The P3 components (P3a and P3b) of the event-related potentials have been analysed in sixteen in-patients with a major depressive episode (DSM III-R) according to their subtype - anxious-agitated and impulsive versus retarded and blunted-affect - assuming that these two opposite dimensions of the depressive mood would rely on different physiopathological mechanisms. The P3a component, which indexes automatic cognitive processes, was dramatically decreased in the retarded and blunted-affect group. This suggests that this special type of cognitive dysfunction in depression is rather related to negative symptoms than to depression itself.

P3 is the most prominent of the electrical potentials of the human electroencephalogram that are sensitive to psychological variables. According to the most influential current hypothesis about its psychological significance [E. Donchin's], the “context updating” hypothesis, P3 reflects the updating of working memory. This hypothesis cannot account for relevant portions of the available evidence and it entails some basic contradictions. A more general formulation of this hypothesis is that P3 reflects the updating of expectancies. This version implies that P3-evoking stimuli are initially unexpected but later become expected. This contradiction cannot be resolved within this formulation. The alternative “context closure” hypothesis retains the concept of “strategic information processing” emphasized by the context updating hypothesis. P3s are evoked by events that are awaited when subjects deal with repetitive, highly structured tasks; P3s arise from subjects' combining successive stimuli into larger units. The tasks in which P3s are elicited can accordingly be classified in terms of their respective formal sequences of stimuli. P3 may be a physiological indicator of excess activation being released from perceptual control areas.

The effect of mild head injury on event-related potential (ERP) correlates of Stroop task performance was explored with the aim of further elucidating the basis of processing impairments after mild head injury. Computer- and card-based Stroop tasks were employed to assess attention function. A sequence of incongruent color words were presented followed by a sequence of congruent color words (printed in congruent colors). Control performance was equivalent on computer- and card-based versions of the incongruent task and faster on the congruent card task than the preceding congruent computer task. The mild head injury group were as fast as controls on the computer-based task but made more errors. However, they were relatively slower on both the congruent and incongruent parts of the card-based task and made more errors in the incongruent task. ERP correlates of computer-based Stroop task performance suggested a greater allocation of attention resources in the incongruent condition in both groups in the form of relatively greater negativity in the latency range 350 to 450 ms with a distribution consistent with the activation of the anterior cingulate gyrus. In addition the mild head injured group showed relatively greater enhancement than the control group in this latency range in both congruent and incongruent conditions. There was, however no evidence of reduced amplitude P1, N1, N2, or P3b deflections. Trails, digit symbol, digit span and auditory verbal learning tests (AVLT) were also administered. Mild head injured participants were impaired only on the AVLT. The finding of greater ERP negativity in the mild head injured group is consistent with greater allocation of attention resources to achieve equivalent performance in the computer-based Stroop task. The present study investigated the effect of target stimulus probability on the P3 component of the respiratory-related evoked potential (RREP). A single respiratory stimulus paradigm was employed where normal breaths served as standard stimuli and occluded breaths presented at various probability levels served as target stimuli. EEG was recorded from 29 channels in young adults in two conditions. In Condition 1, occlusions were presented at target probabilities of 0.5, 0.33, 0.25, 0.2, 0.1, and 0.05, that is, every 2nd, 3rd, 4th, 5th, 10th, and 20th breath. Order of target probability presentation varied randomly. Condition 2 used the same target probabilities, but held breathing rate constant by using a paced respiration task. Results indicated that P3 amplitude was significantly affected by stimulus probability, such that the component showed an inverse relationship with probability. This effect was similar between conditions. However, P3 amplitude reduced and latency increased when the secondary task of paced respiration was introduced. These data suggest that models developed to describe the effect of stimulus probability on the amplitude of visual and auditory P3 components are applicable to the respiratory somatosensory modality.

