The imprinted insulin-like growth factor 2 gene (IGF2) encodes a fetal mitogenic hormone protein (IGF-II) and has previously been shown to be associated with performance in dairy cattle. In this study we assessed genotype-phenotype associations between four single nucleotide polymorphisms (SNPs) located within the bovine IGF2 locus on chromosome 29 and a range of performance traits related to milk production, animal growth and body size, fertility and progeny survival in 848 progeny-tested Irish Holstein-Friesian sires. Two of the four SNPs (rs42196909 and IGF2.g-3815A>G), which were in strong linkage disequilibrium (r2=0·995), were associated with milk yield (P⩽0·01) and milk protein yield (P⩽0·05); the rs42196901 SNP was also associated (P⩽0·05) with milk fat yield. Associations (P⩽0·05) with milk fat percentage and milk protein percentage were observed at the rs42196901 and IGF2.g-3815A>G SNPs, respectively. The rs42196909 and IGF2.g-3815A>G SNPs were also associated with progeny carcass conformation (P⩽0·05), while an association (P⩽0·01) with progeny carcass weight was observed at the rs42194733 SNP locus. None of the four SNPs were associated with body size, fertility and progeny survival. These findings support previous work which suggests that the IGF2 locus is an important biological regulator of milk production in dairy cattle and add to an accumulating body of research showing that imprinted genes influence many complex performance traits in cattle.

The presence of yeasts in milk may cause physical and chemical changes limiting the durability and compromising the quality of the product. Moreover, milk and dairy products contaminated by yeasts may be a potential means of transmission of these microorganisms to man and animals causing several kinds of infections. This study aimed to determine whether different species of yeasts isolated from bovine raw milk had the ability to develop at 37°C and/or under refrigeration temperature. Proteinase and phospholipase activities resulting from these yeasts were also monitored at different temperatures. Five genera of yeasts (Aureobasidium sp., Candida spp., Geotrichum spp., Trichosporon spp. and Rhodotorula spp.) isolated from bovine raw milk samples were evaluated. All strains showed one or a combination of characteristics: growth at 37°C (99·09% of the strains), psychrotrophic behaviour (50·9%), proteinase production (16·81% of the strains at 37°C and 4·09% under refrigeration) and phospholipase production (36·36% of the isolates at 37°C and 10·9% under refrigeration), and all these factors may compromise the quality of the product. Proteinase production was similar for strains incubated at 37°C (16·81% of the isolates) and room temperature (17·27%) but there was less amount of phospholipase-producing strains at room temperature (15·45% of the isolates were positive) when compared with incubation at 37°C (36·36%). Enzymes production at 37°C by yeasts isolated from milk confirmed their pathogenic potential. The refrigeration temperature was found to be most efficient to inhibit enzymes production and consequently ensure better quality of milk. The viability of yeasts and the activity of their enzymes at different temperatures are worrying because this can compromise the quality of dairy products at all stages of production and/or storage, and represent a risk to the consumer.

Terpene composition of ewes’ raw milk from nine commercial flocks was analysed from February to July. Ewes’ diet consisted of concentrate and conserved forage in winter (indoor feeding) and part-time grazing from spring (transition and outdoor feeding). Regardless of the feeding, limonene and β-phellandrene were the most abundant monoterpenes and β-caryophyllene showed the highest concentrations among sesquiterpenes. Terpene content increased in the milks of commercial flocks when animals were reared under grazing management. Monoterpenes were detected in the milks of all the commercial flocks throughout the season, whereas sesquiterpenes were only detected in the milks from flocks grazing on non-cultivated community-owned grasslands in which a higher biodiversity of plant species grew. These preliminary results indicated that β-caryophyllene could be a potential pasture-diet marker in the case of milks from animals grazing a higher biodiversity of plant species but in-depth studies including information on terpene composition of plants ingested by the animals are necessary to evaluate the suitability of β-caryophyllene or another terpenoid compound as pasture biomarker.

