Biobased and biodegradable mulches (BDM) are a potentially sustainable alternative to polyethylene plastic mulch because they can be tilled into the soil at the end of the growing season. However, their degradation rate in the soil is uncertain, limiting their on-farm adoption. The objective of this study was to determine whether organic soil management practices could be leveraged to speed degradation of two potentially BDM across two diverse agroecoregions [Lincoln (LNK) and Scottsbluff (SBF), NE, USA]. Management treatments included compost, compost extract, cover crops, all three of these practices combined and a control. The two mulch types studied were a nonwoven polylactic acid fabric with embedded wood particles (PLA), and a starch-polyester mulch film (BLK). Mulches were applied in spring 2017 for vegetable production and removed in fall after harvest. Recovered mulch was sectioned into squares 10 cm2 and buried in mesh bags for 22 months. Mulch degradation, and soil chemical, physical and biological properties were measured at four times over 2 years. Management treatments applied seasonally across 2 years led to expected changes in soil properties, yet they had no effect on mulch degradation. Instead, mulch degradation was driven by the interaction of location and mulch type. The BLK mulch had degraded by 98% at LNK after 12 months, but only by half after 22 months at SBF. Degradation of PLA after 22 months was similar between locations with 29 ± 4% mulch mass remaining at SBF and 33 ± 4% remaining at LNK. Climate and soil characteristics at each location were strong determinants of mulch degradation. Specifically, soils at LNK were finer textured, lower in pH, higher in soil water content, organic matter and nitrates, and with greater bacterial abundance compared to SBF. The strong location by mulch type interaction observed could inform the development of regionally specific predictive models of degradation.

Biodegradation of chlorpyrifos under the influence of fertilizer application and climate factors such as elevated CO2, temperature and moisture was studied. Soybean was grown in control, inorganic, organic and integrated (both inorganic and organic) fertilized fields. Rhizospheric soils collected during the vegetative growth phase were amended with chlorpyrifos (10 μg/g soil) and incubated under different climate factors. The climate factors were CO2 concentration (400, 800 ppm), temperature (25, 45°C) and moisture-holding capacity (60, 100%). Chlorpyrifos degradation rate varied from 0.28 to 0.65 μg/g soil/d. The abundance of 16S rRNA gene copies of eubacteria varied from 13 × 106 to 7 × 105/g soil. Actinomycetes-specific 16S rRNA gene copies were in the range of 62.5 × 105 to 18.5 × 103/g soil. Microbial abundance was high in organic amended soil and low in control soil irrespective of climate factors. Elevated CO2 and high temperature inhibited (P < 0.05) chlorpyrifos degradation rate and the abundance of 16S rRNA genes of eubacteria and actinomycetes. Chlorpyrifos degradation followed as: organic > integrated > inorganic > control. The degradation rate was positively correlated (P < 0.01) with the soil organic C, available N, water-stable aggregates and mean weight diameter of the soil aggregates of soil. Principal component analysis denoted temperature and fertilizer as the major components of variation. The study highlights that elevated CO2 and temperature affect chlorpyrifos biodegradation; however, the effect can be alleviated by the amendment of organic fertilizer.

The aim of the present study was to map the painting materials, degradation processes, and biological features present on the mural painting in the church of St. Mary in Beram (Croatia) to study their possible interaction and produce information helping the preservation of this valuable painting. The research was conducted on micro samples of painting materials taken from different sites along the painting and the characterization of the present fungal species was carried out. The painting samples, together with observable patinas and degradation products, were studied by optical microscopy (OM), scanning electron microscopy, energy-dispersive spectroscopy (SEM/EDS), Fourier-Transform Infrared spectroscopy, and powder X-ray diffraction. Fungal diversity was studied using cultivation methods followed by OM and SEM analyses in addition to molecular analysis. The results contribute to the characterization of the original painting materials, successively added materials and occurred interventions, to the understanding of degradation progressions and fungal biotransformation processes. A mineral, cumengite, a copper-based pigment extremely rarely used in art, was found. Its occurrence together with barium sulfate, gypsum, and calcium oxalate possibly produced by microbiological activity was studied and information was added regarding the composition of painting materials in St. Mary church mural cycle.

Before wastewaters can be released into the environment, they must be treated to reduce the concentration of organic pollutants in the effluent stream. There is a growing concern as to whether wastewater treatment plants are able to effectively reduce the concentration of micropollutants that are also contained in their influent streams. We investigate the removal of micropollutants in treatment plants by analysing a model that includes biodegradation and sorption as the main mechanisms of micropollutant removal. For the latter a linear adsorption model is used in which adsorption only occurs onto particulates.

