As a potential ultimate cause of microhabitat restriction for ectoparasites of fish, it has been postulated that selection has favoured narrow microhabitats to facilitate mating. The evidence for this hypothesis is evaluated with males and females of the parasitic copepod Lernanthropus cynoscicola, by using methods commonly used in community ecology, and considering the influence of host ontogeny on site selection. No evidence supporting the mating hypothesis was found after comparison of niche measurements between both sexes. Furthermore, aggregation among individuals of the same sex was stronger than among males and females, and the co-occurrence of both sexes did not depart from that expected by chance; also, negative correlations between the intensity of infection of both sexes were observed. Restriction of the microhabitats cannot therefore be due to facilitation of mating. Moreover, both sexes displayed a sequential displacement over the gill arches and differential preferences by specific gill sections in response to an increasing host size, probably as a consequence of a host-size related cline of optimal conditions where fitness is improved. Thus, reproductive benefits other than increased chances of mating may have played a role in the evolution of restricted niches in parasites, and selection may have favoured a narrowing of the niche around sites where fitness is maximized.

The haplosporidian described is found in the Malpighian tubules of the beetle Gonocephalum arenarium in the Cape Peninsula, South Africa.

With the exception of the germination, which must take place in the digestive tract, the life cycle is completed within the Malpighian tubules. Studied over a period of several years, it has been found that the incidence of infection averages about 5 %.

Six Malpighian tubules connect with the gut, and in an infected insect all six are almost always parasitized to the same degree and by the same forms of the parasite.

Intestinal and faecal IgA responses against different developmental stages of Nippostrongylus brasiliensis were studied in Hooded Lister rats vaccinated subcutaneously with infective larvae or intragastrically with 4th-stage larvae, adult nematodes, adult worm somatic and metabolic antigens. It was found that intragastric administration of L4 or adult worms produced as high resistance to challenge as normal infection. Oral vaccination with adult nematode excretory–secretory (ES) and somatic antigens produced 69 and 17% protection respectively. All vaccination regimes induced a considerable IgA response against somatic and metabolic antigens of the intestinal stages. However, no correlation could be seen between the IgA levels and degree of protection. Analysis of intestinal mucosal, intestinal contents and faecal IgA specific to larval or adult antigens suggests that the IgA is probably induced by antigen(s) common to all intestinal stages. Also secretory IgA responses against surface antigens of intestinal L4 did not show stage specificity, although this was detected in sera.

The Trematode parasites of man have been so frequently listed and, for the most part, so fully described in various text-books and treatises on parasitology, pathology and tropical medicine that it might seem superfluous to enumerate them again. My object in doing so is to provide a basis for comparison with the parasites of the Primates, our knowledge of which is still extremely fragmentary, and with the parasites of Mammals in general. It is hoped that the works which are cited will prove to be the most useful or the most authoritative original references available.

The development of the tegument and cercomer of Paricterotaenia paradoxa polycephalic larvae was examined using electron microscopy. Larvae are formed by budding from the inner surface of the tegument of the degenerating hexacanth embryo. A new secondary tegument formed around the larvae is probably produced from the original hexacanth sub-tegumental cells. Microvilli covering the surface of young larvae are converted directly into microtriches, as the larvae develop, by addition of electron-dense material to the proximal part of the microvillus. Remnants of the original microvillus are visible at the distal surface of each new microthrix, but they eventually degenerate. The cercomer homologue is represented by scattered follicular cells, bearing microvilli, lying just within the containing cyst wall. The continuity of tegumentary tissue from one developmental stage to the next is discussed.

During some investigations in which, he has been engaged on the internal parasites of rats in the British Isles, Mr G. C. Dudgeon, of the Wellcome Bureau of Scientific Research, has kindly submitted to the writer examples of all the species of adult Cestodes found.

