Use of isozymes (including allozymes) in studies of population genetics and systematics of seaweeds has increased sufficiently in the last decade to allow some generalization. Only a single locus has been observed for about half the enzymes analysed in seaweeds, compared with 29% in vascular plants. Compared with higher plants, macroalgal species generally have low amounts of electrophoretically detectable genetic variation; the lowest levels of genetic variation found in natural populations are those reported for seaweeds. Nonetheless, seaweeds show an association between levels of genetic diversity as revealed by isozymes and species-specific attributes, such as mating system and predominance of asexual versus sexual reproduction. In systematic studies, isozymes have revealed cryptic species and identified pairs of sibling taxa. The quaternary structure of enzymes appears to be conserved at the phylum level. With the current availability of improved techniques for enzyme electrophoresis and for data interpretation, we expect future studies utilizing isozyme electrophoresis to provide further insight into population and evolutionary processes in seaweeds.

Free-floating Ulva L. biomass in the eutrophic brackish ‘Veerse Meer’ lagoon (southwest Netherlands) consists of four morphologically identified species: U. curvata (Kützing) De Toni, U. lactuca L., U. rigida C. Agardh and U. scandinavica Bliding. U. curvata could be recognized easily because of the characteristic central cavity in the holdfast of the attached plants, the arrangement of cells in rows and the single pyrenoid in each cell. U. rigida was distinguished by the thick thallus and the large number of pyrenoids. The Veerse Meer isolate, however, was slightly different from the isolate from the Oosterschelde estuary (the Netherlands). U. lactuca and U. scandinavica showed a high degree of overlap in thallus thickness and cell size, but U. scandinavica usually possessed more pyrenoids. However, doubts have frequently been expressed about the use of some morphological characters in Ulva taxonomy. To determine the validity of such characters in the identification of Ulva species, the morphological variation within and between morphological species was recorded and a molecular data set generated. To detect possible ecophysiological differences between species, optimum temperatures and salinities for growth were determined experimentally. The sequences of the nuclear ribosomal DNA internal transcribed spacer 2 (ITS2) and flanking regions of U. lactuca, U. rigida and U. scandinavica from the Veerse Meer were all identical, but differed from that of U. rigida from the Oosterschelde estuary. Ulva species from the Veerse Meer were most closely related to U. armoricana and U. rigida from Brittany (2·9% and 3·5% divergence respectively); the difference between U. rigida from the Veerse Meer and from the Oosterschelde estuary was 7·5%. Rooted trees, based on a comparison of these sequences with sequences of other Ulva and Enteromorpha species obtained from the literature, using Monostroma arcticum as outgroup, suggested that Ulva is paraphyletic with respect to Enteromorpha. The optimum temperature for growth of U. curvata was 25 °C; for all other species it was 10 °C. The optimum salinity for growth was 30 °C for all isolates. It is concluded that U. lactuca, U rigida and U. scandinavica from the Veerse Meer are all members of one highly polymorphic species.

Laurencia Lamouroux subgenus Chondrophycus Tokida et Saito was recently raised to generic status as Chondrophycus (Tokida et Saito) Garbary et Harper (Ceramiales, Rhodophyta). The previously unknown morphology and anatomy of two Chondrophycus species, C. undulata (Yamada) Garbary et Harper and C. parvipapillata (C.K. Tseng) Garbary et Harper, were studied in specimens from Korea, Japan and Hawaii. These species share some features, such as a strongly compressed thallus, vegetative axes with two pericentral cells, tetrasporangial production from pericentral cells and spermatangial development of the trichoblast type. However, they differ from each other in the position of the first pericentral cell relative to the trichoblast in axial segments and in the position of the tetrasporangial pericentral cell. In C. undulata, the first pericentral cell is always produced at one side of the basal cell of a trichoblast, whereas in C. parvipapillata it is formed underneath this cell. While tetrasporangia in C. undulata are produced only from three (occasionally four) additional pericentral cells, in C. parvipapillata they are formed by the existing second pericentral cell and two (occasionally three) additional pericentral cells, so the resulting tetrasporangial axial segment has only one sterile pericentral cell rather than two as in C. undulata. C. undulata produces spermatangial branches from two laterals on the suprabasal cell of trichoblasts but one of the two remains partly sterile, whereas in C. parvipapillata a spermatangial branch develops from only one of the two laterals. These two species also differ distinctly from each other in some vegetative features, such as presence or absence of (1) secondary pit connections between epidermal cells, (2) a palisade structure of epidermal cells with conical projections and (3) corps en cerise. The taxonomic implications of these vegetative and reproductive features of Chondrophycus and those of other genera of the Laurencia complex necessitate changes to the generic delineations of Osmundea Stackhouse, Laurencia and Chondrophycus. The required new combinations in Chondrophycus are made, mainly for species from the western Pacific. An infrageneric classification scheme is also proposed for Chondrophycus including Kangjaewonia subgen. nov., Palisada (Yamada) subgen. stat. nov., Yuzurua subgen. nov. and Parvipapillatae sect. nov.

