There are two types of 1-day field tests available for young Swedish Warmblood sport horses; one test for 3-year olds and one more advanced test for 4-year olds. Conformation, gaits and jumping ability are evaluated at both tests. Studies on various genetic parameters were based on about 20 000 tested horses. The data for 4-year olds consisted of 30 years of testing. The aims of the study were to estimate genetic parameters for results from different time periods, and to estimate heritabilities for, and genetic correlations between, traits scored in the two tests. The judgement of traits was shown to have been changed during the 30 years of testing, resulting in changes in higher heritabilities in, and stronger genetic correlations between, later time periods. In the first time period, records showed higher residual and lower genetic variances than in the subsequent time periods. Genetic correlations between traits recorded in the first and last time period deviated considerably from unity. Further studies are needed to investigate how to treat data from the early period in genetic evaluations. Heritabilities were moderate to high for conformation traits (0.24 to 0.58) at both types of tests, except for correctness of legs (0.08). The heritabilities for gait traits were also moderate to high (0.37 to 0.53). For jumping traits, the heritabilities ranged between 0.17 and 0.33. The highly positive genetic correlations (0.82 to 0.99) between corresponding traits tested at the simpler test for 3-year olds and at the ridden test of 4-year olds implied that it would be desirable to include the test results of 3-year olds into the genetic evaluation as breeding values for Swedish Warmbloods for many years has only been based on results from 4-year olds.

Effects of synchronising the availability of amino acids and glucose within a day on protein and energy metabolism were studied in growing pigs. Ten pigs of on average 54 (s.e. 1.0) kg live weight were assigned to each of two dietary treatments (synchronous v. asynchronous nutrient supply) in a change-over design. On the synchronous treatment (SYN), pigs received two balanced meals: one at 0800 h and one at 1600 h. On the asynchronous treatment (ASYN), pigs received virtually all protein at 0800 h and all carbohydrates at 1600 h. The dietary supply of ingredients and nutrients to pigs was similar for both treatments. Pigs were housed individually in respiration chambers. Faecal apparent nutrient digestibility was determined and nitrogen and energy balances were measured. Faecal apparent digestibility of energy, organic matter and non-starch polysaccharides was higher ( P < 0.05) for SYN than for ASYN. The efficiency of utilisation of digestible protein with protein gain was higher ( P = 0.001) for SYN (56.7%) than for ASYN (47.1%). The substantial decrease ( P < 0.05) in respiratory quotient and 13C enrichment of the expired CO2 after the morning meal indicated higher amino acid oxidation for ASYN than for SYN. Heat production and energy retention as fat were not affected by nutrient synchrony. In conclusion, an asynchronous availability of glucose and amino acids within a day increases amino acid oxidation, resulting in a substantial reduction in protein utilisation but with virtually no effect on fat retention.

Epidemiological studies in humans and animal models (including ruminants and horses) have highlighted the critical role of nutrition on developmental programming. Indeed, it has been demonstrated that the nutritional environment during the periconceptional period and foetal development can altered the postnatal performance of the resultant offspring. This nutritional programming can be exerted by maternal and paternal lineages and can affect offspring beyond the F1 generation. Alterations in epigenetic mechanisms have been proposed as the causative link behind the programming trajectories observed in the offspring. Although a clear cause–effect relationship between epigenetic modifications during early development and later offspring phenotype has not been demonstrated in livestock species, strong associations have been reported for some epigenetic marks (e.g. messenger RNA) that are worth exploring as possible predictors of future offspring phenotype. In this review, we shortly describe the main epigenetic mechanisms studied so far in mammals (i.e. mainly in the mouse) thought to be associated with developmental programming, and discuss the few studies available in mammalian herbivores (e.g. cattle) showing the effect of nutrition on epigenetic marks and the associated phenotype. Clearly, there is a need to develop research on nutritional strategies capable of modulating the epigenetic machinery with positive influence on the phenotype of livestock herbivores. This type of research is needed to alleviate the challenges currently faced by the livestock industry (e.g. impaired fertility of high-yielding dairy cows). This in turn will have a positive influence on animal welfare and productivity of livestock enterprises.

