Strawberry is affected by several pests and diseases. Neopamera bilobata is an emerging pest that has been reported by several strawberry growers, usually associated with catfacing symptoms in fruits. We evaluated intercropping garlic or Chinese chives on N. bilobata populations on strawberry crops grown in high tunnels in two experiments. In the first experiment, we evaluated N. bilobata populations on strawberry intercropping with garlic plants (three densities: 8, 16, 24 GP – garlic plant per plot) on the bags by taking 12 samples from December 2015 to April 2017. N. bilobata populations on strawberry were also assessed when Chinese chives were grown under the suspended wooden structures in which strawberry plants are grown (‘undercropping’) (14 samples), in two high tunnels, from November 2016 to March 2017. The number of nymphs and adults on 14 randomly selected fruits per plot were assessed. During the garlic intercropping experiment, the treatments of three densities of garlic reduced N. bilobata populations; however, the 24 GP treatment caused a greater reduction than the 8 GP treatment. Garlic densities reduced N. bilobata populations by 35, 50, and 64% for the 8, 16, and 24 GP treatments, respectively. Chinese chives cultivated under the structures reduced N. bilobata populations by 47%. The results suggest that intercropping garlic or undercropping Chinese chives are suitable tools to be tested in integrated pest management in strawberry crops.

Heat-shock proteins 90 (hsp90s) are a class of molecules able to stabilize a network of ‘client’ proteins that are involved in several processes. Furthermore, recent studies indicated that mutations in the hsp90-encoding gene induce a wide range of phenotypic abnormalities, which have been interpreted as an increased sensitivity of different developmental pathways to hidden/cryptic mutations. In order to verify the role of hsp90 in aphids, we amplified and sequenced the hsp90 gene in 17 lineages of the peach potato aphid Myzus persicae (Sulzer, 1776) looking for the presence of mutations. In particular, we compared lineages with different reproductive modes (obligate vs. cyclical parthenogenesis), propensity to develop winged females and karyotype stability. Differently from the cyclical parthenogenetic lineages that possessed functional hsp90 genes, the seven analysed asexual lineages showed severe mutations (including frameshift and non-sense mutations). In vivo functional assays with the hsp90-inhibitor geldanamycin showed that some lineages with cyclical parthenogenesis may lose their ability to induce sexuales in the absence of active hsp90 revealing the presence of cryptic mutations in their genomes. As a whole, our data suggest that hsp90 could play in aphids a role in buffering hidden/cryptic mutations that disrupt cyclical parthenogenesis.

The development of the Mediterranean flour moth, Ephestia kuehniella (Zeller), was evaluated at 10, 15, 17.5, 20, 22.5, 25, 27.5, 30 and 32.5°C with no lighting. None successfully completed development at 10 and 32.5°C. The total development time from egg to adult emergence was 164, 140, 98, 76, 61, 62 and 50 days, respectively, at the remaining temperatures. The developmental rate of E. kuehniella was described by the common linear model and six non-linear models. The lower temperature threshold for the immature stages and the thermal constant for E. kuehniella were 9°C and 1111 degree-days (DD) to complete development from egg to newly emerged adult. Non-linear models estimated the lower and upper thermal thresholds (Tmin and Tmax) and optimal temperature (Topt). The values of Tmax calculated by three nonlinear models ranged from 34°C to 46°C; Topt for each stage of development varied from 24 and 31°C, consistent with the temperature (30°C) at which the most rapid development occurred. Information on the threshold temperatures for development and thermal requirements can be utilized to predict E. kuehniella population dynamics and phenology and to evaluate optimal temperature conditions for mass rearing in stored products.