The proteolytic activity of a psychrotrophic strain of Burkholderia cepacia isolated from refrigerated raw milk was characterized. Bur. cepacia produced proteolytic activity during growth at refrigeration temperature, with maximum activity at pH 6–7. The enzyme showed relative thermal stability in the range 40–50°C during 25 min, and maintained 80% its initial activity at 76°C/30 s. Milk coagulation assay showed that the crude protease from Bur. cepacia caused coagulation from day 2 for skimmed milk, whereas coagulation was observed from day 5 for whole milk. The adherence of this strain to stainless steel was evaluated, and the substrata had around 107 CFU/cm2 after 15 to 60 min incubation. Results on biofilm development suggest that this bacterium could adhere and to form biofilms even at refrigeration temperatures. These results indicate that Bur. cepacia may represent a potential hazardous to milk and dairy products.

The current work studied four types of binary antagonist/pathogen bacterial culture system, in order to determine the effect of interaction between two strains of Lactobacillus plantarum and two food-borne pathogens, Listeria monocytogenes and Escherichia coli, in whole UHT milk at 37°C. To determine the type of interaction between the two bacterial populations in co-cultures and to evaluate the antagonistic activity of the lactic acid bacteria (LAB) on the pathogenic bacteria, the growth curves, the kinetic parameters, and the pH profiles of mono- and co-cultures were compared. The Lb. plantarum strains showed different bacteriocin-like inhibitory substance (BLIS) production, auto- and co-inducible. The antibacterial effect of neutralized supernatants of mono and co-cultures harvested at different times of incubation was assessed in order to establish the presence of bacteriocin-like inhibitory-substances (BLIS) and their possible relation to the growth inhibition of the pathogen. The LAB reduced the growth of Esch. coli and of List. monocytogenes by 4 and ∼5 log cycles, respectively and influenced other growth kinetic parameters, such as μmax and lag phase, in the different binary combinations. The growth of the LAB was not relevantly altered by simultaneous growth with the pathogenic strains showing an interaction of amensalism. The pattern of inhibition exerted by the LAB on the pathogens was different; Lb. plantarum LB279 inhibited the growth of List. monocytogenes more effectively than that of Esch. coli. The behaviour of Esch. coli in co-culture with Lb. plantarum WS4174 suggested the presence of metabolic crowding in the mechanism of growth suppression. This exploratory study showed the complexity and specific particularities of the inhibition phenomena between bacterial communities.

Flaxseed hull, a co-product obtained from flax processing, is a rich source of n−3 fatty acids but there is little information on digestibility of its nutrients by dairy cows. Four rumen-cannulated multiparous Holstein cows averaging 665±21 kg of body weight and 190±5 d in milk at the beginning of the experiment were assigned to a 4×4 Latin square design with four 28-d experimental periods to determine the effects of feeding monensin and flaxseed hulls on total tract apparent digestibility of nutrients and fatty acids. The four treatments were: (1) diet CO: control with neither flaxseed hulls nor monensin added; (2) diet FH containing 19·8 g flaxseed hulls/100 g dry matter (DM); (3) diet MO with 16 mg monensin/kg DM; (4) diet HM containing 19·8 g flaxseed hulls/100 g DM and 16 mg monensin/kg DM. Diets provided similar amounts of protein and net energy of lactation. Digestibility of crude protein was higher for diets containing flaxseed hulls and for diets supplemented with monensin. Flaxseed hulls supplementation decreased digestibility of acid and neutral detergent fibre. Significantly higher digestibility of ether extract and individual fatty acids was observed for treatments with flaxseed hulls compared with treatments without flaxseed hulls. A combination of flaxseed hulls and monensin did not result in better fatty acid digestibility than when feeding only flaxseed hulls.