The ability of birds to modify dietary phosphorus utilisation when fed with low-phosphorus and calcium (Ca) diets was studied using different sequences of dietary phosphorus and Ca restriction (depletion) and recovery (repletion) during the grower and the finisher phases. A total of 3600 Ross 708 broilers were randomly divided into 10 replicate pens per treatment (60 per pen, six pens per block). Chicks were fed a common starter diet from days 0 to 10, then a grower control diet (C: 0.90% Ca, 0.39% non-phytate phosphorus, nPP), mid-level diet (M: 0.71% Ca, 0.35% nPP) or low Ca and nPP diet (L: 0.60% Ca, 0.30% nPP) from days 11 to 21, followed by a finisher diet C, M or L containing, respectively, 0.85%, 0.57% or 0.48% Ca and 0.35%, 0.29% or 0.24% nPP from days 22 to 37. Six treatment sequences were tested: CC, MM, LL, ML, LC and LM. Bone mineral content by dual-energy X-ray, tibia ash, toe ash weight and tibia breaking strength were measured on days 21 and 37. No significant effect was observed on growth performance throughout the experiment. Diet L reduced bone mineral content, breaking strength, tibia and toe ash by 9%, 13%, 11% and 10%, respectively, on day 21 (compared with diet C, for linear effect, P<0.05). On day 37, bone mineral content, breaking strength, tibia and toe ash remained lower compared with control values (CC v. MM v. LL, P<0.05 for linear and quadratic effects). Mineral depletion duration (ML v. LL) did not affect bone mineral status. Replenishing with the C diet during the finisher phase (LC) restored bone mineral content, tibia ash and toe ash weight better than the M diet did, but not to control levels (CC v. LC v. LM, for linear effect, P<0.05). These results confirm that dietary Ca and nPP may be reduced in the grower phase without affecting final growth performance or breaking strength as long as the finisher diet contains sufficient Ca and nPP. The practical applications of this strategy require further study in order to optimise the depletion and repletion steps.

The experiment compared four different space allowances for growing pigs housed on fully slatted floors. Space allowance (A m2) was defined in relation to live weight (M kg) of the pigs according to the equation A = kM0·67, using values for the constant (it) of 0·024, 0·027, 0·030 and 0·034. Six replicate pens of 12 pigs were housed on each treatment in a controlled environment building. Food was given ad libitum and from 34 kg until slaughter at 85 kg pigs were individually weighed each week and the pen size was adjusted according to the appropriate equation. Increasing space allowance increased live-weight gain (844, 862, 883, 897 (s.e.d. 14·4) g/day). Differences in killing-out proportion reduced the effect on final carcass weight (63·3, 63·9, 64·1, 64·5 (s.e.d 0·82) kg). Daily food intake was unaffected by treatment but food conversion ratio was significantly poorer at the lowest space allowance (2·70, 2·56, 2·60, 2·59 (s.e.d. 0·043) kg/kg). Economic analyses of the results indicated an adverse effect on profitability of space allowance less than that defined by the equation A(m2) = 0·027 M0·67 (kg). A minimum allowance defined by the equation A (m2) = 0·030 M0·67 is recommended in commercial practice for pigs in fully slatted housing.

A study of the effects of low birth weight on muscle cellularity was performed on 48 pedigree Large White pigs selected, from a total of 17 litters, on the basis of their weight at birth. Where possible, the largest male (mean birth weight of 1544 g), smallest male (1135 g), and runt (776 g) littermates were chosen. Fresh frozen, whole mid belly, sections of m. semitendinosus and samples of m. trapezius from each animal were stained for the demonstration of acid pre-incubated myosin adenosine triphosphatase. The use of this stain demonstrated groups of positively stained, slow-contracting, myofibres which were each surrounded by a compliment of negatively stained, fast-contracting, fibres which together constituted ‘metabolic bundles’. The positions of metabolic bundles are indicative of the presence of single primary myofibres in the foetal muscle, all the other myofibres in the metabolic bundles being derived from subsequently formed secondary fibres. Determination of total myofibre number and primary fibre number were made for m. semitendinosus together with an estimation of the secondary to primary fibre-number ratios for both this muscle and for m. trapezius. Low birth weight was associated with a permanently reduced total muscle fibre number, proportionately in the order of 0·19 (P < 0·001) between large and runt littermates. A reduced muscle fibre number was not always associated with low birth weight, but when this was the case it was generated through a reduced secondary to primary fibre-number ratio (P < 0·01). Primary fibre number was not significantly affected in low birth-weight pigs except in extreme cases of runting.

