The effect of niclosamide on the tegument of adult Haplorchis taichui (Trematoda: Heterophyidae) exposed in vitro was observed by scanning electron microscope. Adult worms were incubated in Tyrode's solution containing 0.01, 0.1, 1.0, and 10 μg ml− 1 of niclosamide for 30 min, 1, 6, 12 and 24 h. Control groups were incubated in Tyrode's solution without niclosamide and worms remained active until 24 h. In 0.01 μg ml− 1 of niclosamide, worms showed slightly active movements up to 1 h after incubation, while in 0.1 μg ml− 1 solution a few worms showed only slightly active movements after 30 min. Tegumental changes were determined by scanning electron microscopy. Swelling and blebbing of the tegument were observed on both ventral and dorsal sides. After longer periods, extensive swelling and blebbing of the tegument became more severe and there was a loss of the apical plasma membrane in some regions. Empty spine sockets occurred, and small perforations penetrated the basal lamina, followed by some lesions. Destruction of both surfaces was more pronounced on the posterior compared with the anterior regions.

The process of spermiogenesis in two paramphistomes, Sandonia sudanensis and Basidiodiscus ectorchis from the Nile fish Synodontis schall in Egypt was studied by transmission electron microscopy. Spermiogenesis is characterized by the outgrowth of the zone of differentiation, presenting two basal bodies separated by a microtubule organizing centre, each basal body developing into a flagellum. Proximodistal fusion of these flagella with a median cytoplasmic extension gives rise to the spermatozoon. The mature spermatozoon possesses two axonemes of the 9+‘1’ pattern typical of parasitic helminths. There are few ultrastructural studies on spermiogenesis in paramphistomes, which are considered the most primitive digenetic trematodes. The present study provides new and more detailed information on this process, including the presence of a lateral flange and external ornamentation of the cell membrane. The value of sperm ultrastructure as a taxonomic tool in phylogeny is also discussed.

Angiostrongylus cantonensis can invade the central nervous system, leading to human eosinophilic meningitis or eosinophilic meningoencephalitis. Curcumin is a natural product which has the effects of anti-inflammation, anti-oxidation and anti-carcinogensis, while the administration of curcumin has been reported to possibly relieve the symptoms of meningitis. The present study tested the potential efficacy of curcumin in A. cantonensis-induced eosinophilic meningitis of BALB/c mice. Assay indicators for the therapeutic effect included the larvicidal effect, eosinophil counts and matrix metalloproteinase-9 (MMP-9) activity in angiostrongyliasis. Eosinophils were mildly reduced in treatment groups compared with infected-untreated mice. However, there were no significant differences in larvicidal effects or MMP-9 activity. This study suggests that anti-inflammatory treatment with curcumin alone has low efficacy, but the treatment does not interfere with MMP-9 expression and is not useful for larvicidal effects. The possible reasons include low curcumin across the blood–brain barrier and also those larvae that survive stimulate MMP-9 production, which promotes blood–brain barrier damage, with leukocytes then crossing the blood–brain barrier to cause meningitis. Further studies will be required to test these possibilities.

Patterns of intestinal helminth infections among school-aged children have been assessed in Eko-ende and Ore, as part of an overall assessment of the public health impact of Erinle Dam Reservoir in Osun State, Nigeria. The investigation was carried out between January and May 2005 using the Kato Katz thick smear technique and simple questionnaire for information on the bio-data, knowledge, attitude and practice of individuals towards disease transmission and control. Ascaris lumbricoides, hookworm, Trichuris trichiura, Taenia spp., Strongyloides stercoralis and Schistosoma mansoni were recovered at an overall prevalence of 78.3% among 309 children examined. Ascaris lumbricoides, hookworm, T. trichiura and S. mansoni were the most common infections while S. stercoralis and Taenia spp. were found only among a few children. Infection patterns of the common diseases were age-specific with peaks in the 11–15 age bracket. Children not attending school were significantly (P < 0.05) more at risk of infection than those attending school. Multiple infections were pronounced with over 54% double infections and 6% four parasites or more infections. The need for urgent intervention to arrest the obviously serious public health situation attributable to Erinle Dam is emphasized.