The current study determined whether the pre-implantation conceptus modified endometrial fatty acid concentrations. Oestrus was synchronized in 14 mature lactating cows and embryos were transferred on day 7. Cows were slaughtered 10 d later, with each uterine horn flushed, the pre-implantation conceptus located, and inter-caruncular endometrial tissue collected from the gravid horn (containing the pre-implantation conceptus) and non-gravid horn. Endometrial fatty acid concentrations in the gravid and non-gravid horn were compared using linear models in restricted maxiumum likelihood. Investigations of the correlations among selected fatty acids and trophoblast weight or uterine fluid interferon-tau (IFN-τ) concentrations were also undertaken. The presence of the pre-implantation conceptus had relatively minor effects on endometrial fatty acid concentrations, but the ω6:ω3 ratio was greater and concentrations of stearic and oleic acid were slightly increased in the gravid horn. In the gravid horn, a negative linear relationship between the concentration of arachidonic acid and conceptus weight and IFN-τ concentration in the uterine luminal fluid were observed. In contrast, there was a positive relationship between concentrations of dihomo-γ-linolenic acid in the non-gravid horn and conceptus weight. In conclusion, the presence of the pre-implantation conceptus appears to modulate endometrial fatty acids, as indicated by the differences in endometrial fatty acid concentrations in the gravid and non-gravid uterine horns. The physiological implication of these local effects of the pre-implantation conceptus, on reproductive success requires further investigation.

Although Corynebacterium bovis and coagulase-negative staphylococci are frequently the most commonly isolated bacteria from milk samples submitted for identification of pathogens causing intramammary infection, the individual quarter somatic cell count (SCC) from those samples is most often low. The present study aimed at evaluating the difference in bacteriology results from milk sampled by the standard technique (as recommended by the National Mastitis Council) and by the use of a teat cannula surpassing the teat canal, since C. bovis is often only found in the teat canal. Single quarter milk samples were collected in duplicate from 132 dairy cows on a commercial dairy farm using the standard milk sampling technique and also using a cannula introduced into the teat. Two groups of quarters were sampled: a group that was selected randomly at cow and quarter level and a group that was selected based on having SCC >200 000 cells/ml at the previous milk recording at cow level and on California mastitis test result at quarter level. Bacteriological culture performed on the samples yielded 29 Corynebacterium spp. isolates from the samples collected with the standard technique and 6 isolates from the samples collected with a cannula. Bacteriological culture yielded 73 and 100 culture negative samples respectively with the standard and the alternative sampling technique. A significant difference between the two sampling techniques was observed for recovery of Corynebacterium spp. and for no-growth samples. There was no significant difference in the isolation of Corynebacterium spp. or other bacterial species when using the standard technique before or after sampling with the cannula; thus the observed difference in bacteriology results could not be attributed to a particular sampling order. No significant change was observed overall in individual quarter SCC measured on the sampling day and 7 d later. Our results agree with several studies showing that Corynebacterium bovis often colonizes the teat canal, without causing true intramammary infection.

Prior to processing milk and cream were standardised and homogenised. Skim milk was cross-flow microfiltered (CFMF) prior to treatment with pulsed electric fields (PEF) or high temperature short time (HTST) pasteurization. The effect of temperature of the skim milk and product composition on the efficacy of PEF treatment was determined. The electrical conductivity of the product was related to fat and solids content and increased 5% for every g/kg increase of solids and decreased by nearly 0·7% for every g/kg increase of fat. From the three microbial groups analyzed (mesophilic, coliform, and psychrotroph) in milks differences (P<0·05) in the inactivation of mesophilic microorganisms were observed between the counts following PEF treatment, while HTST pasteurization resulted in higher reductions in all different counts than those obtained after PEF. Increasing the skim milk temperature prior to PEF treatment to about 34°C showed equivalent reductions in microbial counts to skim milk treated at 6°C in half the time. The reductions achieved by a combination of CFMF and PEF treatments were comparable to those achieved when CFMF was combined with HTST pasteurization. A higher reduction in coliform counts was observed in homogenised products subjected to PEF than in products that were only standardised for fat content.