The steady-state solutions of the model are found and their stability is determined as a function of the residence time. In the limit of infinite residence time, we show that the removal of biodegradable micropollutants is independent of the processes of adsorption and desorption. The limiting concentration can be decreased by increasing the concentration of growth-related macropollutants. Although, in principle, it is possible that the concentration of micropollutants is minimized at a finite value of the residence time, this was found not to be the case for the particular biodegradable micropollutants considered.

For nonbiodegradable pollutants, we show that their removal is always optimized at a finite value of the residence time. For finite values of the residence time, we obtain a simple condition which identifies whether biodegradation is more or less efficient than adsorption as a removal mechanism. Surprisingly, we find that, for the micropollutants considered, adsorption is always more important than biodegradation, even when the micropollutant is classified as being highly biodegradable with low adsorption.

The purpose of this study was to develop a simple and economic method for the preparation of porous bioceramics with controllable macrostructure. Raw materials, including very small organic foam balls as the pore-creating reagent, wax (or paraffin) as the solvent, and oleic acid as the surface active agent, were selected along with bioactive materials such as β-TCP ceramic powder as the main component. The selected components were mixed into a slurry at 30–120ºC and shaped into a green body with a hot die-casting machine at 30–90ºC. The green body was then sintered and porous bioceramics were obtained. The main characteristics of porous bioceramics such as weight loss, compressive strength, connection and size of pores, percolation rate of water, apparent porosity and bulk density were measured. The results indicated that the apparent porosity and the specific surface area were large; the pores were connected in three dimensions and the compressive strength was >1.6 MPa. This study demonstrated that the methods used here are simple and effective in generating porous bioceramics with controllable macrostructures.

Elemental behaviour, during the process of weathering of glazed sekishu roof-tiles affected by Lecidea s.lat. sp. (a crustose lichen), was investigated using optical and fluorescence microscopy, field emission scanning electron microscopy (FE-SEM) and transmission electron microscopy. Sekishu roof tiles have an opaque reddish brown glaze on their surfaces which consist of an alkali feldspar-type X-ray amorphous glass recrystallized at 1200°C. Optical and fluorescence microscopy revealed the presence of corrosion pits (at a depth of ∼50 μm) at the lichen-glaze interface. Elemental mapping by FE-SEM identified the concentrations of Ti and Fe in the section of the glazed tile analysed. The behaviour of C was correlated with those elements, suggesting the possibility of biomineralization.

It is UK Government policy to dispose of higher activity radioactive waste through geological disposal into an engineered deep underground geological disposal facility (GDF; DECC, 2014). Those wastes include low-level (LLW) and intermediate-level (ILW) radioactive wastes that are very heterogeneous, containing a range of inorganic and organic materials, the latter including cellulosic items. After closure of the GDF, eventual resaturation with groundwater is expected, resulting in the development of a hyperalkaline environment due to the proposed use of a cementitious backfill. Under these high-pH conditions, cellulose is unstable and will be degraded chemically, forming a range of water-soluble, low molecular weight compounds, of which the most abundant is isosaccharinic acid (ISA). As ISA is known to form stable soluble complexes with a range of radionuclides, thereby increasing the chance of radionuclide transport, the impact of microbial metabolism on this organic substrate was investigated to help determine the role of microorganisms in moderating the transport of radionuclides from a cementitious GDF. Anaerobic biodegradation of ISA has been studied recently in high-pH cementitious ILW systems, but less work has been done under anaerobic conditions at circumneutral conditions, more representative of the geosphere surrounding a GDF. Here we report the fate of ISA in circumneutral microcosms poised under aerobic and anaerobic conditions; the latter with nitrate, Fe(III) or sulfate added as electron acceptors. Data are presented confirming the metabolism of ISA under these conditions, including the direct oxidation of ISA under aerobic and nitrate-reducing conditions and the fermentation of ISA to acetate, propionate and butyrate prior to utilization of these acids during Fe(III) and sulfate reduction. The microbial communities associated with these processes were characterized using 16S rRNA gene pyrosequencing. Methane production was also quantified in these experiments, and the added electron acceptors were shown to play a significant role in minimizing methanogenesis from ISA and its breakdown products.