A stable reference gene is a key prerequisite for accurate assessment of gene expression. At present, the real-time reverse transcriptase quantitative polymerase chain reaction has been widely used in the analysis of gene expression in a variety of organisms. Neoseiulus barkeri Hughes (Acari: Phytoseiidae) is a major predator of mites on many important economically crops. Until now, however, there are no reports evaluating the stability of reference genes in this species. In view of this, we used GeNorm, NormFinder, BestKeeper, and RefFinder software tools to evaluate the expression stability of 11 candidate reference genes in developmental stages and under various abiotic stresses. According to our results, β-ACT and Hsp40 were the top two stable reference genes in developmental stages. The Hsp60 and Hsp90 were the most stable reference genes in various acaricides stress. For alterations in temperature, Hsp40 and α-TUB were the most suitable reference genes. About UV stress, EF1α and α-TUB were the best choice, and for the different prey stress, β-ACT and α-TUB were best suited. In normal conditions, the β-ACT and α-TUB were the two of the highest stable reference genes to respond to all kinds of stresses. The current study provided a valuable foundation for the further analysis of gene expression in N. barkeri.

The white-backed planthopper, Sogatella furcifera (Horváth) (Hemiptera, Delphacidae), has emerged as a serious rice pest in Asia. In the present study, 12 microsatellite markers were employed to investigate the genetic structure, diversity and migration route of 43 populations sampled from seven Asian countries (Bangladesh, China, Korea, Laos, Nepal, Thailand, and Vietnam). According to the isolation by distance analysis, a significant positive correlation was observed between genetic and geographic distances by the Mantel test (r2 = 0.4585, P = 0.01), indicating the role of geographic isolation in the genetic structure of S. furcifera. A population assignment test using the first-generation migrants detection method (thresholds a = 0.01) revealed southern China and northern Vietnam as the main sources of S. furcifera in Korea. Nepal and Bangladesh might be additional potential sources via interconnection with Vietnam populations. This paper provides useful data for the migration route and origin of S. furcifera in Korea and will contribute to planthopper resistance management.

We examined previous reports of Lema praeusta (Fab.) (Coleoptera: Chrysomelidae) as a minor pest of turmeric, eggplant, bottle gourd and pumpkin leaves, but no feeding damage by larvae and adults of L. praeusta were recorded by us on these leaves. We observed feeding by the larvae and adults of L. praeusta on ten species of Commelinaceae plants in no-choice tests. The biology, fecundity and life table parameters of L. praeusta on two Commelinaceae weeds, Commelina benghalensis L. and Murdannia nudiflora (L.) Brenan were determined under laboratory conditions (27 ± 1°C, 65 ± 5% RH and 12L:12D). Total larval development times of L. praeusta were 6.36 ± 0.07 and 7.28 ± 0.11 days (mean ± SE) on C. benghalensis and M. nudiflora, respectively. Adult females lived 106.25 ± 1.17 and 77.65 ± 0.91 days (mean ± SE) on C. benghalensis and M. nudiflora, respectively. Each female laid 272.95 ± 2.39 and 224 ± 1.74 eggs (mean ± SE) during a lifetime on C. benghalensis and M. nudiflora, respectively. The net reproductive rate (Ro), intrinsic rate of increase (rm), generation time (Tc), doubling time (DT) and finite rate of increase (λ) were 136.48, 0.14, 36.17, 5.10 and 1.41 on C. benghalensis, respectively, whereas Ro, rm, Tc, DT and λ were 112, 0.20, 23.64, 3.47 and 1.51 on M. nudiflora, respectively, suggesting that L. praeusta could be a potential biocontrol agent against C. benghalensis and M. nudiflora in the fields of rice, maize, sorghum, soybean, mung bean, peanut and cotton.

The silver fly Leucopis glyphinivora Tanasijtshuk (Diptera: Chamaemyiidae) is an aphidophagous predator during its larval stage. Our study describes the life cycle of L. glyphinivora and evaluates its potential as a biocontrol agent. The development, survival and longevity of L. glyphinivora were observed and compared with those of the aphid gall midge Aphidoletes aphidimyza Rondani (Diptera: Cecidomyiidae). Tests were conducted under laboratory conditions in Petri dishes with the green peach aphid Myzus persicae Sulzer (Hemiptera: Aphididae) on potato leaves. Leucopis glyphinivora has a 42% longer life cycle than A. aphidimyza. Larval development was 27% longer in L. glyphinivora than in A. aphidimyza. Egg hatch rate and pupal emergence rate were similar in both species, but larval survival was 20% lower for L. glyphinivora than for A. aphidimyza. Sex ratio was approximately 50% in both species. A longer development time for L. glyphinivora in the larval stage could be an advantage for biocontrol by providing more time to exert predation pressure against aphids. However, this may be offset by the lower survival rate found in the silver fly larvae.

