The junction between the gregarine trophozoite Nematocystis magna Schmidt and an epithelial cell of its earthworm host Lumbricus terrestris Muller consists of an extensive folding and meshing of the single trilaminar membrane bounding the host cell and the complex folded multilaminar limiting membrane of the trophozoite. They are separated by a structureless intercellular region of some 50 nm in width. Specialized communicating regions or permeable elements were not observed. Transport of a radioactive label across non-junctional and junctional membranes was demonstrated by using tritiated glucose. The attachment organelle of the trophozoite is shown to be capable of allowing metabolites to pass into the cell directly from the host tissue.

Some of the reindeer (Rangifer tarandus) introduced into Scotland from Northern Sweden have become infested with the deer warble fly Hypoderma diana and the deer ked Lipoptena cervi.

Evidence is presented which suggests that the larvae of H. diana proceed to their final site on the back of the host by way of the neural canal, and in this way probably caused the deaths of two reindeer in the winter of 1953–4.

A detailed analysis is presented of the frequency with which various bands of spines occurred on second-stage larvae of H. diana. An analysis of the number of chitinous rings in the two posterior spiracles on second-stage H. diana larvae showed that in any individual larva the two numbers were not independently determined.

On reindeer L. cervi produced puparia in December.

Avian malaria parasites can negatively affect many aspects of the life of the passerines. Though these parasites may strongly affect the health and thus migration patterns of the birds also during autumn, previous studies on avian malaria focused mainly on the spring migration and the breeding periods of the birds. We investigated whether the prevalence of blood parasites varies in relation to biometrical traits, body condition and arrival time in the European Robin (Erithacus rubecula) during autumn migration. We found no sex or age related differences in avian malaria prevalence and no relationship between infection status and body size or actual condition of the birds was found either. However, the timing of autumn migration differed marginally between infected and non-infected juveniles, so that parasitized individuals arrived later at the Hungarian stopover site. This is either because avian malaria infections adversely affect the migration timing or migration speed of the birds, or because later arriving individuals come from more distant populations with possibly higher blood parasite prevalence. The possible delay that parasites cause in the arrival time of the birds during autumn migration could affect the whole migratory strategy and the breeding success of the birds in the next season.

Aspects of vitellogenesis, known to be controlled by juvenile hormone, are adversely affected by Hymenolepis diminuta infection of Tenebrio molitor, in spite of circulating titres of the hormone remaining unchanged. It has therefore been proposed that juvenile hormone binding is disrupted at the tissue site level. Juvenile hormone III binding sites were located in the nuclear, microsomal and post-microsomal supernatant fractions of the follicle cells of Tenebrio molitor. When JH-III binding was quantified for both control and Hymenolepis diminuta-infected beetles, binding in the nucleus and cytosol were found to be largely unaffected. However, microsomal binding was severely disrupted; on days 3 and 6 post-infection, binding was greatly diminished, on day 9 post-infection, binding was slightly reduced and, by day 15, binding was ‘restored’ to that of control insects. Using follicle cell microsomes at day 3 post-infection, previous Scatchard analysis revealed the presence of at least two JH-III binding sites. The first is of higher affinity, Kd = 5·3 x 10·5 x 10−11 mol/mg protein and the second of lower affinity Kd = 7·7 x 10−7 M, Bmax = 9·75 x 10−11 mol/mg protein. A comparison with microsomal binding parameters of follicle cells from non-infected Tenebrio indicated that although the Bmax values were unchanged, the Kd value of the higher affinity site was increased by approximately 5-fold. These data are indicative of a parasite-induced competitive binding inhibitor.

