Efficient seed storage is a shared concern among the growing number of seed banks established for crop improvement or ex situ conservation. Container properties greatly affect seed interactions with the environment and the overall cost and success of seed banking operations. Several material properties contribute to their suitability as seed containers. This paper provides a consolidated list of water vapour permeability properties of thermal plastics commonly used for packaging. Composite packages with layers of film with different properties provide distinct advantages to seed banks. Different seed banks must rank the importance of the various factors depending on their mission and resources. Once the risks, costs and benefits are weighed, an appropriate strategy can be developed that addresses a seed bank's specific needs. Because there are many problems and several solutions, it is likely that strategies will vary among seed banks. This response details variables to consider when selecting seed storage packages, and focuses on water diffusion rates of packages with different compositions. A ‘moisture audit’ will help seed bank operators make informed decisions about packaging.

The ability of orthodox seeds to survive long-term dry storage is a key prerequisite for ex situ seed conservation in genebanks. However, only a few credible observations of seed survival for ≥ 200 years have been reported. In this paper, seed survival is shown for three species under suboptimal storage conditions for a documented time of ≥ 203 years and carbon dated at 218–270 years. Two species that germinated are in the Fabaceae (Liparia sp. and Acacia sp.). A water-impermeable seed coat, and hence an inability to equilibrate with ambient relative humidity, may contribute to long-term survival of these species. The third species that germinated (Leucospermum sp.) does not have a water-impermeable seed coat, and long-term survival may be linked to an oxygen-impermeable barrier in the seed. These results for species from the Cape Floristic Region, South Africa, suggest adaptation for extreme longevity in seeds of species of seasonally dry, Mediterranean environments.

Endo-β-mannanase (EC 3.2.1.78) is involved in the hydrolysis of mannan-type polysaccharides that are present in plant cell walls, especially those of the seed endosperm. The genes encoding the endo-β-mannanases have been studied extensively in tomato (Solanum lycopersicum), and five genes (LeMAN1, LeMAN2, LeMAN3, LeMAN4 and LeMAN5) and/or their products have been isolated and characterized. LeMAN1, LeMAN2 and LeMAN3 are expressed in tomato seeds, LeMAN4 in the fruit and LeMAN5 in the flower. LeMAN5 and LeMAN2 are now considered to be the same gene, and the former is re-designated as LeMAN2*. Transcripts of LeMANs 1, 2 and 3 are detected only in the endosperm of tomato seeds, and their synthesis is promoted by gibberellic acid. LeMAN4, in the fruit, occurs as LeMAN4a and LeMAN4i genes that encode an active or inactive form of endo-β-mannanase, respectively. LeMAN1–4 enzymes encoded by these genes share 80% similarity in amino acid sequence. In tomato, the leucine amino acid present near to the C-terminus of the endo-β-mannanase is the most important for achieving full activity of the enzyme.

A UK seed conservation collection of Anemone nemorosa L. seeds held at the Millennium Seed Bank (MSB) showed low viability in its first post-storage test. Because achenes of A. nemorosa are naturally dispersed when they are green, we tested the hypothesis that seeds may not be fully desiccation tolerant and storable at the time of natural dispersal, and that a post-harvest treatment could increase the proportion of desiccation-tolerant seeds. Achenes harvested at the point of natural dispersal in late May in 2003 and 2004 were either placed immediately on 1% water agar at 20°C (‘laboratory’ treatment), or placed in nylon sachets and buried in leaf litter among plants growing in the wild (‘field’ treatment). Samples were withdrawn at intervals over a period of 168 d and tested for desiccation tolerance (drying to 0.059 g H2O (g DW)− 1) and longevity (controlled ageing at 60% relative humidity and 45°C). An initial increase, followed by a decline, in the proportion of seeds surviving desiccation and in the longevity of both laboratory- and field-treated samples coincided with the development of embryos from globular to heart- and then torpedo-shaped. Developmental arrest was not required for the acquisition of desiccation tolerance, and continued growth and development of the embryo resulted in a loss of desiccation tolerance, similar to that seen in orthodox seeds upon radicle emergence. Furthermore, while A. nemorosa seeds, like many from the Ranunculaceae family, might be described as having morphological or morphophysiological dormancy, this lack of developmental arrest does not fit with the usual concept of dormancy. The implications of these results for the classification systems of seed-storage behaviour and dormancy, and for the long-term conservation of seeds of A. nemorosa, are discussed.