Effects of irradiance and temperature on the Mediterranean red algae Eupogodon spinellus and Eupogodon planus were tested. Growth of both species was saturated at an irradiance of 10–20 μmol m−2 s−1, which is in accordance with their sublittoral habitat. Eupogodon spinellus and E. planus survived permanently at temperatures between 8 and 30 °C. The temperature optimum for growth was 25 °C with suboptimal growth occurring at (10−)15 and 30 °C in both species. At their collection locality (Corsica), potential monthly growth yields would be highest in summer and in winter would be only about 20% of the maximum. Reproductive requirements could be determined only in E. planus. Gametophytes reproduced both in long and in short days but only at 20 °C. Tetrasporophytes reproduced at 15–20 °C but only in short days. Geographic distribution boundaries are not set by growth or survival limits. However, the reproductive requirements of E. planus did account for its restricted distribution in the Mediterranean and on the Canary Islands.

The elemental composition of cells of Anabaena flos-aquae was investigated by energy-dispersive electron probe X-ray microanalysis (10 kV) of mixed phytoplankton samples obtained from different depths within a stratified eutrophic freshwater lake. Preliminary studies had indicated that at 10 kV, X-ray information was derived mainly from the peripheral region (up to 1–2 μm) of cells. Routinely detectable elements (present in at least 50% of cells) included Mg (overall mean 112 mmol kg−1 dry mass), Si (1850), P(313), S (113), Cl (92), K (300) and Ca (56). Peaks of Na and Al were also frequently present. Individual elements showed a wide range of intracellular concentrations at each depth within the lake. Si had a clear bimodal distribution at depths of 1–8 m, indicating both low-Si (mean concentrations 54–290 mmol Si kg−1) and high-Si (2770–3800 mmol Si kg−1) cell subpopulations. Low-Si cells had significantly higher concentrations of other detectable elements compared with high-Si cells, but could not be distinguished from the latter in terms of morphology or stage of cell cycle (comparison of dividing and non-dividing cells). High-Si cells were intermixed with low-Si cells in individual Anabaena colonies and decreased proportionally with depth, ranging from approximately 75% in the surface sample to 10% at 8 m. With the exception of Si, mean elemental concentrations of Anabaena populations were closely similar throughout the epilimnion. These differed from the metalimnion sample, which had significantly higher concentrations of Mg, P, S and K, and a lower concentration of Si. Mean ratios of certain elements (Mg, P, S and K) and ion groups (monovalent/divalent cations) were highly constant throughout the sampled water column. At each depth, most of the cell elements were significantly and positively inter-correlated (Pearson analysis), with Mg, P, S and K typically constituting the major group (factor analysis). Correlations with Si were invariably negative, suggesting a separate (possibly cell wall) location of this element in the cell.

The seasonal colonization and vertical zonation of adpressed and upright algae on Phragmites stems in Lake Belau, Germany, was investigated from November 1988 to October 1991. During the autumns and winters from 1988 to 1990 the dominant adpressed species included the diatoms Epithemia adnata, E. sorex, E. turgida and Rhopalodia gibba, while during the same seasons in 1990 and 1991, Cocconeis placentula var. euglypta was abundant. The upright diatoms were most abundant on the middle sections of the stems, whereas those epiphytes with most of their cell surfaces directly in contact with the substratum constituted the greatest part of the biomass on the lowest sections of the stems. Epithemia spp. and R. gibba attained their highest abundances near the sediment as well as buried beneath the most dense periphyton aggregations, while there were dense populations of C. placentula var. euglypta on the entire surface of the stems. Cocconeis is able to tolerate higher light intensities, with photosynthetically active radiation (PAR) values averaging 38 μmol m−2 s−1, than Epithemia and Rhopalodia, which settle at a significantly lower PAR (26 and 20 μmol m−2 s−1, respectively). The change in dominance among the adpressed species from Epithemia and Rhopalodia to Cocconeis can be explained by changes in the grazing habits of the snail Acroloxus lacustris.

