The effect of two dietary electrolyte balance (dEB, Na+ + K+ – Cl-) levels (–135 and 145 mEq/kg diet) on heat production, energy and nitrogen retention in piglets was assessed. The experiment consisted of a 13-day adaptation period and a 7-day balance period in two open-circuit climate respiration chambers. Nine groups of three (4 weeks old) crossbred barrows were assigned to one of two diets (five and four groups for –135 and 145 mEq/kg dEB diets respectively). During the balance period, diets were provided at 2·3 times the energy requirement for maintenance in two equal meals daily. Total heat production for each group was determined every 9 minutes from the exchange of CO2 and O2. Faeces and urine mixture was quantitatively collected during the balance period to measure energy and nitrogen balance. Total heat production and metabolizable energy costs for maintenance tended (P 0·10) to be higher in the 145 mEq/kg dEB group (681 and 443 kJ/kg0·75 per day respectively) than in the –135 mEq/kg dEB group (660 and 412 kJ/kg0·75 per day respectively). Differences in total heat production between the two dEB groups mainly occurred in the daytime (light period), when significance level was P 0·01. The respiratory quotient and energy retention as fat were numerically (but not statistically significantly) lower in the 145 mEq/kg dEB group compared with –135 mEq/kg dEB. In conclusion, energy balances were similar for both treatments. However in the daytime (light period), piglets needed more energy for maintenance after ingesting a diet with a dEB level of 145 mEq/kg compared to a diet with a dEB level of –135 mEq/kg at a restricted feeding level.

Nutritionists have been discussing whether the dietary supplementation of cyst(e)ine is required as a part of the dietary methionine (Met) in the total sulfur amino acid (TSAA) requirement to achieve optimum performance in broilers. Part of Met is converted to cysteine (Cys) to meet the Cys requirement, especially for feather growth. The TSAA requirement has been determined by using graded levels of free Met in the diet, without supplementation of free cyst(e)ine. It has also been argued that the Met to Cys ratio (Met : Cys) changes with age and even with different Met sources. The objective of this study was to evaluate the two sources of Met, while determining the proportion of Met and Cys in total dietary TSAA that optimize the performance of broilers. A performance assay was carried out in a factorial arrangement (5 × 2) using 1080 broilers from 42 to 56 days of age fed diets having different dietary proportions of Met and Cys (44 : 56, 46 : 54, 48 : 52, 50 : 50 or 52 : 48) while maintaining the same dietary TSAA in the diets. Two synthetic Met sources (dl-Met or l-Met) were used for each of the diets with different dietary Met : Cys ratios. Twenty-one broilers of the same age were fed the diets 44 : 56, 48 : 52 and 52 : 48 by supplementing the diet with L-(15N) Met or L-(15N2) Cystine to study the metabolism of TSAA. No differences were observed between Met sources for feed intake, BW gain and feed conversion ratio (FCR; P > 0.05); however, FCR was numerically improved at 50 : 50 Met : Cys. Regarding TSAA utilization, the conversion of Met to Cys increased with increase in Met : Cys ratios, but the concentration of Met intermediates decreased. Broiler chickens responded to different dietary proportions of sulfur amino acids by altering their sulfur amino acid metabolism, and diets containing 50 : 50 Met : Cys is recommended for broilers of age 42 to 56 days.

