In two separate experiments, the effects of pulsationless milking and milking vacuum on the rate of keratin removal from the teat canal were determined. Sixteen cows were milked with or without pulsation for either a single milking or for eight milkings. Milking without pulsation removed 10% of keratin present before milking, significantly less than milking with pulsation, which removed 32%. After eight milkings (4 d) without pulsation, up to 20% more keratin had accumulated within the teat canal but the rate of keratin regeneration reduced significantly upon return to pulsation milking. In a second experiment, ten cows were milked at 45 or 55 kPa and with or without pulsation. Only the absence of pulsation significantly reduced keratin loss during milking. Keratin loss during milking appears to be controlled by liner compression rather than by the rate of milk flowing through the teat canal. Pulsationless milking may increase penetrability of the teat canal to bacteria by reducing the natural rate of keratin removal during milking and reducing the rate of keratin regeneration.

Mammary tissue (MT) turnover is characterized by programed cell death and remodeling which might be affected by both feeding level and animal species. Thus, twenty-four dairy goats and the same number of sheep were assigned to three homogenous sub-groups per animal species and fed the same diet in quantities which met 70% (FL70), 100% (FL100) and 130% (FL130) of their daily energy and crude protein requirements. Individual MT samples were taken by biopsy from the animals on the 30th and 60th experimental day. The results showed, in the first sampling time, a significant reduction in the mRNA abundance for selected genes involved in programed cell death in both FL 70 fed goats (STAT3 and BECN1) and sheep (CASPASE8 and BECN1) compared with the respective FL100 groups. The FL130, in comparison with the FL100, caused a significant increase in transcripts accumulation of STAT3 gene in both sampling times and CASPASE8 gene in the second sampling time in goat MT, while the opposite happened for the mRNA expression of CASPASE8 and BECN1 genes in sheep MT, but only in the first sampling time. Moreover, a significant up regulation in the mRNA levels of MMP2 gene in MT of FL130 fed sheep was observed. The FL130, in comparison with the FL70, caused an enhancement in the mRNA expression levels of BECN1, CASPASE8, BAX and STAT3 genes in goat MT only. It was also shown that apoptosis and autophagy can be affected simultaneously by the feeding level. Overfeeding affects MT programed cell death and remodeling by a completely different way in goats than sheep. In conclusion, feeding level and animal species have strong effects on both MT programed cell death (apoptosis and autophagy) and remodeling but the molecular mechanisms need further investigation.

Paper-based dietary assessment tools such as food frequency questionnaires (FFQ) and especially dietary screeners are making way for versions that use technology. Amidst low intakes of dairy and dairy-related nutrients in South Africa, and to increase public awareness thereof, we aimed to develop and evaluate the usability of an application (app) to screen for dairy intake in higher income South African adults. In a consultative process, a dairy intake screener (‘Dairy Diary’) was developed as an eight-item quantitative FFQ with four types of commonly consumed local dairy products: milk, maas (fermented milk), yoghurt, and cheese. For each dairy product, usual frequency of consumption and portion size per eating occasion were scored resulting in three risk classes: <1 serving daily; 1≤2 servings daily; ≥2 servings daily. Digitalisation included product- and portion-specific graphics with linkage to risk class-relevant preliminary dairy-related guidance as part of a web-based mobile app. For the evaluation of the usability, the 26-item end-user version of the Mobile Application Rating Scale (uMARS) was used in an online cross-sectional survey (Qualtrics; April 2020). Items were scored on a 5-point Likert-type scale, resulting in three final app scores. From a conveniently recruited sample of 1102, 703 (64%; 81% female; mean age 29.8 ± 11.0 years) were retained for analysis. uMARS-informed descriptive statistics summarise the findings. The uMARS app mean objective quality score (3.9 ± 0.85), app subjective quality score (3.5 ± 0.77), app-specific score (3.6 ± 0.94), and additional question on e-portion (4.3 ± 0.78) met the minimum acceptability score of ≥3.0. For the subscales, the mean score for aesthetics was the highest (4.4 ± 0.82), followed by information (4.3 ± 0.90) and functionality (4.0 ± 1.33). Engagement scored lowest (3.0 ± 1.55). The ‘Dairy Diary’ is a user-friendly screener for dairy intake.