Both the amplitude and latency of P300 vary with changes in stimulus parameters. Stimuli at intensities or pitch separations near threshold evoke a smaller and later P300. P300 is also affected by extraneous stimulus parameters in tasks where stimulus frequency separation is large and stimuli are well above intensity thresholds. For example, the presence of background white noise when tones are suprathreshold and easily detectable has been reported to increase P300 latency. However, the effects of background masking noise on P300 amplitude and scalp topography have not been reported. Subjects performed an oddball task both in the presence and in the absence of background noise. Performance accuracy was unaffected by background noise. P300 showed latency increases when noise was present, but P300 peak amplitude was unaffected. P300 scalp topography was stable across both conditions. P300 latency is affected by background noise, even when performance is not, but amplitude and amplitude topography remain unaffected.

We examined changes of event-related potentials (ERPs) while participants learned stimulus-to-stimulus relations in an S1-S2 task. The design allowed for separating processes of associative learning from nonspecific effects. Participants had to respond to S2 by a left or right key-press dependent on S2 identity (letter W or M). Preparation for S2 could be improved by using the associative information given by S1. The S1 was an arrow pointing to the left or right. In combination with its color, arrow direction was informative about location and identity of S2, but participants were not informed about the relevance of color. Arrows in two of the colors were fully predictive for the S2 whereas the third color gave no valid information. This third stimulus controlled for habituation and procedural learning. Six blocks with 200 trials each and all three S1 colors in random order were presented. Behavioral and ERP differences in each block between “learning” and control trials were used to identify processes of associative learning. Several effects of associative learning were identified indicating the involvement of specific stages of information processing: a continuous increase of P3 amplitude evoked by S1 was accompanied by a decrease of P3 evoked by S2. These changes reflected the modifications of stimulus weights for response selection and the strengthened association between the two stimulus complexes in the time course of learning. The related motor preparation benefited from learning too, expressed in a decrease of CNV amplitude and an increase of LRP amplitude. Finally a decrease of N1 amplitude evoked by S2 indicated the reduced need to allocate spatial attention to the S2 location according to the learned meaning of S1.

Eukaryotic RNase P and RNase MRP are endoribonucleases composed of RNA and protein subunits. The RNA subunits of each enzyme share substantial secondary structural features, and most of the protein subunits are shared between the two. One of the conserved RNA subdomains, designated P3, has previously been shown to be required for nucleolar localization. Phylogenetic sequence analysis suggests that the P3 domain interacts with one of the proteins common to RNase P and RNase MRP, a conclusion strengthened by an earlier observation that the essential domain can be interchanged between the two enzymes. To examine possible functions of the P3 domain, four conserved nucleotides in the P3 domain of Saccharomyces cerevisiae RNase P RNA (RPR1) were randomized to create a library of all possible sequence combinations at those positions. Selection of functional genes in vivo identified permissible variations, and viable clones that caused yeast to exhibit conditional growth phenotypes were tested for defects in RNase P RNA and tRNA biosynthesis. Under nonpermissive conditions, the mutants had reduced maturation of the RPR1 RNA precursor, an expected phenotype in cases where RNase P holoenzyme assembly is defective. This loss of RPR1 RNA maturation coincided, as expected, with a loss of pre-tRNA maturation characteristic of RNase P defects. To test whether mutations at the conserved positions inhibited interactions with a particular protein, specific binding of the individual protein subunits to the RNA subunit was tested in yeast using the three-hybrid system. Pop1p, the largest subunit shared by RNases P and MRP, bound specifically to RPR1 RNA and the isolated P3 domain, and this binding was eliminated by mutations at the conserved P3 residues. These results indicate that Pop1p interacts with the P3 domain common to RNases P and MRP, and that this interaction is critical in the maturation of RNase P holoenzyme.

Identification of the necessary stimulus properties to elicit the stimulus preceding negativity (SPN) has been the impetus for numerous research studies. The current study was conducted to explore the possibility that the SPN is an index of cognitive resource allocation. An auditory warning stimulus (S1) indicated whether an easy or difficult discrimination would occur at S2. The SPN was collected before a nonmotor discrimination task (S2) that consisted of identifying the higher of two bars. To eliminate the influence of motor processing prior to S2, a button press on the side of the higher bar was held until perception of a response cue (S3). Additionally, P3, contingent negative variation (CNV), and behavioral measures were collected to assist in assessing the SPN. Results indicated that although the SPN exhibited increased negativity, no differences were observed based on task difficulty. However, task difficulty did affect P3 data for both the warning tone and the discrimination task, an effect not observed for the CNV. Overall, the data did not support that hypothesis that the SPN provides an index of cognitive demand.

