This study was conducted to determine the effects of different levels of dietary tannin on gastrointestinal tract (GIT) histology and on liver and kidney masses. Five groups of Boer goats were given diets containing 0, 50, 100, 150 and 200 g/kg of tannin for 6 weeks before data collection. Differences in the histopathology of the oesophagus, reticulum, rumen, omasum, abomasum and duodenum were evaluated. Increased dietary tannin levels induced thickening and/or keratinization of epithelial tissue in the reticulum, rumen, omasum and abomasum. Increased tannin levels also resulted in a loss of epithelial cells, erosion of microvilli and shortened villi height in the duodenum, which could impair the absorption of nutrients. Consequently, condensed tannins had a negative effect on the histopathology of the Boer goats.

Hyper-prolific sows nurse more piglets than less productive sows, putting a high demand on the nutrient supply for milk production. In addition, the high production level can increase mobilization from body tissues. The effect of increased dietary protein (104, 113, 121, 129, 139 and 150 g standardized ileal digestible (SID) CP/kg) on sow body composition, milk production and plasma metabolite concentrations was investigated from litter standardization (day 2) until weaning (day 24). Sow body composition was determined using the deuterium oxide dilution technique on days 3 and 24 postpartum. Blood samples were collected weekly, and milk samples were obtained on days 3, 10 and 17 of lactation. Litter average daily gain (ADG) peaked at 135 g SID CP/kg (P < 0.001). Sow BW and back fat loss reached a breakpoint at 143 and 127 g SID CP/kg (P < 0.001). Milk fat increased linearly with increasing dietary SID CP (P < 0.05), and milk lactose decreased until a breakpoint at 124 g SID CP/kg and 5.3% (P < 0.001) on day 17. The concentration of milk protein on day 17 increased until a breakpoint at 136 g SID CP/kg (5.0%; P < 0.001). The loss of body protein from day 3 until weaning decreased with increased dietary SID CP until it reached a breakpoint at 128 g SID CP/kg (P < 0.001). The body ash loss declined linearly with increasing dietary SID CP (P < 0.01), and the change in body fat was unaffected by dietary treatment (P=0.41). In early lactation (day 3 + day 10), plasma urea N (PUN) increased linearly after the breakpoint at 139 g SID CP/kg at a concentration of 3.8 mmol/l, and in late lactation (day 17 + day 24), PUN increased linearly after a breakpoint at 133 g SID CP/kg (P < 0.001) at a concentration of 4.5 mmol/l. In conclusion, the SID CP requirement for sows was estimated to 135 g/kg based on litter ADG, and this was supported by the breakpoints of other response variables within the interval 124 to 143 g/kg.

This study was carried out to determine ruminal dry matter (DM) and crude protein (CP) degradation characteristics of untreated, 2-, 4- and 6-min microwave-treated soya-bean meal (SBM) by using nylon bags and sodium dodecyl sulphatepolyacrylamide gel electrophoresis (SDS-PAGE) techniques. Nylon bags of untreated or treated SBM were suspended into the rumen of three Holstein steers from 0 to 48 h, and data were fitted to non-linear degradation characteristics to calculate effective rumen degradation (ERD). There were significant differences (P < 0·05) for DM and CP degradation parameters between untreated and microwave-treated SBM. Microwave treatments decreased the water-soluble fraction and increased the potentially degradable fraction of CP. The degradation rate of the latter fraction decreased with these treatments. As a consequence, microwave treatments decreased (P < 0·05) ERD of CP. From densitometric scanning, SBM proteins were seen to be composed of two major components; β-conglycinin and glycinin, accounting for proportionately 0·30 and 0·40 of buffer-soluble SBM proteins, respectively. Electrophoretic analysis of untreated, 2-, 4- and 6-min microwave-treated SBM protein residues revealed that two of the subunits of β-conglycinin (α -and α) were degraded completely after 2, 4, 24 and 48 h, respectively, whereas the α subunit of this protein was more resistant to degradation. In untreated SBM, the two subunits of glycinin (acidic and basic polypeptides) were degraded in the middle of the incubation period, but in microwave-treated SBM were not degraded until 48 h of incubation. In vitro digestibility of ruminally undegraded CP of untreated and treated SBM increased (P < 0·05) with increases in rumen incubation time from 8 to 24 h. In conclusion, SBM proteins appeared to be effectively protected from ruminal degradation by a 4-min microwave treatment. SDS-PAGE results indicated that ruminally undegraded protein from untreated SBM was mainly composed of the basic subunit of glycinin, whereas that from microwave-treated SBM was composed of β-conglycinin and both basic and acidic subunits of glycinin.