We investigated four different production methods of Künefe cheese, emphasizing their industrial importance. The four methods used fresh Künefe cheese (FKC), salted Künefe cheese (SKC), Boru type Künefe cheese (BKC) and culture-added processed Künefe cheese (CPKC), all used in the production of Künefe, a syrupy dessert unique to the Middle East. FKC was produced as a result of curd formation from raw milk with rennet and then the fermentation of curd. SKC was manufactured using the salting method in addition to FKC production. BKC was obtained using the dry cooking process with emulsifying salts applied to cheese curd. CPKC was produced by applying heat treatment to raw milk and using starter culture before adding rennet that was different to Boru-type Künefe cheese. The composition, color, meltability, texture and sensory properties of the Künefe cheeses were examined. As a result of statistical analysis, it was determined that the effect of different production methods on the composition, meltability, texture (except springiness and cohesiveness) and sensory properties was significant in all cheeses (P < 0.05). CKPC was the most appropriate cheese in various properties.

Yield gaps in milk production are here defined as the differentials between the actual yield obtained by the dairy farmer and the potential farm yield (production achieved by the top 10% of farmers: Gap 2) as well as the differential between this potential farm yield and the yield registered in the research stations (Gap 1). Assessment of yield gaps provides valuable information on potential production enhancement and drivers behind yield gaps. Milk production can be increased by narrowing the predominant large yield gaps in resource-poor smallholder farming system. Hence, this study assessed the milk yield gap and factors affecting the yield gap in Ri-Bhoi district of Meghalaya, a state located in the north-eastern Himalayan region of India. This research paper provides a scope for exploring the possibilities for improving dairy production in the state as well as contributing to literature through incorporating crucial determinants responsible for milk yield gap. A sample of 81 respondents was drawn purposely from two blocks of the district. The results indicated that the average number of cattle per household was 9.38 in standard animal units. The total yield gap was estimated at 6.20 l (91.06%) per day, composed of 0.80 l (11.76%) per day of yield gap I and 5.40 l (79.30%) per day of yield gap II. This demonstrates that the top performing farms were achieving a production level not dissimilar to that obtained on the research stations, but many were doing far less well. The size of cattle shed, dairy farming experience, concentrate price and human labour were the important determinants of the yield gap. Hence, encouraging the right stocking density of cattle, training on the preparations of home-made concentrates, access to cheap and quality concentrates, incorporating training and experience sharing on proper dairy management practices and use of technology could benefit the dairy farmers of the region.

Milk with 1·5% fat was treated to increase its viscosity to that of whole milk on the assumption that the sensation produced in the mouth, its mouthfeel, would be improved. Selected treated 1·5% fat milks were assessed for similarity to whole milk in taste panel trials. Panelists were chosen because of their ability to distinguish between whole milk and that containing 1·5 % fat by taste in at least one of two tests. Only 34% of the persons screened were suitable. Throughout the trials, the proportion of panelists able to distinguish between the two milks served alone and in tea remained at about 50%, but rose to 80–90% for the milks in coffee. In general, addition of KOH to 1·5% fat milk to pH 7·4 and of 6 mM-citrate overcame about 75 and 50%, respectively, of the difference in mouthfeel between whole and 1·5% fat milk. Whole and 1·5% fat milks were easily distinguishable visually, but the appearance of the latter was improved by adjustment to pH 7·4.

In dry cow therapy (DCT), antibiotics are usually injected direct into the mammary gland after the last milking of the lactation. Most products are designed to eliminate existing intramammary infections (IMI) caused by Staphylococcus aureus and Streptococcus agalactiae and to prevent the establishment of new IMI caused by major pathogens during the early non-lactating period (Nickerson et al. 1999). Systemic therapy has been attempted as a way of improving the cure rates of intramammary treatments of clinical and subclinical mastitis during lactation (Ziv, 1980a; Calvinho et al. 1988; Owens et al. 1988). For systemic therapy to be useful, effective passage of the drug from blood to the foci of infection must be achieved. Maintaining an effective antibacterial concentration in the udder depends on the physicochemical properties of the drug, the dose, the bioavailability of the injected formulation, and the sensitivity of the pathogen (Ziv et al. 1980a, b). The theoretical pharmacokinetic and pharmacodynamic basis for systemic DCT has been reported (Soback, 1988). When used in an attempt to improve cure rates of IMI, subcutaneous injection of tilmicosin at drying-off was ineffective against Staph. aureus IMI (Nickerson et al. 1999) and intramuscular (i.m.) oxytetracycline, in combination with intramammary cephapirin dry-cow treatment did not improve the cure rate for Staph. aureus mastitis (Soback et al. 1990a; Erskine et al. 1994). Nevertheless, i.m. tylosin treatment 2 weeks before the expected day of calving decreased IMI after calving (Zecconi et al. 1999) and subcutaneous norfloxacin nicotinate applied at the cessation of milking proved useful in controlling Staph. aureus infections (Soback et al. 1990a). Moreover, the incidence of this pathogen after systemic DCT may be significantly lower compared with untreated control groups (Soback et al. 1990a, b).