The cabbage aphid, Brevicoryne brassicae L. (Hem: Aphididae), is an important pest of canola that can considerably limit profitable crop production either through direct feeding or via transmission of plant pathogenic viruses. One of the most effective approaches of pest control is the use of biostimulants. In this study, the effects of humic acid, plant growth-promoting rhizobacteria (PGPR), and integrated application of both compounds were investigated on life table parameters of B. brassicae, and the tolerance of canola to this pest. B. brassicae reared on plants treated with these compounds had the lower longevity, fecundity, and reproductive period compared with control treatment. The intrinsic rate of natural increase (r) and finite rate of increase (λ) were lowest on PGPR treatment (0.181 ± 0.004 day−1 and 1.198 ± 0.004 day−1, respectively) and highest on control (0.202 ± 0.005 day−1 and 1.224 ± 0.006 day−1, respectively). The net reproductive rate (R0) under treatments of humic acid, PGPR and humic acid + PGPR was lower than control. There was no significant difference in generation time (T) of B. brassicae among the tested treatments. In the tolerance test, plants treated with PGPR alone or in integrated with humic acid had the highest tolerance against B. brassicae. The highest values of total phenol, flavonoids, and glucosinolates were observed in treatments of PGPR and humic acid + PGPR. Basing on the antibiosis and tolerance analyses in this study, we concluded that canola plants treated with PGPR are more resistant to B. brassicae. These findings could be useful for integrated pest management of B. brassicae in canola fields.

The phenological patterns exhibited by different organisms are known as adaptive responses to the cyclical environmental conditions. However, only a limited number of researches explore which factors are responsible for these phenological patterns in pest species. In the current study, abundance patterns were studied in the phenology of three Spodoptera Guenée, 1852 species, along the 29° latitudinal gradient in South America. The goal was to test whether widely distributed and abundant crop pest species would exhibit different phenological responses to seasonal meteorological variables and host plant availability. To test this, 13 light traps were set up in Brazil to collect adult Spodoptera samples at the time of the new moon, every month, from June 2015 to May 2016. The time of occurrence and intensity of the phenology were determined for each species, employing circular statistics. Both metrics revealed significant variations among the different species, as well as the factors associated with them. Latitude was found to affect the period of occurrence in Spodoptera cosmioides (Walker, 1858) and Spodoptera albula (Walker, 1857), whereas in Spodoptera frugiperda (J. E. Smith, 1797) its effect was evident only in the intensity of its phenology. Further, both meteorological variables and host plant availability in the sampling sites produced predictive models to account for the phenological patterns expressed. These findings suggest that different species of Spodoptera exhibit different adaptive strategies in their life cycles in response to environmental conditions, thus necessitating specific management practices regarding their seasonal population fluctuation.

The oriental fruit fly Bactrocera dorsalis (Hendel) is a destructive insect pest of a wide range of fruit crops. Commensal bacteria play a very important part in the development, reproduction, and fitness of their host fruit fly. Uncovering the function of gut bacteria has become a worldwide quest. Using antibiotics to remove gut bacteria is a common method to investigate gut bacteria function. In the present study, three types of antibiotics (tetracycline, ampicillin, and streptomycin), each with four different concentrations, were used to test their effect on the gut bacteria diversity of laboratory-reared B. dorsalis. Combined antibiotics can change bacteria diversity, including cultivable and uncultivable bacteria, for both male and female adult flies. Secondary bacteria became the dominant population in female and male adult flies with the decrease in normally predominant bacteria. However, in larvae, only the predominant bacteria decreased, the bacteria diversity did not change a lot, likely because of the short acting time of the antibiotics. The bacteria diversity did not differ among fruit fly treatments with antibiotics of different concentrations. This study showed the dynamic changes of gut bacterial diversity in antibiotics-treated flies, and provides a foundation for research on the function of gut bacteria of the oriental fruit fly.

