Given that surgical castration is a painful practice performed on millions of pigs every year, a need to identify novel reliable pain assessment tools exists in order to test anaesthetic and analgesic protocols that may reduce related pain. Two treatments were considered: handling (H) and surgical castration (C). Physiological (cortisol, lactate, glycaemia, rectal and eye temperature) and behavioural variables (latency to move after treatment and alterations in posture and walking) were analysed. Cortisol showed the greatest level in C piglets within 20 min after the surgical procedure and a positive correlation with glucose concentration. Eye temperature was higher in C piglets, and the same difference was detected for rectal temperature 3 h after castration. Behavioural parameters revealed that C piglets had longer latency to move and a higher percentage of them showed alterations in posture and walking. Results of this study showed that, in castrated piglets behavioural and physiological alterations occur mainly in the first 3 h from treatment. Latency to move, alterations in posture and walking, and eye temperature appear to give additional and useful information in piglet pain assessment. However, differently from the behavioural parameters considered, eye temperature involves several manipulations of the animals and a long process to acquire the data.

The nutritional value of a pelleted fattening concentrate mixture, in which heat-sterilized poultry litter (PL) was included at rates of 0, 15, 25 and 35%, was studied. A digestion and nitrogen balance trial was carried out with rams, and a feeding trial was carried out with intact male cattle from the age of 8 mo to slaughter at 14 mo.

Digestibility of crude protein was related inversely to PL content of the diet. This decrease in digestibility was not accompanied by a decrease in the quality of the protein digested.

Gross and digestible energy contents of the diets were related inversely to their PL content. There were no significant differences in blood urea concentration. A concentrate mixture containing 1% urea but otherwise similar to the control, was included in the feeding trial.

There were no significant differences in rate of live-weight or carcass gain between treatments. The differences in degree of fatness of the carcasses tended to be related inversely to PL% in the diet, but they were small and reached significance only with the extreme treatments. Feed intake on PL-containing diets was higher than on the control diet, but feed conversion efficiency was impaired when the proportion of PL exceeded 25%. No significant correlations were found between total volatile fatty acids in rumen liquor and daily live-weight gain.

This study was conducted on 94 Frieswal (5/8 Holstein Friesian 3/8 Sahiwal) crossbred bulls of three different grades, categorized based on their semen freezability visualising Group 1 (consistently freezable semen producer bulls, N = 11), Group 2 (inconsistent freezable, N = 16) and Group 3 (Non freezable, N = 67). Each group was further divided into two classes that is young (up to 30 months) and adult (31 to 70 months) bulls depending upon their age. Sperm morphology was studied by using the eosin-nigrosin staining technique. Bulls age significantly (P < 0.01) affected semen quality and sperm morphology. In adult bulls, semen volume, mass activity and sperm concentration were 36%, 17.56% and 19.6%, respectively, higher than young. Initial progressive motility (%) and livability showed significant (P < 0.01) improvement with the advancement of age (43.37 ± 1.21 and 67.71 ± 1.11, respectively, in young; 53.02 ± 1.11 and 74.17 ± 1.03, respectively, in adult). In young bulls, sperm head, mid piece, tail abnormality and total abnormal sperm percent (12.38 ± 0.92, 4.87 ± 0.24, 11.01 ± 0.60 and 28.26 ± 1.34, respectively) were 1.85, 1.27, 1.20 and 1.44 folds higher than that of their mature stage (6.69 ± 0.64, 3.82 ± 0.32, 9.14 ± 0.64 and 19.66 ± 1.31, respectively). Significant reduction (P < 0.01) in micro cephalic sperm, free heads, bent mid piece, looped mid piece and proximal protoplasmic droplets were observed at mature age as compared with their younger stage. In bulls of consistent freezing category, abnormal sperm heads significantly decreased from 4.40 ± 0.31% to 3.28 ± 0.02% on maturity. Similarly, in inconsistent freezing grade bulls sperm head abnormality (9.28 ± 0.75% to 5.13 ± 1.20%) and total abnormal sperm percent (24.89 ± 1.43 to 18.73 ± 3.40) was decreased over the age. On the contrary, in non-freezing category bulls’ sperm morphology did not show significant (P > 0.05) improvement with age advancement, rather some abnormalities like long slender head, under developed/deformed head, abaxial implantation of mid piece, double mid piece, stump tail and distal protoplasmic droplets tend to increased significantly (P < 0.05) with age of bulls. Results indicated that in potential Frieswal bulls semen quality and sperm morphology were improved from young to mature stage, where as, in poor quality (non-freezing) semen producer bulls neither the morphology nor the semen quality showed any improvement with maturity. It was recommended that crossbred bulls producing more than 25% morphologically abnormal sperms in young age (below 30 months) along with poor progressive motility (<50%) and low sperm concentration (<1000 million/ml) need immediate culling with out any expectation of further improvement in semen quality with age advancement.