Streptococcus uberis do not colonise the teat canal and appear to invade the mammary gland of the dairy cow by direct entry though the canal. When they enter the mammary gland, and the early resulting processes, are unclear. Experimental infusions of the lactating mammary gland have been made to determine outcomes of infection, mastitis and disease. Infusion of 500 cfu bacteria was made immediately after milking (8 and 16 h intermilking interval) and 1, 4 or 12 h prior to milking. A mastitis resulted from all infusions, probably in response to the skim milk carrier. Infusions post milking resulted in clinical mastitis in more than half of the quarters, whereas infusion 1 h premilking created no clinical mastitis. Infusion 4 or 12 h pre milking resulted in the most severe reactions, with all quarters developing moderate to severe clinical mastitis. This was more rapid with the 4 h pre milking group. The results demonstrate that the initial inflammatory response caused by an invasion of the mammary gland is not necessarily protective against establishment of a pathogen, and that especially the response to invasion in the intermilking interval is often insufficient to prevent infection and/or disease.

A chemiluminescent method for determining xanthine oxidase (XOD) activity was developed and applied to the assay of milk enzyme activity using a photomultiplier luminometer. Various kinds of milk and cream samples were analysed for XOD content. In pasteurized milk, XOD activity depended on the fat content and in UHT milk it disappeared owing to the heat treatment. Milk sample preparation was very simple, requiring only homogenization at 40°C followed by a 1[ratio ]10 dilution with UHT (‘XOD-free’) milk. The assay was carried out at 25°C. The response obtained from XOD standard solutions in milk was linear from 0·1 to 500 enzyme units (U) l−1, but for the actual milk samples values ranged only from 1 to 135 U l−1. The detection limit at 2 SD was 0·1 U l−1 in milk, while in buffer it was 100 times lower. The intra-assay and interassay CV for XOD activity in milk were 6–12%.

We investigated whether prepartum levels of serum bone biomarkers are related to the degree of parturient hypocalcaemia and risk of milk fever (MF) in dairy cows with advancing parity. A total of 58 late-pregnant cattle were assigned to four groups: nulliparous, primiparous, multiparous in the 2nd lactation and multiparous in the 3rd–5th lactation. The multiparous cows were further assigned to MF and non-MF groups according to the onset of MF. Serum samples were obtained from the cows during the 3 weeks prepartum to 5 d postpartum period for the measurement of serum calcium (Ca) and three bone biomarkers: tartrate-resistant acid phosphatase isoform 5b (TRAP5b), osteoprotegerin (OPG) and bone isoenzyme of alkaline phosphatase (ALP3). The ratios of OPG to TRAP5b (O/T ratio) and ALP3 to TRAP5b (A/T ratio) were calculated. The data from all cattle showed that the severity of hypocalcaemia at parturition increased with advancing parity/age. The MF cows had elevated serum TRAP5b activity and a decreased O/T ratio after parturition, suggesting an increased number of osteoclasts due to osteoclastogenesis, in response to severe hypocalcaemia. The MF cows showed lower serum ALP3 activity during the 3 weeks prepartum than the non-MF cows, therefore, prepartum osteoblast function was likely weak in the MF cows. During the 2–3 weeks prepartum, serum ALP3 activity and the A/T ratio had moderate associations with the serum Ca concentration at day 0 (day of calving) in the multiparous cows, and receiver operating characteristic curve analysis revealed that ALP3 activity had excellent ability to predict MF. In conclusion, prepartum serum ALP3 activity is a promising biomarker to predict MF in multiparous cows.

