A siliceous sinter collected from Octopus Spring in Yellowstone National Park, USA contains an occluded volcanic rock fragment that has undergone alteration. The sinter piece beyond the fragment is mostly dominated by opal-A with trace amounts of bacterial cells, calcite and detrital quartz. Within the altered rock region, the mineral assemblage is dominated by dioctahedral smectite and quartz with trace amounts of pseudobrookite, ilmenite, rutile and hematite. Onset of opal-CT formation was only found in the outer spicular region of the sinter, which is unexpected given that this outer part represents newest growth. A reaction mechanism is proposed whereby the alteration of feldspar to smectitic clay locally produces excess silica, and alkali metal, and raises pH. As the clay mineral forms, it sequesters ions from pore fluids thereby inhibiting the opal-A phase change to more ordered opal-CT. Ions such as Mg are known to promote the opal-A to opal-CT reaction. Smectite formation therefore may assist microbial-texture preservation processes as excess silica produced increases the rate at which primary opal-A is formed. The altered zone also retains the greatest amount of fixed C and fixed N (operationally defined as C and N retained upon combustion at 450°C). The fixed N probably represents ammonium trapped in the exchangeable interlayer site of the smectite. This fixed N may serve as a potential biological signature of microbial activity in ancient rocks formed in similar environments.

We propose the mathematical notion of information gain as a way of quantitatively assessing the value of biosignature missions. This makes it simple to determine how mission value depends on design parameters, prior knowledge and input assumptions. We demonstrate the utility of this framework by applying it to a plethora of case examples: the minimal number of samples needed to determine a trend in the occurrence rate of a signal as a function of an environmental variable, and how much cost should be allocated to each class of object; the relative impact of false positives and false negatives, with applications to Enceladus data and how best to combine two signals; the optimum tradeoff between resolution and coverage in the search for lurkers or other spatially restricted signals, with application to our current state of knowledge for solar system bodies; the best way to deduce a habitability boundary; the optimal amount of money to spend on different mission aspects; when to include an additional instrument on a mission; the optimal mission lifetime; and when to follow/challenge the predictions of a habitability model. In each case, we generate concrete, quantitative recommendations for optimizing mission design, mission selection and/or target selection.

The identification and quantification of molecules in interstellar space and atmospheres of planets in the solar systems and in exoplanets rely on spectroscopic methods and laboratory work is essential to provide the community with the spectral features needed to analyse cosmological observations. Rotational spectroscopy in particular, with its intrinsic high resolution, allows the unambiguous identification of biomolecular building blocks and biosignature gases which can be correlated with the origin of life or the identification of habitable planets. We report the extension of the measured rotational transition frequencies of dimethylsulphoxide and its 34S and 13C isotopologues in the millimetre wave range (59.6–78.4 GHz) by use of an absorption spectrometer based on the supersonic expansion technique. Hyperfine patterns related to the methyl group internal rotation were analysed in the microwave range region (6–18 GHz) with a Pulsed Jet Fourier Transform spectrometer at extremely high resolution (2 kHz) and reliable predictions up to 116 GHz are provided. The focus on sulphur-bearing molecules is motivated by the fact that sulphur is largely involved in the intra- and inter-molecular hydrogen bonds in proteins and although it is the 10th most abundant element in the known Universe, understanding its chemistry is still a matter of debate. Moreover, sulphur-bearing molecules, in particular dimethylsulphoxide, have been indicated as possible biosignature gases to be monitored in the search of habitable exoplanets.

The question about the stability of certain biomolecules is directly connected to the life-detection missions aiming to search for past or present life beyond Earth. The extreme conditions experienced on extraterrestrial planet surface (e.g. Mars), characterized by ionizing and non-ionizing radiation, CO2-atmosphere and reactive species, may destroy the hypothetical traces of life. In this context, the study of the biomolecules behaviour after ionizing radiation exposure could provide support for the onboard instrumentation and data interpretation of the life exploration missions on other planets. Here, as a part of STARLIFE campaign, we investigated the effects of gamma rays on two classes of fungal biomolecules–nucleic acids and melanin pigments – considered as promising biosignatures to search for during the ‘in situ life-detection’ missions beyond Earth.