Photosynthetic acclimation and the interactions between carbon (C) and nitrogen (N) metabolism have been studied in the red macroalga Gracilaria sp. from Cádiz, Spain, cultured under different inorganic C and N levels. The use of chemostats and buffered medium allowed continuous restoration of the alkaline reserve and constancy of pH during the experiments. The N∶C ratios and phycobiliprotein, chlorophyll a and soluble protein contents decreased when Gracilaria sp. was grown at low N levels. Algae grown in a high inorganic C concentration (5% CO2) displayed a higher soluble carbohydrate concentration and maximum photosynthesis rates but a lower photosynthesic affinity for inorganic C, and lower phycobiliprotein and Rubisco contents, than those cultured at low inorganic C levels (air CO2). The inorganic C enrichment also affected the N uptake and assimilation in Gracilaria sp., causing a decrease in the N uptake rate even under conditions of N sufficiency. These results reflect the significant influence of the inorganic C growth regime on N assimilation in Gracilaria sp.

A new species of Porphyra is described from the south western Cape, South Africa. The gametophyte of Porphyra aeodis sp. nov. grows epiphytically on Aeodes orbitosa (Suhr) Schmitz, and has a seasonal life history that matches that of its host. Although P. aeodis has been confused with P. capensis Kützing in the past, P. aeodis is more similar to the sympatric but epilithic P. saldanhae Stegenga, Bolton et Anderson. There is considerable morphological overlap between P. aeodis and P. saldanhae, although they may be distinguished using a combination of morphological and ecological characters. The taxonomic separation of P. aeodis and P. saldanhae was confirmed using isozyme electrophoresis.

Exposure of Fucus spiralis germlings to precise copper concentrations (0 to 844 nM Cu2+) in chemically defined medium demonstrated a relationship between ultrastructural changes and growth retardation with increasing copper concentration. Electron-translucent vesicles, present in ova, which normally disappear after fertilization, accumulated in germlings exposed to Cu2+ above 10·6 nM, suggesting that copper may inhibit a metabolic pathway involved in cell wall formation which is initiated by fertilization. No membrane damage was observed during the exposure period. During a post-exposure period in copper-free medium, recovery occurred (rhizoid extension, apical hair formation) in germlings previously exposed to concentrations below 106 nM Cu2+ and electron-translucent vesicles became granular and disappeared. It is proposed that the electron-translucent vesicles contain a cell wall precursor and that copper inhibits its incorporation into the cell wall, preventing growth and development of the zygote.

Morphometric analysis and phylogenetic analysis of sequences of the rbcL chloroplast gene (which codes for the large subunit of the ribulose-1,5-bisphosphate carboxylase/oxygenase enzyme) and the nuclear 18S ribosomal RNA (rRNA) gene were carried out on 26 specimens of marine and freshwater Hildenbrandia from North America. Nineteen marine specimens were collected from Alaska to Costa Rica on the Pacific coast and from Newfoundland to Connecticut on the Atlantic coast. Seven freshwater samples were collected from Texas, Costa Rica, St Lucia and Puerto Rico. Three groups of samples were distinguished by morphometric analysis: one containing all freshwater samples (H. angolensis Welwitsch ex West et West), one consisting of a marine sample with parallel tetrasporangial divisions (H. occidentalis Setchell ex Gardner) and one group with non-parallel tetrasporangial divisions (H. rubra (Sommerfelt) Meneghini. These groupings were partially incongruent with those obtained by analyses of the molecular data. Parsimony and distance analyses of the rbcL gene resulted in trees in which Atlantic and Pacific clades were largely resolved. However, an Alaskan sample was included in the Atlantic group, which may indicate a trans-Arctic invasion event. The freshwater samples were paraphyletic for the rbcL gene, among the marine collections, which supports the concept of multiple invasions establishing the freshwater populations in North America. The 18S rRNA gene sequence data indicate that the freshwater samples are monophyletic with the exception of the unresolved position of the H. occidentalis sample. The freshwater samples form a monophyletic clade when multiple outgroups are used. The rbcL data appear to be mutationally saturated above approximately 17% divergence, which makes interpretation of phylogenetic signal among distant groups difficult. This may be a result of the asexual reproduction of the alga.

We recently reported (Gueneau et al., 1998) the existence of a GC-rich region downstream of the trnR gene which was conserved in the plastidial genome of three algae: Thalassiosira weissflogii, Odontella sinensis (Bacillariophyceae) and Porphyra purpurea (Rhodophyceae). We identified this GC-rich region as a possible vestigial ORF and noted additional high similarity to an open reading frame (ORF) in a plastid from a fourth alga, Pavlova lutherii (Haptophyceae); sequence data from P. lutherii did not extend to the TΨC tRNA arm. We now report that this region instead encodes a recently recognized type of RNA known as tmRNA (Fig. 1A).