To reduce nitrogen excretion (NEx) the effect of nitrogen supply on nitrogen retention (NR) and NEx was studied in pregnant sows during three parities. A nitrogen supply of 40 g/day (treatment L) was compared with 62 g/day (treatment C) during day 0 to 85 of pregnancy. In the last month of pregnancy nitrogen supply on treatment L was 50 g/day and on treatment C 74 g/day. Faecal digestible lysine supply followed the same pattern as nitrogen. Nitrogen and energy balances were measured during mid pregnancy (day 50 to 60; no. = 22) and late pregnancy (day 105 to 112; no. = 60). During mid pregnancy treatments had a minor affect on NR and energy metabolism. NEx on treatment L was proportionately about 0·40 lower than on treatment C. During late pregnancy NR was lower on treatment L than on treatment C. NEx on treatment L was proportionately reduced by 0·35 compared with treatment C. In parity 1 the lower NR on treatment L was compensated by a higher fat gain. Results indicate that nitrogen and faecal digestible lysine supply on treatment L were close to minimal requirement in late pregnancy. Assumed values for maintenance requirement of nitrogen (0·45 g N per kg M0·75) and efficiency of ingested nitrogen to NR (0·6) were in line with the results of the balance trials. The observed nitrogen retention of sows, especially during mid pregnancy, permits a reduced nitrogen supply to reduce NEx.

Scans of the m. longissimus and overlying fat at the 12th rib were taken, just prior to slaughter, on 254 lambs using the Scanogram and Danscanner ultrasonic machines. The lambs were from a ram breed trial and comprised six sire breed × five dam breed groups. Mean live weight at slaughter was 44·5 kg.

The precision with which scan measurements predicted comparable carcass measurements and carcass lean content (g/kg lean) was examined within groups with variation in live weight eliminated.

Residual s.d. for the prediction of carcass m. longissimus area from the corresponding scan measurement was 141 and 158 mm2 for the Scanogram and Danscanner respectively (s.d. of carcass m. longissimus area was 162mm2). Residual s.d. for the prediction of lean from the best combination of fat measurements was 32·0 for the Scanogram and 35·8 for the Danscanner (s.d. of lean was 38·5). M. longissimus did not increase the precision of lean prediction.

In a comparable evaluation of the Scanogram, involving 147 lambs, the residual s.d. for the prediction of lean was 24·0 (s.d. = 28·9).

The levels of precision obtained were low and it is debatable whether the ultrasonic techniques examined would be of value for carcass lean prediction in practical applications. They might be of use in selection programmes within nucleus flocks of the more important meat sire breeds or as an aid in experiments for differentiating between animals of widely differing composition.