For sustainable aquaculture, the removal of marine resource ingredients in fish diets is an important objective. While most studies focus on the replacement of fish oil by vegetable oil, little is known on the nutritional effects of presence (which corresponds to the control diet) or absence of dietary fish oil. We studied fatty acid composition of brush-border membranes and digestive enzyme activities of the intestine and measured the expression and activities of several enzymes involved in the hepatic intermediary metabolism of rainbow trout (Oncorhynchus mykiss) fed for 7 weeks with or without fish oil. The diets were pair-fed to ensure that fish fed either diet had comparable carbohydrate and protein intakes. Absence of fish oil significantly reduced growth rate, protein efficiency and plasma lipid components. Activities of intestinal digestive enzymes were significantly decreased in the anterior intestine in fish fed without fish oil. In liver, dietary fish oil removal did not affect the transcript levels or activities of the main enzymes involved in lipogenesis (fatty acid synthase) and fatty acid β-oxidation (3-hydroxyacyl-CoA dehydrogenase), glycolysis or amino acid oxidation. It lowered the expression of the genes coding for gluconeogenic enzymes (glucose-6-phosphatase and phosphoenolpyruvate carboxykinase), but their enzyme activities were not affected. The activities, but not gene expression of lipogenic enzymes, involved in NADPH and malonyl-CoA formation were also modified after fish oil removal as reflected by higher activities of isocitrate dehydrogenase/glucose-6-phosphate dehydrogenase and acetyl-CoA carboxylase enzymes. Overall, our results indicate that the intestinal digestive capacity was strongly modified by dietary fish oil removal, while hepatic intermediary metabolism was only marginally affected, in fed rainbow trout.

One hundred first lactation Friesian dairy heifers were used to investigate the effect of level of nutrition n i late pregnancy and for a limited period around the time of insemination on reproductive performance. The heifers were housed throughout the period of the trial in cubicle yards and all individuals were given controlled amounts of food.

Fifty animals were offered a ‘high’ (H) level of feeding in late pregnancy (for 10 weeks before calving) and 50 were offered a ‘low’ (L) level. After calving, the heifers were re-randomized with half of each pre-calving group being offered either the H or L level of feeding in early lactation (weeks 6 to 18), thus forming four subgroups of 25 heifers: HH, HL, LH and LL. The level of feeding in late pregnancy for all groups increased towards calving. The high plane heifers consumed 83·6 MJ metabolizable energy (ME) per day on average compared with 64·6 MJ ME per day for the low plane heifers, a proportional mean difference of 0·29 (P < 0·001). After calving, heifers consumed an average of 86 MJ ME per day in week 1 rising to 126 MJ ME per day by week 5. Between weeks 6 to 18 of lactation, the high plane group consumed an average of 146·8 MJ ME per day compared with 119·8 MJ ME per day for the low plane group, a proportional mean difference of 0·23 (P < 0001).

There were no significant effects of feeding treatments on milk yield. Fat yield in weeks 6 to 18 was increased by the high level of feeding in lactation (P < 0001), while protein yields in weeks 1 to 5 and n i weeks 6 to 18 were increased by high plane feeding in pregnancy (P < 0·05) and lactation (P < 0·05), respectively. Heifers receiving the high level of feeding in late pregnancy gained significantly (P < 0·001) more weight than those on the lower level of feeding. All groups lost similar amounts of weight during the first 5 weeks after calving, but there were marked differences in the rate of live-weight gain between weeks 6 to 11 of lactation (P < 0·05 to P < 0·001). In particular, the HL group had the lowest rate of live-weight gain, and this was significantly lower than that of the HH group (P < 0·01).