To investigate matrix-specifity of probiotic effects and particularly of the reduction of antibiotics-associated diarrhea, a controlled, randomized, double-blind study was performed, in which 88 Helicobacter pylori-infected but otherwise healthy subjects were given for eight weeks either a) a probiotic fruit yoghurt “mild” containing Lactobacillus acidophilus LA-5 plus Bifidobacterium lactis BB-12, n=30), b) the same product but pasteurized after fermentation (n=29) or c) milk acidified with lactic acid (control, n=29). During week five, a Helicobacter eradication therapy was performed. Helicobacter activity was measured via 13C-2-urea breath tests and antibiotic-associated diarrhoea and other gastrointestinal complaints were recorded by validated questionnaires. In intervention group a, b and c the mean number of days with diarrhoea was 4, 10 and 10 (P<0·05), the frequency of episodes 17%, 7% and 27% (n.s.), and the change in total symptoms score before antibiotics treatment was −1·4±1·1, −1·2±1·1, 2·6±1·1 points/four weeks (P<0·05). All milk products decreased Helicobacter activity by 18 to 45% without significant differences between groups. The observed decrease in Hel. pylori activity seems to be not or not only due to probiotic bacteria but (rather) to components of acidified milk (most probably lactic acid). Fruit-yogurt-like fermented milk products with living probiotic bacteria significantly shorten the duration of antibiotics-associated diarrhoea and improve gastrointestinal complaints. Fruit yogurt-like fermented milk is a matrix suitable for probiotic bacteria.

The effect of freeze-drying on viability and probiotic properties of a microbial mixture containing selected bacterial and yeast strains isolated from kefir grains (Lactobacillus kefir, Lactobacillus plantarum, Lactococcus lactis, Saccharomyces cerevisiae and Kluyveromyces marxianus) was studied. The microorganisms were selected according to their potentially probiotic properties in vitro already reported. Two types of formulations were performed, a microbial mixture (MM) suspended in milk and a milk product fermented with MM (FMM). To test the effect of storage on viability of microorganisms, MM and FMM were freeze-dried and maintained at 4°C for six months. After 180 days of storage at 4°C, freeze-dried MM showed better survival rates for each strain than freeze-dried FMM. The addition of sugars (trehalose or sucrose) did not improve the survival rates of any of the microorganisms after freeze-drying. Freeze-drying did not affect the capacity of MM to inhibit growth of Shigella sonnei in vitro, since the co-incubation of this pathogen with freeze-dried MM produced a decrease of 2 log in Shigella viability. The safety of freeze-dried MM was tested in mice and non-translocation of microorganisms to liver or spleen was observed in BALB/c mice feed ad libitum during 7 or 20 days. To our knowledge, this is the first report about the effect of freeze-drying on viability, in vitro probiotic properties and microbial translocation of a mixture containing different strains of both bacteria and yeasts isolated from kefir.

The objective of this study was to evaluate the effect of dietary supplementation with selenium (Se) and vitamin E (VitE) on milk yield, composition and renneting parameters in dairy Jonica breed goats. Forty multiparous lactating goats fed the same basal diet were randomly allocated to 1 of 2 dietary treatments: for 135 days, the experimental period. Basal diet was composed by ad libitum mixed vetch-oat hay and a definite amount of a pelleted concentrate. Dietary treatments included: (1) the control diet containing background Se and VitE only, and (2) the experimental diet containing sodium selenite as the Se supplement in concentrate (0·20 mg/head per day of Na-selenite and 20 mg/day per head of VitE). During the trial, no significant differences (P>0·05) were registered in dry matter intake and body weight gain among groups. Goats fed Se-VitE diet showed the highest milk production (P<0·05), and fat (P<0·01) and protein (P<0·05) yields and percentages. Somatic cell count were significantly lower in Se-VitE diet compared with control group (P<0·05). Significant differences (P<0·05) were observed in milk renneting properties. Based on our results, it was concluded that dietary supplementation with SE and VitE led to an increase of milk production and quality in Jonica goat, with positively improvement in milk coagulation aptitude.

