Parasites represent a great, unknown component of animal biodiversity. Recent efforts have begun to uncover patterns of infection by helminth parasites in several Neotropical lizards. The present study reports, for the first time, levels of helminth infection in a population of the flat lizard Tropidurus semitaeniatus. One hundred and thirty-nine lizards were examined and evidence of five intestinal helminth species was found, comprising four species of nematodes, one species of cestode and an unidentified encysted larval nematode. The most frequently occurring species was the intestinal nematode Parapharyngodon alvarengai, which did not exhibit differences in prevalence and intensity of infection relative to host sex or age/body size. Furthermore, helminth species richness was not related to host body size.

The migration and distribution of Ancylostoma caninum larvae in the tissues of chickens, infected orally with 1,000 larvae, were studied. Larval yield at necropsy from different organs after digestion with artificial gastric juice revealed a 62·9% recovery four hours after inoculation, followed by a sharp decline to 5·4% at 72 hours. Larvae were found in the heart within four hours, the lungs within eight hours and the liver within 12 to 18 hours but no larvae were recovered from the spleen, kidney or brain. Migration in the muscles of head, neck, thorax and abdomen was detected at 12 hours and was maximal at 36 hours. The establishment of patent infection in the definitive host was studied by feeding infected chicks to hookworm-free pups (one chick/pup) 48 hours, 7 days and 14 days after infection. The mean worm burden at necropsy was highest (15) in the pups fed with chicks 48 hours after infection and was three and nil in the other groups respectively.

The helminthic fauna of a number of Water Deer, Hydropoles inermis from Whipsnade Park has been examined.

Fourteen species of Nematodes were recovered, all of which were new records for this host.

Ostertagia lyrata and O. grühneri are recorded for the first time in England.

Trichostrongylus cervarius n. sp is described.

The importance of arresting the spread of the potato-strain of Heterodera schachtii, which is largely responsible for the serious disease known as “potato sickness,” has frequently been emphasised. The cysts of this nematode, as is well known, can withstand long periods of drought, and can therefore be carried about in a viable condition on any object to which small lumps of soil may become attached. In this way the eelworms may be spread, not only from one field to the next by means of farm implements, but also from one locality to another many miles away in soil adhering to seed potato tubers. Seed potatoes grown on eelworm-infected land, however free from disease in other respects, are therefore highly undesirable for planting on clean land, unless some treatment can be carried out which will kill adhering cysts. It was with the object of discovering some practical means of treating seed potatoes so that, without injury to the tubers, adhering eelworm cysts would be killed, that the experiments recorded here were undertaken.

Human alveolar echinococcosis (AE) is caused by the accidental ingestion of Echinococcus multilocularis eggs. Early detection is essential as surgical resection is the only treatment for a complete cure. However, details are unclear about changes in the antibody response during the initial stages of infection, yet such information is useful for early serodiagnosis. Therefore, a long-term investigation was performed into the time course of the antibody response before ‘positive’ detection. Patient sera were used for enzyme-linked immunosorbent assay (ELISA) and Western blotting (WB) analysis using crude antigens extracted from E. multilocularis protoscoleces. Rats were experimentally infected with AE and similarly analysed by ELISA and WB. Among the markers for diagnoses, the 18 kDa band (main marker) appeared last in the preoperative stages and disappeared first after operation in a WB test. Although the 18 kDa antigen can be useful as a marker for AE diagnosis, it cannot contribute to the detection of some patients before the 18 kDa band appearance. To avoid misdiagnosis, different diagnostic antigens such as the 26–28 and 7–8 kDa bands should also be considered. These bands tend to appear earlier than the 18 kDa band and thus offer the potential for early detection of AE. We first observed changes in the antibody response in a relatively early stage after infection in human AE cases. Notably, changes in the antibody response of two intermediate species were similar. These findings provide valuable information for the early detection of human AE cases in the future.

Previous studies have shown that protein deficiencies can hamper both the course of experimental schistosomiasis and normal development of adult worms. To further investigate this relationship, we compared adult male and female Schistosoma mansoni from malnourished and well-fed mice through morphometric and confocal laser scanning microscopy analysis. Swiss mice were fed protein-deficient diets (8%) and infected subcutaneously with approximately 80 S.mansoni cercariae (BH strain, Brazil). Control mice were fed a standard rodent diet (23% protein). The nutritional status was evaluated by body weight gain and albumin values. Mice were sacrificed 63 days post-infection. Recovered worms were stained with hydrochloric carmine and preserved as whole-mounts for bright-field examination and confocal microscopy. The body weight gain and serum albumin concentrations were significantly lower (P <  0.05) in malnourished mice than in controls. In general, all morphometric values of specimens grown in malnourished mice were lower than those of control mice. Schistosome worms grown in malnourished mice had statistically significant differences (P <  0.05) in the reproductive system and tegument than those grown in mice fed standard diets. In female worms, vitelline glands showed few remaining follicles and ovaries lacked mature oocytes. In male parasites, tubercles were fewer in number on the dorsal surface and testicular lobes presented fewer differentiated germinal cells. In summary, we describe novel data supporting the view that low-protein diets may influence the development of adult worms.