In the work reported in this paper, we attempted to elucidate the nature of the different abscisic acid (ABA) sensitivities presented by developing embryos from sorghum [Sorghum bicolor (L.) Moench] lines with contrasting pre-harvest sprouting (PHS) behaviour (Redland B2, susceptible; IS 9530, resistant). We explored two different hypotheses for a possible mechanism: (1) a different functionality of the ABA signalling pathway, and (2) a different rate of ABA degradation/conjugation in the apoplast of embryos from these genotypes. To assess the first possibility, we used an ABA-responsive gene (Rab17) as a reporter of changes in endogenous ABA content, which were artificially induced in embryos from both genotypes by means of fluridone application immediately after anthesis, to reduce ABA content, and embryo incubation in the presence of ABA. A defect in ABA signalling should be seen as a level of Rab17 expression that is independent of endogenous ABA content. For testing the second possibility at two stages of development, embryos from both lines were isolated and incubated in water for different periods. ABA concentrations in embryos and the incubation media were quantified through radioimmunoassay. In contrast to our findings for the resistant IS 9530 line, Rab17 expression did not respond to changes in ABA levels in sensitive Redland B2 embryos. The ABA degradation/conjugation rates in embryos and incubation media did not show clear differences between sorghum lines for any of the developmental stages analysed. These results suggest that a disruption in the ABA signal transduction pathway in Redland B2 underlies the low ABA sensitivity shown by embryos from this line.

Cotton (Gossypium spp.) is an important source of edible oil and protein meals. Complex genetics and strong environmental effects hinder progress in seed quality trait breeding in this species. The use of molecular markers can improve an understanding of the genetic factors conditioning seed quality traits, and is expected to assist in selection of superior genotypes. This study was conducted to identify quantitative trail loci (QTL) associated with seed physical and nutrient traits in cotton. To achieve this objective, a population of 140 BC1S1 lines developed from a cross between ‘TM-1’ and ‘Hai7124’ was evaluated in 2003 and 2004. A linkage map consisting of 918 markers from this population was used to identify QTL using QTLNetwork-2.0 software. Eleven single QTL were identified for kernel percentage, kernel oil percentage, kernel protein percentage and seven amino acids (Asp, Ser, Gly, Ile, Leu, Phe and Arg). Phenotypic variation explained by each individual QTL ranged from 10.89 to 46.28%. Two epistatic QTL for Cys and Leu were detected, explaining 9.55 and 4.43% of the phenotypic variation. These QTL detected for seed quality traits in cotton are expected to be useful for further breeding programmes targeting development of cotton with improved nutrient quality.

The assumption that seeds imbibe most of the water required for germination as liquid through seed-to-soil contact has been a dominant theme in germination research and seeding technology. Under most conditions, seeds are also exposed to water vapour during imbibition, but the relative contributions of liquid and vapour are difficult to assess. In water uptake models that include vapour, procedures used to estimate potential vapour imbibition have underappreciated the effect of distance on diffusion rate. At the same time, the amount of seed-to-soil contact and the liquid bridge from soil water to the seed tend to be greatly overestimated, considering the soil water contents often found in the field. Most researchers have recorded an approximately equal time to germination at soil water contents ranging from field capacity to nearly permanent wilting point, and little response to bulk density, soil type or seed–soil contact. While hydraulic conductivity decreases by several orders of magnitude as soil water content, bulk density and seed-contact decrease, relative humidity remains near 100%. There are several experiments demonstrating timely germination in water vapour alone. The combined evidence contradicts the assumption that seed–soil contact is important for imbibition of water by seeds. Water vapour should be considered the primary source of water for seeds in unsaturated soils.