Field and laboratory studies were conducted to examine effects of prior butylate use on biodegradation of subsequent applications of butylate and four other carbamothioate herbicides. Bioassays were used to demonstrate reductions of butylate and EPTC activity in four soils preconditioned by annual butylate applications. Combining these herbicides with dietholate, an enzyme inhibitor, prolonged persistence and restored normal herbicidal activity. Expected herbicidal efficacy occurred in adjacent plots with no history of carbamothioate use. Prior applications of butylate resulted in cross-adapted degradation of EPTC but not vernolate, pebulate, or cycloate. When samples of Cecil soil were treated in the laboratory with 14C-butylate, evolution of 14CO2 was significantly higher from soils that had received repeated field applications of butylate. The use of 14C-herbkides could be used to predict problem soils and potential herbicide failure.

Degradation of diphenamid in soil with accelerated degradation and in nontreated control soil and the involvement of soil microorganisms in these processes were investigated. Soil with accelerated degradation and mixed bacterial cultures originating from the same soil degraded diphenamid and its monodemethylated metabolite (diphen M-1) much faster than the control. The bidemethylated derivative (diphen M-2) was degraded much more slowly than diphenamid or diphen M-1. The abundance of fungi capable of degrading diphenamid was similar in the soils with and without accelerated degradation. Degradation of diphenamid by mixed bacterial cultures from a soil with accelerated degradation was much faster than by a culture from nontreated soil and was suppressed by the fungicides thiram and fentin acetate. These two fungicides, as well as the bactericide chloramphenicol, suppressed diphenamid degradation in mixed bacterial cultures. The antifungal cycloheximide and the actinomycete suppressor PCNB did not affect the degradation of diphenamid. Metabolism of 14C-diphenamid in a soil with accelerated degradation and in mixed bacterial cultures originating from the same soil showed similarities with regard to evolution of 14CO2 and production of certain metabolites, while the metabolism of diphenamid by Fusarium, which rapidly degrades diphenamid in cultures, was different with regard to the above parameters. This study suggests that development of accelerated degradation in a soil involves a shift in population and/or in activity of microbial degraders in favor of bacteria. It appears that the accelerated degradation occurred by inducing oxidative reaction which involves demethylation of diphenamid.

Laboratory studies were conducted to investigate the processes of imazaquin and imazethapyr degradation in soil. Microbial degradation of 14C-imazaquin and 14C-imazethapyr was monitored by measuring 14CO2 evolution compared to nonsterile soil. 14CO2 evolution was greatest from carboxyl-labeled imazaquin and imazethapyr compared to ring-labeled imazaquin and imazethapyr. A corn root bioassay indicated nearly complete loss of herbicidal activity in nonsterile soil after 5 mo, but herbicide activity was reduced only 14% in sterile soil. 14CO2 evolution more than doubled when soil temperature increased from 15 to 30 C. Total CO2 production responded similarly. Degradation of imazaquin and imazethapyr increased as soil moisture increased from 15% (-2.4 MPa) to 75% of field capacity (-0.03 MPa).

Accelerated degradation of the herbicide diphenamid (N,N-dimethyl-α-phenylbenzeneacetamide) was investigated in Israeli soils. Repeated application of this herbicide in the soil in the laboratory enhanced its degradation, which increased with an increasing number of applications. After the fourth application nearly 100% of the herbicide was degraded within 5 days of incubation, whereas only slight degradation was observed during the first 25 days in a previously untreated soil. Accelerated degradation was also observed in a soil collected from a field with previous diphenamid treatments. Fumigation with methyl bromide or treating the soil with the fungicide fentin acetate (triphenyltin acetate) was effective in decreasing the accelerated degradation of diphenamid, whereas the fungicides TMTD (tetramethylthiuram disulfide) and TBZ [2-(4-thiazolyl)benzimidazole] were only partially effective in inhibiting accelerated degradation. In the laboratory, fentin acetate prevented the degradation of diphenamid applied to previously untreated soil. Several fungi capable of degrading diphenamid were isolated from soils with or without accelerated degradation.

Mg-based implants have promising applications as biodegradable materials in medicine for orthopedic, dental, and cardiovascular therapies. During wear and degradation microdebris are released. Time-lapse multidimensional microscopy (MM) is proposed here as a suitable tool to follow, in fixed intervals over 24-h periods, the interaction between cells and particles. Results of MM show interactions of macrophages (J774) with the magnesium particles (MgPa) that led to modifications of cell size and morphology, a decrease in duplication rate, and cell damage. Corrosion products were progressively formed on the surface of the particles and turbulence was generated due to hydrogen development. Changes were more significant after treating MgPa with potassium fluoride. In order to complement MM observations, membrane damage as detected by a lactase dehydrogenase (LDH) assay and mitochondrial activity as detected by a WST-1 assay with macrophages and osteoblasts (MC3T3-E1) were compared. A more significant concentration-dependent effect was detected for macrophages exposed to MgPa than for osteoblasts. Accordingly, complementary data showed that viability and cell cycle seem to be more altered in macrophages. In addition, protein profiles and expression of proteins associated with the adhesion process changed in the presence of MgPa. These studies revealed that time-lapse MM is a helpful tool for monitoring changes of biodegradable materials and the biological surrounding in real time and in situ. This information is useful in studies related to biodegradable biomaterials.