Populations of the recently described black oak gall wasp, Zapatella davisae Buffington (Hymenoptera: Cynipidae), have been identified as the cause of extensive tree damage and mortality to black oaks, Quercus velutina Lamarck (Fagales: Fagaceae), in the northeastern United States. Relatively little is known, however, about the distribution, phylogenetic placement, and lifecycle of this important tree pest. Therefore, we conducted next-generation sequencing using the Ion Torrent™ PGM (ThermoFisher Scientific, Inc.) platform to develop genomic resources for the study of Z. davisae and for other closely related species of oak gall wasps. Individual sequence reads were aligned, assembled into unique contigs, and the contigs were then utilized for the in silico isolation and development of microsatellite markers. In total, we screened 36 candidate microsatellite loci, of which 23 amplified consistently (five polymorphic and 18 monomorphic). We then examined whether the polymorphic loci could be used to infer whether populations of Z. davisae from Cape Cod and Nantucket are sexual or asexual by calculating several metrics of genetic diversity that might indicate the mode of reproduction. These included testing for statistical deviations from Hardy–Weinberg equilibrium (HWE) and for linkage disequilibrium (LD), observations for the presence of the Meselson effect, and by calculating the probability that clonal individuals are more prevalent than would be expected in a randomly mating population. While we found significant deviations from HWE and more clonal individuals than expected, our estimates of the Meselson effect were inconclusive due to limited sampling, and we found no evidence of LD. Therefore, the sexual/asexual status of Z. davisae populations remains uncertain.

The sugar beet weevil (SBW), Bothynoderes punctiventris Germar, 1824, is a significant pest in most of Eastern Europe. Here, the SBW is described and its seasonal activity characterized, in terms of its different developmental stages in relation to Julian days (JDs), degree-day accumulations (DDAs), and precipitation, as a key to improving monitoring and forecasting of the pest. The phenology and population characteristics of SBW were investigated in sugar beet fields in eastern Croatia over a 4-year period (2012–2015). By using the degree-day model (lower development threshold of 5°C, no upper development threshold, biofix 1 January), the first emergence of overwintering adults was determined as becoming established when the DDA reached 20. The adult emergence was completed when the DDA reached 428. SBW males emerged first, following which the females dominated the adult population. Overwintering adults were present in the field until early July. In August, adults of the offspring generation began to appear. The eggs laid by the overwintering generation required, on average, 10–15 days to develop into larvae; however, eggs were found in soil samples over a period of 102 days (between JDs 112 and 214). Larvae were present in the soil samples over a period of a maximum of 143 days (the first larvae were established on JD 122 and the last one on JD 265), and pupae were established in the soil over a period of 102 days (between JDs 143 and 245). This study provides important data for understanding SBW population dynamics and developing potential population dynamic models for pest forecasting on a regional scale.

Variation in parasitism risk among hosts can arise from between-patch and within-patch factors, but considerably less information is known about the latter. This study investigated how distributions of the oriental fruit fly Bactrocera dorsalis influenced its parasitism by the pupal parasitoid Dirhinus giffardii in the laboratory. Because B. dorsalis larvae pupate underground, pupation depth was considered as an important factor that affects the risk of parasitism. When the density of B. dorsalis larvae was varied (1, 10, and 100 larvae per arena), average pupation depth increased with the density. When the depth of pupae was manipulated, the rate of parasitism differed by depths. Parasitism at 0 cm differed from the random parasitoid model expectation, but parasitism at 1 cm was not different from the model expectation. Few pupae at 2 cm were parasitized. In another experiment, when pupae were simultaneously presented at 0 cm and 1 cm depths, parasitism at 1 cm was weakened by the presence of puape at 0 cm. These results imply that the density of the host influences pupation depth as well as the distribution of parasitism and plays an important role in host-parasitoid dynamics.