Malaria parasites have had profound effects on human populations for millennia, but other terrestrial vertebrates are impacted by malaria as well. Entire species of birds have been driven to extinction, and many others are threatened by population declines. Recent studies have shown that host-switching is quite common among malaria parasite lineages, and these switches often involve a significant shift in the environment in which the parasites find themselves, including nucleated vs non-nucleated red blood cells and red vs white blood cells. Therefore, it is important to understand how parasites adapt to these different host environments. The mitochondrial cytochrome b (cyt b) gene shows evidence of adaptive molecular evolution among malaria parasite groups, putatively because of its critical role in the electron transport chain (ETC) in cellular metabolism. Two hypotheses were addressed here: (1) mitochondrial components of the ETC (cyt b and cytochrome oxidase 1 [COI]) should show evidence of adaptive evolution (i.e. selection) and (2) selection should be evident in host switches. Overall we found a signature of constraint (e.g. purifying selection) across the four genes included here, but we also found evidence of positive selection associated with host switches in cyt b and, surprisingly, in (apicoplast) caseinolytic protease C. These results suggest that evidence of selection should be widespread across these parasite genomes.

This study assessed the impact of adding community-directed treatment (ComDT) to the routine health facility (HF)-based treatment on prevalence and intensity of soil transmitted helminth (STH) infections among children aged 12 to 59 months. Repeated cross-sectional surveys were conducted among randomly selected children of this age group from the intervention area (HF+ComDT area) and the comparison area (HF area) at baseline (n=986), 12 months (n=796) and 18 months (n=788) follow-up. The prevalence of Ascaris lumbricoides was significantly higher in the HF+ComDT as compared to the HF area at baseline (P=0·048), but not at 12 and 18 months follow-up. At baseline the HF+ComDT area had significantly higher intensities of A. lumbricoides compared to the HF area (P<0·001), but not at 12 and 18 months follow-ups. Prevalence and intensity of hookworm did not differ significantly between treatment arms at any time. Analysis of trends showed a significant decrease in prevalence of A. lumbricoides and hookworm in the HF+ComDT area (P<0·001), of hookworm in the HF area (P<0·05), but not of A. lumbricoides in the HF area. It is concluded that the ComDT approach generally enhanced the treatment effect among under-five year children and that this alternative approach may also have advantages in other geographical settings.

Intraspecific pairing of Haematoloechus medioplexus and Echinostoma revolutum adults and interspecific pairing of H. medioplexus and E. revoltum adults were studied in vitro at 21 ± 1 °C and 30 ± 1 °C in Petri-dish cultures containing an agar substrate and a Locke's 1:1 solution overlay. Salinity and temperature tolerance tests showed that both species survived for 24 h in a Locke's 1:1 solution at 21 ± 1 °C and 30 ±1 °C. For behaviour studies, worms were placed 2 cm apart and then observed at various intervals from 5 min to 4 h. H. medioplexus and E. revolutum showed significant intraspecific pairing at 30±1 °C, whereas only H. medioplexus paired significantly at 21±1°C. Significant interspecific pairing of H. medioplexus and E. revolutum occurred at 21 ±1 °C and 30 ±1 °C. Thin-layer chromatographic analysis showed that H. medioplexus adults released mainly free sterol, along with traces of triacylglycerols and free fatty acids into a non-nutrient medium.

Pooled sample testing (PST) as a strategy for avoiding testing the majority of individual negative samples has been proposed for screening of diseases in low prevalence areas. There has been no standard guideline for PST in screening of Schistosoma japonicum infection of Yunnan, China. To document the optimum pool size with acceptable sensitivity of PST for screening of Schistosoma japonicum infection in this setting, an experimental pooling of each of 31 positive sera by IHA with various numbers of 24 negative sera was done. The results were used to create a statistical model which was subsequently used for simulation to predict sensitivity of the pooled serum tests in the population with varying prevalence and pool size. We found that to keep the sensitivity of PST above 90%, 1:05 should be the maximum dilution, that is, the optimum pool size should not be greater than 6. Antigen will have rather little interference if the prevalence of infection is low e.g. 1% or the antigen:antibody ratio is 1:100 or below. Pooled serum testing by IHA is an acceptable sensitive method for detecting antibody for Schistosoma japonicum infection in this area.