First ovulation occurred significantly earlier in heifers receiving the high level of feeding in pregnancy P ( < 0·01), but there was no effect of treatment on ovulation detection rate, time to first oestrus, time of first insemination or the proportion of wrongly-timed inseminations. Heifers receiving the high level of feeding in lactation had a successful pregnancy rate to first insemination of 0·42 compared with 0·63 for those on the lower level of feeding (P < 0·05), with the greatest difference observed between the HL group (0·72) compared with the HH group (0·35) (P < 0·01). Embryo loss rate was low in the HL group (008) compared with that in the HH group (0·26; P < 0·l). The net result was that the HH group had a median interval from calving to successful pregnancy of 95 days with an interquartile range of 82 days, whereas the figures for the HL group were 75 and 23 days (P < 0·001). Live-weight change at artificial insemination (AI) and level and change in milk yield at AI had independent effects on reproductive performance and as a consequence net energy output showed the strongest relationship with days to successful pregnancy. Heifers with the highest net energy outputs per day at AI had the longest intervals to successful pregnancy (r = 0·408; P < 0·001).

Much emphasis has been put on evaluating alterations in milk composition caused by clinical and subclinical mastitis. However, little is known about changes in milk composition during subclinical mastitis in individual udder quarters with a low-to-moderate increase in milk somatic cell count (SCC). This information is needed to decide whether milk from individual udder quarters with a moderate-to-high increase in milk SCC should be separated or not. The aim of this study was to determine how milk composition in separate udder quarters is affected when cow composite milk has low or moderately increased SCC levels. Udder quarter and cow composite milk samples were collected from 17 cows on one occasion. Milk yield was registered and samples were analyzed for SCC, fat, total protein, whey proteins, lactose, citric acid, non-protein nitrogen (NPN), lactoferrin, protein profile, free fatty acids (FFAs), lactate dehydrogenase (LDH), proteolysis, sodium and potassium. Bacteriological samples were collected twice from all four quarters of all cows. The cows were divided into three groups depending on their SCC at udder quarter level. The first group comprised healthy cows with four udder quarters with low SCC, <50 000 cells/ml; composition was equal when opposite rear and front quarters were compared. In the second and the third groups, cows had one udder quarter with 101 000 cells/ml < SCC < 600 000 cells/ml and SCC > 700 000 cells/ml, respectively. The remaining udder quarters of these cows had low SCC (<100 000 cells/ml). Despite the relatively low average cow composite SCC = 100 000 cells/ml of Group 2, milk from affected udder quarters exhibited lower casein number, content of lactose and β-casein (β-CN), while the content of whey protein, sodium, LDH and α-lactoalbumin (α-la) were higher compared to healthy opposite quarters. In addition to these changes, milk from affected udder quarters in Group 3 also exhibited lower values of potassium and αs1-casein (αs1-CN) and higher values of lactoferrin when compared to milk from opposite healthy quarters. This indicates that even when the SCC in cow composite milk is low, there might exist individual quarters for which milk composition is changed and milk quality impaired.

Exposure of anoestrous ewes to rams induces an increase in LH secretion, eventually leading to ovulation. This technique therefore is an effective, low-cost and hormone-free way of mating sheep outside the breeding season. However, the use of this technique is limited by the variability of the ewes’ responses. In this study, our objective was to understand more completely the origins of this variability and to determine the relative roles of breed, the point in time during anoestrus and the depth of anoestrus on the response to the ‘ram effect’. In the first experiment, the pattern of anoestrus on the basis of the concentration of progesterone determined weekly, was determined in four breeds including two less seasonal (Mérinos d'Arles and Romane), one highly seasonal (Mouton Vendéen) and one intermediate (Île-de-France) breeds. Anoestrus was longer and deeper in Mouton Vendéen and Île-de-France than in Romane or Mérinos d'Arles. In the second experiment, we used the same four breeds and tested their hypophyseal response to a challenge with a single dose of 75 ng gonadotrophin-releasing hormone (GnRH) in early, mid and late anoestrus, and then we examined their endocrine and ovarian responses to the ‘ram effect’. Most (97%) ewes responded to GnRH and most (93%) showed a short-term increase in LH pulsatility following the ‘ram effect’. The responses in both cases were higher in females that went on to ovulate, suggesting that the magnitude of the hypophyseal response to a GnRH challenge could be a predictor of the response to the ‘ram effect’. As previously observed, the best ovarian response was in Mérinos d'Arles at the end of anoestrus. However, there was no relationship between the proportion of females in the flock showing spontaneous ovulation and the response to the ‘ram effect’ of anoestrous ewes from the same flock.