Latrophilin (LPH) is known as an adhesion G-protein-coupled receptor which involved in multiple physiological processes in organisms. Previous studies showed that lph not only involved the susceptibility to anticholinesterase insecticides but also affected fecundity in Tribolium castaneum. However, its regulatory mechanisms in these biological processes are still not clear. Here, we identified two potential downstream carboxylesterase (cce) genes of Tclph, esterase4 and esterase6, and further characterized their interactions with Tclph. After treatment of T. castaneum larvae with carbofuran or dichlorvos insecticides, the transcript levels of Tcest4 and Tcest6 were significantly induced from 12 to 72 h. RNAi against Tcest4 or Tcest6 led to the higher mortality compared with the controls after the insecticides treatment, suggesting that these two genes play a vital role in detoxification of insecticides in T. castaneum. Furthermore, with insecticides exposure to Tclph knockdown beetles, the expression of Tcest4 was upregulated but Tcest6 was downregulated, indicating that beetles existed a compensatory response against the insecticides. Additionally, RNAi of Tcest6 resulted in 43% reductions in female egg laying and completely inhibited egg hatching, which showed the similar phenotype as that of Tclph knockdown. These results indicated that Tclph affected fecundity by positively regulating Tcest6 expression. Our findings will provide a new insight into the molecular mechanisms of Tclph involved in physiological functions in T. castaneum.

The coffee berry borer, Hypothenemus hampei (Ferrari), can survive in residual coffee berries during the inter-harvest period, while new fructification only appears 2–3 months after the last harvest. The dispersal of colonizing females is an adaptation that enables the life cycle of the species to go ahead whenever his flight aptitude allows. This paper focuses on accurately determining the rate of inseminated females ready to reproduce when emerging from residuals berries to colonize new ones, which constitutes a characteristic of the live cycle far from common in Curculionidae. We dissected females caught in traps baited with a mixture of alcohols during the inter-harvest season, females from infested residual berries collected from branches, and virgin females obtained from pupae reared individually in the laboratory. After microscopic preparation with Giemsa stain, spermathecae were observed to identify the physiological status of each specimen. Out of the females found in the traps, 98.4% displayed recent and abundant insemination and 1.6% sporadic insemination. In contrast, in residual berries, most of females were recently inseminated (84.5%), followed by virgin females (10.5%) and older inseminated females (5%). In addition, the flight tests of the virgin females were negative. These results indicate that all colonizing females were inseminated, ready for flying and oviposition, females inside residual berries showed different physiological status, and virgin females could not migrate since they could not flight. The large number of inseminated females inside the residual berries, and the capacity of migrating females to colonize and reproduce, suggest that it is necessary to control residual berries and use traps to stop the dispersal and reproduction of this pest.

The melon fruit fly, Bactrocera cucurbitae (Coquillett), is a serious agricultural pest which has defied the various control measures employed against it. Protease inhibitors present in plants which have the potential to inhibit the growth and development of associated insect pests can be a possible alternative which can be manipulated for developing resistance in plants to the pest. In the present study, winged bean (Psophocarpus tetragonolobus) protease inhibitor isolated through affinity chromatography was explored for its potential to disrupt the development of melon fruit fly, B. cucurbitae. Different concentrations (12.5, 25, 50, 100, 200, and 400 µg ml−1) of the winged bean protease inhibitor (WBPI) were incorporated into the artificial diet of the second instar (64–72 h old) larvae of B. cucurbitae. The WBPI significantly delayed the larval, pupal, and total development period. The percentage pupation and adult emergence of the treated larvae was reduced as compared with control. The activities of major digestive enzymes (trypsin, chymotrypsin, leucine aminopeptidase, and elastase) decreased significantly in the larvae treated with different concentrations (50, 100, 200, and 400 µg ml−1) of WBPI. The findings reveal that the inhibitor holds considerable promise for the management of the melon fruit fly.