Eighty-nine samples of grey wolf (Canis lupus L.) faeces were collected between 2002 and 2004 from two areas in the western Beskidy Mts (south Poland). Helminth eggs were observed in 56.2% of faeces examined. These included: Alaria alata (2.2%), taeniid eggs (11.2%), Toxocara canis (5.6%), Toxascaris leonina (1.1%), Eucoleus aerophilus (14.6%), Ancylostoma caninum (12.3%), Uncinaria stenocephala (37%) and unidentified roundworm eggs of the family Strongyloididae (1.1%). Eucoleus aerophilus is recorded for the first time from Poland. The results are compared with the helminth fauna of other wolf populations in Europe.

Single and double infections of juvenile Omphiscola glabra (Gastropoda: Lymnaeidae) with Paramphistomum daubneyi and/or Fasciola hepatica were carried out to determine the redial burden and cercarial production in snails dissected at day 60 or at day 75 post-exposure (p.e.) in the laboratory at 20°C. The results were compared with those obtained with single-miracidium infections by Fascioloides magna. Compared to F. hepatica, low values were noted at day 75 p.e. for the prevalence of snail infections with P. daubneyi (4.6–8.3% instead of 23.6–25.9%), the total number of free rediae (10.7–17.9 per snail instead of 26.3–34.7), and that of free cercariae (112.8–136.9 per snail instead of 177.8–248.5). Despite a greater number of free rediae at day 75 p.e. (36.2–45.6 per snail), the prevalences of snail infections with F. magna and cercarial production were similar to those noted for F. hepatica. The results concerning F. hepatica and P. daubneyi might partly be explained by a progressive adaptation of O. glabra to sustain the larval development of these digeneans over the years, as this snail is a natural intermediate host of F. hepatica and P. daubneyi in central France since 1995. Compared with the high number of fully-grown rediae of F. magna in O. glabra, cercarial production seemed limited and this might be explained by the presence of high numbers of rediae which reduced the avaibility of nutrients for cercarial differentiation within the snail.

The use of parasites as biological tags in population studies of marine fish in the south-western Atlantic has proved to be a successful tool for discriminating stocks for all species to which it has been applied, namely: Scomber japonicus, Engraulis anchoita, Merluccius hubbsi and Cynoscion guatucupa, the latter studied on a broader geographic scale, including samples from Uruguayan and Brazilian waters. The distribution patterns of marine parasites are determined mainly by temperature–salinity profiles and by their association with specific masses of water. Analyses of distribution patterns of some parasite species in relation to gradients in environmental (oceanographic) conditions showed that latitudinal gradients in parasite distribution are common in the study area, and are probably directly related to water temperature. Indeed, temperature, which is a good predictor of latitudinal gradients of richness and diversity of species, shows a latitudinal pattern in south-western Atlantic coasts, decreasing southwards, due to the influence of subtropical and subantarctic marine currents flowing along the edge of the continental slope. This pattern also determines the distribution of zooplankton, with a characteristic specific composition in different water masses. The gradient in the distribution of parasites determines differential compositions of their communities at different latitudes, which makes possible the identification of different stocks of their fish hosts. Other features of the host–parasite systems contributing to the success of the parasitological method are: (1) parasites identified as good biological tags (i.e. anisakids) are widely distributed in the local fauna; (2) many of these species show low specificity and use paratenic hosts; and (3) the structure of parasite communities are, to a certain degree, predictable in time and space.

In the acanthocephalan Neoechinorhynchus crassus five embryonic envelopes have been identified, including four, which are similar to those in previously studied acanthocephalans and a supplementary one, located between the outer and the second envelopes, which is characteristic of only a few Eoacanthocephala. The terminology of the envelopes in relation to their origin and possible function is discussed.

Parasites have been used successfully as biological tags in population studies, mainly in marine fishes, but also in marine mammals, crustaceans and molluscs. Almost all published information dealing with parasites as biological tags evaluates differences between localities. However, local variability in the component community has not been assessed. In this work, we examined whether local variation of the metazoan parasite fauna of Engraulis ringens, extracted from five independent samples from two nearby localities in northern Chile, can be a factor causing bias in stock identification. Our results show that local variability, as estimated by a single sample, may suffice to represent component community variability with no need for replicated data.

In a cross-sectional study, a total of 5381 slaughtered animals, namely 928 cattle, 243 buffaloes, 3765 sheep and 445 goats were inspected macroscopically for hydatid cysts in northwest Iran, with prevalence values of 38.3%, 11.9%, 74.4% and 20%, respectively, being recorded. Prevalences were higher in females compared with males, but a significant difference (P < 0.001) was only found in sheep and cattle. Most cases which were condemned were seen in the lungs of sheep (13.4%) indicating that sheep are the most important intermediate hosts for Echinococcus granulosus in this area.