The faecal egg count (FEC) and total egg output is reduced in parasite-resistant sheep and breeding sheep for resistance should reduce infective larval contamination of pastures. This research compares lambs and rams from the parasite-resistant flock, Rylington Merinos, with unselected controls at the same level of larval challenge and the same level of food supply in an animal-house environment. The sheep were penned individually. The parasite resistant sheep excreted 0·85-fold less eggs than the control animals after infection with Trichostrongylus colubriformis and Ostertagia circumcincta for 12 or 18 weeks. During the infection period the parasite resistant lambs had higher plasma immunoglobulin concentration and higher ratio of globulin to albumin than the control animals. In the mature rams these genotype differences diminished. They also had lighter mass of the small intestine with a higher proportion of mucosa, but heavier mass of the large intestine. Parasite infection caused a depletion of blood glutathione in the lambs given food at maintenance or 1·5 × maintenance, and the parasite-resistant sheep (both lambs and rams) also had lower glutathione concentration. Maintaining good body nutrient reserve before the infection reduced the egg output in the early stage of the infection. Food intake had little effect on worm egg production in young sheep given food at maintenance or above. It is suggested that an improvement of nutrition, sulphur-containing amino acids in particular, is required to recover the loss of productive performance caused by the infection.

In each of two trials, three groups of eight cows, with condition scores of 1-5 to 2, 2-5 to 3 and 3-5 to 4 at calving, were given a complete diet for the first 16 weeks of lactation. Food intake, milk yield, live weight and condition score were measured, and related to levels of plasma glucose and free fatty acids, serum insulin, prolactin and progesterone in weekly blood samples, in an attempt to explain differences in food intake.

In both trials, levels of plasma free fatty acids were high after calving for all cows and declined rapidly until week 8, after which they were fairly constant. Glucose and insulin levels increased after calving, producing curves similar to that for dry-matter intake in the first 8 weeks of lactation. Prolactin levels fluctuated too markedly to provide useful information. Progesterone levels increased until week 7 and then fluctuated at 3-week intervals coincident with oestrus. Dry-matter intake was negatively correlated with free fatty acid levels and positively correlated with insulin in trial 1, and with glucose and progesterone in both trials.

Resilience is the ability of an animal to return soon to its initial productivity after facing diverse environmental challenges. This trait is directly related to animal welfare and it plays a key role in fluctuations of livestock productivity. A divergent selection experiment for environmental variance of litter size has been performed successfully in rabbits over ten generations. The objective of this study was to analyse resilience indicators of stress and disease in the divergent lines of this experiment. The high line showed a lower survival rate at birth than the low line (−4.1%). After correcting by litter size, the difference was −3.2%. Involuntary culling rate was higher in the high than in the low line (+12.4%). Before vaccination against viral haemorrhagic disease or myxomatosis, concentration of lymphocytes, C-reactive protein (CRP), complement C3, serum bilirubin, triglycerides and cholesterol were higher in the high line than in the low line (difference between lines +4.5%, +5.6 µg/ml, +4.6 mg/ml, +7.9 mmol/l, +0.3 mmol/l and +0.4 mmol/l). Immunological and biochemical responses to the two vaccines were similar. After vaccination, the percentage of lymphocytes and CRP concentration were higher in the low line than in the high one (difference between lines +4.0% and +13.1 µg/ml). The low line also showed a higher increment in bilirubin and triglycerides than the high line (+14.2 v. +8.7 mmol/l for bilirubin and +0.11 v. +0.01 mmol/l for triglycerides); these results would agree with the protective role of bilirubin and triglycerides against the larger inflammatory response found in this line. In relation to stress, the high line had higher basal concentration of cortisol than the low line (+0.2ng/ml); the difference between lines increased more than threefold after the injection of ACTH 1 to 24, the increase being greater in the high line (+0.9 ng/ml) than in the low line (+0.4 ng/ml). Selection for divergent environmental variability of litter size leads to dams with different culling rate for reproductive causes and different kits’ neonatal survival. These associations suggest that the observed fitness differences are related to differences in the inflammatory response and the corticotrope response to stress, which are two important components of physiological adaptation to environmental aggressions.