This study tested the hypotheses that machine milked dairy sheep have a high prevalence of teat-end hyperkeratosis (TEH), which contributes to udder health problems. A random sample of 1360 milking ewes from 28 dairy sheep farms was monitored. Milking procedures, milking parlour characteristics and maintenance were recorded during a designated on farm audit; records were obtained through observations and interviews with farmers. Number of ewes/milker, ewes/milking unit and milkings/milking unit were calculated. Vacuum level, pulsation rate and ratio were measured. Four combinations of vacuum level and pulsation rate were defined; <40 kPa and <150 cycles/min (VP1), <40 kPa and ≥150 cycles/min (VP2), ≥40 kPa and <150 cycles/min (VP3), ≥40 kPa and ≥150 cycles/min (VP4). California Mastitis Test (CMT; scores 0–4) was done on all ewes. Then the udder of each ewe was assessed for TEH (scores 1–4) and ewes were assigned into three groups according to TEH severity (no or mild, medium and severe TEH). Severe TEH (scores 3 and 4) prevalence at teat-level was ca. 13%. TEH severity was associated with the combination of vacuum level and pulsation rate; ewes milked with VP4 combination were more likely to have a one-level increase on TEH severity compared to ewes milked with VP2 and VP3 combinations. More ewes/milker and less ewes/milking unit increased the likelihood of a one-level increase on TEH severity. Finally, ewes with severe TEH were more likely to have a one-level increase on CMT score. Therefore, our hypotheses that TEH is prevalent in dairy ewes and contributes to udder health problems were confirmed. Additionally, farmers can reduce TEH prevalence by optimising the way they milk and their milking parlour.

Bovine κ-casein glycomacropeptide (GMP) is a sialic acid containing glycopeptide, which is considered as a health promoting compound found in cheese whey. The study described in this research communication was undertaken to determine whether GMP with undetectable level of contaminating protein or phenylalanine can be isolated from bovine whey fraction using batch anion exchange technique with chitin as an adsorbent. A soluble whey fraction (SWF) prepared from 1 g whey protein isolate (WPI) was mixed with a slurry of 1 g chitin, and the mixture was incubated at pH 3.0. After incubation, the mixture was filtered, and the residue obtained (containing chitin-GMP complex) was washed with water and eluted stepwise with 0.5 M NaCl and 2.0 M NaCl. Most of GMP (corresponding to 75.8% of total sialic acid recovered) was eluted with 0.5 M NaCl. The recovered GMP accounted for 5.4% dry weight of WPI (or 18.9% dry weight of SWF). Amino acid analysis showed that there was no detectable level of contaminating amino acids including phenylalanine, histidine, arginine and tyrosine in the GMP fraction. It was concluded that the batch anion exchange method with chitin developed in this study can be used for the isolation of high purity GMP from bovine SWF.

The aim of this study was to evaluate the effect of the sprinkler cycle and flow rate on physiological, behavioural, and productive responses in dairy buffaloes. Nine Nili Ravi lactating buffaloes were subjected to three sprinkler cycles and two flow rates using a double replicated 3 × 3 Latin square design. The flow rates were 1.25 and 2 l/min, and the sprinkler cycles (minutes water on/off, number of cycles/h) were: 3/3, 10 cycles; 3/6, 7 cycles and 3/9, 5 cycles. The showering was applied from 0800 till 1630 h daily. In the first square of 21 d, each of the three sprinkler cycles was applied using a 1.25 l/min flow rate for 7 d per cycle. In the later square, the same treatments (sprinkler cycles) were applied using the 2 l/min flow rate. The average temperature humidity index during the study period was 85.7 ± 3.8 (Mean ± sd). The result showed that the 3/3 treatment group had lower body temperature and respiration rate than the other groups. The buffaloes in the 3/3 group produced 0.5 and 0.7 kg more milk with 1.4 and 2.4% more fat than the 3/6 and the 3/9 treatment groups, respectively. Similarly, the 2 l/min flow rate had a lower core body temperate and respiration rate and higher milk yield than the 1.25 l/min group. The 3/3 showering cycle with a 2 l/min flow rate appeared effective in improving physiological responses and milk yield in dairy buffaloes.

Though dairying is as old as New Zealand itself, and as far back as 1855 some 785 cwt. of butter and 406 cwt. of cheese were exported, it was not until about the last decade of the nineteenth century that it assumed the importance of an industry. Prior to that time butter and cheese were made on farms and sold locally, neither creameries nor factories had been established, and the quality of that produced was as varied as the skill and care of the farm womenfolk, who, for the most part, were responsible for the manufacture of the butter or the cheese. At that time dairying was an art and not a science. While clean hygienic conditions were applauded, they were not considered essential. Neither the local nor the export trade bad reached such dimensions that grading or standardisation was necessary. Moreover, these factors were not even considered in an age when each butter or cheese maker endeavoured, by skilful practice, to build up a personal reputation which commanded a premium when the produce was disposed of.

