Water deficiency is one of the most severe abiotic stresses in rainfed dry lands and limits crop productivity. Exogenous applications of salicylic acid (SA) have been applied to mitigate the adverse effects of water-deficit stresses, but the relative efficacy of different derivatives of SA in enhancing water-deficit tolerance along with the underlying physio-biochemical mechanism and yield of crops is not well documented. Field experiments were conducted to ascertain the relative efficacy of exogenous application of three plant bioregulators (PBRs) [SA, thiosalicylic acid and 5-sulfosalicylic acid (SSA)], each at three concentrations (0.5, 1.0 and 1.5 mM), on the growth, physio-biochemical characteristics and yield of cluster bean under rainfed conditions. Based on a 2-year field experiment, the application of PBRs enhanced yield (from 8 to 16%). The yield enhancement with the application of PBRs was associated with elevated water content (from 9 to 17%), membrane stability (from 12 to 18%) and antioxidant enzyme activity (from 12 to 33%) and reduced lipid peroxidation (from −15 to −34%) in leaves. The effects of PBRs were conditionally type and concentration dependent. The application of SSA at a rate of 1 mM was more effective in enhancing water-deficit tolerance and improving the yield of cluster bean under water shortage conditions. This study provides empirical evidence of the potential for the application of SA and its derivatives to enhance crop yields under drought conditions. The results have direct implications for sustainable crop production for similar regions of the world facing water deficits.

Salicylic acid (SA), a phytohormone, has been considered to be a key regulator mediating plant defence against pathogens. It is still vague how SA activates plant defence against herbivores such as chewing and sucking pests. Here, we used an aphid-susceptible wheat variety to investigate Sitobion avenae response to SA-induced wheat plants, and the effects of exogenous SA on some defence enzymes and phenolics in the plant immune system. In SA-treated wheat seedlings, intrinsic rate of natural increase (rm), fecundity and apterous rate of S. avenae were 0.25, 31.4 nymphs/female and 64.4%, respectively, and significantly lower than that in the controls (P < 0.05). Moreover, the increased activities of phenylalanine-ammonia-lyase, polyphenol oxidase (PPO) and peroxidase in the SA-induced seedlings obviously depended on the sampling time, whereas activities of catalase and 4-coumarate:CoA ligase were suppressed significantly at 24, 48 and 72 h in comparison with the control. Dynamic levels of p-coumaric acid at 96 h, caffeic acid at 24 and 72 h and chlorogenic acid at 24, 48 and 96 h in wheat plants were significantly upregulated by exogenous SA application. Nevertheless, only caffeic acid content was positively correlated with PPO activity in SA-treated wheat seedlings (P = 0.031). These findings indicate that exogenous SA significantly enhanced the defence of aphid-susceptible wheat variety against aphids by regulating the plant immune system, and may prove a potential application of SA in aphid control.

Genome editing with the CRISPR/Cas (clustered regularly interspaced short palindromic repeats/CRISPR associated protein) system allows mutagenesis of a targeted region of the genome using a Cas endonuclease and an artificial guide RNA. Both because of variable efficiency with which such mutations arise and because the repair process produces a spectrum of mutations, one needs to ascertain the genome sequence at the targeted locus for many individuals that have been subjected to mutagenesis. We provide a complete protocol for the generation of amplicons up until the identification of the exact mutations in the targeted region. CRISPR-finder can be used to process thousands of individuals in a single sequencing run. We successfully identified an ISOCHORISMATE SYNTHASE 1 mutant line in which the production of salicylic acid was impaired compared to the wild type, as expected. These features establish CRISPR-finder as a high-throughput, cost-effective and efficient genotyping method of individuals whose genomes have been targeted using the CRISPR/Cas9 system.

Putative glyphosate-resistant sourgrass was collected to determine its resistance level and to evaluate its metabolic profile after resistance. Although accumulation of shikimic acid is known to occur in glyphosate-susceptible populations, differences in the ability of resistant (R) and susceptible (S) biotypes to accumulate quinic acid, salicylic acid, and aminomethylphosphonic acid (AMPA) have been studied to a lesser extent. Our objective was to confirm glyphosate resistance in sourgrass and to understand the metabolic profile of these plants in response to the herbicide. Greenhouse experiments were carried out from January 2016 to June 2018. There were no significant differences in glyphosate translocation in the plants. No metabolism of glyphosate to AMPA was observed; therefore, metabolism of glyphosate to AMPA is not a mechanism in R biotypes. S biotypes showed higher concentrations of shikimic acid and quinic acid before glyphosate and accumulated less of both secondary acids in treated leaves 72 h after glyphosate application. Resistant biotypes showed higher concentrations of salicylic acid before glyphosate application.