A study was conducted to evaluate the effects of pre partum (PRE) and post partum (POST) dietary energy and nutrient supply (E) and their interactions on feed intake, performance and energy status in dairy cows of three breeds. In this experiment, the effects of three energy and nutrient supply levels (low (L), medium (M), high (H)), both pre-calving and post-calving, were investigated, using a 3×3 factorial arrangement of treatments. In both phases (84 days pre- and 105 days post-calving) E levels applied to a total of 81 multiparous cows of breeds Simmental (SI), Brown Swiss (BS) and Holstein–Friesian (HF; n=27 for each breed), were 75%, 100% and 125% of recommendations of the German Society of Nutrition Physiology (GfE). Dry matter intake (DMI) was restricted, if energy intake exceeded target values. Pre partum DMI and energy intake were different as designed, liveweight and body condition score (BCS) of SI cows were higher, but EB was lower, compared to BS and HF cows. Milk yield and composition were influenced by all three main experimental factors (EPRE, EPOST, breed). Energy-corrected milk yield was 25.6, 28.6 and 30.1 kg/day for LPRE, MPRE and HPRE as well as 21.5, 30.1 and 32.6 kg/day for LPOST, MPOST and HPOST, respectively. Numerically, only for milk protein content the interactions EPRE×EPOST and EPRE×breed reached significance. Impact of energy supply pre-calving was more pronounced when cows had lower energy supply post-calving and vice versa. On the other hand, milk yield response of cows to energy supply above requirements was greater for cows that were fed on a low energy level pre partum. Impact of energy level pre partum was higher for HF cows, showing that their milk production relies to a greater extent on mobilization of body reserves. Increasing energy supply pre partum led to a more negative energy balance post partum, mainly by increasing milk yield and content, whereas feed intake was slightly reduced. Increasing energy supply post partum enhanced milk yield as well as milk protein and lactose content. Calculated energy balance corresponded well with liveweight and BCS change. Response of milk yield to increasing energy supply followed the principle of diminishing returns, since energy was increasingly partitioned to body retention. Increasing energy supply pre partum enhances milk yield and content post partum, but exacerbates negative energy balance and its consequences.

The aim of this study was to investigate the BW and immune status of lambs reared under natural conditions or under artificial conditions fed two different colostrum amounts. In this study, 60 lambs were randomly divided into groups according to treatment. Twenty lambs remained with their dams (natural rearing (NR) group). Forty lambs were removed from their dams at birth. Lambs were bottle-fed with a pool of sheep colostrum, receiving either 4 g of IgG/kg of BW at birth (C4 group) or 8 g of IgG/kg of BW at birth (C8 group). The total colostrum amount was equally divided into three meals at 2, 14 and 24 h after birth. After this period, lambs were bottle-fed a commercial milk replacer. Blood plasma sample analysis and BW recordings were carried out before feeding at birth and then at 1, 2, 3, 4, 5 and 20 days after birth. Another blood sample analysis and BW recording was carried out when animals reached 10 kg of BW. During weaning (30 days), sampling was carried out every 5 days. Blood plasma was used to determine the concentrations of IgG and IgM and the complement system activity – total and alternative pathways. The NR group showed greater BW than the C4 and C8 groups during milk feeding period, whereas the C4 and C8 groups had greater BW than the NR group at the end of weaning period. The C8 and NR groups had greater plasma IgG and IgM concentrations than the C4 group during milk feeding period. In addition, C4 and C8 groups showed similar IgG concentrations and greater IgM concentrations than the NR group at the end of the weaning period. Complement system activity was greater in the NR group than in the C4 and C8 groups during the first 3 days after birth. In conclusion, lambs fed amounts of colostrum equivalent to 8 g of IgG/kg of BW showed similar immune variables compared to lambs reared under natural conditions, obtaining a greater BW at the end of the weaning period. Nevertheless, this study shows that not only the colostrum amount but also the management during the milk feeding and weaning period, such as stress produced by dam separation, milk quality and suckling frequency, can affect the final immune status of lambs.