This study evaluated different female-selective genotyping strategies to increase the predictive accuracy of genomic breeding values (GBVs) in populations that have a limited number of sires with a large number of progeny. A simulated dairy population was utilized to address the aims of the study. The following selection strategies were used: random selection, two-tailed selection by yield deviations, two-tailed selection by breeding value, top yield deviation selection and top breeding value selection. For comparison, two other strategies, genotyping of sires and pedigree indexes from traditional genetic evaluation, were included in the analysis. Two scenarios were simulated, low heritability (h2 = 0.10) and medium heritability (h2 = 0.30). GBVs were estimated using the Bayesian Lasso. The accuracy of predicted GBVs using the two-tailed strategies was better than the accuracy obtained using other strategies (0.50 and 0.63 for the two-tailed selection by yield deviations strategy and 0.48 and 0.63 for the two-tailed selection by breeding values strategy in low- and medium-heritability scenarios, respectively, using 1000 genotyped cows). When 996 genotyped bulls were used as the training population, the sire’ strategy led to accuracies of 0.48 and 0.55 for low- and medium-heritability traits, respectively. The Random strategies required larger training populations to outperform the accuracies of the pedigree index; however, selecting females from the top of the yield deviations or breeding values of the population did not improve accuracy relative to that of the pedigree index. Bias was found for all genotyping strategies considered, although the Top strategies produced the most biased predictions. Strategies that involve genotyping cows can be implemented in breeding programs that have a limited number of sires with a reliable progeny test. The results of this study showed that females that exhibited upper and lower extreme values within the distribution of yield deviations may be included as training population to increase reliability in small reference populations. The strategies that selected only the females that had high estimated breeding values or yield deviations produced suboptimal results.

Methane (CH4) is a greenhouse gas (GHG) produced and released by eructation to the atmosphere in large volumes by ruminants. Enteric CH4 contributes significantly to global GHG emissions arising from animal agriculture. It has been contended that tropical grasses produce higher emissions of enteric CH4 than temperate grasses, when they are fed to ruminants. A number of experiments have been performed in respiration chambers and head-boxes to assess the enteric CH4 mitigation potential of foliage and pods of tropical plants, as well as nitrates (NO3−) and vegetable oils in practical rations for cattle. On the basis of individual determinations of enteric CH4 carried out in respiration chambers, the average CH4 yield for cattle fed low-quality tropical grasses (>70% ration DM) was 17.0 g CH4/kg DM intake. Results showed that when foliage and ground pods of tropical trees and shrubs were incorporated in cattle rations, methane yield (g CH4/kg DM intake) was decreased by 10% to 25%, depending on plant species and level of intake of the ration. Incorporation of nitrates and vegetable oils in the ration decreased enteric CH4 yield by ∼6% to ∼20%, respectively. Condensed tannins, saponins and starch contained in foliages, pods and seeds of tropical trees and shrubs, as well as nitrates and vegetable oils, can be fed to cattle to mitigate enteric CH4 emissions under smallholder conditions. Strategies for enteric CH4 mitigation in cattle grazing low-quality tropical forages can effectively increase productivity while decreasing enteric CH4 emissions in absolute terms and per unit of product (e.g. meat, milk), thus reducing the contribution of ruminants to GHG emissions and therefore to climate change.