This study investigated the functional significance of the N2 response to novel stimuli. In one condition, background, target, and deviant stimuli were simple geometric figures. In a second condition, all stimulus types were unfamiliar/unusual figures. In a third condition, background and target stimuli were unusual figures and deviant stimuli were simple shapes. Unusual figures, whether they were deviant, target, or background stimuli, evoked larger N2 responses than their simple, familiar counterparts. N2 elicited by an unusual background stimulus was larger than that evoked by simple, deviant stimuli, a pattern opposite that exhibited by the subsequent P3. Deviance from immediate context had limited influence over N2 amplitude. The results suggest that novelty N2 and novelty P3 reflect the processing of different aspects of “novel” visual stimuli. The novelty P3 is particularly sensitive to deviation from immediate context. In contrast, the novelty N2 is sensitive to deviation from long-term context that renders a stimulus unfamiliar and difficult to encode.

This study investigated the relationship between resistive load magnitude, load magnitude estimation, and the respiratory-related evoked potential. In Part 1, 10 healthy subjects estimated the magnitude of five inspiratory resistive loads. Two subjects were shown to have a markedly reduced slope of the magnitude estimation-resistive load relationship and were suggested to be “poor perceivers” of respiratory stimuli. In Part 2, evoked potentials were recorded from the same 10 subjects using the same resistive loads as Part 1. A log-log plot of the group averaged P1 amplitudes showed a linear relationship with resistive load. Aberrant P3 components were seen in the 2 poor perceiving subjects and one of the 2 showed no late response. In the other 8 subjects, P3 varied as a function of resistive load, being augmented to larger loads. These results provide evidence that P3 may be a key index of the perception of respiratory sensitivity and effort.

The present study assessed the effects of occlusion duration and attention on components of the respiratory-related evoked potential (RREP). Twenty-nine channel evoked potential recordings were obtained from 12 young adults exposed to a pseudorandom sequence of 100-, 200-, 400-, and 800-ms inspiratory occlusions, under attend and ignore conditions. Results demonstrated that the duration of an inspiratory occlusion does not affect RREP components systematically, highlighting the importance of the onset of the occlusion in producing the cortical responses. Attention resulted in augmentation of the N1, P2, and P3 components but did not affect the early latency Nf and P1 components. P1, N1, and P3 occurred with shorter latencies in the attend condition. One subject with poor duration estimation ability displayed substantially delayed P3 latency. This result highlights the relationship between P3 and perception of respiratory somatosensory information.

In the present study it was investigated whether the smaller P3s in attention-deficit hyperactivity disorder (ADHD) children are caused by a shortage of capacity underlying P3 processes or whether they are due to a capacity allocation problem. Also, effects of methylphenidate on these processes were investigated. Performance and event-related potentials (ERPs) of 14 ADHD and 14 control children were measured using an irrelevant-probe technique. Three types of task irrelevant visual probes (standards, deviants, and novels) were presented against the background of two visual tasks that varied in task difficulty. The parietal P3 wave was measured in response to task stimuli and probes. ADHD subjects made significantly fewer correct detections than normal controls in both the easy and the hard tasks. Controls showed an enhanced P3 to task-relevant stimuli in the hard task, whereas ADHD children did not. Probe (novel) P3 amplitudes decreased from the easy to the hard task to the same extent in both groups. Methylphenidate enhanced the percentage of correct responses and task P3 amplitudes in both the easy and the hard task but probe P3 amplitudes were not influenced by methylphenidate. It was concluded that ADHD children do not suffer from a shortage in attentional capacity; rather, the evidence is in favor of a problem with capacity allocation. Furthermore, methylphenidate had enhancing effects on performance and ERPs, but did not improve the capacity-allocation deficit.