Fertilization is an intricate cascade of events that irreversibly alter the participating male and female gamete and ultimately lead to the union of paternal and maternal genomes in the zygote. Fertilization starts with sperm capacitation within the oviductal sperm reservoir, followed by gamete recognition, sperm–zona pellucida interactions and sperm–oolemma adhesion and fusion, followed by sperm incorporation, oocyte activation, pronuclear development and embryo cleavage. At fertilization, bull spermatozoon loses its acrosome and plasma membrane components and contributes chromosomes, centriole, perinuclear theca proteins and regulatory RNAs to the zygote. While also incorporated in oocyte cytoplasm, structures of the sperm tail, including mitochondrial sheath, axoneme, fibrous sheath and outer dense fibers are degraded and recycled. The ability of some of these sperm contributed components to give rise to functional zygotic structures and properly induce embryonic development may vary between bulls, bearing on their reproductive performance, and on the fitness, health, fertility and production traits of their offspring. Proper functioning, recycling and remodeling of gamete structures at fertilization is aided by the ubiquitin–proteasome system (UPS), the universal substrate-specific protein recycling pathway present in bovine and other mammalian oocytes and spermatozoa. This review is focused on the aspects of UPS relevant to bovine fertilization and bull fertility.

It has become common practice in pig fattening production systems to castrate young boar piglets without the use of anaesthesia. In this study, we examined whether or not CO2 gas is capable of inducing an acceptable anaesthetic state during which castration can be performed. The first step was to identify the most promising CO2/O2 mixture. Based on the results from this first experiment, a mixture of 70% CO2 + 30% O2 was chosen for further investigation as a potential anaesthetic during the castration of young piglets. Thereby, it was established whether the duration and depth of anaesthesia were acceptable for castration where the animal has to be insensible and unconscious. Physiological effects were assessed based on electroencephalogram (EEG) and electrocardiogram (ECG) measurements, blood gas values and behavioural responses. During the induction phase, the only typical behaviour the piglets exhibited when exposed to the 70/30 gas mixture was heavy breathing. All piglets (n = 25) lost consciousness after approximately 30 s according to the EEG. Heart rate decreased slowly during the induction phase, a serious drop occurred when piglets lost their posture. Immediately after this drop, the heart rate neared zero or showed a very irregular pattern. Shortly after loss of posture, most animals showed a few convulsions. None of the animals showed any reaction to castration in behaviour and/or on the EEG and ECG. On average, the piglets recovered within 59 s, i.e. EEG returned to its pre-induction pattern and piglets were able to regain a standing position. After 120 s, heart rate returned to pre-induction levels. In order to explore the usage range of CO2 concentration, 24 piglets were exposed to 60% CO2 + 20% O2 + 20% N2 for up to 30 s after loss of consciousness (as registered on EEG), and castrated after removal from the chamber. Sixteen of the 24 animals showed a reaction to the castration on the EEG. To establish the maximum time piglets survive in 70% CO2 + 30% O2, five piglets were placed in this mixture for 3 min. Two of them died. After that, four piglets were placed in this mixture for 2 min after unconsciousness, one died after 2 min. It was concluded from this study that it is possible to anaesthetise piglets with a mixture of 70% CO2 + 30% O2, but that there are limits to its safety in terms of CO2 concentration and duration of exposure. Before implementation for practical use, further research is essential to assess the limits of gas concentration and exposure times.

The influence of environmental stimuli considered biologically relevant for nest building sows, on nursing and suckling behaviour and piglet growth was investigated. Effects of floor type (beach sand v. concrete) and substrate type (straw feeder v. no straw feeder) were examined in a 2 × 2 factorial experiment with nine replicates of four multiparous sows. The sows were kept individually in roofed 7·6 m2 ‘get-aw ay-pens’ from 1 week pre-partum until 13 to 15 days post-partum. The behaviour of sows and piglets were video recorded for 24 h on days 0, 3, 6 and 12 post-partum.