The modern hyper-prolific sow gives birth to more piglets than she has functional teats (in the following called supernumerary piglets). The aim of the present study was (1) to investigate the production consequences of hyper-prolific sows rearing supernumerary piglets equal to the mean live-born litter size, and (2) investigate whether potential negative effects on survival and growth could be alleviated by providing access to milk replacer and/or providing easier access to the udder (by loose housing). At day 1 (D1) postpartum (pp), 93 litters were standardised to 14 or 17 piglets (LS14/LS17) after which no piglets were moved between sows leading to decreased litter size if piglets died. Litters were provided with or without milk replacer in milk cups (+MILK/−MILK), and sows were either crated or loose housed (CRATE/LOOSE) in a 2 × 2 × 2 factorial design. Piglet mortality was higher in LS17 compared to LS14 (P < 0.01; OR = 2.0), higher in −MILK compared to +MILK (P = 0.01; OR = 1.2) and higher in LOOSE compared to CRATE (P = 0.02; OR = 1.8). This study showed that sow rearing of supernumerary piglets while supplying with milk replacer can increase piglet survival. It also showed that early mortality before piglets learned to drink milk replacer posed a challenge using this automatic milk replacer system. An interaction between access to milk replacer and the standardised litter size D1 affected litter weight (P < 0.01) and piglet weight day 28 (D28) (P = 0.03). The highest litter weight D28 was found in LS17 +MILK (P < 0.01) but with a lower individual piglet weight than in LS14 −MILK. Piglet weight D28 was higher in LS14 −MILK compared to LS17 regardless of access to milk replacer. Heterogeneity in piglet weight within litters D28 was larger in LS17 (P = 0.03) but could be reduced with +MILK in CRATE (P < 0.01). No effects were found on sow weight loss and feed intake (P > 0.05). In conclusion, the results showed that sows cannot rear the supernumerary piglets without further management interventions to reduce mortality. Supplying supernumerary piglets equal to the mean live-born litter size of hyper-prolific sows with milk replacer can from results of this study be an alternative strategy to the use of nurse sows.

Effects of a two-diet system combined with ad libitum lactation feeding of sows on food consumption, sow live weight (LW), backfat depth, condition scoring, rebreeding interval, symptoms of agalactia and culling were studied on 60 sows followed for four parities. Feeding regimes were: CR, conventional i.e. restricted during gestation and lactation; SA, a simplified diet offered at a restricted level during gestation and a conventional diet ad libitum during lactation; CA, conventional during gestation and the same diet ad libitum during lactation. During lactation CR sows consumed 5·9 kg food daily (71 MJ metabolizable energy (ME)), whereas SA and CA sows consumed on average 7·0 kg (85 MJ ME), the difference being larger in multiparous than in primiparous sows and larger in the first than in later weeks of lactation. From the second farrowing onward sows on CR treatment were lighter and had thinner backfat and lower condition scores than sows on CA treatment, with sows on SA treatment in between, i.e. approaching the CR sows in the gestation periods and the CA sows in the lactation periods. At first service LW was 130 kg and backfat thickness 14 mm. During the fourth lactation LW averaged 190 kg in CR sows and around 220 kg for SA and CA sows, and backfat thickness was 11 mm in CR sows and around 16 mm in SA and CA sows. SA sows tended to rebreed earlier after weaning than did CR or CA sows. CA sows and in particular SA sows had higher incidences of agalactia symptoms and higher rectal temperatures post partum than CR sows. The culling rates did not differ between feeding regimes, although the predominant causes for culling did, being shoulder lesions and abscesses in thin CR sows and leg disorders in SA and CA sows. At the end of the experiment, however, more SA sows than others were retained in the experimental herd. It was concluded that the SA feeding regime was superior in terms of sow performance, despite an increased incidence of agalactia symptoms.

Six ruminal cannulated Texel sheep were used to assess the dose response and the effect of live yeasts (Levucell® SC, Saccharomyces cerevisiae CNCM I-1077) on the prevention of induced ruminal latent acidosis. The sheep received, in a replicated 3×3 Latin-square design, an acidotic diet (wheat +lucerne hay, 60:40 (dry matter (DM) basis); starch: 410 g/kg DM) without yeast (control group; L0 treatment), supplemented with 0·2 g/day yeast (4×109 colony-forming units (c.f.u.) per day corresponding to producer recommendations; L1 treatment) or with 2 g/day yeast (4×1010 c.f.u. per day; L10 treatment). The following measurements were carried out: food intake, ruminal pH, ruminal volatile fatty acids (VFA), lactate and ammonia (NH3) concentrations, protozoal and lactate-utilizing bacterial counts, relative proportions of two main bacteria implicated in lactate metabolism (a lactate-producing species, Streptococcus bovis, and a lactate-utilizing species, Megasphaera elsdenii) using specific 16S-rRNA-targeting oligonucleotide probes, activities of lactate dehydrogenase (LDH) and of polysaccharidases involved in plant cell wall (xylanase, carboxymethylcellulase) and starch (amylase) degradation. The acidotic diet (L0) induced a butyric (12 mol per 100 mol total VFA) rather than lactic (<4 mmol/l) ruminal latent acidosis. Ruminal pH, fermentative patterns, lactate-metabolizing bacteria (concentration, LDH activity) and polysaccharidase activities were similar between treatments (P>0·1). Yeast supplementation tended to increase ruminal protozoal population (P<0·1) and intake of animals (P<0·1). The recommended yeast concentration (L1) was sufficient to ensure these effects. The yeast's action may differ according to the nature and function of the micro-organisms involved and the type of fermentative pattern favoured (protozoa/butyrate v. lactate-metabolizing bacteria/lactate and propionate) during ruminal acidosis in sheep.