Martin's (1946) seed classification system has 10 types based on embryo and endosperm characteristics and two additional types based on seed size: dwarf (0.3–2.0 mm) and micro ( ≤ 0.2 mm). He listed 17 families and 12 genera (in five other families) as having dwarf seeds. Our recent discovery of morphophysiological dormancy in dwarf seeds of several taxa of Campanulaceae and one of Gentianaceae prompted an evaluation of dwarf seeds. Martin's paper contains 37 families with one to several small (0.3–2.0 mm) seeded species that he did not list as being dwarf. Comparison of Martin's dwarf families and the 37 small-seeded non-dwarf families revealed no consistent differences between the two groups in endosperm texture, seed-coat anatomy, embryo morphology, class of seed dormancy or phylogenetic position. Also, Martin's dwarf seeds include a variety of embryo morphologies. Consequently, we have revised Martin's key to seed types. The dwarf category has been removed and the micro category replaced by ‘undifferentiated’ to reflect the state of the embryo in fresh seeds. Further, the key now includes linear fully developed, linear underdeveloped, spatulate fully developed and spatulate underdeveloped seed types, which Martin illustrated but did not include in his key. In the revised key, all seeds are distinguished on the basis of embryo and endosperm characteristics.

Red rice (Oryza sativa L.) dispersal units (florets) were dry-afterripened for 0–8 weeks and subsequently incubated at 30°C in polyethylene glycol (PEG) solutions with water potentials from 0 to − 1.6 MPa. Germination percentages and rates increased with dry-afterripening and water potential of the incubation medium. The seed population exhibited a normal distribution of base water potentials (Ψb, i.e. minimum water potential allowing germination) among individual seeds, characterized by three parameters: the hydrotime constant (θH), the mean base water potential (Ψb) and the standard deviation of the base water potential distribution (σΨb). Changes in germination during afterripening could be described by modifications of such parameters, particularly Ψb, which was employed to derive an index, DH(ARX = Ψb(ARX) − Ψb(ARN), where DH(ARX) represents a measure of dormancy of the seed population (in MPa) based on the hydrotime model, Ψb(ARX) is the mean base water potential of the seed population at any afterripening time X, and Ψb(ARN) is the mean water potential of the non-dormant (fully afterripened) population. The introduction of this index permitted interpretation of afterripening as a measurable reduction in the dormancy status of the seed, with progressive acquisition of both full germinative capacity and maximum germination rate, as anticipated by the hydrotime model. Moreover, secondary dormancy was induced proportionally to the reduction in water potential in the dark. Susceptibility to secondary dormancy induction was defined through DI(ARX), an index analogous to DH(ARX). These indices revealed that, in red rice, both breaking of primary dormancy and the inducibility of secondary dormancy followed decay kinetics with different sensitivities to the duration of dry storage.

In this study, we characterized two sunflower (Helianthus annuus L.) lines with differential sensitivity to drought, the sensitive line B59 and the tolerant line B71. Using both lines, we compared the content of endogenous jasmonates (JAs) in dry and imbibed seeds from plants grown under irrigation and drought. Jasmonic acid (JA), 12-oxo-phytodienoic acid (OPDA), 11-hydroxyjasmonate (11-OH-JA) and 12-hydroxyjasmonate (12-OH-JA) were detected in dry and imbibed sunflower seeds. Seeds from plants grown under drought had a lower content of total JAs and exhibited higher germination percentages than seeds from irrigated plants, demonstrating that environmental conditions have a strong influence on the progeny. OPDA and 12-OH-JA were the main compounds found in dry seeds of both lines. Imbibed seeds showed an enhanced amount of total JAs with respect to dry seeds produced by plants grown in both soil moisture conditions. Imbibition triggered a dramatic OPDA increase in the embryo, suggesting a role of this compound in germination. We conclude that JAs patterns vary during sunflower germination and that the environmental conditions experienced by the mother plant modify the hormonal content of the seed progeny.