The abomasal and small intestinal helminth fauna of three species of captive gazelles (Gazella dama mhorr, G. cuvieri and G. dorcas neglecta) kept in captivity in Almería (southeast Spain) have been studied, and the following species were identified: Nematodirus spathiger, N. filicollis, N. helvetianus, Camelostrongylus mentulatus, Trichostrongylus vitrinus, T. probolurus, T. colubriformis, Ostertagia ostertagi, O. harrisi, Teladorsagia (Ostertagia) circumcincta, and T. (Ostertagia) davtiani. Camelostrongylus mentulatus and N. spathiger were the most prevalent and abundant parasites. Ostertagia ostertagi, O. harrisi, N. helvetianus, and T. (Ostertagia) davtiani were identified for the first time in the genus Gazella. In addition, O. harrisi and Trichostrongylus probolurus are new records for Spain.

The development of lymphoid cells reactive to tapeworm-associated antigens during the course of Hymenolepis diminuta rejection from mice was studied using an in vitro tapeworm extract (TWE)-induced cell proliferation culture system. Mice infected with three cysticercoids on day 0 developed three adult worms by day 7 but worms were rejected by day 21 post-infection. Concomitant with worm rejection was the development of TWE-sensitized lymphoid cells which responded by proliferation when stimulated in vitro with TWE. Sensitized cells were detected in gut-associated mesenteric lymph nodes but were not detected in spleen, axillary lymph nodes, or peyer's patches of infected mice, or in lymphoid organs of non-infected mice. These studies suggest that rejection of H. diminuta from mice is associated with the activities of gut-associated, tapeworm antigen-sensitized immune cells localized in the mesenteric lymph nodes.

May I say, in the first place, how highly honoured I am by the invitation of the Royal Faculty of Physicians and Surgeons of the City in which I received my early medical training to give the sixth Finlayson Memorial Lecture.

A comparison is made of the variations in the mean size (length), reduced weight, oxygen uptake, metabolic rate and the number of contained fully formed metacercariae undergoing autolysis, in mature daughter sporocysts of Microphallus pygmaeus, in sea water, artificial sea water and in a chemically defined nutrient medium (modified medium 199). The work indicates that the sporocysts begin to degenerate almost immediately in the non-nutrient media but have a higher metabolic activity and remain healthy for up to 36 days in the nutrient medium.

The ultrastructure of the flame cells, capillaries, collecting tubes, excretory bladder, excretory atrium, caudal vesicle, lateral caudal ducts and excretory pores of cercariae of Bucephaloides gracilescens (Rudolphi, 1819) Hopkins, 1954 and Prosorhynchus squamatus Odhner, 1905 (Digenea: Bucephalidae) is described. Both species are essentially similar except for some details. The terminal parts of the protonephridia have all the structural features that are typical of trematodes. The collecting tubes in the cercarial body are composed of cells that are wrapped around the lumen. The main collecting tubes are joined to the excretory bladder syncytium by septate junctions. Features of P. squamatus excretory bladder epithelium indicate that it is involved in secretory activity, but this is not the case in B. gracilescens. In both species the luminal surface of the excretory bladder epithelium is increased by lamellae, and the basal plasma membrane forms invaginations. In the bladder syncytium of P. squamatus both apical lamellae and basal invaginations are more developed and mitochondria are also more numerous. The excretory atrium is lined by a syncytium with nucleated cytons located in the surrounding parenchyma. The atrium lining is not continuous with the body tegument and possesses specific secretory inclusions and a thick glycocalyx. Septate junctions connect the atrium syncytium to the excretory bladder epithelium at its anterior end and to the syncytial excretory epithelium lining the caudal vesicle and the lateral caudal ducts at its posterior. In the excretory pores the caudal duct syncytium is joined to the tegument by septate desmosomes.

