each spine has a serrated edge with 16 to 20 sharp points, and measures about 20 μm in width and 30 μm in height. In the mid-region the spines increase in size (up to 54 μm in width and 58 μm in height) and number, especially towards the lateral aspect of the body. Towards the posterior end the spines progressively decrease in both size and number. The tegumental surface between the spines appears highly corrugated with transverse folds alternating with grooves. At higher magnifications the surface of each fold is further increased with a meshwork of small ridges separated by variable-sized pits or slits. There are three types of sensory papillae on the surface. Types 1 and 2 are bulbous, measuring 4–6 μm in diameter at the base with nipple-like tips, and the type 2 also have short cilia. Type 3 papillae are also bulbous and of similar size but with a smooth surface. These sensory papillae usually occur in clusters, each having between 2 and 15 units depending on the region of the body. Clusters of papillae on the lateral aspect (usually types 1 and 2) and around the suckers (type 3) tend to be more numerous and larger in size. The dorsal side of the body exhibits similar surface features, but the spines and papillae appear less numerous and are smaller. Corrugation and invaginations of the surface are also less extensive than on the ventral side of the body.

Collections of flounder, Platichthys flesus, at two sites on the tidal River Thames in 1994 and 1995 have, for the first time, revealed the sympatric occurrence of the freshwater and marine/estuarine strains of the acanthocephalan parasite Pomphorhynchus laevis. This natural co-occurrence of the strains has been employed to compare infection levels and a range of parasite attributes of the two strains under conditions of sympatry. At both Lots Road (Chelsea) and Tilbury the marine/estuarine strain was present at far higher infection levels than the freshwater form. In a detailed comparison of worms from Tilbury flounder, a range of differences was revealed between the two strains. In single strain infections in individual fish, freshwater and marine/estuarine worms had distinct but overlapping gut microhabitat use patterns, with the former having a central intestinal bias and the latter a bias for the posterior region of the gut. In mixed strain infections, niche contraction occurred so that no overlap occurred. Freshwater worms were larger and had more eggs, more ovarian balls, and a higher percentage of fully developed eggs than the marine/estuarine worms. These differences are thought to reflect intrinsic, presumably genetically determined, differences between the two strains as they occurred in the same fish host species collected at the same place and time. Apparent differences in strain reproductive potential in flounder in the tidal Thames are discussed in the context of previous studies and the intermediate host segment of the parasite life cycle.

Histopathology of the lung and total IgE in serum were compared in toxocariasis and allergic asthma murine models using BALB/c and C57BL/6 mice. Infection with Toxocara canis resulted in both strains of mice in marked histological changes and increased levels of total serum IgE. The ovalbumin (OVA) sensitization/challenge treatment for the induction of allergic asthma resulted in similar histological changes in BALB/c and, to a less extent, in C57BL/6 mice. Serum IgE levels of OVA-treated C57BL/6 mice were low. Histological changes observed included perivascular infiltration with eosinophils and mononuclear cells, peribronchiolitis, alveolitis and mucus production. Although these changes in addition to increased IgE production did occur in T. canis-infected C57BL/6 mice they were more pronounced in BALB/c mice. Thus, BALB/c mice appear to be the most appropriate strain of mice to perform studies on the possible connection between infection with T. canis and allergic asthma.

Species co-occurrences and interspecific associations between intensity of infection were studied in helminth communities of three populations of brown trout, Salmo trutta, from northern Italy. Of the eight helminth species, only four were common enough to be included in the analyses: Pomphorhynchus laevis, Acanthocephalus anguillae, Echinorhynchus truttae and Cyathocephalus truncatus. The observed frequencies of co-occurrences of the different species, based on presence/absence data, did not differ from those predicted by a null model derived from prevalence data. However, the intensity of infection (number of individuals per fish) of different helminth species were generally, but not always significantly, negatively correlated in tests of pairwise associations. Variation in fish sizes and its effect on infection levels, and whether or not two helminth species used the same or different intermediate hosts, had no influence on these findings. Of the few significant negative associations found between pairs of helminth species, none was found in more than one fish population. This suggests that interspecific associations may be condition-dependent: even in apparently similar localities, the same combinations of helminth species show different associations. Without evidence of replicability, it is almost impossible to conclude to the consistent role of competition between any pair of helminth species in the field.