Recent findings suggest there is a complex interaction between the IGF system and the inflammatory immune response. The objective of this study was to determine whether gene expression of growth factors (IGF-1, IGF-binding protein-3 (IGFBP-3) and growth hormone receptors (GHR)) in the liver is associated with gene expression of immunomodulators in the liver, including C-reactive protein (CRP), serum amyloid A (SAA), haptoglobin (Hp), interferon-α (IFN-α), IFN-γ, tumour necrosis factor-α (TNF-α), interleukin (IL)-1β, IL-6, IL-10 and IL-18, as well as with the presence of Salmonella spp., Lawsonia intracellularis, Brachyspira spp., enterotoxigenic Escherichia coli, methicillin-resistant Staphylococcus aureus, swine influenza virus, and porcine reproductive and respiratory syndrome virus (PRRSV) in nursery pigs (n=74) from commercial farms (n=4). Gene expression was quantified using reverse transcription quantitative-PCR (RT-qPCR) and the data were modelled using logistic regression methods. Pigs with elevated IGF-1 expression were less likely to have increased expression of TNF-α (odds ratio (OR)=0.14, P<0.01) and IL-18 (OR=0.19, P<0.05), and less likely to be colonized with PRRSV (OR=0.03, P<0.01). Pigs with increased expression of IGFBP-3 were more likely to have elevated IL-6 expression (OR=8.5, P<0.05). It was also observed that IGFBP-3 and IGF-1 were significantly associated when Hp expression was low (OR=30, P<0.05), but this association was not significant when Hp expression was high (P=0.54). Pigs with increased expression of GHR were less likely to have elevated expression of SAA (OR=0.01, P<0.05) and IL-1β (OR=0.03, P<0.05), but more likely to have increased expression of CRP (OR=290, P<0.01). Overall, there appears to be an inverse association between the hepatic expression of the IGF system (IGF-1, IGFBP-3, GHR) and certain cytokines (IL-1β, IL-18, TNF-α) and acute-phase proteins (SAA, Hp).

The carcass characteristics of young adult female pigs (gilts) slaughtered after weaning their first litters have been investigated using three groups of 19 Landrace × (Landrace×Large White) gilts. In two treatment groups gilts were mated at their pubertal oestrus and suckled their piglets for 5 to 11 and 35 to 42 days respectively. These gilts were slaughtered on average 10 days after weaning. A third control group consisted of unmated gilts slaughtered at 118 kg live weight.

There was no significant difference in the killing-out percentage for the three groups. The carcasses of the gilts which farrowed contained significantly less fat (P<0·001) than those of the unmated controls. The carcasses of the farrowed gilts had significantly lighter middle sections (P<0·001) and significantly heavier shoulders (P<0·001) than the controls.

Despite the variations in tissue yield and distribution, the yield of prime joints for the gilts which had farrowed was only 0·62 percentage units less than that of the controls.

No commercially significant variations between groups in meat quality were observed.

The nutrient partitioning of growing pigs was altered through nutrition and castration in order to investigate the relationship between serum insulin-like growth factor 1 (IGFl) and protein deposition and serum cholesterol and fat deposition. In a 2 × 2 factorial experiment 18 entire and 18 castrated male pigs, of 20 kg initial live weight, were given either 2·25 (low) or 3·4 (high) times maintenance energy requirements to a scale based on live weight. Nitrogen and energy balances were measured over 7-day periods when the pigs reached about 30, 60 and 90 kg. Fasting blood samples were taken at each weight and serum was analysed for IGFl and insulin and total- and high density lipoprotein (HDL)-cholesterol. Protein and fat deposition, IGFl and cholesterol concentrations were higher for pigs given the high, as opposed to the low, feeding level. Protein deposition and IGFl concentrations were higher for entire males, as opposed to castrated males, whereas fat deposition and cholesterol concentrations were higher in castrated than in entire males. IGFl and protein deposition increased with age for entire but not for castrated males. Conversely, fat deposition increased with age in castrated but not in entire males. There was no effect of age on serum cholesterol. Serum IGFl was correlated with protein deposition at 30, 60 and 90 kg (r = 0·40, r = 0·63 and r = 0·67; P < 0·05, P < 0·002 and P < 0001 respectively, no. = 36). Serum total cholesterol and low density lipoprotein (LDL)-cholesterol were correlated with fat deposition at 60 and 90 kg (r = 0·65 and r = 0·54; both P < 0·001 for total cholesterol; r = 0·66 and r = 0·50; both P < 0·001 for LDL-cholesterol). Insulin levels were similar for pigs in all treatment groups. It is concluded that serum IGFl and cholesterol may give a useful indication of protein and fat deposition in pigs of between 60 and 90 kg live weight.