This study indicates that induced disease resistance might be useful to control branched broomrape. Strains of the rhizosphere bacteria Pseudomonas spp. (Proradix®), salicylic acid derivates (Bion®), and extracts of the algae Ascophyllum nodosum L. (Goemar Fruton Spezial®) can decrease branched broomrape infection to 80%. Results suggest that agents working as elicitors for resistance in other plant–pathogen interactions by induced systemic resistance or by systemic acquired resistance also could reduce branched broomrape infestation. These findings suggest that activation of immune responses before infection of plants could be an innovative control method for parasitic weeds.

Vaccinations are the mainstay of western preventive medicine, and they have been used to protect some crops against disease and insect pests. We consider rice as a model for protection using induced resistance since it is one of the most important staple crops and there have been significant new developments in: cross-resistance among rice insects, chemical pathways involved in induced resistance, sequencing the rice genome and expression of genes conferring resistance against rice insect pests. Insect attack has been found to cause lesions that kill planthopper eggs and early stages of gall midges. Damaged plants released volatiles that made them less likely to be chosen by planthoppers and more attractive to parasitoids. Chemical elicitors have been developed for dicotyledonous plants and these can induce resistance in rice, although rice does not fit models developed to explain signalling in dicots. For example, salicylic acid did not increase in rice after infection by pathogens and did not appear to be the mobile signal for induced resistance against pathogens although it was involved in induced responses to phloem-feeding insects. Jasmonic acid acted as a signal in some induced responses to pathogens as well as chewing insects. Many of the genes associated with induced resistance in rice have recently been mapped, and techniques are being developed to incorporate them into the genome of cultivated varieties. Attempts to control insect pests of rice will affect interactions with pathogens, predators and parasites, and other organisms in this agroecosystem.

Salicylic acid (SA) is a natural phenolic compound known as the active principle of aspirin. Its presence in vegetal sources suggests that fruit and vegetable (FV) consumption could produce measurable SA serum concentrations in human subjects not taking aspirin. The aim of this study was to investigate the relationship between FV intake and circulating SA in healthy subjects. Thirty-eight volunteers (twenty-two males and sixteen females) were recruited from an Italian university campus. They recorded their food intake for 7 d to evaluate dietary consumption and, in particular, FV intake; fasting blood samples were taken on the morning of the eighth day to measure SA serum concentration, using a sensitive stable isotope dilution and GC–MS method. Median SA serum concentration was 0·124 μmol/l (range 0·028–0·295). Circulating SA was significantly related to FV consumption, both to the mean daily intake (r2 0·13, P = 0·03) and to the last day intake (r2 0·16, P = 0·01). The subjects in the highest FV intake quartile in the preceding day (>4·75 servings) had significantly higher SA concentrations than in the lowest quartile ( < 2·3 servings) (median concentrations 0·188 and 0·112 μmol/l, respectively; P = 0·04). This study proved that, after overnight fast, human subjects not taking aspirin display circulating SA in amounts related to the FV consumption. It is therefore possible that the beneficial effects of regular FV consumption in man could also depend on low chronic SA exposure.

Acetylsalicylic acid (aspirinTM; 2-acetoxybenzoic acid) has been used for >100 years for pain relief and to treat inflammatory conditions and fevers. More recently, regular intake has been associated with decreased incidence of certain cancers, particularly colon cancer. After absorption aspirin is very rapidly hydrolysed to salicylic acid (2-hydroxybenzoic acid). The anti-cancer effects of aspirin may be a result of salicylic acid reducing the transcription of prostaglandin H2-synthase and thereby the synthesis of pro-inflammatory and potentially-neoplastic prostaglandins. Salicylic acid is widely present in plants and functions as a hormonal mediator of the systemic acquired resistance response to pathogen attack and environmental stress. Thus, it is present in a large range of fruit, vegetables, herbs and spices of dietary relevance. Consequently, the recognised effect of consuming fruit and vegetables on lowering risk of colon cancer may be partly attributable to salicylates in plant-based foods. The present review discusses which types of fruit and vegetables are the richest source of salicylates and whether they are sufficiently released from the food matrix to modify the key cellular events associated with the pathogenesis of colon cancer.