The germination of developing seeds is very uncommon and is generally associated with deficiencies in abscisic acid (ABA) synthesis or sensitivity. This paper examines the quantitative relationship between the inhibition of precocious germination and endogenous ABA in the embryonic axis (ABAa) of hydrated soybean [Glycine max (L.) Merr.] seeds, isolated after the completion of histodifferentiation and before the beginning of dehydration, as well as the magnitude and evolution of axis sensitivity to endogenous ABA during that period. Developing seeds harvested at 25, 30, 35, 40 and 45 d after anthesis (DAA) were subjected to incubation or washing to induce changes in ABA content. ABA content was measured by radioimmunoassay, using a monoclonal antibody against free ABA. Germinability was measured as the time to 50% germination (t50). Washing and incubation induced eight- and twofold increases, respectively, in the rate of ABAa decline compared with the in planta ABAa decline. The threshold ABAa for inhibition of precocious germination (ABAc) increased slightly from 25 to 40 DAA [1.15–1.66 μg ABA (g DW)− 1]. This contrasted with the substantial decline in ABAa [10.90–2.07 μg ABA (g DW)− 1] during the same period, and indicated that sensitivity to endogenous ABA of hydrated seeds was initially high and diminished slowly during development. The relationship between (ABAa–ABAc) and t50 was linear for immature seeds incubated before and after washing. Below the ABAc, there were no differences in the t50 of 25–45 DAA seeds. The ABAa contribution to the control of precocious soybean seed germination was evident, although other potentially interacting factors were also present.

Orthodox, desiccation-tolerant seeds lose desiccation tolerance during germination. Here, we quantify the timing of the loss of desiccation tolerance, and explore the implications of this event for seed mortality and the shape of germination progress curves for pioneer tree species. For the nine species studied, all seeds in a seedlot lost desiccation tolerance after the same fixed proportion of their time to germination, and this proportion was fairly constant across the species (0.63–0.70). The loss of desiccation tolerance after a fixed proportion of the time to germination has the implication that the maximum number of seeds in a seedlot that can be killed by a single drying event during germination (Mmax) increases with an increasing time to 50% germination (t50) and an increasing slope of the germination progress curve. Consequently, to prevent the seed population from becoming highly vulnerable to desiccation-induced mortality, species with a greater t50 would be expected to have a shallower germination progress curve. In conclusion, these data suggest that the loss of desiccation tolerance during germination may constitute a significant, but previously unexplored, source of mortality for seeds in seasonal environments with unpredictable rainfall.

The stimulatory role of 3-methyl-2H-furo[2,3-c]pyran-2-one, a smoke-derived butenolide, on germination and post-germination events is well documented. Previous studies have involved germinating seeds in the continuous presence of the compound. However, commercial growers cannot exploit the potential of the butenolide for large-scale production of crops due to limited availability and environmental constraints. The present study was undertaken to assess the potential of the butenolide as a priming agent of tomato (Solanum esculentum Mill.) seeds. Flow cytometry data revealed that butenolide-primed seeds had a higher percentage of nuclei at the 4C stage than water-primed seeds. Emergence of the radicle was much faster in the primed seeds. After 36 h of imbibition, a higher percentage of the butenolide-primed seeds (22%) exhibited radicle emergence compared to the water-primed seeds (12%). While butenolide-primed seeds initially germinated more rapidly than either water-primed or unprimed seeds in a 48-h period, water-imbibed seeds reached a similar germination level as the butenolide-primed seeds by 60 h of incubation. The butenolide-primed seeds produced significantly (P ≤ 0.05) more vigorous seedlings than water-primed seeds or seeds in the continuous presence of either water or butenolide. A gradual decrease in the seedling vigour index was recorded for both water and butenolide-primed seeds with increased seed storage at room temperature. Nevertheless, the vigour index was significantly greater in the butenolide-primed seeds than the water-primed seeds. Vigour indices were significantly (P ≤ 0.05) higher for the butenolide-primed seeds under various stress conditions (salinity, osmoticum or temperature) compared to control or water-primed seeds. Results of the present study suggest that the butenolide is a good seed-priming agent. Additionally, primed seeds retained the promotive effect for a considerable time. This was also the case for tomato seeds under various simulated field stress conditions.