The latency from termination of farrowing until perceptible milk ejection tended to be shorter for experienced sows (parity 4 to 5) kept on sand than on concrete floors (1207 (s.e. 109) min; no. = 7 v. 1725 (s.e. 123) min; no. = 6, respectively; F2,8 = 3·93, P < 0·07) and for experienced sows with access to a straw feeder than without access to a straw feeder (1257 (s.e. 113) min; no. = 8 v. 1666 (s.e. 132) min; no. = 5, respectively; F2,8 = 3·56, P < 0·08). On days 3 and 6 a longer duration of suckling was found for sows on sand v. concrete floors (384 (s.e. 20) v. 327 (s.e. 16) s on day 3; F1,19 = 5·6, P < 0.03 and 377 (s.e. 14) v. 318 (s.e. 13) on day 6; F1,21 = 9·09; P < 0·01, respectively) and with straw feeder v. without straw feeder (385 (s.e. 20) v. 326 (s.e. 18) s on day 3; F1,19 = 5·11, P < 0.04 and 372 (s.e. 14) v. 323 (s.e. 13) on day 6; F1,21 = 6·21; P < 0·03, respectively). On day 3, a tendency for a sand × straw feeder interaction (F11,8 = 3·58, P < 0.08), showed that sows without environmental stimuli terminated more sucklings than sows with access to a straw feeder and sows kept on sand (32 (s.e. 7) %, 16 (s.e. 4) % and 13 (s.e. 5) %, respectively). Similarly, on day 6 a significant sand × straw feeder interaction (F316 = 7·15, P < 0·01) led to increased frequency of foreleg rowing for sows without environmental stimuli compared with the three other treatments (14 (s.e. 2) v. 7 (s.e. 1), 5 (s.e. 1) and 6 (s.e. 1) foreleg rowings per h, respectively). During the stay in the experimental pens, the growth rate of piglets kept on sand tended to be higher than for piglets kept on concrete floors (261 (s.e. 33) v. 240 (s.e. 30) g per piglet per day; F1,25 = 3·38; P < 0·08).

The results of the present experiment indicate that provision of biologically relevant stimuli affect the nursing and suckling behaviour of sows and piglets. The reduced termination of sucklings, the reduced frequency of foreleg rowing, the increased duration of suckling as well as the earlier development of suckling behaviour might have been advantageous for the early milk intake of the piglets.

We evaluated the efficiency of selection for body weight in a cooperative village breeding program for Menz sheep of Ethiopia under smallholder farming system. The design of the program involved organizing villagers in a cooperative breeding group to implement selective breeding of their sheep. The program was jump-started through a one-time provision of elite rams from a central nucleus flock, but subsequent replacement rams were selected from within the village flocks. We also evaluated body weight trends in a village where cooperative breeding was not implemented and individual farmers managed their flocks under traditional breeding practices. Under traditional breeding practices, genetic progress over 8 years either stagnated or declined in all the weights recorded. In the cooperative villages, selection differentials of 2.44 and 2.45 kg were achieved in 2010 and 2011 selection seasons, respectively. Birth weight, 3-month weight and 6-month weight increased, respectively, by 0.49, 2.29 and 2.46 kg in the third-generation lambs over the base generation. Improved rams supplied from the central nucleus flock gave an initial genetic lift of 14.4% in the 6-month weight. This was higher than the gain achieved from selection in the village flocks, which was 5.2%. Our results showed that village-based genetic improvement in body weights under smallholder conditions could be feasible if appropriate designs are adopted and that commencing with elite central nucleus rams help jump-start village-based programs.