An enterotoxigenic Escherichia coli (ETEC) vaccine designed to prevent diarrhoea was inoculated into dairy cows, and the occurrence of clinical mastitis was investigated for 2 years. Half of 480 cows in five farms were subcutaneously inoculated with ETEC vaccine (Imocolibov) twice with a 1-month interval in 2007 and 2008. Fisher's exact test and survival (time to event) analysis with the log-rank test were used to compare vaccinates and controls. In 2007, there was no significant difference in the incidence rate of mastitis between vaccinate (20·3%) and control (17·1%) cows. The rate of death or culling due to mastitis was lower in vaccinated cows (7·4%) than in control cows (29·2%, P=0·07, Fisher's exact test; P=0·02, log-rank test). In 2008, there was no significant difference in both the incidence rate of mastitis and the rate of death or culling due to mastitis. Milk productivity was compared between vaccinates and controls in one farm. Multi-way analysis of variance (ANOVA) was performed for the amount of 4% fat-corrected milk, and there was no significant difference between vaccinates and controls. These results suggest that ETEC vaccine inoculation reduces death or culling due to mastitis, whereas no preventive effect on the development of mastitis was observed.

This study evaluated the effects of probiotic Dahi administration in ageing mice on macrophage and lymphocyte functions. Probiotic Dahi were prepared by co-culturing in buffalo milk (3% fat) Dahi bacteria (Lactococcus lactis ssp. cremoris NCDC-86 and Lc. lactis ssp. lactis biovar diacetylactis NCDC-60) along with Lactobacillus acidophilus LaVK2 (La-Dahi) or combined Lb. acidophilus and Bifidobacterium bifidum BbVK3 (LaBb-Dahi). Four groups of 12 mo old mice were fed for four months, with the supplements (5 g/day) of buffalo milk (3% fat), Dahi, La-Dahi and LaBb-Dahi, respectively, in addition to basal diet, and a fifth group that received no supplements served as control. The immune functions of young mice (4 mo old) were also compared with those of ageing adult mice (16 mo old). The production of nitric oxide and cytokines IL-6 and TNF-α declined and that of immunosuppressive prostaglandin E2 (PGE2) increased by stimulated peritoneal and splenic macrophages in ageing mice, compared with their young counterparts. The proliferation of stimulated splenocytes diminished and the production of IL-2 decreased and that of IL-6 and TNF-α enhanced in ageing compared with young mice. Feeding ageing mice with La-Dahi or LaBb-Dahi improved peritoneal macrophage functions stimulating nitric oxide and IL-6 and diminishing PGE2 production. Feeding La-Dahi or LaBb-Dahi also improved lymphocyte functions stimulating their proliferation and production of IL-2 in ageing mice. To conclude, the probiotic La-Dahi and LaBb-Dahi are effective in reversing age related decline in immune functions in mice.

An investigation of the shape of the lactation curve and the mastitis incidence was conducted to identify whether management interventions of the lactation curve constitute a potential for reducing incidence of mastitis at herd level. Lactation curves were estimated to describe the variation of daily milk yield during the 305-d lactation period in Norwegian Red cows. Associations between mastitis incidence at herd level and lactation curve characteristics such as production level at onset of lactation, magnitude and time of peak milk yield, and increase and decrease of milk yield rates were studied. Data from 250 303 lactations occurring during 2005 and 2006 from 14 766 herds were obtained from the Norwegian Dairy Herd Recording System. Besides veterinary treatments, the records included information on monthly test-day milk yields. The shapes of the lactation curves at herd level were parameterized using a modified Wilmink model in two separate mixed model analyses. In the first analysis a subset of lactations with no records of veterinary treatments was used. Lactation curves from herds with high (>0·31 cases/305-d lactation) and low (<0·07 cases/305-d lactation) herd mastitis incidence rate were parameterized and compared for three separate strata of parity. The result showed that high herd mastitis incidence rate was associated with a low intercept (P<0·05), a steep slope before peak milk yield (P<0·01) and a rapid decline after peak milk yield (P<0·01). In the second analysis a subset of high-yielding lactations with veterinary treatments of mastitis only and lactations with no records of veterinary treatment were compared. This was done to investigate whether the findings at herd level were also reflected at cow level. These results showed that lactation curves from lactations with mastitis cases were associated with a steep slope before peak milk yield (P<0·05) in second and later parities and a rapid decline after peak milk yield (P<0·01) in all three parity groups.

