The mammalian rod bipolar, for which only one class has been identified, has been described as being hyperpolarizing by some investigators and depolarizing by others. We now report the effects of 2-amino-4-phosphonobutyrate (APB), a potent blocker of depolarizing bipolar cells, on visual behavior in the dark-adapted monkey. While in mesopic and photopic conditions only the monkeys' ability to detect incremental stimuli is impaired, under scotopic conditions all light mediated response in the monkey is eliminated. Assuming APB is acting on rod bipolars in the same fashion as it does on cone bipolars, we conclude that the primate rod bipolars all depolarize to light and that the ON and OFF channels are formed by the amacrine cell network.

The distribution of GABA-like immunoreactivity in the cat retina was studied through the use of preembedding immunocytochemistry for light microscopy and by postembedding immunogold techniques for electron microscopy. Staining was observed in both inner and outer plexiform layers. Approximately 30% of the somata in the amacrine portion of the inner nuclear layer were immunoreactive and included amacrine and interplexiform cells. Horizontal cells and a subpopulation of cone bipolar cells were also stained. In the ganglion cell layer, staining was observed in both small- and medium-sized neurons. GABA-labeled amacrine cells were presynaptic to somata of amacrine cells and to dendrites of amacrine, bipolar, and ganglion cells. Bipolar cells were a major target, receiving more than 60% of all labeled synapses in the inner plexiform layer. Many of these contacts were reciprocal synapses. These findings support a major role for GABA-labeled amacrines in providing feedback inhibition to bipolar cells in the inner retina.

We have examined the development of microglia in the rat retina, using a peroxidase-conjugated lectin derived from Griffonia simplicifolia. Retinas were studied from animals aged from E(embryonic day)12, just after the invagination of the optic cup and prior to the closure of the optic fissure, to adulthood. The lectin also proved a sensitive label for the endothelial cells of the developing retina. Our results provide some support for the view that microglia are derived from the monocyte-macrophage series of blood cells. At E12, most labeled cells were found at the vitreal surface, suggesting that they had come from the hyaloid circulation, while some had entered the retina and appeared to be migrating towards its ventricular surface. From E14 to early postnatal ages, most labeled cells had processes and resembled the amoeboid microglial cells described in silver carbonate staining studies (Ling, 1982). The number of labeled cells rose from about 700 to E14 to a peak of about 27,000 at P(postnatal day)7, and fell to about 19,600 by P12. As early as E16, a regularity was apparent in the distribution of microglial cells over the surface of the retina, the cells tending to avoid each other. Microglial cells are found throughout the thickness of the very young retina, but as the layers of the retina differentiate, they are increasingly restricted to the inner half of the retina. Our findings indicate that microglia enter the retina well before the period of neuronal death, making it unlikely that they invade the retina solely in response to cell death. Our results confirm however that, once in the retina, microglia become associated with, and appear to phagocytose, the pyknotic debris which appears during the period of neuronal death. They also become closely associated with the retinal vasculature. In the adult, the intensity of the labeling of microglia was much reduced. Those cells which were labeled appeared more differentiated, resembling the “resting microglia” described in earlier studies.

Normal, regenerating, and developing optic nerves of the goldfish were studied utilizing a monoclonal antibody (mAb) 1E1T which has specificity for Müller cells in the retina, radial ghal cells in the tectum, and non-neuronal cells in the optic nerve.

Sections of the normal optic nerve revealed longitudinally oriented chains of non-neuronal cells, that were 4–8 cells long The number of chains in the normal nerve was very few. In addition, short acellular septa, probably the connective tissue septa, were also labeled with mAb 1E1T. Sections of crushed optic nerves showed an increase in the antigen recognized by mAb 1E1T within the septa and new septa were now visualized. Furthermore, the existing septa were longer and extended the length of the optic nerve. The formation and elongation of the septa occurred as early as 3 day postcrush. Between 3 and 11 d postcrush, there was heavy labeling of the septa and a large accumulation of non-neuronal cells at the crush site. At 3 months postcrush, the accumulation of non-neuronal cells labeled by mAb 1E1T were no longer visible but heavy labeling of the septa was still apparent.