There is considerable debate as to the optimal light intensities for growing chickens. This is influencing regulations and industry practices. The present study examines the preference of broiler chickens for light intensity. A choice system was developed to allow determination of the preferences of broiler chickens for light intensity. This system had three light proof pens each with feeders or waterers but different light intensities. There was a connecting transit pen with a light intensity of 1 to 2 lux. This allowed birds access to the pens each with feeders or waterers. There were markedly more chickens observed in the pens each with feeders or waterers and a light intensity of 20 lux than 5 lux. Moreover, more feed was consumed in the 20 lux pens than 5 pens. There were also high numbers of chickens in the transit compartment with its low light intensity (1 to 2 lux) and no feeders or waterers. Broiler chickens exhibited a preference for 20 lux light intensity for feeding compared to 5 lux light intensity. The present study supports the view that there should be a light intensity of at least 20 lux for the areas around the feeders and also suggests that light intensity may be reduced in other areas for resting and other activities.

To alleviate adverse effects of heavy metal toxicity, diverse range of removing methods have been suggested, that is usage of algae, agricultural by-products and microorganisms. Here, we investigated lead (Pb) biosorption efficacy by two lactic acid bacteria species (LABs) in broiler chickens. In an in vitro study, Pb was added to culture medium of LABs (Lactobacillus pentosus ITA23 and Lactobacillus acidipiscis ITA44) in the form of lead acetate. Results showed that these LABs were able to absorb more than 90% of Pb from the culture medium. In follow-up in vivo study, LABs mixture was added to diet of broiler chickens contained lead acetate (200 mg/kg). Pb exposure significantly increased lipid peroxidation and decreased antioxidant activity in liver. The changes were recovered back to normal level upon LABs supplementation. Moreover, addition of LABs eliminated the liver tissue lesion and the suppressed performance in Pb-exposed chicks. Analysis of liver and serum samples indicated 48% and 28% reduction in Pb accumulation, respectively. In conclusion, results of this study showed that L. pentosus ITA23 and L. acidipiscis ITA44 effectively biosorb and expel dietary Pb from gastrointestinal tract of chickens.

This review brings together research findings on cervical relaxation in the ewe and its pharmacological stimulation for enhancement of the penetration needed for transcervical insemination and embryo transfer. On the basis that the success of artificial insemination is the percentage of ewes lambing, a review is made of recent research aimed at understanding and minimising the sub-lethal effects of freezing and thawing on the viability of spermatozoa, their membrane integrity and their ability to migrate through cervical mucus, as these characteristics have a major influence on fertility, particularly when semen is deposited, artificially, in the os cervix. Milestones of achievement are given for transcervical intrauterine insemination, embryo recovery and transfer and the birth of lambs of pre-determined sex, firstly following intracytoplasmic sperm injection, then laparoscopic intrauterine insemination using highly diluted flow-cytometrically sorted fresh semen and subsequently by os cervix insemination using sexed semen that had been frozen and thawed. Diversity of research endeavour (applied, cellular, molecular), research discipline (anatomy, histology, immunology, endocrinology) and research focus (cell, tissue, organ, whole animal) is embraced within the review as each has significant contributions to make in advancing recent scientific findings from the laboratory into robust on-farm transcervical insemination and embryo transfer techniques.