Intact seeds (seed+endocarp) from freshly harvested fruits of Prunus campanulata were dormant, and required 4–6 weeks of warm followed by 8 weeks of cold stratification for maximum germination percentage. Removing both endocarp and seed coat, however, promoted germination in a high percentage of non-stratified seeds. Treatment of intact, non-stratified seeds with gibberellic acid (GA3) was only partially effective in breaking dormancy. However, GA3 promoted germination of non-stratified seeds in which the endocarp (but not the seed coat) had been removed. The order of abscisic acid (ABA) concentration in fresh seeds was endocarp > seed coat > embryo, and its concentration in endocarp plus seed coat was about 6.2-fold higher than that in the embryo. Total ABA contents of seeds subjected to warm and/or cold moist stratification were reduced 6- to 12-fold. A higher concentration of GA4 was detected in embryos of non-dormant than in those of dormant seeds. Fluridone, a carotenoid biosynthesis inhibitor, was efficient in breaking dormancy of Prunus seeds. Paclobutrazol, a GA biosynthesis inhibitor, completely inhibited seed germination, and the inhibitory effect could be partially reversed by GA4, but not by GA3. Thus, dormancy in P. campanulata seeds is imposed by the covering layers. Dormancy break is accompanied by a decrease in ABA content of the covering layers and germination by an increase of embryonic GA4 content.

We studied seasonal variation in density and species richness of seeds in the 0–10 cm soil depth layer in primary, secondary and pine forests, and in shrubland and grassland in the Shilin Stone Forest Geographical Park, Yunnan, SW China. Soil samples were collected four times during the year at 3-month intervals. Seeds from 119 species were identified by germination tests in the soil samples. Density and species richness of seeds of herbaceous plants were greater than woody plants at all five sites throughout the year. Sampling time and site differences had significant effects on the mean number of species and on seed-bank density. Mean number of species per sample increased from February, reached the highest value in May, decreased to the lowest value in August and then increased in November. An exception was in the primary forest, where the highest number of species was found in February. Mean seed-bank density peaked in May at all five sites, and no significant differences were found between densities in February, November and August, except for the primary forest. The peak in seed-bank density in May might be due to dispersal of new seeds of spring-fruiting species, combined with persistence of seeds dispersed in previous years. This seasonal variation of individual species was due primarily to differences in species phenology rather than to differences between sites. Four seasonal seed-bank strategies were identified: two transient and two persistent. At all sites, similar numbers of seeds of herbaceous species were found between seasons, but the number of species of trees and shrubs decreased in August.

Morphological traits of diaspores that can predict their potential attachment to animal coats may help to model epizoochory in plant populations and communities. The present study tested the role of seed mass, shape and the presence of dispersal structures in the attachment potential of a sample of 14 abundant species in Mediterranean grassland and shrubland, using sheep and cattle coats that were shaken mechanically using a standardized protocol. We also tested recently proposed predictive models for the attachment potential of diaspores. Attachment potential measured on cattle hide was low in comparison with sheep wool for all types of diaspores. Differences between vertically and horizontally positioned coats were significant only for cattle, in which attachment potential was higher in vertically positioned coats. Seed weight was highly significant to predict attachment potential in sheep coats, but yielded significant results only in the case of vertically positioned cattle coats. In both cases, light seeds were best retained. Shape yielded marginally significant results only in the case of horizontally positioned cattle coats. In this case, elongated seeds seemed to be best retained. The presence of appendages in diaspores was significant only for sheep, in which attachment potential was higher for seeds with appendages. Recently proposed predictive models for the attachment potential were found to be highly robust for predicting this parameter in sheep coats, and thus support their generalization for this type of coat.