The present study characterizes the main deterioration mechanisms affecting the early 17th frescoes of Casa de Fresco, the only known example in Portugal of a semi-underground leisure room richly decorated with a balcony over a water well. Frescoes from the vault are at risk due to salt weathering and biodeterioration. The aim of the research was identification of the deterioration materials, determination of their origin, and their effect on the frescoes before future intervention. Scanning electron microscopy with an energy-dispersive X-ray detector (SEM-EDS) was used to determine salt morphology and microanalysis. The mineralogical characterization was performed by X-ray powder diffraction, complemented with µ-Raman and µ-Fourier transform infrared spectroscopy. Biological assessment was evaluated with optical microscopy and SEM-EDS. Bacterial and fungal isolation and identification were performed using standard culture media and methods according to Bergey’s Manual of Systematic Bacteriology and from the Compendium of Soil Fungi. The results show that Ca and Ca-Mg carbonates from the paint renderings are the predominant salt species affecting the site. Bacterial strains from the genera Bacillus and Pseudomonas and fungal strains from the Cladosporium spp. and Penicillium spp. were isolated in the salt formations, within and between the mortar layers. Azurite, malachite, and smalt paint layers are the most affected by the weathering conditions.

This article reports the structure and morphology of the in vivo interface between implants composed of either α tricalcium phosphate (αTCP) or αTCP doped with 3.0 wt% dicalcium silicate (αTCPss) ceramic, and natural bone of rabbit tibias. Both interfaces developed a new bone layer in direct contact with the implants after 4 and 8 weeks of implantation. The specimens were examined using analytical scanning and transmission electron microscopy, up to the lattice plane resolution level. Degradation processes of the implants developed at the interfaces encouraged osseous tissue ingrowth into the periphery of the material, changing the microstructure of the implants. The ionic exchange initiated at the implant interface with the environment was essential in the integration process of the implant, through a dissolution–precipitation–transformation mechanism. The interfaces developed normal biological and chemical activities and remained reactive over the 8-week period. Organized collagen fibrils were found at the αTCPss/bone interface after 4 weeks, whereas a collagen-free layer was present around the Si-free αTCP implants. These findings suggest that the incorporation of silicate ions into αTCP ceramic promotes processes of the bone remodeling at the bone/αTCPss interface, hence the solubility rate of the αTCPss material decreased.

Two new approaches were examined, aimed at increasing survival of the house fly (Musca domestica L.) larvae under mass-rearing conditions of a biodegradation facility: modification of the larval substrate and dispersal of the eggs during inoculation. The two types of pig manure used in this study (manure with sawdust and manure without sawdust) differed in terms of larval survival and nutritional value for the house fly larvae. Larval survival in manure without sawdust in the control treatment was low (46.8 ± 2.1%) and its nutritional value for the larvae were high. Addition of 5.7% of previously biodegraded manure did not significantly affect larval survival (52.3 ± 1.9%), but larval development was faster and the pupae were significantly smaller (14.28 ± 0.4 mg) compared to the control (16.29 ± 0.5 mg). Using alternative substrate for incubation of eggs and first-instar larvae significantly increased larval survival (63.3 ± 3.3%) and decreased the mean weight of produced pupae (14.39 ± 0.71 mg). Overall, the weight of recovered biomass in the alternative substrate treatment increased by 14.3 kg ton−1 of manure compared to the control. Larval survival in manure with sawdust was generally higher than 70%, but its nutritional value for the larvae was lower than in manure without sawdust. Dispersal of eggs over the surface of manure with sawdust significantly affected larval survival and mean weight of pupae. Larval survival was significantly lower (59.2 ± 4.0%) and pupae were significantly heavier (18.45 ± 0.8 mg) when eggs were applied to a small area on the manure surface (spot treatment), as compared to diagonal, Z-line and multiple zig-zag dispersal (72.5 ± 2.4 to 74.6 ± 3.0% and 14.76 ± 0.6 to 15.97 ± 0.6 mg, respectively). No significant differences were observed in larval survival or mean weight of pupae when comparing the diagonal, Z-line and multiple zig-zag dispersal patterns. Implementation of the techniques which improve larval survival and increase the weight of produced fly biomass may decrease demand for production of house fly eggs and, therefore, reduce the maintenance costs of adult colony, as well as increase the revenue earned by selling the products.