Laboratory experiments indicated that third-stage larvae of T. axei and T colubriformis were able to penetrate soil to a depth of 12 in. or more, although most larvae remained in the upper 2 in. No geotaxis was observed.

Heavy downflows of water caused severe mortalities but did not affect the rate of movement of surviving larvae through the soil. Very limited random lateral movement occurred in turf, being more pronounced in the presence of faeces. Prolonged desiccation was extremely lethal to infective larvae. The activity of larvae was directly and linearly proportional to normal temperatures. The average activity of groups of larvae declined with time.

The expansion in the volume of research activity over the last 100 years and the acceleration of technological change that underpins the nature and, to a lesser extent, the quality of the science we do, means that those in science today have little time, or indeed opportunity, for reflection on some of the pioneering efforts within their fields. From the first discovery of schistosome parasites in Egypt in 1851 by Theodor Bilharz, research on schistosomiasis has provided valuable insights into many wider aspects of biology and medicine and has attracted an array of colourful and dynamic scientists who have made major advances through elegant experimentation.

Mice infected with Toxoplasma gondii manifest a rapid decline in serum thyroxine (T4) levels. To locate the locus of the hypothyroxinaemia, the integrity of the pituitary–thyroid axis of infected mice was assessed by a thyrotropin-releasing hormone (TRH) assay. A rise in serum T4 after inoculation of TRH implies the release of thyrotropin (thyroid-stimulating hormone) from a functionally intact pituitary. Administration of a single, large-dose (1 μg) bolus of TRH to infected mice induced a positive, although subnormal, T4 response. In contrast, when infected mice were pre-treated with a series of low-dose (5 ng) pulses of TRH prior to the bolus challenge, the T4 response was markedly enhanced. We suggest that the multiple inoculations of low-dose pulses of TRH ‘primed’ the pituitary (and secondarily the thyroid) and led to replenishment of their readily available hormone reserves and the heightened response to stimulation. These observations indicate that the locus of thyroid dysfunction is in the hypothalamus, not the pituitary or thyroid, and apparently involves impairment of the pulsatile release of TRH.

The nematodes recorded in the present paper were obtained from fishes caught on some of the deep-sea fishing grounds to the west of Ireland, namely, the Porcupine Bank and Irish Atlantic Slope, and from the Irish Sea. The writer is indebted to Mr S. J. Hughes and Mr J. Llewellyn for the collection of material from the deep-sea fishing grounds, excursions having been made for the purpose in a commercial trawler in August 1938 and July 1939. The majority of the nematodes were collected by Mr S. J. Hughes during the 1939 excursion, the first excursion being devoted mainly to the collection of trematodes and cestodes. A record of the trematode and cestode parasites obtained during both years has already been published (Rees & Llewellyn, 1941). Nematodes from fishes in the Irish Sea have been obtained at intervals from 1936 onwards.

Sarcocystis spp. are intracellular coccidian parasites which infect domestic and wild animals and birds, resulting in considerable economic losses in production animals, and public health concerns worldwide. Sarcocystis spp. have an indirect life cycle where wild and/or domestic canine species primarily serve as definitive hosts and several domestic and wild animals (such as camels) act as intermediate hosts. In Northern Africa, the Middle East, Central Asia and China, camel meat is preferred due to cultural and religious traditions as well as its lower cholesterol/fat content than other red meat. However, camel meat quality could be downgraded by the presence of sarcocysts. To date, two Sarcocystis spp. have been reported from camels, including Sarcocystis ippeni (forms microscopic sarcocysts) and Sarcocystis cameli (forms both macroscopic and microscopic sarcocysts). Sarcocystosis is usually asymptomatic, though significant pathogenic effects have also been reported in camels. Despite the high occurrence of sarcocystosis in camels, little is known about various aspects of the disease in these animals. This paper provides a comprehensive review of the existing knowledge on the taxonomy, pathogenesis, epidemiology and diagnosis of Sarcocystis spp. infecting camels and it also highlights areas for further research that could enhance our understanding about sarcocystosis in camels.