To assess the processing stages involved in attention shifting and response selection tasks in children, we recorded event-related potentials (ERPs) and performance measures during a variant of the Posner paradigm. Subjects responded to visual targets, either preceded by a spatial cue (valid = same side; invalid = opposite side) or presented uncued. Valid targets evoked high-amplitude P1 responses, single-peaked P3s, and the shortest reaction times (RTs). Invalidly cued stimuli evoked delayed RTs, resulting in part from incorrectly oriented attention (decreased P1) leading to delayed target processing (belated N2-P3). Invalid targets also evoked a positive slow wave attributed to prolonged response selection due to cue/target incompatibility. Uncued stimuli elicited the longest RTs, unexplained by deficits in target detection or response selection, which likely resulted from a deficit in motor preparation due to the lack of warning signal. This method may be applied in clinical settings to disentangle selective processing deficits in target detection, response selection, or motor preparation stages.

In this study, we examined changes in the event-related potential (ERP) to stimuli with and without reproductive significance occurring during the menstrual cycle. Eleven spontaneously cycling women were tested during three menstrual phases (menses, ovulatory phase, luteal phase) differing in plasma concentrations of gonadal hormones. ERPs were recorded while subjects were presented with slides showing pictures from four different stimulus categories (sexual stimuli, babies, people occupied with body care, ordinary people). Slides were presented randomly in the context of two tasks, requiring either affective processing (i.e., to judge the emotional content of a slide as positive, neutral, or negative) or structural processing (i.e., to estimate the number of parallel thin lines inserted in each picture). Menstrual phase primarily affected a late positive component (LPC) peaking 550–600 ms poststimulus. The effects were as follows: (i) During the ovulatory phase, amplitude of the LPC to sexual stimuli was larger than that evoked by the other stimulus categories. (ii) This relationship was not apparent during the other menstrual phases or (iii) during the ovulatory phase when the task required structural processing. The ovulatory increase in LPC positivity to sexual stimuli suggests a greater valence of these stimuli during a phase of increased sexual desire. The data indicate a specific effect of the menstrual cycle on the processing of sexual stimuli that increases with deeper emotional processing.

Event-related potentials (ERPs) were recorded during a go/no-go reaction time (RT) task in which subjects responded to rare target tones and withheld response to frequent tones. In a predictable condition, a rare visual stimulus signalled the impending occurrence of a rare tone, whereas a frequent visual stimulus signalled that a frequent tone would be presented. The rare visual stimuli elicited P3, associated with violations of expectations, whereas the rare tones, being predictable, did not. The rare tones elicited the mismatch negativity (MMN), a component associated with preattentive deviance detection, despite the fact that they were expected. RT was faster in the predictable condition than in another condition in which the tones were not predictable. The P3 and RT data indicate, respectively, that subjects anticipated and were ready to respond to the target tones. The MMN result indicates that immediately before target tones, the preattentive system underlying the MMN was set for frequent tones, being unaffected by the information available to the higher order system. Thus, the higher order cognitive system associated with P3 and the lower order cognitive system associated with the MMN were prepared simultaneously for opposite events.

The purpose of the study was to examine the hypothesis that distractibility is a fundamental characteristic of mild closed head injury (MHI). The claim that cognitive symptoms in MHI are due to a mild type of frontotemporal injury was also investigated. Cognitive event-related potentials (ERPs), accuracy and reaction time to target stimuli in a dichotic listening paradigm, and neuropsychological test results were studied in patients with MHI (N = 15), patients with verified frontal lobe damage (N = 10), and healthy controls (N = 13). Information processing reflecting target detection (N2, P3b) and sustained selective attention (processing negativity) was studied. The MHI and frontal patients did not differ on behavioral measures, except that the MHI group had significantly longer reaction times to target stimuli in the ERP task. Both patient groups had deviant ERPs compared with controls, but their ERP patterns differed in important respects. Contrary to expectations, the MHI patients had the most abnormal ERPs. They showed significantly smaller N2 and Nd amplitudes than frontal patients and controls, indicating that the mediating cognitive mechanisms were not equivalent in MHI and frontal injury. The data suggest that MHI patients allocated less processing resources to the task than either the control subjects or the patients with frontal lobe damage.