The fluid layer within the cuticle of the nematode Nippostrongylus brasiliensis contains haemoglobin. The absorbance spectra of the haemoglobins extracted from whole nematodes were measured spectrophoto-metrically and the spectra of cuticular haemoglobin in individual living nematodes was obtained by microdensitometry. Adult female nematodes were confined under cover-slips in oxygenated saline and measurements of optical density of a small area of the cuticular haemoglobin were taken at 10 sec intervals. The optical density decreased over a period of 4 min at 543 nm and increased at 553 nm indicating deoxygenation of the cuticular haemoglobin; irrigation with fresh saline restored the haemoglobin to its oxygenated state. This demonstrates that cuticular haemoglobin loads and unloads oxygen in the living animal. Isoelectric focusing showed that mature nematodes have haemoglobins which are isoelectric around pH 7·0 and at pH 9·8. Isolated cuticular fluid contained the haemoglobin which was isoelectric at pH 9·8, but no other haemoglobin, and immature adults, which do not have cuticular haemoglobin, contained only those haemoglobins which were isoelectric at pH 7·0. Sodium dodecyl sulphate polyacrylamide gel electrophoresis and column chromatography showed that the molecular weight of the cuticular haemoglobin sub-unit is 16·9–17·5 × 103 Daltons and that the haemoglobin exists primarily as a tetrameric molecule with a molecular weight of 65–75 × 103 Daltons. The presence of cuticular haemoglobin may allow adult Nippostrongylus to exploit oxygen-deficient areas within the environment of the rat's intestine where it would otherwise be unable to survive for prolonged periods.

Haplosporidian protist parasites are a major concern for aquatic animal health, as they have been responsible for some of the most significant marine epizootics on record. Despite their impact on food security, aquaculture and ecosystem health, characterizing haplosporidian diversity, distributions and host range remains challenging. In this study, water filtering bivalve species, cockles Cerastoderma edule, mussels Mytilus spp. and Pacific oysters Crassostrea gigas, were screened using molecular genetic assays using deoxyribonucleic acid (DNA) markers for the Haplosporidia small subunit ribosomal deoxyribonucleic acid region. Two Haplosporidia species, both belonging to the Minchinia clade, were detected in C. edule and in the blue mussel Mytilus edulis in a new geographic range for the first time. No haplosporidians were detected in the C. gigas, Mediterranean mussel Mytilus galloprovincialis or Mytilus hybrids. These findings indicate that host selection and partitioning are occurring amongst cohabiting bivalve species. The detection of these Haplosporidia spp. raises questions as to whether they were always present, were introduced unintentionally via aquaculture and or shipping or were naturally introduced via water currents. These findings support an increase in the known diversity of a significant parasite group and highlight that parasite species may be present in marine environments but remain undetected, even in well-studied host species.

The period 1875–1925 was remarkable in the history of parasitology partly because of the number of significant discoveries made, especially the elucidation of important life cycles, and partly because of the achievements of the clinicians and scientists who made these discoveries. What is remarkable is that so many of these individuals were Scots. Preeminent in this pantheon was Patrick Manson, who not only discovered the mosquito transmission of filarial worms but was instrumental in directly encouraging others to make significant discoveries in the fields of malaria, Guinea worm disease (dracunculiasis), onchocerciasis, loiasis and schistosomiasis and, indirectly, sleeping sickness and leishmaniasis. This chapter describes and discusses the contributions made by Douglas Argyll-Robertson, Donald Blacklock, David Bruce, David Cunningham, Robert Leiper, William Leishman, George Low, Patrick Manson, Muriel Robertson and Ronald Ross together with short biographical notes.

Parasites sometimes expand their host range and cause new disease aetiologies. Genetic changes can then occur due to host-specific adaptive alterations, particularly when parasites cross between evolutionarily distant hosts. Characterizing genetic variation in Cryptosporidium from humans and other animals may have important implications for understanding disease dynamics and transmission. We analyse sequences from four loci (gp60, HSP-70, COWP and actin) representing multiple Cryptosporidium species reported in humans. We predicted low genetic diversity in species that present unusual human infections due to founder events and bottlenecks. High genetic diversity was observed in isolates from humans of Cryptosporidium meleagridis, Cryptosporidium cuniculus, Cryptosporidium hominis and Cryptosporidium parvum. A deviation of expected values of neutrality using Tajima's D was observed in C. cuniculus and C. meleagridis. The high genetic diversity in C. meleagridis and C. cuniculus did not match our expectations but deviations from neutrality indicate a recent decrease in genetic variability through a population bottleneck after an expansion event. Cryptosporidium hominis was also found with a significant Tajima's D positive value likely caused by recent population expansion of unusual genotypes in humans. These insights indicate that changes in genetic diversity can help us to understand host-parasite adaptation and evolution.