Dirofilaria immitis is a parasitic nematode that survives in the circulatory system of suitable hosts for many years, causing the most severe thromboembolisms when simultaneous death of adult worms occurs. The two main mechanisms responsible for thrombus formation in mammals are the activation and aggregation of platelets and the generation of fibrin through the coagulation cascade. The aim of this work was to study the anticoagulant potential of excretory/secretory antigens from D. immitis adult worms (DiES) on the coagulation cascade of the host. Anticoagulant and inhibition assays respectively showed that DiES partially alter the coagulation cascade of the host and reduce the activity of the coagulation factor Xa, a key enzyme in the coagulation process. In addition, a D. immitis protein was identified by its similarity to the homologous serpin 6 from Brugia malayi as a possible candidate to form an inhibitory complex with FXa by sodium dodecyl sulfate polyacrylamide gel electrophoresis and mass spectrometry. These results indicate that D. immitis could use the anticoagulant properties of its excretory/secretory antigens to control the formation of blood clots in its immediate intravascular habitat as a survival mechanism.

Labelled antigens sharing determinants with both membrane and cytoplasmic fractions of Plasmodium knowlesi were detected in culture medium after in vitro incubation of schizont-stage parasites previously pulse-labelled with [3H]isoleucine. Release of antigens occurred only during schizont rupture and merozoite re-invasion. Antigenic material accounted for up to a third of the total trichloroacetic acid-insoluble radioactivity released by the cells. Absorption experiments indicated that approximately two-thirds of this antibody-precipitable material shared determinants with parasite membrane antigens, with a similar quantity sharing determinants with cytoplasmic proteins. Using antisera derived against 2 different antigenic variants of P. knowlesi, no evidence for the release of variant-specific antigens was obtained. Centrifugal analysis revealed that the majority of the radio-isotope labelled antigens were particulate in nature; however, some appeared to exist in true solution.

An amoeba was found in the faeces of ten out of fourteen English goats from three different localities.

The amoeba, of which a detailed description is given, is morphologically identical with Entamoeba debliecki of the pig and is therefore included in the same species.

A statistical analysis shows that the caprine strains of E. debliecki comprise two races distinguishable from one another by the size of their cysts but identical in all other features.

The caprine amoeba appears to be non-pathogenic.

In view of their bearing upon the systematic position of the parasite of the goat, a critical review is given of the data concerning the various amoebae previously recorded from the pig, goat and some other ruminants.

Photoleter anterouterus Fischthal & Nasir, 1974, from Pelecanus occidentalis Linnaeus and Pelecanus erythrorhynchos Gmelin from Florida, USA and the ventrogenital complexes of Photoleter gastrophilus (Kossack, 1910) from Delphinus delphis Linnaeus from Queensland, Australia and of Phocitrema fusiforme Goto & Ozaki, 1930 from Alopex lagopus (Linnaeus) and Phoca vitulina Linnaeus from Alaska are redescribed. The genus Photoleter Odhner, 1914 is emended and compared with Phocitrema Goto & Ozaki, 1930 and both genera are compared with members of, and as a result transferred from the family Opisthorchiidae Looss, 1899 to, the subfamily Centrocestinae Looss, 1899, family Heterophyidae Odhner, 1914.

A new method of computing the infectivity index of microfilariae (mf) for the mosquito population is proposed using the estimated mf density distribution in the human population. The observed density distribution is considered a compound of the Poisson and the gamma distributions. The former distribution describes the probability of a specimen containing a specified number of mf and the latter describes the density distribution of mf in the host population. The mf infectivity index is the probability that a blood meal will include at least 1 mf, conditional on the population-density distribution of mf as specified by the gamma distribution. Actual data indicate that this population-density-based infectivity index can be considerably different from the conventional index based on the survey-density distribution. The level of the carrier rate of mf in a survey is greatly influenced, apart from the sample variation, by the average volume of blood taken from each person. The rate computed on the estimated population-density distribution of mf is convertible to any base amount of blood.