The effects of a quantitative feed restriction on the digestive physiology of the young rabbit remain largely unclear. Several digestive functions were thus analysed in the rabbit after weaning, using a monofactorial design that produces a linear reduction of the intake, from ad libitum (AL group) to 80%, 70% and 60% of AL (I80, I70 and I60). The restriction programme was applied by giving a daily meal during 21 days after weaning (34 days), and then a 4-day transition period was managed where the feed intake was fixed at 80% of the AL group, before to be fed ad libitum till 69 days of age. The young rabbit quickly adapted to the restriction programme, since within 4 days after weaning they ate totally their ration within 6–7 h after the feed distribution at 8:00, while AL animals consumed 75% of their feed between 15:00 and 8:00. From 55 to 59 days old, rabbits of I70 and I60 groups reached the intake of the I80 group within 1 day, and then the feed intake of restricted animals increased progressively without over-eating. From 54 to 69 days old, the intake of the four groups did not differ and averaged 143.7 g/day per rabbit. During restriction, the live weight and the weight gain decreased linearly with the restriction level. From 55 to 69 days, the weight gain increased linearly according to the restriction level previously applied, but the final weight of restricted rabbits remained lower than AL ones (−3%, −5% and −7%, respectively, for I80, I70 and I60). After 7 days of restriction, the digestibility was not significantly affected by the restriction level, except for crude protein that presented a slightly higher (+1.5 unit, P = 0.05) coefficient in I70 and I60 groups. The mean retention time (MRT) of particles increased by 50% for restricted animals (mean: 26.2 h for I80 and I60) compared to the AL ones, while that of the liquid phase (three times longer than the particles) was linearly and moderately increased with restriction (+20% between AL and I60). In restricted groups, the caecal pH was lower (−0.3 unit, P < 0.05) and could be related to their higher volatile fatty acid (VFA) concentration (+16 mmol/l compared to AL, P < 0.05). The fermentation pattern, ammonia concentration and the caecal bacterial fibrolytic activity remained similar among treatments, although the butyrate proportion tended to be higher in restricted animals. Impact of feed restriction on performances and digestive health is reported in the second part of this study.

Milk is a source of bioactive molecules with wide-ranging functions. Among these, the immune properties have been the best characterised. In recent years, it has become apparent that besides the immunoglobulins, milk also contains a range of minor immune-related proteins that collectively form a significant first line of defence against pathogens, acting both within the mammary gland itself as well as in the digestive tract of the suckling neonate. We have used proteomics technologies to characterise the repertoire of host-defence-related milk proteins in detail, revealing more than 100 distinct gene products in milk, of which at least 15 are known host-defence-related proteins. Those having intrinsic antimicrobial activity likely function as effector proteins of the local mucosal immune defence (e.g. defensins, cathelicidins and the calgranulins). Here, we focus on the activities and biological roles of the cathelicidins and mammary serum amyloid A. The function of the immune-related milk proteins that do not have intrinsic antimicrobial activity is also discussed, notably lipopolysaccharide-binding protein, RNase4, RNase5/angiogenin and cartilage-glycoprotein 39 kDa. Evidence is shown that at least some of these facilitate recognition of microbes, resulting in the activation of innate immune signalling pathways in cells associated with the mammary and/or gut mucosal surface. Finally, the contribution of the bacteria in milk to its functionality is discussed. These investigations are elucidating how an effective first line of defence is achieved in the bovine mammary gland and how milk contributes to optimal digestive function in the suckling calf. This study will contribute to a better understanding of the health benefits of milk, as well as to the development of high-value ingredients from milk.