In this work, we address the difficulty of reliably identifying traces of life on Mars. Several independent lines of evidence are required to build a compelling body of proof. In particular, we underline the importance of correctly interpreting the geological and mineralogical context of the sites to be explored for the presence of biosignatures. We use as examples to illustrate this, ALH84001 (where knowledge of the geological context was very limited) and other terrestrial deposits, for which this could be properly established. We also discuss promising locations and formations to be explored by ongoing and future rover missions, including Oxia Planum, which, dated at 4.0 Ga, is the most ancient Mars location targeted for investigation yet.

Lipids are among the organic substances that can work as biosignatures, indicating life in an environment. We present an experimental investigation concerning analysis of lipids from a microbial source deposited on the Mars Global Simulant (MGS-1) regolith by geomatrix-assisted laser desorption/ionization-mass spectrometry (GALDI-MS). Our results indicate that lipids from intact microbial cells of a black yeast strain can be detected in these mimetic samples of Martian soil. These lipid molecules are predominantly associated with the occurrence of adducts in the GALDI-MS spectra. The results can be helpful in the planning of future planetary missions.

Can multicellular life be distinguished from single cellular life on an exoplanet? We hypothesize that abundant upright photosynthetic multicellular life (trees) will cast shadows at high sun angles that will distinguish them from single cellular life and test this using Earth as an exoplanet. We first test the concept using unmanned aerial vehicles at a replica moon-landing site near Flagstaff, Arizona and show trees have both a distinctive reflectance signature (red edge) and geometric signature (shadows at high sun angles) that can distinguish them from replica moon craters. Next, we calculate reflectance signatures for Earth at several phase angles with POLDER (Polarization and Directionality of Earth's reflectance) satellite directional reflectance measurements and then reduce Earth to a single pixel. We compare Earth to other planetary bodies (Mars, the Moon, Venus and Uranus) and hypothesize that Earth's directional reflectance will be between strongly backscattering rocky bodies with no weathering (like Mars and the Moon) and cloudy bodies with more isotropic scattering (like Venus and Uranus). Our modelling results put Earth in line with strongly backscattering Mars, while our empirical results put Earth in line with more isotropic scattering Venus. We identify potential weaknesses in both the modelled and empirical results and suggest additional steps to determine whether this technique could distinguish upright multicellular life on exoplanets.

The search for life in the Universe is a fundamental problem of astrobiology and modern science. The current progress in the detection of terrestrial-type exoplanets has opened a new avenue in the characterization of exoplanetary atmospheres and in the search for biosignatures of life with the upcoming ground-based and space missions. To specify the conditions favourable for the origin, development and sustainment of life as we know it in other worlds, we need to understand the nature of global (astrospheric), and local (atmospheric and surface) environments of exoplanets in the habitable zones (HZs) around G-K-M dwarf stars including our young Sun. Global environment is formed by propagated disturbances from the planet-hosting stars in the form of stellar flares, coronal mass ejections, energetic particles and winds collectively known as astrospheric space weather. Its characterization will help in understanding how an exoplanetary ecosystem interacts with its host star, as well as in the specification of the physical, chemical and biochemical conditions that can create favourable and/or detrimental conditions for planetary climate and habitability along with evolution of planetary internal dynamics over geological timescales. A key linkage of (astro)physical, chemical and geological processes can only be understood in the framework of interdisciplinary studies with the incorporation of progress in heliophysics, astrophysics, planetary and Earth sciences. The assessment of the impacts of host stars on the climate and habitability of terrestrial (exo)planets will significantly expand the current definition of the HZ to the biogenic zone and provide new observational strategies for searching for signatures of life. The major goal of this paper is to describe and discuss the current status and recent progress in this interdisciplinary field in light of presentations and discussions during the NASA Nexus for Exoplanetary System Science funded workshop ‘Exoplanetary Space Weather, Climate and Habitability’ and to provide a new roadmap for the future development of the emerging field of exoplanetary science and astrobiology.