In the process of developing a generic Hazard Analysis and Critical Control Point (HACCP)-like system for securing animal health and welfare in organic egg production, an expert panel analysis was used to perform the initial hazard analysis. Eighteen advisers and researchers in organic egg production were included in the expert panel. In a series of four questionnaires, the expert panel first scored 34 health and welfare problems seen in Danish organic egg production. Based on scorings of severity and occurrence, 10 problems were selected for further analysis. The experts subsequently suggested and scored risk factors for those problems and finally suggested control points, alarm values indicating the need for corrective actions in order to control the risk factors and monitoring frequencies of these. The 10 selected problems were hunger, thirst, piling, crop impaction, blackhead, pasteurellosis, bone fractures, cannibalism, predators and red mites. A total of 154 different risk factors were suggested for these problems. The 41 risk factors which rated highest in a combined scoring of importance and occurrence were selected for further analysis. There was a high degree of consensus between experts when scoring both problems and risk factors. The level of consensus, as defined by an interquartile range ⩽1, was 79% to 100% when scoring the health and welfare problems (scale 1–5) and 77% to 95% when scoring the risk factors (scale 1–4). On average, 5.8 control points were suggested for every risk factor. Alarm values were often not detailed enough to be of practical significance and further analysis is needed in order to define these. The experts were highly diverse in their suggested monitoring frequencies and establishment of monitoring schemes should be part of developing the farm specific systems. An expert panel analysis based on questionnaires was a useful tool during the first steps of developing a HACCP plan, conducting a hazard analysis and suggesting control points. However, care should be taken regarding the panel’s size and fields of expertise in order to assure that the panel fully covers the field of interest throughout the study. A further development of the hazard analysis into a farm-specific HACCP system would include assembling an on-farm HACCP team consisting of farmers and advisors for the practical application of control points, alarm values and monitoring schemes. The results from the fourth questionnaire would be useful as input for this work.

The objectives of this study were to determine the effect and mode of action of Saccharomyces cerevisiae (YST2) on enteric methane (CH4) mitigation in pigs. A total of 12 Duroc×Landrace×Yorkshire male finisher pigs (60±1 kg), housed individually in open-circuit respiration chambers, were randomly assigned to two dietary groups: a basal diet (control); and a basal diet supplemented with 3 g/YST2 (1.8×1010 live cells/g) per kg diet. At the end of 32-day experiment, pigs were sacrificed and redox potential (Eh), pH, volatile fatty acid concentration, densities of methanogens and acetogens, and expression of methyl coenzyme-M reductase subunit A gene were determined in digesta contents from the cecum, colon and rectum. Results showed that S. cerevisiae YST2 decreased (P<0.05) the average daily enteric CH4 production by 25.3%, lowered the pH value from 6.99 to 6.69 in the rectum, and increased the Eh value in cecum and colon by up to −55 mV (P<0.05). Fermentation patterns were also altered by supplementation of YST2 as reflected by the lower acetate, and higher propionate molar proportion in the cecum and colon (P<0.05), resulting in lower acetate : propionate ratio (P<0.05). Moreover, there was a 61% decrease in Methanobrevibacter species in the upper colon (P<0.05) and a 19% increase in the acetogen community in the cecum (P<0.05) of treated pigs. Results of our study concluded that supplementation of S. cerevisiae YST2 at 3 g/kg substantially decreased enteric CH4 production in pigs.