“Sustained” visual units, i.e. nonerasable (R1) and erasable (R2) units as commonly described, were recorded in the superficial layers of the rostral optic tectum of Rana esculenta and their neuronal activity (expressed as the mean firing frequency ) was quantitatively analyzed The mean value of the exponent α of the velocity function of the erasable R1 units was found to be 0.46 and the constant k to be about 24.7. The optimal stimulus diameter was equal to 2.6 deg. For the whole population of the erasable units, the exponent α was found to be between 0.36–0.75 (mean value ≈ 0.56, i.e. largely different from the values currently reported for such units) and was independent of the stimulus diameter (D = 0.6–6 deg). However, the constant k of the velocity function (range: 8.2–37.7) varied with D. Most of the erasable units (called “mixed R1-R2” units) therefore showed an R1-type velocity function with an exponent α ≈ 0.5 and a constant k ≈ 25 but reacted to other parameters as did “typical” R2 units: (i) their neuronal response was related to the stimulus diameter by a logarithmic function with a maximum obtained for D = 2.4 deg; and (ii) a variation of about 4–17% on either side of the average value of was observed, independent either of the stimulus velocity or of the diameter of the stimulus. Results are compared to known quantitative data, and the use of stimulus-response relationships as a tool for classification is discussed.

The normal postnatal development, the influence of age, and the effects of visual deprivation on the dopamine system in the retina of rhesus monkeys were examined. The lowest level of retinal dopamine was found at birth. By 3–4 weeks of age, the dopamine concentration had more than doubled. This level remained relatively constant in the retinas of older infants and of adult monkeys up to 34 yr of age. The level of the dopamine metabolite 3,4-dihydroxyphenylacetic acid (DOPAC) and the activity of tyrosine hydroxylase did not significantly change as a function of age during the postnatal life span.

Monocular occlusion of newborn or infant monkeys for 1–15 months with opaque contact lenses resulted in decreases in the retinal concentrations of dopamine and DOPAC relative to the concentrations in the same animals' unoccluded eyes. Occlusion also resulted in a lower level of tyrosine hydroxylase activity in the retina. Monocular eyelid suture from birth to 15 months of age resulted in less consistent alterations of retinal dopamine and DOPAC levels; tyrosine hydroxylase activity, however, was consistently reduced by lid suture. Thus, dopamine synthesis and metabolism, and the ontogenetic increase of the retinal dopamine level of rhesus monkey are reduced by light deprivation.

The spatial region involved in vernier acuity (integration region) was behaviorally determined in cats and compared with published human data and cortical scaling parameters for the two species. Integration region was estimated from vernier thresholds obtained with: (1) gaps of various sizes between the contiguous ends of the offset contours (gap effects); and (2) irrelevant contours placed at various distances from the vernier offset (interference effects). The gap studies showed that thresholds decreased as the gap grew from 0 to 4–7 min but then increased progressively with separations greater than 7 min. In the interference studies, thresholds were increased 2–6 times when the irrelevant contours were located within 40 min of the offset. Comparing these data with available human measures showed large species differences. Converting the interference distances for both species into cortical distances using published estimates of cortical magnification factor did not resolve the discrepancy. However, a comparison of the cat data with published human data, taken at an eccentricity at which the estimated human cortical magnification factor equals that of the cat, i.e. 7 deg, showed the effects of gap size and interference distance on vernier thresholds to be remarkably similar between the two species. This similarity supports the hypothesis that positional acuity depends upon mechanisms in striate cortex, and also suggests that such acuity may be mediated by a generic neural mechanism common to mammals.