The aim of this trial was to study the concentration of zinc (Zn), iron (Fe), copper (Cu), manganese (Mn), selenium (Se), cobalt (Co) and iodine (I) in milk and blood serum of lactating donkeys, taking into account the effects of lactation stage and dietary supplementation with trace elements. During a 3-month period, 16 clinically healthy lactating donkeys (Martina-Franca-derived population), randomly divided into two homogeneous groups (control (CTL) and trace elements (TE)), were used to provide milk and blood samples at 2-week intervals. Donkeys in both groups had continuous access to meadow hay and were fed 2.5 kg of mixed feed daily, divided into two meals. The mixed feed for the TE group had the same ingredients as the CTL, but was supplemented with a commercial premix providing 163 mg Zn, 185 mg Fe, 36 mg Cu, 216 mg Mn, 0.67 mg Se, 2.78 mg Co and 3.20 mg I/kg mixed feed. The concentrations of Zn, Fe, Cu, Mn, Se, Co and I were measured in feeds, milk and blood serum by inductively coupled plasma-MS. Data were processed by ANOVA for repeated measures. The milk concentrations of all the investigated elements were not significantly affected by the dietary supplementation with TE. Serum concentrations of Zn, Fe, Cu Mn and Se were not affected by dietary treatment, but TE-supplemented donkeys showed significantly higher concentrations of serum Co (1.34 v. 0.69 μg/l) and I (24.42 v. 21.43 μg/l) than unsupplemented donkeys. The effect of lactation stage was significant for all the investigated elements in milk and blood serum, except for serum manganese. A clear negative trend during lactation was observed for milk Cu and Se concentrations (−38%), whereas that of Mn tended to increase. The serum Cu concentration was generally constant and that of Co tended to increase. If compared with data reported in the literature for human milk, donkey milk showed similarities for Zn, Mn, Co and I. Furthermore, this study indicated that, in the current experimental conditions, the mineral profile of donkey milk was not dependent on dietary TE supply.

Three concentrate mixtures were compared in a replicated 3 × 3 Latin square feeding trial with cows in early lactation to evaluate the effect of inclusion of tallow. The concentrates were based on barley and soya bean meal (223 g crude protein per kg dry matter) with either no tallow addition (control group) or with the addition of a protected tallow premix (0·75 spray-dried formaldehyde-treated tallow, 0·25 casein) or an unprotected tallow premix (0·75 spray-dried tallow, 0·25 casein), both added at 138 g/kg. Each concentrate was given at a rate of 8 kg per cow per day and silage was offered ad libitum. The inclusion of unprotected tallow in the concentrate did not significantly change milk yield, fat concentration, lactose concentration or live weight compared with those in the control group. The inclusion of the protected tallow supplement resulted in significantly higher yields of milk (22·9 kg/day), fat (0·95 kg/day) and lactose (1·05 kg/day) compared with those of the control group (21·3 kg/day, 0·79 kg/day, 0·95 kg/day), whilst milk protein concentration was significantly reduced. A digestibility trial with steers showed that, when hay was given with the diets, inclusion of tallow in the concentrate significantly increased the digestible and metabolizable energy concentration of the diets compared with the control diet, but dry matter, energy and fibre digestibility was reduced.

The objective of this study was to measure the effect of feeding two total mixed rations (TMRs), differing in their roughage content and in vitro dry matter (DM) digestibility, on the physiological response and energy balance of lactating cows. The partitioning of metabolizable energy intake (MEI) between heat production (HP) and retained energy (RE) of cows held under hot weather conditions and external evaporative cooling was measured. In all, 42 lactating cows were divided into two similar sub-groups, each of 21 animals, and were fed either a control (CON) ration containing 18% roughage neutral detergent fiber (NDF) or an experimental (EXP) TMR containing 12% roughage NDF and used soy hulls as partial wheat silage replacer. The in vitro DM digestibility of the CON and EXP TMR was 75.3% and 78.6%, respectively (P < 0.05). All cows were cooled by evaporative cooling for 2 adaptation weeks plus 6 experimental weeks under hot weather conditions. The EXP diet reduced rectal temperature and respiratory rate of the cows while increasing their DM intake (DMI) from 23.1 to 24.7 kg/cow per day, milk yield from 41.9 to 44.2 kg and yield of energy-corrected milk from 38.7 to 39.7 kg, as compared with the CON group. Cows fed the EXP TMR had increased RE in milk and body tissue, as compared with the CON group, but the diets had no effect on the measured HP that was maintained constant (130.4 v. 130.8 MJ/cow per day) in the two groups. The measured MEI (MEI = RE + HP) and the efficiency of MEI utilization for RE production were also similar in the two dietary groups.