Feeding dietary supplements containing trans-10, cis-12-conjugated linoleic acid (t10,c12-CLA) has been shown to induce milk fat depression in cows, ewes and goats. However, the magnitude of the response is apparently less pronounced in lactating goats. The objective of this study was to evaluate the effects of increasing doses of CLA methyl esters (CLA-ME) on milk production, composition and fatty-acid profile of dairy goats. Eight Toggenburg goats were separated in two groups (four primiparous and four multiparous) and received the following dietary treatments in a 4×4 Latin Square design: CLA0: 45 g/day of calcium salts of fatty acids (CSFA); CLA15; 30 g/day of CSFA+15 g/day of CLA-ME; CLA30: 15 g/day of CSFA+30 g/day of CLA-ME; and CLA45: 45 g/day of CLA-ME. The CLA-ME supplement (Luta-CLA 60) contained 29.9% of t10,c12-CLA; therefore, the dietary treatments provided 0, 4.48, 8.97 and 13.45 g/day of t10,c12-CLA, respectively. Feed intake, milk production, concentration and secretion of milk protein and lactose, body condition score and body weight were unaffected by the dietary treatments. Milk fat secretion was reduced by 14.9%, 30.8% and 40.5%, whereas milk fat concentration was decreased by 17.2%, 33.1% and 40.7% in response to CLA15, CLA30 and CLA45, respectively. Secretions of both de novo synthesized and preformed fatty acids were progressively reduced as the CLA dose increased, but the magnitude of the inhibition was greater for the former. There was a linear reduction in most milk fat desaturase indexes (14:1/14:0, 16:1/16:0, 17:1/17:0 and 18:1/18:0). Milk fat t10,c12-CLA concentration and secretion increased with the CLA dose, and its apparent transfer efficiency from diet to milk was 1.18%, 1.17% and 1.21% for CLA15, CLA30 and CLA45 treatments, respectively. The estimated energy balance was linearly improved in goats fed CLA.

The present study aimed to evaluate the effect of dietary cation–anion difference (DCAD) on ruminal fermentation, total apparent digestibility, blood and renal metabolism of lactating dairy cows. Sixteen Holstein cows were distributed in four contemporary 4×4 Latin Square designs, which consisted of four periods of 21 days and four treatments according to DCAD: +290; +192; +98 and −71 milliequivalent (mEq)/kg dry matter (DM). Ruminal pH and concentrations of acetic and butyric acid increased linearly according to the increase of DCAD. Similarly, NDF total apparent digestibility linearly increased by 6.38% when DCAD increased from −71 to 290 mEq/kg DM [Y=65.90 (SE=2.37)+0.0167 (SE=0.0068)×DCAD (mEq/kg DM)]. Blood pH was also increased according to DCAD, which resulted in reduction of serum concentrations of Na, K and ionic calcium (iCa). To maintain the blood acid–base homeostasis, renal metabolism played an important role in controlling serum concentrations of Na and K, since the Na and K urinary excretion increased linearly by 89.69% and 46.06%, respectively, from −71 to 290 mEq/kg DM. Changes in acid–base balance of biological fluids may directly affect the mineral composition of milk, as milk concentrations of Na, K, iCa and chlorides were reduced according to blood pH increased. Thus, it can be concluded that the increase of DCAD raises the pH of ruminal fluid, NDF total apparent digestibility, and blood pH, and decreases the milk concentration of cationic minerals, as well as the efficiency of Na utilization to milk production.

Multi-tier aviary systems, where conveyor belts below the tiers remove the manure at regular intervals, are becoming more common in organic egg production. The area on the tiers can be included in the net area available to the hens (also referred to as usable area) when calculating maximum indoor stocking densities in organic systems within the EU. In this article, results on live weight, health measures and perching are reported for organic laying hens housed in a multi-tier system with permanent access to a veranda and kept at stocking densities (D) of 6, 9 and 12 hens/m2 available floor area, with concomitant increases in the number of hens per trough, drinker, perch and nest space. In a fourth treatment, access to the top tier was blocked reducing vertical, trough, and perch access at the lowest stocking density (D6x). In all other aspects than stocking density, the experiment followed the EU regulations on the keeping of organic laying hens. Hen live weight, mortality and foot health were not affected by the stocking densities used in the present study. Other variables (plumage condition, presence of breast redness and blisters, pecked tail feathers, and perch use) were indirectly affected by the increase in stocking density through the simultaneous reduction in access to other resources, mainly perches and troughs. The welfare of the hens was mostly affected by these associated constraints, despite all of them being within the allowed minimum requirements for organic production in the EU. Although the welfare consequences reported here were assessed to be moderate to minor, it is important to take into account concurrent constraints on access to other resources when higher stocking densities are used in organic production.