Growth of Trypanosoma cruzi as colonies on solid medium has not been widely used as an experimental procedure. We therefore sought to establish a reliable and routine plating method. The optimal results were achieved with a matrix of 0·65% low melting point agarose onto which epimasigotes from the mid-to-late logarithmic phase of growth were spread. Colonies could be isolated after incubation for 21 days in a humidified 5% CO2 environment at 28°C. Plating efficiencies in the range of 40% were obtained by this method and clones could be recovered into liquid medium or onto blood-agar slopes with a high success rate. The procedure has also been adapted for the isolation of genetically transformed clones after electroporation of epimastigotes with either plasmid or cosmid vectors. This was best achieved by inclusion of the electroporated cell inoculum in a 0·6% agarose overlay containing G418 as the selective drug, on top of a 0·8% agar base. Transformation efficiencies were as high as 10−5 cells per μg of DNA. A reliable plating method for T. cruzi will have many applications and is a significant step towards the use of ‘shotgun transformation’ to generate libraries of T. cruzi recombinants.

Our understanding of the neurobiology of the Platyhelminthes has come in large part from free-living turbellarians. In addition to providing considerable information about the capabilities of the rudimentary nervous system present in all members of the phylum, turbellarians have provided the most definitive information about the variety of ion channels present in the membranes of neurones and muscle cells, and about the physiology and pharmacology of those channels. Furthermore, preparations of single, viable muscle cells have provided some of the most conclusive evidence about the variety of transmitters present, and the types of response they evoke. Here, we review what is known about the physiology and pharmacology of the turbellarian neuromuscular system. Particular attention is given to the triclad flatworm Bdelloura Candida, the best studied species in this respect, but other species are included where relevant.

We have investigated the metabolic disturbances in 2 murine models of blood-stage malaria, Plasmodium chabaudi and Plasmodium yoelii. Blood glucose, plasma insulin and parasitaemia were measured in normal and infected mice before and after treatment with diazoxide, adrenaline, Sandostatin and quinine. Severe hypoglycaemia and marked hypersinsulinaemia developed during both infections. A single injection of diazoxide (25 mg/kg i.p.) or adrenaline (0·03 mg s.c.) lowered insulin concentrations in normal mice, reversed the hypoglycaemia in both infections and significantly reduced the hyperinsulinaemia in P. chabaudi-infected mice (P < 0·0001). Higher doses of Sandostatin (500 μg/kg s.c.) were required to reverse hypoglycaemia. Quinine (25 mg/kg i.p.) significantly increased blood glucose in normal and infected mice (P < 0·001) and no hypoglycaemia was observed in mice with normal blood glucose for more than 3 h. This study shows that the major cause of hypoglycaemia in murine malaria is hyperinsulinaemia rather than high consumption of glucose by host and parasites or chemotherapy with quinine, and that hypoglycaemia can be reversed by correcting the hyperinsulinaemia.

Unlike in other acanthocephalans, the overian balls of Centrorhynchus corvi are complex and are composed of 24–30 ovarian ball units. Each ovarian ball unit consists of three structural and functional units – the oogonial syncytium, developing oogenetic cells and the supporting syncytium – complementary to the ovarian balls of other acanthocephalans. Three metamorphic stages of the nuclei in the oogonial syncytium are described, depending on the nuclear morphology, chromatin structure and appearance of the nucleolus. Third-stage nuclei bulge out at the periphery of the oogonial syncytium and are surrounded by its cytoplasm and a thin membrane. Ultimately these are separated from the oogonial syncytium to from oogonia containing small amounts of cytoplasmic components derived from the oogonial syncytium. Nuclei of the young oogonia, oognia at the budding stage, and 3rd-stage nuclei of the oogonial syncytium all possess nucleoli and are similar also in their nuclear dimensions and cytochemical affinities. Nuclear resemblances, and cytoplasmic similarities of the oogonia and oogonial syncytium with regard to the presence of lipids, RNA and proteins support the concept of the origin of oogonia from the oogonial syncytium.