Investigations into the existence of life in other parts of the cosmos find strong parallels with studies of the origin and evolution of life on our own planet. In this way, astrobiology and paleobiology are married by their common interest in disentangling the interconnections between life and the surrounding environment. In this way, a cross-point of both sciences is paleometry, which involves a myriad of imaging and geochemical techniques, usually non-destructive, applied to the investigation of the fossil record. In the last decades, paleometry has benefited from an unprecedented technological improvement, thus solving old questions and raising new ones. This advance has been paralleled by conceptual approaches and discoveries fuelled by technological evolution in astrobiological research. In this context, we present some new data and review recent advances on the employment of paleometry to investigations on paleobiology and astrobiology in Brazil in areas such biosignatures in Ediacaran microbial mats, biogenicity tests on enigmatic Ediacaran structures, research on Ediacaran metazoan biomineralization, fossil preservation in Cretaceous insects and fish, and finally the experimental study on the decay of fish to test the effect of distinct types of sediment on soft-tissue preservation, as well as the effects of early diagenesis on fish bone preservation.

We propose that retinal-based phototrophy arose early in the evolution of life on Earth, profoundly impacting the development of photosynthesis and creating implications for the search for life beyond our planet. While the early evolutionary history of phototrophy is largely in the realm of the unknown, the onset of oxygenic photosynthesis in primitive cyanobacteria significantly altered the Earth's atmosphere by contributing to the rise of oxygen ~2.3 billion years ago. However, photosynthetic chlorophyll and bacterio chlorophyll pigments lack appreciable absorption at wavelengths about 500–600 nm, an energy-rich region of the solar spectrum. By contrast, simpler retinal-based light-harvesting systems such as the haloarchaeal purple membrane protein bacteriorhodopsin show a strong well-defined peak of absorbance centred at 568 nm, which is complementary to that of chlorophyll pigments. We propose a scenario where simple retinal-based light-harvesting systems like that of the purple chromoprotein bacteriorhodopsin, originally discovered in halophilic Archaea, may have dominated prior to the development of photosynthesis. We explore this hypothesis, termed the ‘Purple Earth,’ and discuss how retinal photopigments may serve as remote biosignatures for exoplanet research.

We suggest testing S isotopes as biomarkers for Mars. An analogous robust biosignature has recently been proposed for the forthcoming exploration of the icy surface of Europa, and in the long term for the exploration of the surfaces of other icy moons of the outer solar system. We discuss relevant instrumentation for testing the presence of life itself in some sites, whether extinct or extant in order to complement a set of other independent biosignatures. We pay special attention to the possible early emergence of sulphate-metabolizing microorganisms, as it happened on the early Earth. Fortunately, possible sites happen to be at likely landing sites for future missions ExoMars and Mars 2020, including Oxia Planum and Mawrth Vallis. We suggest how to make additional feasible use of the instruments that have already been approved for future missions. With these instruments, the proposed measurements can allow testing S isotopes on Mars, especially with the Mars Organic Molecule Analyzer.

Several space missions, such as JWST, TESS and the very recently proposed ARIEL, or ground-based experiments, as SPHERE and GPI, have been proposed to measure the atmospheric transmission, reflection and emission spectra of extrasolar planets. The planet atmosphere characteristics and possible biosignatures will be inferred by studying planetary spectra in order to identify the emission/absorption lines/bands from atmospheric molecules such as water (H2O), carbon monoxide (CO), methane (CH4), ammonia (NH3), etc. In particular, it is important to know in detail the optical characteristics of gases in the typical physical conditions of the planetary atmospheres and how these characteristics could be affected by radiation driven photochemical and biochemical reaction. The main aim of the project ‘Atmosphere in a Test Tube’ is to provide insights on exoplanet atmosphere modification due to biological intervention. This can be achieved simulating planetary atmosphere at different pressure and temperature conditions under the effects of radiation sources, used as proxies of different bands of the stellar emission. We are tackling the characterization of extrasolar planet atmospheres by mean of innovative laboratory experiments described in this paper. The experiments are intended to reproduce the conditions on warm earths and super earths hosted by low-mass M dwarfs primaries with the aim to understand if a cyanobacteria population hosted on a Earth-like planet orbiting an M0 star is able to maintain its photosynthetic activity and produce traceable signatures.