Introduction. En Tunisie, les dattes de type Deglet Nour sont réputées pour leur couleur jaune doré et elles présentent une valeur marchande élevée ; elles constituent la majorité des dattes exportées vers l’Europe. L’objectif de nos travaux a été d’étudier l’effet de l’oxygène et de la température sur la dégradation de la couleur de ce type de dattes durant la période d’entreposage. Matériel et méthodes. L’évolution de la couleur de dattes du type Deglet Nour stockées sous humidité relative constante (75 %), à trois températures différentes [(10, 25 et 37) °C] et en présence de trois concentrations d’oxygène [(0, 20 et 40) %] a été suivie chaque semaine durant 35 jours à l’aide de la mesure des paramètres L* et b* de l’échelle Hunter Lab. Résultats. La température a joué un rôle déterminant dans la dégradation de la couleur des dattes durant l’entreposage et l’effet de l’oxygène a été important quoique secondaire. Conclusion. La couleur des dattes au cours de leur stockage se révèle pouvoir être prédite à partir du seul paramètre b* de l’échelle Hunter Lab. Dans le cas des dattes conditionnées sous différentes concentrations en oxygène et à différentes températures, la perte de la couleur initiale du fruit a été davantage expliquée par une augmentation de la température ambiante que par la variation de la concentration en oxygène du milieu de conservation. La dégradation de la couleur serait causée par des brunissements non enzymatiques et enzymatiques oxydatifs.

To better understand the molecular mechanisms involved in the biodegradation of hydrocarbon compounds from the “Erika” oil-spill, we have studied the ability of Marinobacter hydrocarbonoclasticus strain sp 17 to cope with hexadecane as sole carbon and energy source. Growth kinetics of cultures shifted from acetate to hexadecane revealed the presence of a 20 hours adaptation phase. Changes in global protein expression in response to hexadecane was analyzed by two-dimensional gel electrophoresis. Of the 370 proteins detected 42 had their expression level altered in presence of hexadecane indicating that alkane adaptation may involve many cellular functions.

Antarctic soil chronically exposed to gas-oil was analysed in order to isolate and study the growth conditions of hydrocarbon degrading bacteria. Soil samples taken near the shoreline in Jubany Station (King George Island, South Shetland Islands) were used as inoculum in liquid culture media with crude oil as sole carbon source. A psychrotrophic Acinetobacter strain was isolated and selected for further investigations. Effects were studied of temperature, initial pH, NaCl concentration and different chemical structure of the hydrocarbon on growth. Degradation rate was determined with n-dodecane and n-hexadecane. Growth of Acinetobacter ADH-1 showed no differences at an initial pH of 7.0, 7.5 and 8.0. Optimum temperature ranged between 25–30°C but the strain was capable of growing on n-dodecane at 4°C. Growth was observed in the presence of 3.5% NaCl. A decrease in the surface tension values was observed in the culture broth during the first 20 h of incubation (from 68 din cm−1 to 31 din cm−1). This proved to be related to the cellular fraction of the culture. The study shows that Acinetobacter ADH-1 is a psychrotrophic bacteria able to grow with hydrocarbons as sole carbon and energy source and could be potentially useful to design bioremediation processes in temperate and cold climate areas.

The prospect of the deliberate environmental release of genetically manipulated microorganisms has given rise to a great deal of polemic. Amongst the rational scientific concerns are those concerned with the fate of the released bacteria, the fate of the recombinant genes that they carry, the selective pressures acting upon them in different environmental situations and the long term effects on the environment and human health. All recombinant DNA is carried by vectors (plasmids, transposons or bacteriophage or remnants of these). Thus the way in which recombinant constructions are made may itself lead to potential biosafety concerns, irrespective of the host bacterium and the recombinant DNA fragment of primary interest. The purpose of the present review is to assess progress in improved vector design aimed at eliminating risks due to the way recombinant vectors are constructed. Improved vector constructions include the avoidance of the use, or removal, of antibiotic resistance genes, the use of defective transposons rather than plasmids in order to reduce horizontal transfer and the development of conditionally lethal suicide systems. More recently, new site-specific recombination systems have permitted transposon vectors to be manipulated following strain construction, but before environmental release, so that virtually all recombinant DNA not directly involved in the release experiment is eliminated. Such bacteria are thus pseudo-wild type in that they contain no heterologous DNA other than the genes of interest.