Levels of haptoglobin and Pig-major acute phase protein (MAP) were analysed in animals from a commercial herd receiving or not a diet enriched with an additive. The group receiving the additive exhibited a decrease in haptoglobin after 3 weeks, suggesting that a better health status has been established, together with an improvement in total body weight and average daily gain. In contrast, Pig-MAP does not significantly change under these conditions. Aujeszky live modified vaccination, which is compulsory in Spain, did cause a significant increment in haptoglobin serum concentration although it did not affect Pig-MAP. The response of acute phase proteins to vaccination was similar in both control and additive-treated groups. Interleukins (IL)-1β and IL-6 was below the detection limits in most of the animals. In conclusion, this study shows that haptoglobin serum concentration, but not Pig-MAP, is a good biomarker to monitorize production parameters and for monitoring Aujeszky modified live vaccine in pigs reared under standard commercial conditions.

In a previous study, we showed that access to willow fodder decreased somatic cell counts (SCC) in the milk of local Mamber goats grazing in brushland at the end of lactation. To test whether the consumption of willow affects the cells of the immune system, Alpine crossbred dairy goats grazing in the same environment were either offered free access to freshly cut willow fodder (W, n = 24) or not (C, n = 24) for 2 weeks. The willow fodder contained 7.5 g/kg DM of salicin. The other major secondary compounds were catechin, myricitrin, hyperin and chlorogenic acid (2.2, 2.6, 1.0 and 0.75 g/kg DM, respectively). Udder health status was determined before the experiment, and each of the two groups included five (W) or six (C) goats defined as infected, as established by microbial cfu in milk, and 19 (W) or 18 (C) non-infected goats. Goats ingested, on average, 600 g of DM from willow (25% of food intake), resulting in minor changes in dietary quality compared to the controls, as established by faecal near-IR spectrometry. Throughout the 2 weeks of experiment, differences between groups in dietary CP contents were minor and affected neither by infection nor by access to willow; the dietary percentage of neutral detergent fibre (NDF) decreased in C and increased in W; dietary acid detergent fibre (ADF) increased; and the dietary tannin contents decreased for both treatments. However, milking performance and milk quality attributes in both W and C goats were similar. Initial SCC and milk neutrophil (cluster of differentiation (CD)18+ and porcine granulocyte (PG)68) cell counts were higher in infected than in non-infected goats; counts decreased significantly in W but not in C uninfected goats. The percentage of CD8+ T-cells increased in all C goats, while in the W group, a significant increase was found only for infected goats. The consumption of willow mitigated an increase in CD8+ in blood and triggered an increase in CD8+ in milk, suggesting an immune-regulatory effect independent of udder status. To our knowledge, this is the first report of a direct nutraceutical effect of fodder ingestion on the immune status of goats.

Feeding a diet with a small amount of hay and a high proportion of concentrate given in large meals entails a risk of colic to horses as this can impact the hindgut microbial ecosystem. To counteract this potential negative effect, one feeding strategy is to modify the distribution sequence of concentrate and hay. The purpose of the present study was to assess the impact of feeding the concentrate meal before or with the hay meal on fermentative parameters postprandial variations in caecum and right ventral colon contents and faeces. We focused on the comparison of pH value and volatile fatty acid (VFA) concentrations between the three compartments. Four horses fitted with two cannulas in the caecum and right ventral colon were fed a 100% hay diet (H) for a 3-week period, and then a 40% concentrate–60% hay diet for two consecutive periods of 3 weeks each. Concentrate and hay were offered simultaneously in two equal meals at 0800 and 1700 h (HC1) during one period, or separately in two equal meals at 0800 and 1600 h for the concentrate and at 1000 and 1730 h for hay (HC2) during the second one. During the 2nd week of each diet period (H, HC1, HC2), caecal, right ventral colonic and faecal samples were collected before (0) and at 2, 4, 6 and 8 h after the morning meal. pH values and VFA concentrations were measured. Adding concentrate to the hay was associated with a decrease of pH value in the caecum, right ventral colon and faeces and an increase of faecal VFA concentrations (P<0.01). An impact of the concentrate feeding sequence was observed on caecal pH and faecal propionate concentration (P<0.001) but none on the other parameters investigated. In the caecum and right ventral colon, pH values decreased and VFA concentrations increased within the range of 4 to 8 h after the morning meal. Our data suggested that the variation of faecal pH could be an indicator of those of the caecal and right ventral colonic pH with a delay of 2 h. The present study showed that hay and commercial pelleted could be safely distributed simultaneously, thus making easier every day horse feeding.

1. Two experiments were made. In Experiment 1, 124 ram, wether and ewe lambs were allocated to four equal groups as they reached a live-weight of 30 lb. Group A received no hexoestrol; group B were implanted with a 15 mg. pellet of hexoestrol and lambs of group C were given a second pellet of 15 mg. 35 days after the first. Group D were implanted with three 5 mg. pellets simultaneously in three different sites. The lambs were creep-fed while running with their dams on pasture and were killed as they reached a live-weight of 95 lb.