Samples of soil and recently deposited sediments were collected from the river bed, bank face and bank tops of two lengths (reaches) of the River Frome, Dorset, UK and one reach of the River Tern, Shropshire, UK. Soil propagule bank samples were collected in May 2003, and depositional samples were collected subsequently over four consecutive 4-month periods between June 2003 and October 2004. The samples were subjected to three emergence trials under drained, waterlogged and submerged conditions. Significantly more seedlings germinated in the drained than waterlogged trial, and waterlogged than submerged trials. Drained, waterlogged and submerged trials identified 186, 76 and 37 species, respectively. Six species identified in the waterlogged trials were not identified in the drained trials, and five species in the submerged trials were not found in the drained trials. Submerged trials added two species to the drained and waterlogged results. Application of detrended correspondence analysis (DCA) to average species abundance data, for the sampled hydrological habitats (bed, bank face, bank top) within the soil propagule bank and four depositional data sets, generated different results for the three trials. The drained treatment revealed significant differences between sites, seasons and hydrological habitats, whereas the waterlogged and submerged treatments presented an increasingly homogeneous view of the samples. Our results confirm other propagule bank emergence comparisons and extend them to depositional samples, demonstrating that the strong environmental sieves imposed by waterlogging and submergence restrict emergence of numerous terrestrial, wetland and even some aquatic species that were successfully identified using the drained conditions.

Germination and dormancy breaking requirements were studied in Selinum carvifolia (L.) L. and Angelica sylvestris L. (Apiaceae). Seeds of these two species have an underdeveloped embryo and are morpho-physiologically dormant. The embryo does not start to grow until physiological dormancy is broken by cold stratification. Incubating seeds at fluctuating temperatures in the light, after cold stratification, had a stimulating effect on embryo growth and seed germination. Seeds of S. carvifolia and A. sylvestris have non-deep simple morphophysiological dormancy (MPD), since gibberellic acid (GA3) could substitute for cold stratification. This is the first report of non-deep simple MPD that is broken by cold stratification in the Apiaceae. Under natural conditions, physiological dormancy is broken by low temperature conditions during winter. Embryo growth and germination occur in a short time interval when temperatures start rising in early spring. Due to the fact that multiple environmental signals regulate dormancy, seedling emergence in these species is timed very accurately in spring.

Seeds vary widely in the sensitivity of germination to oxygen (O2) partial pressure, depending upon the species, temperature, dormancy state and physiological status of the seeds. Most analyses of the O2 sensitivity of germination have focused on final germination percentages and estimated the O2 percentage in air that is required to reduce germination to a given percentage (usually 50%). In contrast, we have applied a population-based threshold model utilizing time courses of germination to quantify three parameters related to seed germination sensitivity to O2 availability: the median base (or threshold) O2 percentage, the standard deviation of O2 thresholds among seeds in the population, and an oxygen–time constant that relates O2 percentage to germination timing. The model fits germination responses accurately across a wide range of O2 concentrations. The response to O2 was logarithmic in all cases, with the O2 percentage required for 50% germination ranging from 21% to as low as 0.005%, depending upon the species, the temperature and the seed dormancy level. Modelling indicated that some seeds can adapt to low O2 percentages and shift their thresholds to lower values over time. Lower temperatures decreased the minimum O2 threshold, as did after-ripening. Seed priming generally reduced the oxygen–time constant and increased the standard deviation of germination responses, but had relatively little effect on the O2 sensitivity per se. The population-based threshold model can be used to quantify the O2 sensitivity of seed germination and to predict germination rates and percentages when O2 availability is limiting.