Enzyme-linked immunosorbent assays (ELISAs) which detect specific antibodies to fluke cysteine proteinases have provided good sensitivity and specificity for the immunodiagnosis of trematode diseases. To detect specific antibodies without the need for purified proteinase antigens, an immunocapture assay using Protein A was applied for the immunodiagnosis of paragonimiasis and fascioliasis. ELISA plate wells were coated with Protein A, incubated with diluted patient sera, then incubated with a preparation containing fluke cysteine proteinases, excretory–secretory (ES) products of adult Paragonimus westermani or Fasciola sp. The activity of fluke cysteine proteinases bound on the wells was measured by adding fluorogenic peptidyl substrate, Z-Phe-Arg-MCA or Boc-Val-Leu-Lys-MCA. This assay detected specific immunoglobulin G to cysteine proteinases of P. westermani and Fasciola sp. by measuring proteinase activity on the plate wells. Patient sera showed significant high values of proteinase activity when the wells were treated with the respective homologous ES products, whereas the sera had low values after treatment with the heterologous ES products. The sera of patients with other parasitoses and uninfected healthy individuals also showed low values after treatment with the above fluke ES products. Thus, Protein A immunocapture assay, which detected IgG specific for fluke cysteine proteinases, provided a high sensitivity and specificity for immunodiagnosis of paragonimiasis and fascioliasis.

Oesophagostomum bifurcum larvae, cultured from human stools collected in northern Ghana, were used to establish experimental infections in monkeys. A patent infection was established in a rhesus monkey (Macaca mulatta) and this infection was used to generate larvae to inoculate additional monkeys. In all, 17 animals were inoculated. Thirteen of 15 animals developed antibodies to the infection between 19 and 62 days post inoculation (PI); two animals had a positive response before inoculation. Four of ten animals developed patent infections between 88 and 134 days and passed eggs in the faeces. Egg shedding was consistent in only one animal, but at low levels of one or two eggs per 2 mg direct smear, and extended over a 400 day period. In the other three animals, egg shedding was sporadic and of only 2–4 weeks duration. In seven animals necropsied between 19 and 22 days PI, one to 17 early fourth-stage larvae were recovered from nodules in the bowel wall; in an eighth animal examined at 314 days, six immature adult worms (early fifth stage) were recovered from nodules in the bowel wall. The morphological features and growth of these recovered larvae are described. Three animals were inoculated with larvae that had been dried for one week at 28°C; two animals began shedding eggs at 128 and 134 days PI, respectively. The present results suggest that the parasite obtained from humans is poorly adapted to lower primate hosts, and supports the concept that Oesophagostomum bifurcum found in humans and monkeys in the same geographical region of northern Ghana and Togo are distinct and that the infections in humans are not likely to represent zoonotic infections acquired from monkeys.

Benzimidazole carbamates (mebendazole, albendazole and fenbendazole) are the most commonly used anthelmintic drugs for the treatment of larval toxocariasis (Toxocara canis) in paratenic hosts. However, the bioavailability of these drugs for tissues is very low due to their extremely low solubility, resulting in the administration of relatively high doses over a long period. To overcome this problem, neutral, negatively or positively charged and stabilized liposome drug carriers were examined in the chronic phase of T. canis infections in mice each orally inoculated with 1000 eggs. Moreover, liposomized albendazole and fenbendazole were co-administered with liposomized immunomodulator glucan. The highest efficacy of both drugs, evaluated 4 weeks after treatment, was recorded after their subcutaneous administration (ten doses of 25 mg kg-1) in stabilized liposomes and intramuscular co-administration of liposomized glucan (two doses of 5 mg kg-1). Fenbendazole was more effective in muscles (91.5%) whereas albendazole was more effective in the brain (92.2%). Liposomes with incorporated benzimidazole carbamate anthelmintics provide sustained drug-release reservoirs and can considerably enhance drug efficacy. Moreover, despite suppression by T. canis antigens, stimulation of the immune system by the immunomodulator glucan potentiates the effects of these antiparasitic drugs.

Over the last century in the uplands of Scotland, the extent of heather moorland which supports high densities of mountain hares Lepus timidus has diminished and has gradually been replaced by large-scale commercial forestry plantations or expanding natural woodlands. The potential impact of such a change in land use on host–parasite interactions was investigated by comparing the intensity and prevalence of infection of hares by parasites in two separate habitats: a large hare-fenced young forestry plantation and the adjacent open moorland. Carcasses were collected in November 1990 from within both habitats and after the woodland had been enclosed for nine months. Age, sex, fatness (kidney fat index) and degree of infection of hares were noted. Two parasites were recorded: the nematode Trichostrongylus retortaeformis and the cestode Mosgovoyia pectinata. Clear differences in the intensity of infection of adults occupying the different habitats had occurred in the nine months since woodland enclosure. Adult mountain hares in the woodland had levels of infections approaching four times that observed in hares occupying the open moorland and although not significant, the prevalence of infection was greater in hosts inhabiting the woodland than the open moorland. It is suggested that the parasite–host relationship differs between the two habitats and as heather-dominated moorland landscapes become more fragmented with the increasing establishment of woodlands, the impact of parasites on the life history strategies of mountain hares needs to be reconsidered.