A mouse-derived strain of Rodentolepis ( = Hymenolepis) nana was transferred to the Mongolian jird Meriones unguiculatus. It was found that M. unguiculatus has low susceptibility to R. nana mouse isolates. Likewise, adaptation of the parasite to jird hosts, in the absence of dexamethasone treatment, was not demonstrable, at least during ten-passage trials. Nevertheless, the parasite was able to establish, grow and develop to gravid adults in M. unguiculatus treated daily with dexamethasone. Prepatent periods in dexamethasone-treated jirds in ten-passage series ranged from 10 to 17 days post-infection (DPI), the average being 12 days, and the patent periods lasted from 18 to 30 DPI, with an average of 25 days. The population pattern of faecal egg output in immunosuppressed jirds suggests that under a daily dexamethasone treatment protocol, the parasite is able to maintain egg production as long as treatment is sustained.

In the present paper, recent results obtained on the use of different distributions observed in larval species of Anisakis, genetically identified by means of allozyme markers, for stock characterization of demersal (Merluccius merluccius), small (Trachurus trachurus) and large pelagic (Xiphias gladius) finfish species in European waters, are reviewed and discussed. Several species of Anisakis were identified in the three fish hosts: A. simplex (s.s.), A. physeteris, A. typica, A. ziphidarum,A. pegreffii, A. brevispiculata and A. paggiae. Canonical discriminant analysis performed on all the samples of the three fish species collected in areas comprising their geographical range, according to the different species of Anisakis identified, showed distinct fish populations in European waters. In all the three fish hosts, the pattern of distribution of Anisakis larvae allowed discrimination of Mediterranean stocks from Atlantic stocks. In the case of swordfish, the possible existence of a southern Atlantic stock separated from a northern one is also suggested. Congruence and discordance with the population genetic data inferred from allozyme markers on the same samples of the three fish species are also discussed.

During an investigation on strongyloidiasis in AIDS patients from Iran, direct smears of stools revealed many eggs and larvae of Rhabditis axei, which were subcultured to confirm identification.

The relative importance of ecology and evolution as factors determining species richness and composition of the helminth communities of fish is a matter of current debate. Theoretical studies use host–parasite lists, but these do not include studies on a temporal or spatial scale. Local environmental conditions and host biological characteristics are shown to influence helminth species richness and composition in four fish species (Eugerres plumieri, Hexanematichthys assimilis, Oligoplites saurus, and Scomberomorus maculatus) in Chetumal Bay, Mexico. With the exception of H. assimilis, the helminth communities had not been previously studied and possible associations between environmental and host biological characteristics as factors determining helminth species richness and composition using redundancy analysis (RDA) are described. Thirty-four helminth species are identified, with the highest number of species (19 total (mean = 6.3 ± 2.1)) and the lowest (9 (4.0 ± 1.0)) occurring in H. assimilis and S. maculatus, respectively. The larval nematodes Contracaecum sp. and Pseudoterranova sp. were not only the helminth species shared by all four host species but also were the most prevalent and abundant. Statistical associations between helminth community parameters and local ecological variables such as host habitat use, feeding habits, mobility, and time of residence in coastal lagoons are identified. Phylogeny is important because it clearly separates all four host species by their specialist parasites, although specific habitat and feeding habits also significantly influence the differentiation between the four fish species.

The present study focuses on the helminth parasite community of the wild rabbit in a sand dune area in Portugal over a 5-year period. The influence of host sex and year on the composition of the helminth community is assessed, along with the potential effect of the detected helminths on host body condition. The basic structure of the helminth community comprises Mosgovoyia ctenoides, Graphidium strigosum, Trichostrongylus retortaeformis, Nematodiroides zembrae and Passalurus ambiguus. Mean intensities of G. strigosum varied between years. General G. strigosum intensities were also found to vary according to both year and host sex, but not according to the interaction of both factors. When assessing the effect of helminths on rabbit body condition (expressed by the kidney fat index), higher burdens of M. ctenoides, a cestode that presents a relatively large body mass, were found to induce a reduction in rabbit condition.