We develop a polarimetry-based remote-sensing method for detecting and identifying life forms in distant worlds and distinguishing them from non-biological species. To achieve this we have designed and built a bio-polarimetric laboratory experiment BioPol for measuring optical polarized spectra of various biological and non-biological samples. Here we focus on biological pigments, which are common in plants and bacteria that employ them either for photosynthesis or for protection against reactive oxygen species. Photosynthesis, which provides organisms with the ability to use light as a source of energy, emerged early in the evolution of life on Earth. The ability to harvest such a significant energy resource could likely also develop on habited exoplanets. Thus, we investigate the detectability of biomolecules that can capture photons of particular wavelengths and contribute to storing their energy in chemical bonds. We have carried out laboratory spectropolarimetric measurements of a representative sample of plants containing various amounts of pigments such as chlorophyll, carotenoids and others. We have also measured a variety of non-biological samples (sands, rocks). Using our lab measurements, we have modelled intensity and polarized spectra of Earth-like planets having different surface coverage by photosynthetic organisms, deserted land and ocean, as well as clouds. Our results demonstrate that linearly polarized spectra provide very sensitive and rather unambiguous detection of photosynthetic pigments of various kinds. Our work paves the path towards analogous measurements of microorganisms and remote sensing of microbial ecology on the Earth and of extraterrestrial life on other planets and moons.

The biosignatures of life on Earth do not remain static, but change considerably over the planet's habitable lifetime. Earth's future biosphere, much like that of the early Earth, will consist of predominantly unicellular microorganisms due to the increased hostility of environmental conditions caused by the Sun as it enters the late stage of its main sequence evolution. Building on previous work, the productivity of the biosphere is evaluated during different stages of biosphere decline between 1 and 2.8 Gyr from present. A simple atmosphere–biosphere interaction model is used to estimate the atmospheric biomarker gas abundances at each stage and to assess the likelihood of remotely detecting the presence of life in low-productivity, microbial biospheres, putting an upper limit on the lifetime of Earth's remotely detectable biosignatures. Other potential biosignatures such as leaf reflectance and cloud cover are discussed.

Spring and evaporite deposits are considered two of the most promising environments for past habitability on Mars and preservation of biosignatures. Manitoba, Canada hosts the East German Creek (EGC) hypersaline spring complex, and the post impact evaporite gypsum beds of the Lake St. Martin (LSM) impact. The EGC complex has microbial mats, sediments, algae and biofabrics, while endolithic communities are ubiquitous in the LSM gypsum beds. These communities are spectrally detectable based largely on the presence of a chlorophyll absorption band at 670 nm; however, the robustness of this feature under Martian surface conditions was unclear. Biological and biology-bearing samples from EGC and LSM were exposed to conditions similar to the surface of present day Mars (high UV flux, 100 mbar, anoxic, CO2 rich) for up to 44 days, and preservation of the 670 nm chlorophyll feature and chlorophyll red-edge was observed. A decrease in band depth of the 670 nm band ranging from ∼16 to 80% resulted, with correlations seen in the degree of preservation and the spatial proximity of samples to the spring mound and mineral shielding effects. The spectra were deconvolved to Mars Exploration Rover (MER) Pancam and Mars Science Laboratory (MSL) Mastcam science filter bandpasses to investigate the detectability of the 670 nm feature and to compare with common mineral features. The red-edge and 670 nm feature associated with chlorophyll can be distinguished from the spectra of minerals with features below ∼1000 nm, such as hematite and jarosite. However, distinguishing goethite from samples with the chlorophyll feature is more problematic, and quantitative interpretation using band depth data makes little distinction between iron oxyhydroxides and the 670 nm chlorophyll feature. The chlorophyll spectral feature is observable in both Pancam and Mastcam, and we propose that of the proposed EXOMARS Pancam filters, the PHYLL filter is best suited for its detection.