A protoscoleces/vesicles in vitro maintenance test with assessment of viability by eosin exclusion was used to evaluate the quantitative and qualitative activities of isoprinosine, its active component inosine and the dipeptide methylester L-Phe-Phe-OMe on isolated protoscoleces of Echinococcus multilocularis for 24 and 48 h. Isoprinosine and inosine showed dose- and time-dependent activity, the latter displaying a more rapid effect than the former. A high activity was shown with L-Phe-Phe-OMe, when compared to praziquantel. Ultrastructural alterations were much more striking with L-Phe-Phe-OMe, with an effect similar to that of praziquantel, whereas the chemotherapeutic activity of inosine and isoprinosine appeared to be directed against a metabolic target, with a lethal effect not immediately visible at the ultrastructural level. Thus, the previously reported in vivo activities of these drugs result largely from a direct effect on the parasite.

The high prevalence of Toxocara in cats, dogs and foxes results in the contamination of soil with infective eggs of Toxocara spp. which are found in soil samples from public and private places worldwide. In Poland the most contaminated areas were city backyards where 38–53% of soil samples were positive, especially in the spring. Human exposure to infection with Toxocara spp. was proportional to the prevalence of eggs in the samples examined but soil texture was not a critical factor in the degree of soil contamination. Eggs of Toxocara spp. placed on the ground penetrated a sand soil profile slowly. Their presence in the superficial layer of soils and the role of earthworms are instrumental in the dissemination of Toxocara eggs.

A preliminary purification has been carried out by continuous elution electrophoresis of a 49.5 kDa protease of crude extracts from Dicrocoelium dendriticum eggs. The enzyme showed a high capacity to degrade the collagen derivative azocoll at acidic pH. Although it is necessary to carry out further experiments to confirm any physiological role, this protease could be implicated in penetration mechanisms.

Effects of the anthelmintics, pyrantel and levamisole, on egg development of Angiostrongylus costaricensis were studied in vitro. After 7 days, about 80% of eggs developed to first-stage larvae in Ham's F-12 medium with 10% foetal calf serum under 5% CO2. Significant inhibition of development was caused by pyrantel (10-9–10-8 g ml-1) and levamisole (10-9–10-8 g ml-1) (Mann-Whitney U-test; P<0.05), and none of the eggs developed to first-stage larvae in higher concentrations of these anthelmintics (10-7 g ml-1). Furthermore, incubation with these drugs at 10-8 g ml-1 for at least 3 h or at 10-4 g ml-1 for 1 h caused irreversible effects on egg development.

A dichloromethane extract of the seeds of Millettia thonningii (Leguminosae) which contains a mixture of isoflavonoids (predominately robustic acid, alpinumisoflavone and dimethylalpinumisoflavone) is known to have larvicidal activity towards the miracidia and cercariae of schistosomes and to possess significant molluscicidal activity. The present investigation has assayed the effects of this extract on the electron transport systems of isolated rat liver mitochondria. The extract was found to inhibit mitochondrial electron transport at Site I (NADH dehydrogenase) at concentrations of 30–159 mg l-1. Although the extract is not as potent an inhibitor at Site I as rotenone, a known inhibitor of NADH dehydrogenase, such observations could explain the molluscicidal and schistosomicidal activity of dichloromethane extracts of the seeds of M. thonningii.

The prevalence of toxocariasis was evaluated for 445 dogs, 105 cats and 92 foxes from the Poznan region during 1997–1998. Forty one cats were infected (39%), 140 dogs (32%) and 15 red foxes (16%). Toxocara canis was found most frequently in puppies up to 3 months old (58%) and T. cati in kittens 4–6 months old (64%). Toxocariasis was much more prevalent amongst adult foxes (14%) than adult dogs (3%). In contrast to cats, female dogs and foxes were less infected than males. The present study suggests that cats may constitute an underestimated risk of transmission of Toxocara spp. to humans and the progressive synatropization of red foxes may also increase the sources of environmental contamination with Toxocara eggs.