In Experiment 2, 20 weaned ram lambs were allocated to a hexoestrol-implant group or a control group as they reached a live-weight of 40 lb. They were fed a pelleted ration indoors and were slaughtered at a live-weight of 110 1b.

2. Length of teats and weight of the pituitary were significantly increased for all lambs treated with hexoestrol as compared with their controls. Excessive mammary development and one case of rectal prolapse were observed i n treated ewe lambs, while 3 of the treated rams in Experiment 2 had scrotal oedema.

3. The rate of live-weight gain was greater for all hexoestrol-implanted lambs in Experiment 1 but less for the implanted lambs on Experiment 2 as compared with their respective controls. The differences just failed to attain significance at the 5% level.

4. Dressing percentage was significantly reduced by hexoestrol treatment i n Experiment 1. On the other hand treated ram lambs in Experiment 2 had significantly higher dressing percentages and appeared to be fatter than the controls.

5. Weights of testes and seminal vesicles in the ram and of the ovary in the ewe were decreased by hexoestrol treatment. Weights of seminal vesicles in the wether and of oviducts and uterus in the ewe were increased.

An experiment was performed to measure the response of young pigs to dietary tryptophan (TRP) concentrations and environmental temperatures. Seventy-two entire male Large White ✕ Landrace pigs were assigned to one of six dietary treatments (2·90 (T1), 2·46 (T2), 2·01 (T3), 1·57 (T4), 1·12 (T5) g/kg and T5 + supplemented TRP (T6)) and one of three temperature treatments (20, 25 and 30°C) at a mean starting live weight of 14·38 (s.e. 0·201)kg. Animals were given ad libitum access to food until a final weight of 26·42 (s.e. 0·479) kg. There were no significant interactions between temperature and dietary TRP on any production variable. There was a significant (P < 0·05) quadratic improvement in the rate of live-weight growth (ADG) as the concentration of dietary TRP increased and as the temperature decreased. However, the response to increasing dietary TRP was independent of the environmental temperature. Maximum ADG was attained on T2 (0·498 (s.e. 0·023) kg/day) and at 20ºC (0·412 (s.e. 0·024) kg/day). Final live weight was a significant (P < 0·001) covariate for ADG and food intake (FI) responses. With TRP as a precursor for serotonin, a neurotransmitter that regulates appetite, it was anticipated that food intake would be affected with decreasing dietary TRP levels. However, there was no response in daily food intake to decreasing TRP concentration. This lack of response in appetite to dietary TRP may have been a result of an increasing TRP to large neutral amino acid ratio, which is known to correlate with an increase in serotonin synthesis. Total heat loss followed a similar response to FI. The gain per unit of food consumed was significantly (P < 0·001) reduced as the TRP content of the diet was decreased. The most efficient treatments were T1 (506 (s.e. 1·90) g gain per kg food) and T2 (495 (s.e. 23·2) g gain per kg food) while the worst was T5 (237 (s.e. 22·3) g gain per kg food). There were significant quadratic responses to dietary TRP in protein content of the empty body (P < 0·05) and the rate of protein retention (PR) (P < 001) but only PR was affected by temperature (P < 001). Both temperature (P < 0·05) and dietary TRP (P < 0·001) had a significant effect on the lipid content of the body but only temperature affected the rate of lipid retention, with a significantly (P < 0·001) lower rate at 30 oC. The efficiency of TRP utilization improved with increasing temperature. It was lowest at 20ºC (0·60 g TRP per kg protein) and highest at 30ºC (0·86 g/kg), while the mean efficiency for pigs between 14 and 26 kg live weight, at thermoneutrality (25°C), was close to 0·71 g/kg.