The free oogonia divide mitotically in the cellular zone where they undergo a single mitotic division and the resulting oocytes enter the prophase of meiosis. I. The meiotic primary oocytes in contrast to the permeiotic primary oocytes of other acanthoscephalans enter the growth phase which is closely accompanied by the accumulation of various ooplasmic components such as basophilic yolk nucleus which is composed of RNA, proteins, lipoproteins and phospholipids. Histochemical features of the oogonial and supporting syncytium are described.

Samples of approximately five wild rabbits were obtained each week over the 4-year period 1967–70, and their intestines examined for adult tapeworms. Pour species of anoplocephalinae were found, Cittotaenia pectinata, C. denticulata, C. ctenoides and Andrya cuniculi. C. pectinata was found in 29% of the 968 rabbits examined, C. denticulata in 14%, C. ctenoides in 12% and A. cuniculi in 6%. There were 63 double infections, of which 44 involved A. cuniculi, and three triple infections. The most frequent number of worms per infection was one for C. pectinata and A. cuniculi and two for C. denticulata and C. clenoides, although the mean numbers for these species were respectively 7·2, 5·3, 2·7 and 1·8.A. cuniculi, and to a lesser extent C. ctenoides, were very restricted in distribution within the study area, occurring only on the higher, chalk downland (250–550 ft A.S.L.). This finding implies that individual rabbits usually spend their lives within relatively small areas.Considering all the material a seasonal trend in host infection rate was apparent only with C. denticulata, which increased in abundance from July to a peak in September and then declined gradually to a low level, lasting from April to June. Infections, including small specimens of any of the four species, were rare from February to June and it is presumed that infected intermediate hosts were correspondingly rare during this period. Except for C. denticulata, which was found more frequently in male rabbits than females, the sex of the host had no influence on its likelihood of infection. However, except for C. pectinata, the age of the host was important. Thus, C. denticulata and A. cuniculi were significantly more frequent in juveniles than adults whilst the reverse was true for C. ctenoides. Small (‘young’) specimens of C. denticulata and A. cuniculi were more frequent in juvenile than adult rabbits, but the difference was not significant for the other two species. It is concluded that only C. ctenoides survives for more than a year and that this species and C. pectinata are more likely to infect rabbits that are adult than are the other two species.

A description is given of the life history and pathogenicity of a species of Eimeria, derived from a single oocyst infection in a coccidia-free bird, and subsequently identified as E. meleagridis.

There are two asexual generations followed by the sexual generation. The first schizogony cycle occurs in the small intestine and the second and sexual generations are restricted to the caeca.

The organism is not pathogenic but the sexual stages produce marked caseous lesions in the caeca. It is immunologically distinct from a pure strain of E. adenoeides and cannot be transmitted to chickens.

The hatching organ of Lipeurus columbae Linn. is described. It is very similar to that already described in sucking lice, and unlike that of any other group of insects. This resemblance is further evidence for the affinity of the Mallophaga and Siphunculata.