Rats are known to be relatively resistant to infection with Echinococcus multilocularis. However, when rats are inoculated with the parasite tissues, E. multilocularis proliferates slowly at first but after 6 months the cysts increase in size considerably and contain large numbers of protoscoleces. As rats survive for 18 months or longer, approximately 100 ml of packed protoscoleces can be produced from each rat. A comparison of the antigenicity of the protoscoleces and microvesicles by immunoblot methods showed that both Em18 and Em16 are shared components between both protoscoleces and microvesicles, although the latter have some additional antigenic components. In antigens prepared from protoscoleces, the banding patterns around Em18 were much simpler than those from microvesicles. Therefore, for serodiagnosis of E. multilocularis, antigens should be carefully prepared from protoscoleces rather than microvesicles from the rat.

A unique human case of alveolar echinococcosis was described in 1979 from the Klatovy district of the Czech Republic. However, there were no previous epidemiological studies in this area focusing on detection of the source of infection – Echinococcus multilocularis adults producing eggs. During the period June 1997 to April 1999, 29 out of a total of 46 (63.3%) red foxes (Vulpes vulpes) in the Klatovy district and one of four foxes (25.0%) in the Pilsen South district were found to be infected with adult worms of E. multilocularis. No E. multilocularis adults were found in other animals from the Klatovy district (i.e. three specimens of Martes martes, two Martes foina, one Mustela erminea, two Meles meles and one Felis catus f. domestica). An examination of faecal samples from 55 dogs (Canis familiaris) from the Klatovy district resulted in the detection of E. multilocularis DNA in one (1.8%) sample. The present results support the possibility that human alveolar echinococcosis previously described in the Czech Republic had the character of an autochthonous infection. There are also indications of a potential risk of infection to humans.

The genetic diversity within the genus Trichinella was studied using cleavage fragment length polymorphism (CFLP) analysis. The CFLP method generates specific fingerprints based on single nucleotide mutations. By this method the amplified intergenic regions of the 5S rRNA genes of the eight different genotypes of Trichinella were analysed. The CFLP pattern of T. spiralis was completely different compared with the sylvatic species T. britovi, T. nativa, T. nelsoni, and the genotypes Trichinella T5, Trichinella T6 and Trichinella T8. The T. pseudospiralis intergenic region can be differentiated by size from the other species of Trichinella.

Aminopeptidase-like activities in crude whole body extracts of the free-living nematode Caenorhabditis elegans and the plant parasitic soybean cyst nematode Heterodera glycines were examined. General characteristics including pH optima, heat lability, and inactivation of enzyme by organic solvent were the same for the two species. All developmental stages of H. glycines exhibited activity. In older females, activity was present primarily in the eggs. Affinity for the substrate L-alanine-4-nitroanilide was the same regardless of the stage examined, and was similar for the two species (Km=2.3±0.3 mM FOR C. ELEGANS AND 2.9±0.2 Mm for H. glycines). Nearly all (>95%) of C. elegans aminopeptidase-like activity was present in the soluble fraction of the extract, while H. glycines activity was distributed between the soluble and membrane fractions. Specific activities of the soluble enzymes were highest in C. elegans and H. glycines juveniles. The C. elegans enzyme was susceptible to a number of aminopeptidase inhibitors, particularly to amastatin and leuhistin, each of which inhibited aminopeptidase-like activity more than 90% at 90 μm. In H. glycines, aminopeptidase-like activity was inhibited 39% by amastatin at 900 μm. The apparent molecular weight of the soluble C. elegans enzyme is 70–80 kDa. Some activity in H. glycines is present in the 70–80 kDa range, but most activity (80–90%) is associated with a very high molecular weight (>240 kDa) component.

The role of the red fox Vulpes vulpes in the dissemination of eggs of Toxocara canis into the environment is considered with reference to female worm fecundity and egg output in the faeces of infected foxes collected from four localities in southern England. A significant positive correlation was found between female worm size and the number of eggs in the uterus but there was no significant relationship between T. canis worm numbers and egg output in fox faeces. Reliable estimates of worm burdens in foxes could not, therefore, be determined from faecal egg counts alone. The highest mean egg output of 2145.0 epg recorded from adult foxes indicated that fox cubs are not necessarily the main sources of environmental contamination with T. canis eggs. Saturated magnesium sulphate was found to be a more effective flotation solution than zinc sulphate and sodium chloride for recovering eggs from fox faecal samples.

The prevalence of Spirocerca lupi in 260 privately owned dogs with different life and hunting styles in Greece was based on the examination of randomly taken faecal samples using Teleman's sedimentation technique. The dogs did not demonstrate any clinical signs of spirocerciasis. Although the prevalence was 10%, it was found to be significantly higher in trace hunting dogs (21%), than in scent hunting dogs (5%) and household pets (0%). There was no relationship between prevalence and age/sex of dogs. The impact of life and hunting styles on the prevalence of S. lupi in the dog and that of the faecal examination technique, are discussed.