The objective of this study was to investigate the effects of a flaxseed-supplemented diet on archaeal abundance and gene expression of methanogens in the rumen of dairy cows. In all, 11 non-lactating dairy cows were randomly divided into two groups: group A (five cows) and B (six cows). The two diets fed were: (1) the control diet, a conventional dry cow ration; and (2) the flaxseed-supplemented diet, the conventional dry cow ration adjusted with 12.16% ground flaxseed incorporated into the total mixed ration. A cross-over experiment was performed with the two groups of cows fed the two different diets for five 21-day periods, which included the first adaptation period followed by two treatment and two wash out periods. At the end of each feeding period, rumen fluid samples were collected via rumenocentesis and DNA was extracted. Quantitative PCR was utilized to analyze the gene abundance of 16S ribosomal RNA (16S rRNA) targeting the ruminal archaea population and the mcrA gene coding for methyl coenzyme-M reductase subunit A, a terminal enzyme in the methanogenesis pathway. Results demonstrated a 49% reduction of 16S rRNA and 50% reduction of mcrA gene abundances in the rumen of dairy cows fed the flaxseed-supplemented diet in comparison with those fed the control diet. This shows flaxseed supplementation effectively decreases the methanogenic population in the rumen. Future studies will focus on the mechanisms for such reduction in the rumen of dairy cattle, as well as the relationship between methanogenic gene expression and methane production.

Quantitive collection of night-time urine every 2nd week was used to elucidate the temporal relationship between changes in melatonin production, weight gain, and priming of the winter pelage under natural light conditions and under a long photoperiod (18L: 6D) imposed at different times in four groups of female mink during autumn. In mink maintained outdoors under the natural photoperiod (group 1), melatonin excretion was 1·80 (s.d. 0·80) ng per night (no. = 3) at the beginning of September, thereafter it began to decrease slowly until November when it was only 0·62 (s.d. 0·32) ng per night. These mink had a normal autumn moult during the first half of October and the winter pelage was mature at the end of November. When mink maintained outdoors were transferred to 18L: 6D on 7 September (group 2), excretion of melatonin decreased promptly, autumn moult was disrupted and winter fur priming delayed. Long photoperiod after mid October (group 3), did not interfere with pelage maturation despite the decrease in melatonin production. Mink implanted with melatonin on 14 July (group 4) showed very high melatonin excretion in early September. In these animals the winter pelage growth was unaffected by the long-day conditions; the pelage matured in mid October. Body weight increased in mink of all groups during autumn. This increase levelled off or a weight loss (group 2) was seen along with the final hair maturation. Activation of hair follicles occurred during a 4 to 6 week period. According to these results, melatonin is the photoperiodic signal to autumnal weight increase and autumn moult but seems not to be necessary for later pelage growth and maturation.

Forty-two piglets were used to study the effects of heavy and light birth weight, and of four levels of feeding, ranging from 37 to 91 g dry matter per kg M0 75 per day, on growth between 5 days of age and 6·5kg live weight, and on body composition, and the cellularity of muscle and subcutaneous adipose tissue, at the latter weight. The effects of birth weight and level of feeding to 6·5 kg on performance between 6·5 and 11 -5 kg, and on body composition at 11·5 kg, were also investigated.

Light birth weight piglets grew more slowly to 6·5 kg than those of heavy birth weight (P < 0·05). However, birth weight had no effect on food conversion efficiency or on body composition.

Each increment in feeding level resulted in increases in growth rate (P < 0·01), body fat content (P < 0·01) and average fat-cell diameter (P < 0·01) at 6·5 kg, but in decreases in body protein and water (P < 0·01).

The weight of the m. semitendinosus at 6·5 kg was unaffected by either birth weight or level of feeding. However, the deoxyribonucleic acid content of the muscle at both 6·5 and 11·5 kg was less in light birth weight piglets fed at the lowest level prior to 6·5 kg (P < 0·05). These piglets also exhibited poorer growth performance subsequent to 6·5 kg than their heavier birth weight counterparts.

Nevertheless, piglets of both light and heavy birth weight, fed at the lowest level to 6·5 kg, grew faster and more effficiently between 6·5 and 11·5kg (P < 0·05), and were leaner at 11·5kg than those previously fed at the highest level (P < 0·01). Average fat-cell diameter at 11·5kg was also less in pigs fed at the lowest level prior to 6·5kg (P < 0·01). Total deoxyribonucleic acid in subcutaneous adipose tissue at 11·5kg was unaffected by either birth weight or level of feeding prior to 6·5 kg.

Scottish Blackface (BF) and East Friesland × Scottish Blackface (EFX) ewes rearing single or twin lambs grazed for 14 weeks of lactation on two different pasture types in two consecutive years. Mean daily milk production, live-weight change and herbage intake of the ewes and live-weight gain of the lambs were measured each week.