Our previous light-microscopic study indicates that unilateral retinal impulse blockage with tetrodotoxin (TTX) causes a reversible decrease of cytochrome oxidase (CO) in alternating rows of metabolically active zones (puffs) in the adult macaque striate cortex (Wong-Riley & Carroll, 1984b). The goal of the present study was to determine if TTX blockade adversely affects all neurons or only a subpopulation of neurons within the puffs. Three major neuronal types were identified based on mitochondrial CO activities and morphological characteristics. Type A neurons were the most prevalent, consisting of small pyramidal and nonpyramidal neurons that received only symmetrical axosomatic synapses. They had little cytoplasm and relatively low levels of CO activity, and showed the least change with TTX treatment. Type B cells were medium-to-large pyramidal neurons that received exclusively symmetrical axosomatic synapses and were moderately reactive for CO. Impulse blockage caused a decrease in mitochondrial size and packing density, but somal size remained within the control range. Type C cells were medium-sized nonpyramidal neurons contacted by both asymmetrical and symmetrical axosomatic synapses. They contained abundant darkly reactive mitochondria and presumably are metabolically the most active. This cell type suffered the greatest decrease in somal size and packing density of mitochondria, particularly the darkly reactive ones. A rare fourth cell type, type D, was a small, darkly reactive nonpyramidal variety that gave rise to somatodendritic synapses. Their low occurrence prevented statistical analysis under normal and TTX-treated conditions. These data indicate that retinal impulse blockade is most detrimental to the metabolically most active neurons in the adult primate cortical puffs. The alterations are not permanent, because the effects of TTX are fully reversible (Carroll & Wong-Riley, 1987).

Unilateral retinal impulse blockage with tetrodotoxin (TTX) induces reversible shrinkage and decreased cytochrome oxidase (CO) activity in alternate rows of supragranular, CO-rich puffs in the adult macaque striate cortex (Wong-Riley & Carroll, 1984b: Carroll & Wong-Riley, 1987). The present study extended the findings to the electron-microscopic (EM) level to determine if various neuropil profiles in control puffs exhibit heterogeneous levels of CO activity, and whether specific processes were more susceptible to intravitreal TTX than others.

Within the neuropil of control puffs, 60% of the total mitochondrial population resided in dendrites, and the majority of dendritic mitochondria were highly reactive for CO. Axon terminals forming symmetrical synapses also contained darkly reactive mitochondria, whereas those forming asymmetrical synapses possessed very few and mainly lightly reactive mitochondria. Unmyelinated axon trunks, myelinated axons, and glia all exhibited low levels of CO activity. Synaptic count revealed a 3:1 ratio of asymmetrical to symmetrical synapses.

Intravitreal TTX for 2–4 weeks adversely affects dendrites and symmetrical terminals much more so than other neuropil processes. There was a general decrease in darkly and moderately reactive mitochondria and an increase in lightly reactive mitochondria throughout the puffs, especially in dendrites. This indicates that afferent blockade is more detrimental to processes of higher metabolic activity. Changes also differed between central and peripheral regions of puffs, and indications of axonal and synaptic reorganization were more evident in the latter. Thus, stabilization of neuronal structure and synapses appears to be activity-dependent even in the adult. A working model of these metabolic and morphological responses to chronic TTX is proposed.

The two main receptor subtypes for 5-hydroxytryptamine (5HT) were measured and localized in visual cortical areas of macaque monkey. [3H]5HT was used to label all 5HT-1 receptor subtypes and [3H]ketanserin was used to label 5HT-2 receptors. Both receptor types could be demonstrated in membranes prepared from macaque primary visual cortex. The specificity of these ligands for 5HT-1 or 5HT-2 receptors was demonstrated by the pharmacological profile of inhibitors of the specific binding. 5HT-1A receptor sites were detected by displacement experiments and by direct labeling with [3H]8-hydroxy-2(di-n-propylamino) tetralin 8OH-DPAT. Receptor autoradiography showed that the distribution of these receptor subtypes varied from one part of visual cortex to another. 5HT-1 receptors, labeled with [3H]5HT were present in several bands through layer IV of primary visual cortex with the densest band seen in and above layer IVA: another band was in lower layer VI. The band in layer VI was predominantly 5HT-1A sites. There were two main bands of 5HT-2 receptor sites, the most prominent around the IV/V boundary, and the other extending from layer IVA upwards. Adjacent areas showed 5HT receptors in a broad band corresponding to layer IV. 5HT-1A sites were found in superficial layers of adjacent areas, except V2. These layering patterns did not correspond precisely with cytoarchitectonic layering, nor with the pattern of 5HT-containing presynaptic fibres in published reports. It is important, therefore, in considering the role of the 5HT-containing neurons in cortical function to take account not only of the anatomy of the presynaptic terminals, but also of the postsynaptic receptors upon which the released transmitter will act, and their location within the cortex.