The objective of this experiment was to investigate the effect of four perennial ryegrass cultivars: Bealey, Astonenergy, Spelga and AberMagic on the milk yield and milk composition of grazing dairy cows. Two 4 × 4 latin square experiments were completed, one during the reproductive and the other during the vegetative growth phase of the cultivars. Thirty-two Holstein–Friesian dairy cows were divided into four groups, with each group assigned 17 days on each cultivar during both experiments. Within each observation period, milk yield and milk composition, sward morphology and pasture chemical composition were measured. During the reproductive growth phase, organic matter digestibility (OMD) was greater for Bealey and Astonenergy (P < 0.001; +1.6%). AberMagic contained a higher stem proportion (P < 0.01; +0.06) and a longer sheath height (P < 0.001; +1.9 cm). Consequently, cows grazing AberMagic recorded a lower milk yield (P < 0.001; −1.5 kg/day) and a lower milk solids yield (P < 0.001; −0.13 kg/day). During the vegetative growth phase, OMD was greater (P < 0.001; +1.1%) for Bealey, whereas the differences between the cultivars in terms of sward structure were smaller and did not appear to influence animal performance. As a result, cows grazing Bealey recorded a higher milk yield (P < 0.001; +0.9 kg/day) and a higher milk solids yield (P < 0.01; +0.08 kg/day). It was concluded that grass cultivar did influence milk yield due to variations in sward structure and chemical composition.

Because not all animal factors influencing profitability can be included in total merit breeding indices for profitability, the association between animal total merit index and true profitability, taking cognisance of all factors associated with costs and revenues, is generally not known. One method to estimate such associations is at the herd level, associating herd average genetic merit with herd profitability. The objective of this study was to primarily relate herd average genetic merit for a range of traits, including the Irish total merit index, with indicators of performance, including profitability, using correlation and multiple regression analyses. Physical, genetic and financial performance data from 1131 Irish seasonal calving pasture-based dairy farms were available following edits; data on some herds were available for more than 1 year of the 3-year study period (2007 to 2009). Herd average economic breeding index (EBI) was associated with reduced herd average phenotypic milk yield but with greater milk composition, resulting in higher milk prices. Moderate positive correlations (0.26 to 0.61) existed between genetic merit for an individual trait and average herd performance for that trait (e.g. genetic merit for milk yield and average per cow milk yield). Following adjustment for year, stocking rate, herd size and quantity of purchased feed in the multiple regression analysis, average herd EBI was positively and linearly associated with net margin per cow and per litre as well as gross revenue output per cow and per litre. The change in net margin per cow per unit change in the total merit index was €1.94 (s.e. = 0.42), which was not different from the expectation of €2. This study, based on a large data set of commercial herds with accurate information on profitability and genetic merit, confirms that, after accounting for confounding factors, the change in herd profitability per unit change in herd genetic merit for the total merit index is within expectations.

Rumen microbiome profiling uses 16S rRNA (18S rRNA, internal transcribed spacer) gene sequencing, a method that usually sequences a small portion of a single gene and is often biased and varies between different laboratories. Functional information can be inferred from this data, but only for those that are closely related to known annotated species, and even then may not truly reflect the function performed within the environment being studied. Genome sequencing of isolates and metagenome-assembled genomes has now reached a stage where representation of the majority of rumen bacterial genera are covered, but this still only represents a portion of rumen microbial species. The creation of a microbial genome (bins) database with associated functional annotations will provide a consistent reference to allow mapping of RNA-Seq reads for functional gene analysis from within the rumen microbiome. The integration of multiple omic analytics is linking functional gene activity, metabolic pathways and rumen metabolites with the responsible microbiota, supporting our biological understanding of the rumen system. The application of these techniques has advanced our understanding of the major microbial populations and functional pathways that are used in relation to lower methane emissions, higher feed efficiencies and responses to different feeding regimes. Continued and more precise use of these tools will lead to a detailed and comprehensive understanding of compositional and functional capacity and design of techniques for the directed intervention and manipulation of the rumen microbiota towards a desired state.