Three groups of lambs of about 25 kg live weight were given a concentrate diet either on its own (eight lambs) or supplemented with 10 g/kg ammonium chloride (eight lambs) or 20 g/kg sodium bicarbonate (12 lambs). At about 45 kg live weight the lambs were killed and their body composition was determined. The composition of their gains was also determined using information obtained from a fourth group of lambs (eight lambs) which were killed at the start of the experiment. Lambs given the ammonium chloride diet showed a reduction in blood pH while those given the sodium bicarbonate diet showed an increase to levels seen in lambs given forage diets. There were also marked effects on calcium (Ca), phosphorus (P) and magnesium retention with retentions being reduced in those given the ammonium chloride diet and increased in those given the bicarbonate diet. The average rates of retention of Ca and P in lambs given the bicarbonate diet were 9·1 and 5·9 g/kg empty-body weight gain and are comparable with rates seen in lambs given forage diets. These results suggest that dietary induced changes in blood acid-base status is a major factor contributing to the lower rates of retentions of these minerals in lambs given cereal-based diets.

Pregnancy-associated glycoproteins (PAGs) are a large grouping of placental proteins that belong to the aspartic peptidase gene family. Although useful to detect pregnancy in ruminant species, the function of these molecules is unclear. Several PAGs expressed by trophoblast binucleate cells can enter the maternal circulation, suggesting that they could have a systemic role in altering maternal physiology. The objective of this work was to examine whether these circulating placental antigens were important in pregnancy by actively immunizing ewes against them. PAGs were purified by pepstatin-affinity chromatography and conjugated to the immunogenic protein, keyhole limpet hemocyanin (KLH). Ewes were immunized with PAG–KLH conjugate (n = 22) or with KLH alone (n = 9), and bred to intact rams. Blood samples, collected on Day 0 (day of estrus), Day 10, Days 15 to 25 and weekly throughout pregnancy, were analyzed for PAG by an ELISA. On Day 30, pregnancy was confirmed by ultrasound. Ewes immunized against PAG–KLH produced a range of reactive anti-PAG titers, whereas all immunized ewes had high anti-KLH immunoreactivity. PAGs became detectable in the anti-KLH (control) ewes at Day 21.6 ± 2.2 of pregnancy. Those ewes immunized against PAGs (n = 7), that had very low immunoreactivity toward PAGs, had measurable PAG by Day 22.9 ± 1.3, and their PAG serum profiles throughout pregnancy did not differ from the controls. Those exhibiting moderate to high anti-PAG immunoreactivity (n = 15), had significantly lower PAG concentrations than controls, with antigen not becoming detectable until Day 48.1 ± 15.6. The decrease in circulating PAG in the immunized animals did not correlate with changes in pregnancy rates, lamb number or lamb birth weight. These results suggest that while PAGs may play a role in maintaining pregnancy, their major contribution is likely to be at the fetal–maternal interface. Their actions at extra-placental sites are presumably of more secondary importance.

The effects of inbreeding on the gross financial returns from ezues have been studied in a flock of sheep comprising three hill breeds and Ihe crosses among them. Each of these groups mis represented In non-inbred sheep (purebred and crossbred, designated O2, and F2, respectively) and four classes of inbreeding (inbreeding coefficients (F): 0·25, 0·375, 0·50 and 0·59). A second non-inbred class ums represented by crosses of inbred lines.

Gross lifetime income per eive derived from the sale of lamps for slaughter, cast ezves and wool was reduced by inbreeding in an almost linear fashion (between the O2/F2 and the F 0·50 class) at a rate of £1·27 (at 1992 prices) for ench 0·01 (percentage point) increase in the level of inbreeding — an overall reduction to 0·35 of the O2/F2 level. Line crosses gave a better return than the other non-inbred sheep. When expressed in terms of ewe metabolic body weight (M0·73) the decline with inbreeding was less —an overall reduction to 0·4 of the O2/F, income. Inbreeding of Ihe lamb caused a further decline in output independent of the inbreeding of the ezve.