In the first year the ewes grazed a ryegrass/clover sward. There were significant differences in milk production associated with breed of ewe and number of lambs reared. Peak yields, achieved in the 3rd or 4th week, reached 2·25, 2·5 and 3·25 kg/day for BF ewes with a single lamb and EFX ewes with single or twin lambs, respectively. Corresponding yields in the 14th week were 0·5, 1·35 and 1·15 kg/day. Live weight of ewes in all groups declined during the first 2 weeks but recovered by week 6. Herbage intake increased in all groups until the 8th week and then declined much more rapidly in the BF than in the EFX ewes. Lamb growth was related to milk intake to give weaning weights of 31·5, 39·0 and 29·0 kg, respectively.

In the second year, when ewes grazed a hill pasture dominated by an AgrostislFestuca community, differences in milk production, though significant, were smaller than in the previous year. Peak yields were 2·1, 2·25, 2·25 and 3·1 kg/day, respectively, for BF ewes with single or twin lambs and EFX ewes with single or twin lambs. At 14 weeks, yields had fallen to between 0·6 and 1·0 kg/day. The patterns of live-weight change and food intake were similar to those in the previous year. The EFX ewes consumed more food than BF ewes throughout lactation, particularly in the second half. Lamb growth rate was again related to milk supply and was lower than in the previous year. Weaning weights were 31·0, 25·5, 34·5 and 27·0 kg for single or twin lambs reared by BF or EFX ewes, respectively.

To study the activation of caspase-9 and its potential influence in conditioning, longissimus thoracis (LT), semitendinosus (STN) and psoas minor (PMi) muscles were used to analyze the ratio of pro-apoptotic bax to anti-apoptotic bcl-2 in fresh tissues and observe the changes in ATP, cytosolic cytochrome c and caspase-9 activity levels during storage at 4°C. Caspase-9 activity at 5 h is higher than the activity at 0 and 24 h in the muscles (P<0.001). The ATP content decreased between 0 and 3 h, between 8 and 14 h in the PMi and LT muscles (P<0.0001), whereas between 0 and 5 h, between 8 and 14 h in the STN muscle (P<0.0001). There is 60.2%, 55.3% and 43.1% available ATP in the STN, LT and PMi muscles at 5 h, respectively. The cytosolic cytochrome c level increased during 5 and 24 h storage in the LT and PMi muscles (P<0.0001), during 5 and 96 h in the STN muscle (P<0.0001). The cytosolic cytochrome c at 24 h (P<0.001) and ratio of bax to bcl-2 (P<0.05) was higher in the PMi than in other muscles. We concluded that the increase in cytosolic cytochrome c and available intracellular ATP should be responsible for the increase in caspase-9 activity; the activation of caspase-9 could be limited by the subsequent depletion of ATP; the postmortem release level of cytochrome c could be determined by the ratio of bax to bcl-2 in fresh tissues.

The wool growth cycle in three rams and three ewes was studied by taking monthly wool and skin samples for periods up to 3 years. The percentage of inactive follicles and of medullated fibres was recorded, fleece length was measured and change in fibre diameter and pigmentation between summer and winter was investigated.

Casting of the fleece appeared to depend more on the thinning of fibres than on the proportion with brush ends, casting in some instances taking place with only 30% brush ends, while not in others with as much as 70% follicle inactivity.

Follicles began to become inactive in both sexes in September following the autumnal equinox. The secondary follicles had a greater proportion of inactivity (mean of 45% compared with 20% in the primaries) but tended to regrow earlier, and regrowth in the rams tended to begin before that in the ewes. Regrowth as a whole began from January to March. Subsidiary cycles of inactivity of about 2 months' duration were evident.

Maximum growth in length occurred between April and October. A difference in mean diameter of only a few microns could be detected between summer and winter. Maximum medullation occurred in July in the rams and August in the ewes. Decrease in pigmentation in winter was brought about by a lowering of the density of pigment granules within the fibres.

The effect of gradually replacing barley with swedes or potatoes on intake of digestible dry matter and on the volatile fatty acid (VFA) composition of liquor was measured in an experiment with eight female and four castrated male sheep approximately six months old. The total intake of digestible dry matter was greatest when rolled barley was given at two-thirds of estimated maximal intake and roots given ad libitum. The intake of digestible dry matter was reduced to 80% of the highest intake when all the barley was substituted with swedes and to 70% of the highest intake when all the barley was substituted with potatoes. Replacing barley with roots had little or no influence on VFA composition.