Recent and proposed robotic missions to Mars are equipped with implements to expose or excavate fresh material from beneath the immediate surface. Once brought into the open, any organic molecules or potential biosignatures of present or past life will be exposed to the unfiltered solar ultraviolet (UV) radiation and face photolytic degradation over short time courses. The key question, then, is what is the window of opportunity for detection of recently exposed samples during robotic operations? Detection of autofluorescence has been proposed as a simple method for surveying or triaging samples for organic molecules. Using a Mars simulation chamber we conduct UV exposures on thin frozen layers of two model microorganisms, the radiation-resistant polyextremophile Deinococcus radiodurans and the cyanobacterium Synechocystis sp. PCC 6803. Excitation–emission matrices (EEMs) are generated of the full fluorescence response to quantify the change in signal of different cellular fluorophores over Martian equivalent time. Fluorescence of Deinococcus cells, protected by a high concentration of carotenoid pigments, was found to be relatively stable over 32 h of Martian UV irradiation, with around 90% of the initial signal remaining. By comparison, fluorescence from protein-bound tryptophan in Synechocystis is much more sensitive to UV photodegradation, declining to 50% after 64 h exposure. The signal most readily degraded by UV irradiation is fluorescence of the photosynthetic pigments – diminished to only 35% after 64 h. This sensitivity may be expected as the biological function of chlorophyll and phycocyanin is to optimize the harvesting of light energy and so they are readily photobleached. A significant increase in a ∼450 nm emission feature is interpreted as accumulation of fluorescent cellular degradation products from photolysis. Accounting for diurnal variation in Martian sunlight, this study calculates that frozen cellular biosignatures would remain detectable by fluorescence for at least several sols; offering a sufficient window for robotic exploration operations.

Rock coatings have been observed on Mars by Mars Pathfinder, Viking and the Mars Exploration Rovers. Although rock varnish has been studied for its potential as a biosignature, other types of rock coating have been largely ignored. In Kärkevagge, Swedish Lapland, sulphate crusts, aluminium glazes and Fe/Mn films occur with mineralogies mimicking those observed on the surface of Mars. Molecular analysis and scanning electron microscopy (SEM) were used to investigate the bacterial communities associated with these rock coatings. Molecular techniques revealed differences in community structure and metabolisms associated with the production of secondary minerals between the three coating types. SEM analysis showed evidence of encrustation in mineral coatings in the Fe/Mn films and aluminium glazes, and evidence of abundant microbial communities in all three coating types. These observations provide evidence for bacterial participation in the genesis of rock coatings. For astrobiology on Mars, rock coatings are an attractive biosignature target scientifically and logistically: they are surface environments easily accessible by rovers, endoliths are afforded protection from surface conditions, and evidence of life could potentially be preserved through biomineralization and lithification. This study describes the bacterial communities from rock coatings compatible with martian mineralogy, explores the potential for biologically facilitated rock-coating formation, and supports rock coatings as targets of astrobiological interest on Mars.

The future biosphere on Earth (as with its past) will be made up predominantly of unicellular micro-organisms. Unicellular life was probably present for at least 2.5 Gyr before multicellular life appeared and will likely be the only form of life capable of surviving on the planet in the far future, when the ageing Sun causes environmental conditions to become more hostile to more complex forms of life. Therefore, it is statistically more likely that habitable Earth-like exoplanets we discover will be at a stage in their habitable lifetime more conducive to supporting unicellular, rather than multicellular life. The end stage of habitability on Earth is the focus of this work. A simple, latitude-based climate model incorporating eccentricity and obliquity variations is used as a guide to the temperature evolution of the Earth over the next 3 Gyr. This allows inferences to be made about potential refuges for life, particularly in mountains and cold-trap (ice) caves and what forms of life could live in these environments. Results suggest that in high latitude regions, unicellular life could persist for up to 2.8 Gyr from present. This begins to answer the question of how the habitability of Earth will evolve at local scales alongside the Sun's main sequence evolution and, by extension, how the habitability of Earth-like planets would evolve over time with their own host stars.

If life occurs elsewhere in the Solar System, there is a strong likelihood that it occurs in a deep biosphere beneath the planetary surface. The evidence for methane in the martian atmosphere has drawn attention to the possible role of serpentinites in fuelling a deep biosphere through the generation of hydrogen and/or methane. Serpentinites represent a good target for the search for biosignatures in a range of reaction products. Isotopic measurements in each of methane, sulphide and carbonate in serpentinites can help determine evidence of biological activity. We show that ancient terrestrial serpentinites retain methane that could be subject to the measurement of carbon and hydrogen isotopes. There is, therefore, potential to sample serpentinites on Mars and test for evidence of life in the deep geological record of Mars.

The ability to differentiate abiotic organic material from material of a biological origin is a critical task for astrobiology. Mass spectrometry and spectroscopy provide key tools for advancing this task and are two techniques that provide useful and highly complementary compositional information independent of a specific biochemical pathway. Here we address some of the utility and limitations of applying these techniques to both orbital and in situ exploration of icy moons of the outer solar system.