Hassall and Stiles (1892) described this delicate parasite and gave it the name Strongylus rubidus. Our knowledge of it has recently been increased by Hall (1921) who has shown that it is not a member of the genus Strongylus, as represented by S. equinus of the horse, but is a member of the family Trichostrongylidæ. He has created a new genus Hyostrongylus for its reception and has devoted a large part of his paper to the consideration of a male accessory organ which serves as a guide for the spicules and to which he gives the name telamon.

We make new morphological observations not previously reported for the old acanthocephalan Macracanthorhynchus hirudinaceus (Pallas, 1781) Travassos, 1917, described for the first time about 240 years ago. Our specimens were collected from the wild boar, Sus scrofa Linn., in Ukraine in 2005. We provide comparative morphometrics with other populations from Ukraine, Union of Soviet Socialist Republics, Europe, Brazil and the United States of America. Our specimens from the Ukraine had the smallest trunk (110–120 mm long), longest and thickest hooks, with the third hook being the largest, largest eggs, and a proboscis wider than long. We document the morphology of the proboscis, apical organ, hooks, hook roots, sensory pores, micropores, and eggs with scanning electron microscopy for the first time. We also provide chemical analysis of hooks and eggs using energy-dispersive X-ray analysis, discuss its micropores, and provide a new molecular profile based on 18S rDNA from a European population for the first time. Edge of hook tips feature high levels of calcium and phosphorous but the high level of sulphur is mostly found in the cortical layer of eggs. One new partial 18S rDNA sequence (482 nt length) was generated from an adult specimen of M. hirudinaceus. We present the first 18S rDNA published sequence for this cosmopolitan acanthocephalan obtained from Europe. The amplified region corresponded to the approximate middle region of the small subunit ribosomal ribonucleic acid gene, which is ~1800 nt in length. This molecular contribution is especially valuable in light of the extreme scarcity of genetic information about species of Macracanthorhynchus and of the family Oligacanthorhynchidae as a whole.

To improve diagnostic tools, immunotherapies and vaccine development for trichinellosis surveillance and control there is a need to understand the host immune responses induced during infection with Trichinella zimbabwensis, a tissue-dwelling nematode. In this study, we sought to determine immune responses induced in mice during T. zimbabwensis infection. The parasite strain used (Code ISS1209) was derived from a naturally infected crocodile (Crocodylus niloticus) and is the main Trichinella species prevalent in southern Africa. Sixty 6- to 8-week-old female BALB/c mice were randomly assigned to two equal groups: T. zimbabwensis-infected (n= 30) and the non-infected control group (n= 30). Levels of serum tumour necrosis factor-alpha (TNF-α), interleukin-10 (IL-10), interleukin-4 (IL-4) as well as parasite-specific IgM, IgG, IgG1, IgG2a, IgG2b and IgG3 antibody responses were determined using enzyme-linked immunosorbent assay (ELISA). The cytokines and antibodies provided information on T-helper 1 (Th1)- and Th2-type, T-regulatory and antibody responses. Results showed that during the intestinal stage of infection, higher levels of parasite-specific IgM, IgG, IgG1 (P <  0.05) and IL-10 and TNF-α (P <  0.001) were observed in the Trichinella-infected group compared with the non-infected control group. In the parasite establishment and tissue migration phases, levels of IgG1 and IgG3 were elevated (P <  0.001), while those of IgM (P <  0.01) declined on days 21 and 35 post infection (pi) compared to the enteric phase. Our findings show that distinct differences in Th1- and Th2-type and T-regulatory responses are induced during the intestinal, tissue migration and larval establishment stages of T. zimbabwensis infection.

Fasciola hepatica is the causative agent of fascioliasis, one of the most economically important helminth diseases of livestock worldwide. Traditionally, fascioliasis has been controlled by the strategic use of fasciolicidal drugs, but the emergence of resistant parasites has spurred an interest in developing vaccines as an alternative means of control. Most vaccine studies to date have evaluated conventional antigens, which are exposed to the host's immune system during the course of a natural infection. ‘Hidden’ antigens have proven to be effective vaccine candidates in other parasite species, most notably the blood-feeding nematode parasite, Haemonchus contortus, and tend to be expressed in the intestine or gut of the parasite. Fasciola hepatica is known to ingest large quantities of blood and may be vulnerable to this approach. Most, if not all, of the candidate antigens identified thus far have been membrane-bound glycoproteins which were solubilized by detergents. Here, we have attempted to employ lectins to select gut-associated glycoproteins from complex mixtures of somatic extracts of adult F. hepatica. We have conducted a comprehensive lectin-binding screen on adult histological sections with a panel of 16 fluorescently labelled lectins. Seven of the lectins bound to molecules within the gastrodermis but also bound to a range of other tissues. Within the gut tissues, jacalin and peanut lectins bound selectively to the gut lamellae and gastrodermal cells, respectively. These lectins were then used to isolate proteins from the integral membrane protein component of the adult fluke. Both lectins showed selectivity for relatively simple subsets of proteins compared to the original crude extracts.