In two experiments involving a total of 48 individually-fed pigs on experiment from 20 to 90 kg live weight, the addition per kg of diet of 250 mg Cu, provided either by copper sulphate or by cuprous chloride, similarly and significantly improved daily gain and feed: gain ratio. Neither of the supplements had any significant effect on carcass measurements but both markedly and similarly increased the copper content of the liver at slaughter weight.

The relationship among parasitological parameters, abomasal size and body size measurements was investigated in lambs following an experimental infection with Haemonchus contortus. In total, 100 lambs from five different genotypes (German Merino (GM), Texel × GM, Suffolk × GM, German Blackhead Mutton × GM and Ile de France × GM) were experimentally infected with 5000 infective third stage larvae of H. contortus at the time of weaning at 12 weeks of age. Four and six weeks after infection, individual faecal samples were collected for estimation of faecal egg counts (FECs). Furthermore, wither height, shoulder width, heart girth, loin girth and body length were taken at 18 weeks of life. Lambs were slaughtered and necropsied 7 weeks post-infection, and worm counts, abomasal volume and surface area were determined. Positive correlations were found between different body size parameters, body weight and abomasal sizes. FEC and worm counts were not significantly correlated either with body size parameters or with abomasal size. The mean worm burden was higher in GM than in crossbred lambs. There was no significant difference in abomasal size between GM and crossbred lambs. The results suggest that the variations between animals in worm burden following an experimental infection with H. contortus (worm resistance) are not influenced by body size parameters or abomasal sizes. Therefore, other factors, including genetic-based differences in resistance, must cause these findings between and within breeds.

1. An anatomical explanation of the observed differences between double-muscled and phenotypically normal cattle was sought by detailed dissection of the sides of six double-muscled Santa Gertrudis steers and six Brangus steers.

2. Relative to carcass weight the double-muscled steers had a significantly greater proportion of muscle and a significantly lower proportion of bone and fat than the Brangus steers. Relative to muscle plus bone weight the muscle proportion was greater and the bone proportion lower in the double-muscled steers.

3. The musculature of the double-muscled steers exhibited a gradient of hypertrophy: 12 muscles (234g/kg of total muscle weight) were grossly hypertrophied, 56 muscles (565g/kg) were hypertrophied to a lesser degree and 28 muscles (195g/kg) showed either a minor degree of hypertrophy or no hypertrophy.

4. The grossly hypertrophied muscles were generally the larger, superficial muscles of the proximal pelvic limb and shoulder area. The muscles showing least hypertrophy were mostly small muscles located in the deeper areas of the carcass, particularly around the spine and in the proximal hindlimb, and in the proximal and distal forelimb.

5. Abnormalities of shape in double-muscled cattle may be attributed largely to altered proportions of total muscle and total fat, and to changes in weight distribution within the musculature.

The present study examines the size distribution of ovine steroidogenic and non-steroidogenic luteal cells throughout pregnancy. Cells were isolated from corpora lutea collected from early (< 8 weeks), mid (9 to 14 weeks) or late (15 to 18 weeks) stages of pregnancy. Cells were stained for 3β-hydroxysteroid dehydrogenase (3β-HSD) activity, a marker for steroidogenic cells. Both 3β-HSD positive and β-HSD negative cells covered a wide spectrum of size ranging from 7 to 37 μm in diameter. There was a significant increase (P > 0·01) in mean diameter of non-steroidogenic luteal cells as pregnancy progressed. Mean diameter of 3β-HSD negative cells increased from 17·8 (s.e. 0·4) μm in the corpus luteum of early stage of pregnancy to 22·4 (s.e. 0·3) μm in the corpus luteum of advanced pregnancy. However, there was no significant increase in the mean diameter of 3β-HSD positive cells. Corpora lutea obtained from early stages of the pregnancy contained more steroidogenic cells than the cells obtained from mid and late pregnancy (P < 0·01). Percentage of 3β-HSD negative cells had increased 2·07-fold by 18 weeks of pregnancy when compared with the early stage of pregnancy. In contrast, percentage of 3β-HSD positive cells had decreased to 50% of starting values during the same period (P < 0·05). These results indicate that the ovine corpus luteum of pregnancy is morphologically dynamic over the course of pregnancy. Steroidogenic activity of luteal cells may decrease as pregnancy progresses, especially activity of the large luteal cells.