Infections with gastrointestinal (GI) nematodes are amongst the most prevalent worldwide, especially in tropical climates. Control of these infections is primarily through treatment with anthelmintic drugs, but the rapid development of resistance to all the currently available classes of anthelmintic means that alternative treatments are urgently required. Cysteine proteinases from plants such as papaya, pineapple and fig are known to be substantially effective against three rodent GI nematodes, Heligmosomoides polygyrus, Trichuris muris and Protospirura muricola, both in vitro and in vivo. Here, based on in vitro motility assays and scanning electron microscopy, we extend these earlier reports, demonstrating the potency of this anthelmintic effect of plant cysteine proteinases against two GI helminths from different taxonomic groups – the canine hookworm, Ancylostoma ceylanicum, and the rodent cestode, Rodentolepis microstoma. In the case of hookworms, a mechanism of action targeting the surface layers of the cuticle indistinguishable from that reported earlier appears to be involved, and in the case of cestodes, the surface of the tegumental layers was also the principal location of damage. Hence, plant cysteine proteinases have a broad spectrum of activity against intestinal helminths (both nematodes and cestodes), a quality that reinforces their suitability for development as a much-needed novel treatment against GI helminths of humans and livestock.

Studies conducted in the 1980s suggested that parasites were useful in distinguishing adjacent populations of Atlantic cod Gadus morhua in the north-western Atlantic. During the mid-1980s, cod populations began to decline, culminating in closure of the fishery in 1992. A marked decrease of cod off the coast of Labrador was coincident with climatic shifts, including a decline of bottom sea temperature, and virtual absence of capelin Mallotus villosus, its main food prey. These changes in some components of the ecosystem were independent of anthropogenic influences. An initial survey of cod parasites in 2000 from coastal Labrador revealed a dramatic decline in abundance of metazoans and also a change in dominance of species of digenetic trematodes compared to 1980–1981. Comparison of metazoan parasites in three other populations of cod inhabiting the Grand (northern and southern) and St. Pierre Banks, where there was no apparent evidence of environmental changes but only commercial over-exploitation, revealed no major alterations in abundance or dominance between 1980–1981 and 2003. The results of this current study suggest that environmental changes in the food web affected the abundance of metazoan parasites only off coastal Labrador but did not influence the use of the parasites to distinguish between cod populations inhabiting the Grand or St. Pierre Banks.

The composition of the abomasal helminth fauna and parasite diversity were studied in 298 red deer collected during 1997–2000 from three different sectors (Bolzano, Trento and Sondrio provinces) of the Stelvio National Park, one of the main protected areas of north-eastern Italy. The association between parasite burdens and geographical areas of the hosts was assessed using the negative binomial regression. A variety of abomasal helminths, both host specific and generalist, was found in all sectors. The most commonly observed parasites were the Spiculopteragia spiculoptera morph spiculoptera and Ostertagia leptospicularis morph leptospicularis, with prevalences of 79.5% and 40.9%, respectively. The minor morphs S. spiculoptera morph mathevossiani (prevalence 31.9%) and O. leptospicularis morph kolchida (18.8%) occurred at lower prevalences. Teladorsagia circumcincta morph circumcincta, Marshallagia marshalli morph marshalli and Haemonchus contortus were rarer, at prevalences of 1, 1.3 and 1.3%, respectively. Deviance analysis of the negative binomial regression model shows that the geographical area is significantly related to parasite burdens (P = 0.001). Prevalences of hosts with parasites were greater in the Sondrio (odds ratio = 1.31; 95% confidence interval: 0.16–10.85) and smaller in the Trento (odds ratio = 0.62; 95% confidence interval: 0.20–1.96) sectors with respect to Bolzano, but these differences were not statistically significant. Possible cross-infections by more generalist parasites between wild and domestic animals were also suggested, as deer and domestic ruminants (Bovinae, Caprinae and Ovinae) used the same feeding areas of the park sectors.

Five species of monogeneans were recovered from the gill filaments of 181 chub mackerel, Scomber japonicus, from the Madeiran waters of the Atlantic Ocean, Portugal, during 2004/2005. The monogenean Pseudokuhnia minor showed the highest prevalence (98.68%) and a mean intensity of 28.23, followed by Kuhnia scombri (prevalence of 43.71% and mean intensity of 2.69) and K. scombercolias (prevalence of 39.1% and a mean intensity of 1.81). Kuhnia sprostonae and Grubea cochlear were rare, occurring in only one and five fish hosts respectively. No correlation between fish host length and mean intensity of infection with the three most abundant monogeneans was found. However, significant differences in prevalence and abundance were found in relation to date of sampling for P. minor, and all parasites were aggregated in their distribution.