Bulk tank ewe's milks with low (<500,000 ml−1), medium (1,000,000–1,500,000 ml−1) and high (>2,500,000 ml−1) somatic cell counts (SCC) from three breeds were used to manufacture hard ewes’-milk cheese. Physico-chemical analysis and capillary electrophoresis of fresh cheeses and cheeses that had been ripened for 1, 2, 3 and 6 months were carried out. The results showed that high SCC levels in milk affected the moisture content of only freshly made cheeses and the pH, fat content and fat acidity of ripened cheeses. Regarding proteolysis, the levels of all β-CNs in freshly made cheeses were significantly lower as the SCC values increased and the Castellana breed was the most affected by SCC levels because a significant decrease in all α-CNs was also observed as SCC levels rose. Analysis of the casein profile by principal component analysis (PCA) revealed that there were no clear differences according the SCC up to the third month. However in the third and sixth months cheeses with low levels of SC were closely grouped and characterised by the highest levels of intact caseins. Regarding the effect of breed, the results point to a more intense proteolytic activity in the Assaf breed, whose more matured cheeses showed the highest content of casein proteolytic fragments.

The effects of the caprine αs1-casein (CSN1S1) polymorphisms on milk quality and cheese yield have been widely studied in French and Italian goat breeds. Much less is known about the consequences of κ-casein (CSN3) genotype on the technological and coagulation properties of goat milk. In the current study, we have performed an association analysis between polymorphisms at the goat CSN1S1 and CSN3 genes and milk coagulation (rennet coagulation time, curdling rate and curd firmness) and technological (time to cutting of curd and cheese yield) properties. In this analysis, we have included 193 records from 74 Murciano-Granadina goats (with genotypes constituted by different combinations of alleles B, E and F of the gene CSN1S1 and alleles A and B of the gene CSN3) distributed in three herds, which were collected bimonthly during a whole lactation. Data analysis, using a linear mixed model for repeated observations, revealed significant associations between CSN1S1 genotypes and the rate of the curdling process. In this way, milk from EE goats had a significantly higher curdling rate than milk from BB individuals (P<0·05). Contrary to previous experiments performed in French breeds, cheese yield was not significantly different in BB, EE and EF goats. Moreover, we have shown that CSN3 genotype has a significant effect on the rennet coagulation time (BB>AB, P<0·05) but not on cheese yield. No interaction between the CSN1S1 and CSN3 genotypes was observed.