Lactobacilli have been shown to inhabit the proximal regions of the digestive tract of many species of animals, and some of these lactobacilli have the ability to adhere to and colonise the epithelium of the oesophagus (Tannock, Blummershine and Archibald, 1987) and stomach (Fuller and Turvey, 1971; Tannock et al., 1982), whilst others appear to be associated with the small intestine (Tannock et al., 1982). The mechanisms by which these bacteria colonise their habitats is still unclear.In this study the adhesion of lactobacilli, isolated from the small intestine of pigs, to porcine small intestinal cells was investigated.

For the Mpwapwa cattle breed in Tanzania, the efficiency of various breeding schemes including an open nucleus was investigated by model calculations. Artificial insemination and intensive recording of production are assumed to be applied in a nucleus which is the main breeding unit. As a pre-nucleus, associated herds with less intense data recording serve as the basis to screen superior cows for nucleus replacements, provide the capacity for progeny testing, and operate as bull multipliers for commercial herds.

The criteria of efficiency were genetic gain and profit from selection for a dual purpose breeding objective (milk and beef) in a total population of 10 000 cows. Introducing a two breeding tier scheme through separating all recorded cows into a nucleus and pre-nucleus leads to an increase of the genetic gain rather than the profit. Further improvement is obtained by introduction of artificial insemination in pre-nucleus herds. The nucleus size should not exceed about 5% of the cow population and an optimum size of the pre-nucleus is about 15%. Opening the nucleus to replacement cows coming from the pre-nucleus affects the aggregate genetic gain very little although it can be recommended if milk yield is to be mainly improved or if the total profit is taken into account. Furthermore, the nucleus should be opened if there is little difference between the heritabilities in the nucleus and pre-nucleus and also in order to avoid detrimental effects of inbreeding and genotype × environment interaction.

Thirty one-year-old Charolais × Holstein-Friesian (CH) and Hereford × Holstein-Friesian (HH) steers averaging about 268 kg live weight were used to study the effect of increasing slaughterweight from the traditional 454 kg (1000 lb) to 544 and 635 kg (1200 and 1400 lb) live weight. The steers were fed a growing ration of grain, silage and hay until they weighed 68 kg (150 lb) below the assigned slaughter weight, followed by a finishing ration of grain mixture and maize silage given ad libitum. Average daily gain on the growing ration was 0·91 kg in CH, 14% higher than that in HH steers, while on the finishing ration HH steers, finished at 544 and 635 kg, made higher gains. CH steers were 7·9% more efficient than HH steers in feed conversion. Feed conversion efficiency (total dig. nutrients (kg) per kg live-weight gain) decreased by 10% and 19% respectively as slaughter weight increased to 544 and 635 kg. Live measurements showed that CH steers were generally longer, taller and wider at the hip, whereas HH steers had a greater depth of chest and width of shoulder. CH steers, slaughtered at different weights, had a higher percentage of round and shank, while HH steers had a higher percentage of all other wholesale cuts. Carcass measurements were generally higher in CH than in HH steers. CH steers had a higher percentage of muscle and bone, and lower fat, in the 12th rib. They also had larger areas of m. longissimus thoracis. Fat deposition over and in that muscle was much higher in HH than in CH steers. Under the conditions of this study, 544 kg was found to be the optimum slaughter weight for both groups. Slaughtering steers at 635 kg was found to be economically disadvantageous.