Acephalic cysticercus (Ac), a rarely developed multilobulated and nonencysted form of larval Taenia, causes hydrocephalus or adhesive arachnoiditis in the ventricles and subarachnoidal space that often lead to fatal outcome in affected patients. Ac has been proposed to originate from T. solium on the basis of morphological features, while no molecular data supporting the presumption have been available. In the present study, we investigated the immunological properties as well as molecular characteristics of Ac that was obtained surgically from 6 patients. Immunoblotting of the cyst fluid from Ac samples demonstrated the constitutive expression of a T. solium metacestode (TsM) 10 kDa protein. Specific antibodies against the truncated 10 kDa protein, which appears to be species specific for TsM cysticercosis, were detected in both serum and cerebrospinal fluid samples of Ac patients. Nucleotide sequences of mitochondrial cytochrome c oxidase subunit I (COI) and NADH dehydrogenase subunit 1 (ND1) genes of Ac were almost identical to those of T. solium but differed substantially from those of the other Taenia species. In phylogenetic analysis, Ac clustered with T. solium in a well-supported clade. Our results strongly suggest that Ac may have originated from T. solium.

1. 2:4-diamino-6:7-diisopropylpteridine has an antimalarial index of 1:40, when tested against Plasmodium gallinaceum.

2. 2:4-diamino-1′-methylindolo-(2′:3′-6:7)-pteridine showed no activity against P. gallinaceum in doses tolerated by the chick.

3. Proguanil- and sulphadiazine-proguanil-resistant strains of P. gallinaceum were cross-resistant to 2:4-diamino-6:7-diphenylpteridine.

4. Proguanil- and sulphadiazine-proguanil-resistant strains of P. gallinaceum showed no cross-resistance to 2:4-diamino-6:7-diisopropylpteridine, but were slightly hypersensitive to this compound.

Previous work is discussed and described. The numbers of larvae present in a square yard of soil can be estimated by the use of caged guinea-pigs or by light traps. The infestation observed on the ears of wild rabbits gives a rough index of the population present in a given area.

Using a modified light trap, gorged larvae can be collected from their hosts.

Adults can be extracted from light, friable soils by flotation in water and from any type of soil by the Salt-Hollick soil-sampling machine.

The harvest mite occurs throughout the British Isles. It seems possible that climate and the geological origin of the soil do not affect harvest mite abundance. They are, however, less numerous on heavy clays.

Larvae are most abundant in late summer and autumn. Adults remain constant in numbers throughout the year.

The larvae appear to infest most warm-blooded vertebrates. They do not attack at night. Nymphs have been observed sucking the contents of nymphophanes.

Adults have been found at a depth of 3 ft. They appear to come nearer the surface in warm, moist weather, but are driven down by drought, heavy rain and ground frost.

A total of 28 components were detected in the free amino acid (FAA) pool of hydatid fluid from primary and secondary equine cysts, secondary ovine cysts and host plasma. Examination of data from equine cysts revealed that the majority of FAAs were present in significantly greater concentrations in secondary cysts, glycine being over 30 times more concentrated. Values for total carbohydrates and glucose did not, however, differ significantly and total protein content was greater in primary cysts. Comparison of the (FAA) pool of secondary equine and ovine cysts revealed strain variation. It was demonstrated that most FAAs were more concentrated in hydatid fluid than in the corresponding host plasma, many concentration ratios exceeding 10. The possible contribution that mediated amino acid transport across the cyst wall and parasite amino acid metabolism makes to the composition of the FAA pool was discussed. No significant plasma aminoacidaemia was associated with infection.

Previous work has shown that the surface of infective larvae of parasitic nematodes will not bind the fluorescent lipid analogue 5-N(octadecanoyl)aminofluorescein (AF18) until after exposure of the parasite to mammalian tissue-culture conditions. In this study, culture media which are permissive or non-permissive for the acquisition of lipophilicity for AF18 were altered in order to examine possible stimuli involved. This showed that external alkaline pH and high sodium ion concentration were highly stimulatory. The internal signalling pathways which may be involved in the surface alteration were then examined using agents which are known to affect intracellular signalling in mammalian cells. The results indicated that elevation of cGMP levels was stimulatory whereas inhibition of a putative Na+/H+ antiporter or calcium mobilization was inhibitory, and it is argued that high intracellular levels of cAMP may be inhibitory. Whilst the precise effects of the agents used on nematode cells remain to be established, these results provide a framework for the examination of the processes involved in the modification of the nematode surface which takes place immediately after the infection event.