Dioxins are lipophilic compounds with a small molecular weight and are highly persistent, bioaccumulative and toxic. Dioxin detoxification is associated with an increased production of reactive oxygen species (ROS). In physiological conditions the body is protected against ROS and their toxic products by a wide range of antioxidant systems. We hypothesize that the imbalance between ROS production, associated with dioxin exposure, and the antioxidant defence capacity, may lead to oxidative stress, with consequent increased consumption of antioxidants and accumulation of toxic compounds in blood and tissues. The objective of this study was to evaluate the effect of exposure to dioxins on the plasma redox status of lactating buffalo cows. To this aim, the major liposoluble (retinol and α-tocopherol) and water-soluble (ascorbate) antioxidants, the superoxide dismutase (SOD) and glutathione peroxidase (GPx) activity, the total antioxidant capacity (TAC), as well as specific protein oxidation markers (protein bound carbonyls and nitro-tyrosine) and lipid oxidation markers (hydroperoxides), were chosen as indices of blood redox status. The concentration of antioxidants, protein-bound carbonyls (PC), nitro-tyrosine (N-Tyr), and hydroperoxides (LPO), the SOD and GPx activity, and the TAC were measured in plasma samples obtained from buffalo cows exposed to environmental levels of dioxins higher (n=21, group A) or lower (n=29; group B) than those permitted. Plasma titres of antioxidants, as measured by HPLC, and the total antioxidant capacity, as measured by trolox equivalents capacity, were higher in group B than in A. Similarly, SOD and GPx activities were higher in group B than in A. Conversely, plasma levels of PC, N-Tyr and LPO, as measured by ELISA, were higher in group A than in B. Our results suggest that exposure to dioxins impairs the plasma antioxidant defence system of lactating buffalo cows, and that metabolic processes associated with dioxin detoxification might induce or enhance oxidation of protein and lipids. This adverse effect on blood redox status might have negative implications for animal health and reproduction, and might compromise animal welfare.

Recombinant bovine somatotropin (rbST) is used to increase milk yield in cows, but it has been forbidden in some countries and in the EU. However, rbST misuse represents a concern in both bovine and buffalo dairy production. A number of studies on rbST treatment have been performed on bovines, but there are few data on buffaloes. In this study, we treated eight lactating buffaloes with biweekly injections of a slow-release formulation of rbST, for five cycles of administration, and analysed total ST and insulin-like growth factor 1 (IGF-1) variations in serum and IGF-1 in milk. The aim was to assess their power as potential indicators of rbST-treatment. Blood was collected on days 2, 5, 9 and 14 of each cycle, and milk on days 2, 9 and 14 of cycles 2 and 5. Results showed an extraordinary increase in ST levels on day 2 in treated animals, followed by a rapid decrease over the following days, while a significant increase in IGF-1 was observed both in serum and in milk throughout most of each cycle. These results suggest that serum ST levels are a good indicator of treatment. However, the rapid decrease after the peak limits the useful period of sample collection.

Fatty acids, vitamins and minerals in milk are important for the human consumer, the calf and the cow. Studies indicate that milk from organic and conventional dairy herds may differ in these aspects. The aim of this study was therefore to investigate whether there are differences in the fatty acid composition and concentration of vitamins and selenium in milk between organic and conventional herds in Sweden. Bulk tank milk was sampled in 18 organic and 19 conventional dairy herds on three occasions during the indoor season 2005–2006. Herd characteristics were collected by questionnaires and from the official milk recording scheme. Multivariable linear mixed models were used to evaluate the associations between milk composition and type of herd, while adjusting for potential confounders and the repeated observations within herd. In addition to management type, variables included in the initial models were housing type, milk fat content, herd size, average milk yield and time on pasture during summer. The median concentration of conjugated linoleic fatty acids (CLA) was 0·63% in organic compared with 0·48% in conventional herds, the content of total n-3 fatty acids was 1·44% and 1·04% in organic and conventional milk, respectively, and the content of total n-6 fatty acids was 2·72% and 2·20% in organic and conventional milk, respectively. The multivariable regression models indicated significantly higher concentrations of CLA, total n-3 and n-6 fatty acids in organic milk and a more desirable ratio of n-6 to n-3 fatty acids, for the human consumer, in organic milk. The multivariable models did not demonstrate any differences in retinol, α-tocopherol, β-carotene or selenium concentrations between systems. Median concentrations of α-tocopherol were 0·80 μg/ml in organic and 0·88 μg/ml in conventional milk, while for β-carotene the median concentrations were 0·19 and 0·18 μg/ml, respectively; for retinol, the median concentration was 0·32 μg/ml in both groups; the median concentrations of selenium were 13·0 and 13·5 μg/kg, respectively, for organic and conventional systems.