Multi-sire mating of a mob of ewes is commonly used in commercial sheep production systems. However, ram mating success (defined as the number of lambs sired by an individual) can vary between rams in the mating group. If this trait was repeatable and heritable, selection of rams capable of siring larger numbers of lambs could reduce the number of rams required for mating and ultimately lead to increased genetic gain. However, genetic correlations with other productive traits, such as growth and female fertility, could influence the potential for ram mating success to be used as a selection trait. In order to investigate this trait, parentage records (including accuracy of sire assignment) from 15 commercial ram breeding flocks of various breeds were utilised to examine the repeatability and heritability of ram mating success in multi-sire mating groups. In addition, genetic and phenotypic correlations with growth and female fertility traits were estimated using ASReml. The final model used for the ram mating success traits included age of the ram and mating group as fixed effects. Older rams (3+years old) had 15% to 20% greater mating success than younger rams (1 or 2 years of age). Increasing the stringency of the criteria for inclusion of both an individual lamb, based on accuracy of sire assignment, or a whole mating group, based on how many lambs had an assigned sire, increased repeatability and heritability estimates of the ram mating success traits examined. With the most stringent criteria employed, where assignment of sire accuracy was >0.95 and the total number of lambs in the progeny group that failed to have a sire assigned was<0.05, repeatability and heritability for loge(number of lambs) was 0.40±0.09 and 0.26±0.12, respectively. For proportion of lambs sired, repeatability and heritability were both 0.30±0.09. The two ram mating traits (loge(nlamb) and proportion) were highly correlated, both phenotypically and genetically (0.88±0.01 and 0.94±0.06, respectively). Both phenotypic and genetic correlations between ram mating success and growth and other female fertility traits were low and non-significant. In conclusion, there is scope to select rams capable of producing high numbers of progeny and thus increase selection pressure on rams to increase genetic gain.

A deterministic, dynamic model was developed, to enable predictions of phosphorus (P) digested, retained and excreted for different pig genotypes and under different dietary conditions. Before confidence can be placed on the predictions of the model, its evaluation was required. A sensitivity analysis of model predictions to ±20% changes in the model parameters was undertaken using a basal UK industry standard diet and a pig genotype characterized by British Society Animal Science as being of ‘intermediate growth’. Model outputs were most sensitive to the values of the efficiency of digestible P utilization for growth and the non-phytate P absorption coefficient from the small intestine into the bloodstream; all other model parameters influenced model outputs by <10%, with the majority of the parameters influencing outputs by <5%. Independent data sets of published experiments were used to evaluate model performance based on graphical comparisons and statistical analysis. The literature studies were selected on the basis of the following criteria: they were within the BW range of 20 to 120 kg, pigs grew in a thermo-neutral environment; and they provided information on P intake, retention and excretion. In general, the model predicted satisfactorily the quantitative pig responses, in terms of P digested, retained and excreted, to variation in dietary inorganic P supply, Ca and phytase supplementation. The model performed well with ‘conventional’, European feed ingredients and poorly with ‘less conventional’ ones, such as dried distillers grains with solubles and canola meal. Explanations for these inconsistencies in the predictions are offered in the paper and they are expected to lead to further model development and improvement. The latter would include the characterization of the origin of phytate in pig diets.

The potential of using low glucosinolate full-fat rapeseed meal (FFR) in the diets of early-weaned piglets was investigated in this experiment. FFR (containing 17 (xmol total glucosinolates and 2·8 mg erucic acid per g of whole seed) was used on an isonitrogenous basis at either 200 g<kg or 400 g<kg to replace full-fat soya-bean meal. Four replicates of three contemporary pen groups of 10 piglets weaned between 3 and 4 weeks of age were randomly allocated at a mean initial weight of 8·41 (s.e. 1·105) kg to the three dietary treatments. The experimental diets were given for 3 weeks. Inclusion of FFR significantly depressed food intake, daily live-weight gain and food conversion. There were no significant differences between the performance of piglets given either 200 or 400 g FFR per kg. Intakes of the diets containing FFR per unit of metabolic body weight as a proportion of the control diet decreased from about 0·85 to 0·75 from the 1st to the last week of the feeding period. This suggested that the acceptability of diets containing FFR decreased with time, possibly due to the presence of unpalatable and perhaps antinutritive factors which may include glucosinolates although present at relatively low concentrations. It was concluded that the feeding value of FFR would need to be improved by development of oilseed rape varieties with much lower levels of glucosinolates.