Coomansus batxatensis sp. nov., recorded from Vietnam, is described and illustrated and its phylogenetic relationship within the Mononchida is analysed. The molecular data (18S and 28S ribosomal DNA) are provided for the new species. The new species is characterized by small body size (body length, L = 0.7–0.9 mm); buccal cavity sub-rectangular in shape, flattened at base, 21–24 × 12–13 μm or 1.9 (1.7–2.0) times as long as wide; posterior position of dorsal tooth apex (59–63% from the base of buccal cavity); pars refringens vaginae with faint and small (2.5 × 1.7 μm) teardrop-shaped pieces, short pars distalis vaginae; and males with short spicules (50–51.5 μm) with rounded head and conical blade part. The new species is close to Coomansus parvus but differs from it by the smaller buccal cavity size, more posterior position of the dorsal tooth apex, longer tail and presence of males. An updated identification key to Coomansus species and a compendium of all the species known are presented.

A total of 301 blue whiting, Micromesistius poutassou Risso, 1826, ranging in length from 17 to 28 cm, from Motril Bay (Mediterranean coast, south Spain) were examined for anisakid nematodes, as these fish are common items in the Spanish Mediterranean diet. Three anisakid species were morphologically identified with a total prevalence of 10.63%. Anisakis simplex s.l. Rudolphi, 1809 had a prevalence value of 6.65%, compared with 2.66% for A. physeteris Baylis, 1923 and 2.33% for Hysterothylacium aduncum Rudolphi, 1802. Variations in prevalence values with season and host size are discussed. Allozyme markers (leucine aminopeptidase-1) were used to identify anisakid nematodes assigned to the A. simplex complex and all examined larvae were found to correspond genetically to A. pegreffii Nascetti et al., 1986.

Two groups of Galba truncatula and two groups of Lymnaea natalensis were experimentally infected with Fasciola gigantica to determine if snail species had an influence on the redial burden and cercarial shedding of this trematode when snails of both species were infected with the same isolate of miracidia. In the two groups used for the study of redial burden, the total number of free rediae was significantly higher at day 49 post-exposure in L. natalensis than in G. truncatula. In the groups used for cercarial shedding, the life-span of cercaria-shedding snails and those of infected snails which died without cercarial emission, and the duration of the prepatent period were significantly longer in L. natalensis than those noted in G. truncatula. However, the mean numbers of shed cercariae did not significantly differ and showed no differences in their daily distribution throughout the shedding period. These results demonstrate that G. truncatula might be the principal intermediate host of F. gigantica in Egypt, at least in the areas where this lymnaeid species lives.

A new scheme of clarifying clinical forms of toxocariasis is proposed to include: (i) systemic forms: classical VLM and incomplete VLM; (ii) compartmentalized forms: ocular and neurological toxocariasis; (iii) covert toxocariasis; and (iv) asymptomatic toxocariasis. The following markers are helpful in defining clinical forms namely, patient characteristics and history, clinical symptoms and signs, positive serology, eosinophilia and increased levels of IgE. Amongst the available drugs albendazole is the most commonly used, although other benzimidazole compounds have a similar efficacy. The recommended dose of albendazole is 15 mg kg-1 body weight daily for 5 days and in some cases with VLM syndrome the treatment needs to be repeated. An evaluation of treatment efficacy can be made by observing a rise in eosinophilia within a week followed by any improvement in clinical symptoms and signs, lower eosinophilia and serological tests taken over a period of at least 4 weeks. In addition to clinical rationales for the specific treatment of VLM and OLM, preventive treatment needs to be considered bearing in mind the increasing risk of larvae localizing in the brain during the course of an infection. To reduce migration of Toxocara larvae a single course of albendazole is suggested in cases where eosinophilia and serology are at least moderately positive.

During late summer in 1995 to 1997, repeated outbreaks of maculopapular skin eruptions were noted on the legs of children after wading in the pond in the Family Park in Laugardalur, Reykjavík, Iceland. Clinical symptoms developing on the legs resembled those of cercarial dermatitis. An examination of Lymnaea peregra snails from this pond and from the adjacent Lake Tjornin resulted in detection of previously undescribed schistosome cercariae. This is the first report of schistosomes in Iceland and also the most northern occurrence of these parasites in Europe.