The milking characteristics of conventional clusters were compared with individual teatcups or clusters fitted with valves in the short milk tubes without external air admission to the liners. Individual teatcups fitted with non-return valves had peak milk flow rates 13% higher than conventional teatcups. A daily cross-over experiment involving 36 cows compared clusters fitted with non-return valves to conventional clusters. Valved clusters milked 18% faster, showed significantly better teatcup stability and 3% higher machine milk yields, but 48% higher strip yields were recorded. Clusters with non-return valves were difficult to remove after milking because the valve closed when the vacuum supply was shut off, thereby maintaining the vacuum level under the teats. When the nominal plant vacuum level for teatcups with non-return valves was reduced by 10 kPa to compensate for the higher effective milking vacuum level, the valved teatcups had peak flow rates 20% lower than the conventional teatcups. Clusters fitted with a modified valve, which allowed some reverse flow when closed (a ‘leaky’ valve), milked significantly faster than conventional clusters, had 33% lower strip yields and were easy to remove after milking.

Bacteriophage active against a lactic streptococcus was first isolated in this laboratory in February 1935. In May of the same year some batches of phage for Streptococcus cremoris strain HP were prepared by lysing the bacteria in skim-milk culture and preparing a Seitz-filtered whey as described by Whitehead & Cox(1). At that time it was not the practice to adjust the reaction of phage filtrates and the pH of the batches prepared in 1935 was not recorded. It was probably between 5·2 and 6·1 (Hunter & Whitehead (2)). Samples of four different preparations of the phage were filled into sterile tubes and sealed off in a blow-pipe flame. Two of the samples, sealed on 24 May 1935, had titres of 107 (as determined by spotting of standard loops of dilutions on a bacterial mat) and the other two, sealed on 16 July 1935 had titres of 106. The sealed tubes were stored in the refrigerator at 4–7° C.

The performance of khoa, a local heat-concentrated milk product, as a milk solids source in ice cream was tested by replacing 250, 500, 750 and 1000 g/kg of the condensed whole milk in control ice cream. Proximate composition was little affected by the inclusion of khoa at any level tested. At levels of inclusion > 250 g/kg the viscosity of both fresh and aged experimental mixes was significantly (P < 0·05) increased compared with controls. Some mixes containing khoa appeared slightly off-white, but there were no overall differences from control. Control and experimental mixes were identical in whipping capacity and overrun. In organoleptic evaluation, the colour score of the ice creams declined significantly (P < 0·05) when 750 or 1000 g/kg of the condensed milk was replaced with khoa. Ice creams containing khoa were equivalent or superior to controls in flavour. In objective tests, the experimental ice creams showed significantly (P < 0·05) faster meltdown compared with control, but no differences were found in subjective assessments. We conclude that khoa can be used successfully as a milk solids source for ice cream without any serious adverse effects on quality, the only problem being the dull colour.

While using an acid degree value test during work on lipolysis in milk, difficulty was encountered in extracting the fat from the milk of certain cows, especially milk of high butterfat content and milk from newly calved cows. This difficulty was overcome by increasing the quantity of reagent used. A study was made of the quantities of reagent required for milks of different butterfat content and for colostrum. No effect on the acid degree value occurred as a result of increasing the quantity of reagent. A modification of the test is recommended, using an increased quantity of reagent.

As a member of Committees of the Empire Marketing Board and subsequently of the British Standards Institution, the author has had experience in methods of milk testing which has led to the discovery of a simplified test for butterfat, which may be of value in filling some of the gaps in the application of the present known methods.

1. The addition of 1 µg. menadione/ml. to milk had no effect on the souring of the milk at 37° C. With higher concentrations of menadione the effect on the keeping quality at 37° C. varied with different milks. In some milks the clot-on-boiling stage was delayed by 1 hr. with 10 µg. menadione and by 2 to over 5 hr. with different milks containing 100 µg. menadione per ml. Some milks, however, showed no alteration in the keeping quality at 37° C. with as much as 1000 µg. menadione/ml.

2. The response of individual milks to the presence of menadione appeared to be related largely to the bacterial flora of the milk, The relative insensitivity of Gram negative rods, including coliform bacteria and of enterococci, to menadione was confirmed. Milks containing large numbers of coliform bacteria showed the least and those free of such bacteria the greatest response to menadione.

3. Menadione fed to milking cows at the rate of 100 mg·/day for a period of 3 weeks had no effect on the keeping quality of the milk when it was held at 37, 22 or 15·5° C. The time of incubation required for the milk to attain a titratable acidity equivalent to 2 ml. N/9-NaOH, to become unstable to 68 % alcohol or to boiling showed no difference between the milk from the cows given menadione and that from the control cows.

4. The menadione had no demonstrable effect on the appetite or the well-being of the cows nor on their milk yield.

5. When as much as 100 μg. menadione was given per cow per day, no menadione was detected in the milk under conditions suitable for the detection of 1 μg. menadione/ml.milk.
